A deep-learning pipeline to extract clinical gait metrics from single-camera recordings.

Single camera markerless motion capture is a low-cost, accessible motion capture technology that could be used to improve clinical decision making. However, current models lack the spatial and anatomical fidelity required for research-grade analysis, which prevents accurate modelling of 3D human gait dynamics. The purpose of the current study was threefold, 1) to develop a means of predicting 3D gait kinematics using consumer-grade video inputs, 2) to assess the accuracy of the method across a variety of ecologically relevant perturbations such as camera location, body size, and clothing chara

High-fat diet attenuates estradiol's efficacy on cognitive function and hippocampal synaptic plasticity in middle-aged ovariectomized mice.

Loss of circulating ovarian hormones during menopause is associated with increased risk for cognitive decline in women. Although estradiol has been shown to enhance memory and synaptic function in preclinical studies, clinical studies of hormone therapy in menopausal women are less clear, and its efficacy may be diminished in the context of metabolic dysfunction. This study investigated whether high-fat diet-induced obesity impairs the beneficial effects of estradiol on hippocampus-dependent memory and synaptic plasticity in middle-aged, ovariectomized female mice. Female C57BL/6J mice were fe

Structural features of the hippocampus covary with memory-guided attention depending on the side of hippocampal sclerosis.

Past experiences stored in long-term memory (LTM) provide a valuable resource for making predictions that shape perception and guide goal-directed behavior. Contents from the high-capacity LTM system guide contextual selective attention to enhance sensory and higher-order processing of memory-predicted targets, in a process known as LTM-guided attention. While this essential cognitive function is believed to depend on the hippocampus, evidence is still scarce. In this study, we used a neuropsychological approach to test LTM-guided attention in the context of isolated hippocampal pathology and

Research on LSTM-based spatial target trajectory forecasting enhanced by attention mechanisms.

To address the strong dependence of space object orbit prediction on physical models and initial conditions, as well as the difficulty of completely eliminating prediction errors, this study proposes a satellite orbit prediction correction method that integrates an attention mechanism with a long short-term memory (LSTM) network. Taking the LAGEOS satellite as the research object, the proposed method uses position error, velocity, and acceleration features extracted from historical orbital data to train a deep learning model for predicting one-day-ahead orbital errors and correcting the SGP4 o

A single extracellular glycan links AMPA receptor gating to synaptic plasticity and memory persistence.

Persistent synaptic plasticity is essential for memory, yet its stabilizing mechanisms remain incompletely understood. AMPA-type glutamate receptors (AMPARs) mediate most fast excitatory transmission in the brain, but how extracellular posttranslational modifications regulate their functional dynamics in vivo remains unclear. Here, we identify a single endogenous N-linked glycan at Asn401 of the AMPAR GluA1 subunit as a regulator that links receptor gating, membrane microdomain organization, and memory persistence. Loss of this glycan markedly reduced AMPAR desensitization and promoted resensi

Memory traces of duration and location in the right intraparietal sulcus.

Time and space form an integral part of every human experience, and for the neuronal representation of these perceptual dimensions, previous studies point to the involvement of the right-hemispheric intraparietal sulcus and structures in the medial temporal lobe. Here we used multi-voxel pattern analysis (MVPA) to investigate long-term memory traces for temporal and spatial stimulus features in those areas. Participants were trained on four images associated with short versus long durations and with left versus right locations. Our results demonstrate stable representations of both temporal an

RLBindDeep: A ResNet-LSTM based novel framework for protein-ligand binding affinity prediction.

The prediction of the binding affinity of proteins and ligands in computational drug discovery with high accuracy is critical when evaluating the effectiveness of potential therapeutic compounds. This research work introduces RLBindDeep, a novel deep learning architecture based on the amalgamation of the ResNet and LSTM architectures, for improved accuracy in predicting protein-ligand binding affinities. Most traditional methodologies utilizing conventional molecular docking techniques suffer from poor accuracy owing to semi-flexible modeling approaches and limited considerations of complex in

Deep Learning for Scalp-Level Nonlinear Source Separation in Electroencephalogram: A Comparative Evaluation of Spatiotemporal Architectures.

