We aimed to compare the cytogenetic and molecular analyses in the assessment of imatinib mesylate response in patients suffering the chronic phase of chronic myelocytic leukemia who were refractory to alpha-interferon treatment. In this study, we examined the effect of thrombopoietin (TPO) on the aggregation of platelets from 40 patients with myeloproliferative disorders (MPDs), including 17 patients with chronic myelogenous leukemia in the chronic phase (CML-CP), 10 with polycythemia vera, 10 with essential thrombocythemia, and three with myelofibrosis. A patient with Meige syndrome associated with spasmodic torticollis was treated with benztropine mesylate (Cogentin) at doses of 12-16 mg daily. Marked suppression of both oromandibular dystonia and the torticollis was obtained. Mild impairment of recent memory was the major side effect. Peripheral anticholinergic side effects were controlled by the concomitant administration of ambenonium chloride (Mytelase) 15 mg daily.

Amenorrhea and galactorrhea developed in a female patient with tuberous sclerosis. There was no evidence of a pituitary tumor; she had an abnormal EEG, and computed tomographic scan showed multiple intracerebral calcifications but no lesions in the pituitary gland or hypothalamus. She had fixed hyperprolactinemia that was unresponsive to protirelin, chlorpromazine, levodopa, bromocriptine mesylate, or estrogen. The circulating prolactin may be of pituitary origin or may possibly be secreted ectopically by a hamartoma.

Although imatinib mesylate therapy has dramatically improved the prognosis of patients in the advanced phases of chronic myeloid leukemia, room for improvement remains. Tyrosine kinase inhibitors are undergoing evaluation as second-line therapy for patients with imatinib-resistant disease. Kantarjian et al. recently demonstrated that once-daily dasatinib 140 mg is well tolerated and achieves a high response rate.

PURPOSE OF REVIEW: Chronic myelogenous leukemia was once the most common indication for unrelated allogeneic hematopoietic stem cell transplantation but has become dramatically less so during the past 5 years. This procedure remains an option in chronic myelogenous leukemia but its use will become more sparing and selective. The synthetic routes used were (a) reaction of the 16alpha,17-disubstituted 21-mesylate with lithium chloride and (b) reaction of the 16alpha-substituted 17,21-cyclic ortho ester with triphenylmethyl chloride. AIM: The COVID-19 pandemic caused by infection with the novel coronavirus SARS-CoV-2 is urging the scientific community worldwide to intense efforts for identifying and developing effective drugs and pharmacologic strategies to treat the disease. Many of the drugs that are currently in (pre)clinical development are addressing late symptoms of the disease. This review focuses on potential pharmacologic intervention at an early stage of infection which could result in less-infected individuals and less cases with severe COVID-19 disease due to reduced virus entry into the cells.

Fatigue is the most commonly reported and most disabling of all post-polio sequelae (PPS). Bromocriptine mesylate (Parlodel) was employed in a placebo-controlled trial in five survivors of paralytic polio who continued to report moderate to severe daily fatigue after complying with the conservative treatments prescribed for PPS.
