Evaluation of cefiderocol trailing and other unclear susceptibility testing endpoints for Acinetobacter spp.

UNLABELLED: Broth microdilution (BMD) susceptibility testing with Acinetobacter spp. can be challenging with skipped wells and trailing observed for some agents. This also applies to cefiderocol, where isolates can display different growth patterns, such as trailing, regrowth, or hazy growth. To better understand the growth patterns observed with cefiderocol BMD testing, the content of wells was evaluated microscopically. Analysis of the trailing, hazy, and regrowth wells above the MIC endpoints showed disturbed growth with long filamentous and spherical cells characteristic of cell envelope i

Persenone A from Avocados Exhibits Potent Anti-Fungal Activity against Coccidioides.

Coccidioidomycosis, or Valley fever, is a fungal respiratory disease caused by Coccidioides immitis or Coccidioides posadasii. Clinical manifestations range from asymptomatic infection to chronic pulmonary disease. Despite the increasing endemic range and affected population, therapeutic strategies display moderate efficacy and adverse side effects. This study explores California's rich botanical diversity as a source for agents that inhibit the early stages of Coccidioides development. We established a screening protocol containing a colorimetric metabolism assay and microscopic imaging. We t

Antifungal effects of thiosemicarbazone-chalcones on Aspergillus, Candida and Sporothrix strains.

The frequency of opportunistic fungal infections has been increasing, impacting agriculture, food, and health sectors. In this work, four thiosemicarbazone-chalcones (TC) were synthesized and evaluated by the radial diffusion method against filamentous fungi. All TCs were effective against Aspergillus parasiticus, especially the fluor-substituted one, with radial growth inhibition of 62,9% and 74,4% at the lower (0.209 µmol/ml) and highest (1.670 µmol/ml) concentrations tested, respectively. On the other hand, the non-substituted derivative was the most active against A. carbonarius, inhibitin

In vitro activity of novel antifungals, natamycin, and terbinafine against Fusarium.

The genus Fusarium includes several trans-kingdom pathogens, infecting plants, animals, and humans, with widespread occurrence. Invasive Fusarium infections are often destructive due to their prevalence in critically ill patients. Furthermore, severe superficial infections, like keratitis or endophthalmitis with potential loss of vision, are rising in incidence globally. Infections are difficult to treat due to the intrinsic resistance of Fusarium against echinocandins and often high minimal inhibitory concentrations (MICs) for azoles. In this article, we assessed the in vitro activity of the

Antimicrobial Activity of Chalcones with a Chlorine Atom and Their Glycosides.

Chalcones, secondary plant metabolites, exhibit various biological properties. The introduction of a chlorine and a glucosyl substituent to the chalcone could enhance its bioactivity and bioavailability. Such compounds can be obtained through a combination of chemical and biotechnological methods. Therefore, 4-chloro-2'-hydroxychalcone and 5'-chloro-2'-hydroxychalcone were obtained by synthesis and then glycosylated in two filamentous fungi strains cultures, i.e., Isaria fumosorosea KCH J2 and Beauveria bassiana KCH J1.5. The main site of the glycosylation of both compounds by I. fumosorosea K

Targeting Candida albicans O-acetyl-L-homoserine sulfhydrylase (Met15p) in antifungal treatment.

Fungal infections are a serious threat to public health as they are becoming increasingly frequent. A major problem stems also from a rising fungal resistance to currently available antifungal therapies, therefore novel molecular targets are highly desirable. Exploration of enzymes participating in the biosynthesis pathways of essential amino acids such as L-methionine (L-Met) may provide new insights into pharmaceutical development. The MET15 gene from Candida albicans, encoding O-acetyl-L-homoserine sulfhydrylase (Met15p), an enzyme catalyzing the second step in that pathway, was cloned and

[How to read new antimicrobial susceptibility testing results?].

HOW TO READ NEW ANTIMICROBIAL SUSCEPTIBILITY TESTING RESULTS? Antimicrobial susceptibility testing (AST) results (formerly 'susceptible', 'intermediate' and 'resistant') are now expressed with the new susceptibility categories 'susceptible, standard dosing regimen', 'susceptible, increased exposure' and 'resistant'. These new categories introduce a paradigm shift in the way clinicians have to interpret AST results: uncertainties (previously embedded in the 'intermediate' category) now have to be dealt with exclusively by the laboratory, independently from the clinical categorization itself, an

Novel quinoxaline compound against extended-spectrum beta-lactamases producing bacteria.

OBJECTIVE: Extended-spectrum beta-lactamases (ESBLs) targeting beta-lactam antibiotics pose a major healthcare challenge. Carbapenems are known to be less impacted. However, the emergence of carbapenem-resistant strains can add further complexity to this existing challenge. With slow drug discovery and rapid resistance, repurposing existing drugs is crucial. This research study aims to provide insight into the antimicrobial effectiveness of 3-hydrazinoquinoxaline-2-thiol against diverse clinical ESBL-producing isolates. MATERIALS AND METHODS: The broth microdilution assay was conducted on a t

Pseudomonas Acts as a Reservoir of Novel Tigecycline Resistance Efflux Pump tmexC6D6-toprJ1b and tmexCD-toprJ Variants.

Several variants of the plasmid-carried tigecycline resistance gene cluster, tmexCD-toprJ, have been identified. This study characterized another novel variant, tmexC6D6-toprJ1b, located on the chromosome of environmental-origin Pseudomonas mendocina. TMexC6D6-TOprJ1 mediates resistance to multiple drugs, including tigecycline. The promoter activity of tmexC6D6-toprJ1b and negative transcriptional repression by the upstream regulator tnfxB6 are crucial for the expression of tmexC6D6-toprJ1b. tmexC6D6-toprJ1b was found in the plasmids or chromosomes of different Pseudomonas species from six cou

Production of new ent-hardwickiic acid derivatives by microbial transformation and their antifungal activity.

Ent-hardwickiic acid is the major compound of Copaifera pubiflora Benth oleoresin traditionally used in Brazilian folk medicine as an antimicrobial agent. Microbial transformation of ent-hardwickiic by Cunninghamella elegans ATCC 10028b resulted in two and five antifungal derivatives (four new ones) produced in the Czapek modified and Koch's K1 media, respectively. The derivatives were isolated and their structures were determined by spectral analysis, namely 1D/2D NMR and HR-ESIMS. All compounds were tested for cytotoxic and antifungal activities and they were not cytotoxic to the tested cell
