Modeling Properties and Interaction Mechanisms of PMMA and Cellulose Nanocomposite Materials via Molecular Dynamics.

Polymer nanocomposites are highly tunable materials with broad-reaching applications due to the diverse properties imbued through interactions between polymers and nanofillers. The vast combinatorial space of polymer composition, nanofiller types, sizes, mixture ratios, processing conditions, and use-cases necessitates improved modeling capability of the materials to help isolate the molecular mechanisms of interaction and predict the resultant potential properties. Poly methyl-methacrylate (PMMA) and cellulose nanocomposites are a potential combination of strength, biocompatibility, and avail

Cuproptosis-related molecular alterations in a neonatal rat model of hypoxic-ischemic brain damage.

BACKGROUND: Copper homeostasis imbalance is closely linked to the progression of various neurological disorders. Cuproptosis, a recently identified form of cell death caused by abnormal intracellular copper accumulation, has an unclear relationship with hypoxic-ischemic brain damage (HIBD). This study aims to characterize cuproptosis-associated molecular changes in the pathological process of HIBD in neonatal rats. METHODS: A neonatal rat HIBD model was established using the modified Rice-Vannucci method. Cerebral blood flow was measured with laser Doppler flowmetry, while infarct volume was

Design, synthesis, anticancer activity and molecular modeling of new pyrazoline derivatives as telomerase inhibitors.

Telomerase is an enzyme that rebuilds chromosomal ends called telomeres, allowing cells to keep dividing. Blocking telomerase is an important strategy in cancer treatment, as tumor cells are then forced to continue dividing without repairing their telomeres, leading to progressive telomere shortening, genomic stress, and ultimately the cessation of cell division or death, thereby slowing tumor growth and enhancing the effect of chemotherapies or targeted therapies. Twelve new pyrazoline derivatives were synthesized and evaluated for their cytotoxicity against MCF-7 and MDA-MB-231 breast adenoc

Molecular surveillance and predictive risk modelling of avian influenza virus in wild birds in Egypt.

Avian influenza viruses (AIVs) continue to circulate and pose a persistent threat to animal and public health in Egypt, a country located along major migratory birds' flyways that enable repeated viral incursions. In this study, a total of 1,087 wild birds representing 19 species were sampled to assess the role of migratory wild birds in virus introduction and spread. Six species were found positive for AIV, with Eurasian teal showing the highest prevalence (12.3%). The H5 subtype was found predominant, representing 70.3% of positive cases, whereas H9 was detected in 9.9% of positive cases. Mo

Spaceflight alters molecular networks linked to diverse human diseases in a single cellular model.

The International Space Station provides an opportunity to study the impact of spaceflight on gene expression and possible links to human health. Our study investigates global changes in messenger RNA (mRNA) abundance in the THP-1 cell line, a monocyte-macrophage lineage known for plasticity and immune reprogramming features. We identified pathways positively enriched with genes affecting muscle and cardiac contraction, neuronal system, and sensory perception. Available computational models identified links with health issues, including cardiac, neurological, muscular, and renal disorders and

Coarse-Grained Molecular Modeling of Pore Network Evolution in Particle- and Surfactant-Laden Emulsions.

Macropore-infused nanocomposite emulsion thermosets (MINETs) are porous materials formed from emulsions containing high loadings of surfactants and nanoparticles, such as silica, activated carbon, alumina, and zinc oxide dispersed in the fluid mixture. After high-shear mixing, phase separation leads to the formation of a stable porous solid with a pore size that scales with the particle size. The mechanisms governing this pore network formation remain unclear. Here, we develop a coarse-grained molecular dynamics model to investigate phase separation and pore evolution in four-component systems

Lasiodiplodia theobromae as a Model Latent Pathogen: Molecular Mechanisms, Global Impact, and Sustainable Management.