Electroencephalography (EEG) is a common technique to measure field potentials of various brain regions, and event-related potentials (ERPs) reflect stimulus- or response-locked brain responses that are spatially distributed across the scalp due to volume conduction. However, accurately capturing stimulus specific ERPs from EEG remains a major challenge due to complex mixtures of ERPs from multiple stimulus sources. This study addresses the challenge of separating nonlinearly mixed ERPs in EEG data using synthetic and experimental datasets. We simulated synthetic ERP datasets with known ground

An information-theoretic evaluation framework for CNN-LSTM-based Alzheimer's disease classification from structural MRI.

Early detection of Alzheimer's disease (AD) is important because of its progressive impact on cognitive function. This study presents a CNN-LSTM-based framework for three-class AD classification from structural MRI, with the primary contribution being a post-hoc information-theoretic evaluation strategy rather than a new network architecture. Experiments were conducted using 827 ADNI subjects, including normal controls (NC: 340), mild cognitive impairment (MCI: 307), and AD (180). To mitigate data scarcity and improve training diversity, GAN-based augmentation was applied only to the training

Pattern separation memory requires Cerebellin 4 - Neogenin 1 signaling at dentate gyrus synapses.

The ability to avoid confusion between similar episodic memories enables organismal survival and fitness. This evolutionarily conserved differentiation process of memories as distinct representations is known as pattern separation. A central role for the entorhinal cortex→dentate gyrus (EC→DG) circuit in pattern separation memory is well established, but the molecular mechanisms that enable this circuit to mediate pattern separation memory are incompletely understood. We previously found that a trans-synaptic protein complex formed by presynaptic Cerebellin-4 and postsynaptic Neogenin-1 is sel

Early sepsis prediction using a hybrid LSTM-GAT model: a study on the PhysioNet 2019 dataset.

OBJECTIVE: Sepsis is a potentially fatal systemic response to infection, in which early clinical intervention is critical to reduce mortality. This study presents a hybrid deep learning model that combines temporal and structural information from clinical data to improve early sepsis prediction. METHODS: We used data from the PhysioNet/Computing in Cardiology Challenge 2019 to predict sepsis onset up to 12 hours in advance. We developed a hybrid model integrating long short-term memory (LSTM) networks and graph attention networks (GAT) to capture temporal dynamics and intervariable relationsh

CNN-BiLSTM-Based Hybrid Deep Learning for Multi-Metric Anomaly Detection and Mitigation in Secure IoMT Healthcare WBANs.

Wireless Body Area Networks (WBANs) have become an essential component of modern Internet of Medical Things (IoMT) healthcare systems, enabling continuous monitoring of patient physiological signals through wearable sensors. Despite their advantages, WBAN environments remain highly prone to cyber threats, privacy breaches, and single points of failure. To address these risks, this work proposes a Hybrid Multi-Metric Anomaly Detection (HM-MAD) framework deployed on the NodeMCU-32S platform with BLE 5.0 connectivity for secure continuous glucose monitoring (CGM) data transmission. The detection

Intelligent fall risk prediction and real-time warning system for elderly care based on multimodal deep learning and wearable sensor fusion.

Falls remain a leading cause of injury-related morbidity and mortality among adults aged 65 and older, and these human and economic costs keep rising as populations age worldwide. Most existing risk assessment tools rely on periodic clinical evaluations or single-sensor wearable devices, neither of which tracks the dynamic, multifactorial nature of fall risk throughout daily life. In this paper we describe the design, implementation, and pilot-level field validation of an integrated system that pairs a multi-sensor wearable unit (triaxial accelerometer, gyroscope, insole pressure array, and wr

Accurate classification and prediction of knee osteoarthritis based on Al-Biruni Earth Radius metaheuristic optimizer and LSTM classifier.