Lasiodiplodia theobromae (Botryosphaeriaceae) is a cosmopolitan ascomycete that endophytically colonizes over 500 plant species and switches to necrotrophy under host stress. In mango (Mangifera indica), it causes stem-end rot, dieback, gummosis, and collar rot, driving substantial pre- and postharvest losses worldwide. This review integrates five thematic areas: (i) multilocus phylogenetics (ITS, tef1-α, tub2, and rpb2) resolving cryptic species within the L. theobromae species complex; (ii) disease epidemiology centered on latent colonization of inflorescences and stress-triggered activation

Discovery of novel BCAT2 inhibitors: design, synthesis, in vitro evaluation, and molecular modeling studies.

Branched-chain amino acid transaminase 2 (BCAT2) is a promising therapeutic target for metabolic diseases and cancers such as pancreatic ductal adenocarcinoma. In this study, we report the discovery of novel and potent inhibitors against this enzyme. Through a structure-based strategy integrating scaffold hopping, core simplification, and hydrophobic cavity filling, three series of compounds (A, B, and C) were designed and synthesized. Several compounds in series C exhibited nanomolar inhibitory potency (C10, IC50 = 44 nM; C11, IC50 = 54 nM). Key structure-activity relationships were elucidate

Integrating AI and Molecular Modeling for Structural Prediction of a Closed-Pore State of the hERG Channel.

The voltage-gated potassium channel hERG (Kv11.1) plays a central role in cardiac repolarisation by mediating the rapid delayed rectifier K+ current (IKr). Blockage of hERG by small molecules can lead to delayed repolarisation, QT interval prolongation, and potentially fatal arrhythmias, making the channel a critical focus in drug safety screening. Despite extensive pharmacological and electrophysiological characterization, a complete structural understanding of hERG gating remains limited by the absence of an experimentally determined closed-state structure. Here, we use AI-based structural m

A patient-derived mouse model reproduces molecular, neurological, and sleep symptoms of SHINE syndrome.

SHINE syndrome is a rare neurodevelopmental disorder caused by mutations in DLG4, which encodes the postsynaptic scaffolding protein PSD-95. Key symptoms include sleep problems, hypotonia, intellectual disability, neurological disorders, and epilepsy, hence the name 'SHINE.' Here, we developed and characterized a mouse model of SHINE syndrome carrying the patient-derived DLG4V692Wfs*12/+ variant associated with a severe form of the disorder. The mutant transcript escapes nonsense-mediated decay but results in reduced PSD-95 protein expression, faithfully reproducing the molecular phenotype obs

A Novel Model System to Identify Cellular and Molecular Defects Underlying Rare Genetic Disorders.

Ankyloblepharon-ectodermal defects-cleft lip/palate (AEC) is a disorder caused by autosomal-dominant mutations in the TP63 gene. AEC is characterised by the presence of severe and painful skin erosions that can take years to heal. Current treatment options for these devastating lesions are limited, highlighting the need for new therapeutic strategies. We previously generated keratinocytes from patient-derived induced pluripotent stem cells (iPSC-K) and identified defects in several cell adhesion complexes, including desmosomes, hemidesmosomes and focal adhesions. In the present study, we devel

Explainable Artificial Intelligence (XAI) and Molecular Modeling Techniques to Discover Putative HER2 Inhibitors.

Breast cancer is one of the prominent reasons of death in women. HER2 is a promising target to counter breast cancer. In the current research, a structure-based pharmacophore model was generated to map and screen CMNPD, a comprehensive database of marine natural products. The two compounds (CMNPD30448 (hit1) and CMNPD7060 (hit2)) displayed better LibDock scores than the reference co-crystallized ligand. These compounds demonstrated stable molecular dynamics results conducted for 500 ns with stable root mean square deviation (RMSD) at 0.3 nm, stable radius of gyration (Rg) and root mean square

Molecular dynamics modeling of the Candida albicans membrane for the discovery of membrane-binding antifungal peptides.

Fungal infections caused by Candida albicans (C. albicans) pose a significant clinical challenge, especially due to the rise of multidrug-resistant strains. Antifungal peptides (AFPs) represent promising alternatives because of their high efficacy, low toxicity, and reduced risk of resistance. In this study, we integrated computational modeling and experimental validation to identify novel AFPs targeting the fungal membrane. Four candidate peptides-HIV-F9170 (GWEALKYLWNLLQYW), PinA (WRWWKWWK), PinB (WPTKWWK), and XYP1 (KIKWFKAMKSIAKFIAKDQLKKHL)-were evaluated using AlphaFold2 structure predict

Non-severe burn injury leads to sustained molecular changes in the brain in a murine model.

Paediatric burn patients, including those with non-severe burns, have an increased risk of admission to hospital for mental health conditions for many years after the burn, even in children too young at the time of the burn to remember the incident. This study aimed to investigate the long-term physiological impact of non-severe burn injuries and non-burn trauma (NBT) on the brain in mice to understand whether there is a sustained impact of such injuries on the brain that may be linked to the increased mental health morbidity observed in patients. Mice were exposed to either a non-severe burn

Supervised machine learning and molecular docking modeling to identify potential Anti-Parkinson's agents.

Parkinson's disease is a neurodegenerative condition that affects the brain's neurons, and causes malfunction of nerve cells and their death. A neurotransmitter called dopamine interacts with the part of the brain in charge of coordination and movement. In general, the brain produces less dopamine as Parkinson's disease worsens; therefore, it becomes harder to control the movements. In this study, a dataset collected from CHEMBL library was applied to build four machine learning models using three different descriptors functions to determine the best models with the best features and suggest t

Molecular view of the interactions between the amphiphilic drug propranolol hydrochloride and model lipid membranes.

HYPOTHESIS: Many clinically used drugs are amphiphiles, yet the effect of their amphiphilicity on their therapeutic activity is poorly understood. As cell membranes are the first barrier a drug encounters on route to its site of action, we propose that an amphiphilic drug's membrane activity will impact its clinical efficacy. EXPERIMENT: Comprehensive quantitative detail on the membrane interactions of the amphiphilic β-blocker drug, (±) propranolol has been investigated, using a synergistic combination of surface area-pressure measurements, ellipsometry, Brewster angle microscopy and selecti

Mediation analysis of the molecular phenotypes in a severe MASH-like liver injury mouse model.

Metabolic dysfunction-associated steatohepatitis (MASH), a severe form of fatty liver disease, is a leading cause of cirrhosis and lacks effective therapies. Understanding the molecular mediators of disease progression remains a critical gap. This study aimed to investigate the roles of molecular phenotypes as mediators of MASH disease features in a diet-induced mouse model. Data used for these analyses were from a previous study where male and female CC042 mice were fed either a control or high-fat, high-sucrose (HF/HS) diet for 20, 40, or 60 weeks. Associations and mediated relationships bet

Modeling the Mutational Effects on Biochemical Phenotypes of SARS-CoV-2 Using Molecular Fields.

The ongoing evolution of SARS-CoV-2 has given rise to variants with enhanced transmissibility and pathogenicity, many of which harbor mutations in the receptor-binding domain (RBD) of the viral spike protein. These mutations often confer increased viral fitness and immune evasion by modulating interactions with the human ACE2 receptor (hACE2) and escaping neutralizing antibodies. Accurate prediction of the functional consequences of such mutations-particularly their effects on receptor binding and antibody escape-is critical for assessing the public health threat posed by emerging variants. Valproate Exposure as an In vitro Model for Studying Morpho-Molecular Features of ASD: A Systematic Review.

INTRODUCTION: Autism Spectrum Disorder (ASD) is a complex neurodevelopmental disorder with a strong genetic and environmental basis. It frequently causes social and communication deficits, as well as repetitive behaviours. Valproic acid (VPA) has been shown to induce autisticlike features in animal models when administered during critical development periods. However, not much is known about its effect on cells to replicate ASD characteristics in vitro. OBJECTIVE: This review explores in vitro VPA models to elucidate the molecular and morphological characteristics of ASD, emphasizing their po