Knee osteoarthritis (KOA) is one of the worst varieties of arthritis. If not treated right away, it might lead to knee replacement. For this reason, early KOA detection is crucial for optimal therapy. This work tested and improved deep learning (DL) algorithms for predicting and identifying KOA. The suggested approach was evaluated utilizing an available dataset that included preprocessing approaches such as scaling and normalization. The Google-BER-LSTM hybrid model was explicitly designed to improve classification accuracy. The proposed binary optimization approach and other comparable metho

Guanosine protects against behavioural and mitochondrial bioenergetic alterations after mild traumatic brain injury.

Traumatic brain injury (TBI) constitutes a heterogeneous cerebral insult induced by traumatic biomechanical forces. Mitochondria play a critical role in brain bioenergetics, and TBI induces several consequences related with oxidative stress and excitotoxicity clearly demonstrated in different experimental model involving TBI. Mitochondrial bioenergetics alterations can present several targets for therapeutics which could help reduce secondary brain lesions such as neuropsychiatric problems, including memory loss and motor impairment. Guanosine (GUO), an endogenous neuroprotective nucleoside, a

Information loss over time defines the memory defect of propofol: a comparative response with thiopental and dexmedetomidine.

BACKGROUND: Sedative-hypnotic drugs impair memory, but details regarding the nature of this effect are unknown. The influences of propofol, thiopental, and dexmedetomidine on the performance of a task that isolates specific components of episodic memory function were measured. METHODS: Working (1 intervening item, 6 s) and long-term memory (10 intervening items, 33 s) were tested using auditory words in a continuous recognition task before and during drug administration. Eighty-three volunteer participants were randomly assigned to receive a constant target concentration of drug or placebo, p

Phenylbutyric acid protects against spatial memory deficits in a model of repeated electroconvulsive therapy.

Repeated electroconvulsive therapy (rECT) is widely applied in the treatment of refractory depression. Among the side effects of rECT, memory impairment is noticeable and needs effective protection. In this study, by employing a recognized repeated electroconvulsive shock (rECS) rat model, we found that rECS induced the significant spatial memory retention deficits with the simultaneous decreases in long-term potential (LTP), enhanced excitable postsynaptic potentials (EPSP), population spike (PS) and input/output curve in perforant pathway-dentate gyrus (PP-DG), but had no obvious neuron loss

Synaptic potentiation requires PARP1 activation: prevailing concepts are revisited.

The therapeutic potential of PARP inhibitors in neurodegenerative diseases of the central nervous system is widely accepted. This prevailing concept is challenged by more recent findings, unveiling the role of PARP1 activity in synaptic long-term potentiation, synaptic plasticity and long-term memory. So, PARP1 inhibition, though intended to prevent memory deterioration, actually interferes with stimulation-induced synaptic plasticity and long-term memory. On the other hand, PARP inhibitors could be useful for preventing mental disorders associated with memory retrieval.

Alzheimer's Disease Risk Factor Pyk2 Mediates Amyloid-β-Induced Synaptic Dysfunction and Loss.

Dozens of genes have been implicated in late onset Alzheimer's disease (AD) risk, but none has a defined mechanism of action in neurons. Here, we show that the risk factor Pyk2 (PTK2B) localizes specifically to neurons in adult brain. Absence of Pyk2 has no major effect on synapse formation or the basal parameters of synaptic transmission in the hippocampal Schaffer collateral pathway. However, the induction of synaptic LTD is suppressed in Pyk2-null slices. In contrast, deletion of Pyk2 expression does not alter LTP under control conditions. Of relevance for AD pathophysiology, Pyk2-/- slices

Neonatal exposure to sevoflurane caused cognitive deficits by dysregulating SK2 channels and GluA2-lacking AMPA receptors in juvenile rat hippocampus.

Anesthetics exposure to neonates leads to impairment of hippocampal synaptic plasticity and cognitive functions later in life. This phenomenon complies with the concept of metaplasticity: a priming stimulation can affect induction of synaptic plasticity mins or days later. We aimed to understand whether small conductance Ca2+-activated potassium channel type2 (SK2) and subunit composition of AMPA receptors are altered and contribute to sevoflurane-induced metaplasticity. To fulfill this goal, we exposed neonatal rats (postnatal day 7) to 2% sevoflurane for 2 h (sevoflurane rats) and examined s
