Foetal ultrasound measurement imputations based on growth curves versus multiple imputation chained equation (MICE).

BACKGROUND: Ultrasound measures are valuable for epidemiologic studies of risk factors for growth restriction. Longitudinal measurements enable investigation of rates of change and identification of windows where growth is impacted more acutely. However, missing data can be problematic in these studies, limiting sample size, ability to characterise windows of vulnerability, and in some instances creating bias. We sought to compare a parametric linear mixed model (LMM) approach to multiple imputation in this setting with multiple imputation by chained equation (MICE) methodology. METHODS: Ultr

Longitudinal beta-binomial modeling using GEE for overdispersed binomial data.

Longitudinal binomial data are frequently generated from multiple questionnaires and assessments in various scientific settings for which the binomial data are often overdispersed. The standard generalized linear mixed effects model may result in severe underestimation of standard errors of estimated regression parameters in such cases and hence potentially bias the statistical inference. In this paper, we propose a longitudinal beta-binomial model for overdispersed binomial data and estimate the regression parameters under a probit model using the generalized estimating equation method. Non-Parametric Mediation Analysis of Non-Markov Illness-Death Model.

The illness-death model is widely used to characterize disease progression over time. Previous work focuses either on estimation without causal interpretation or on causal interpretation under a strong Markov assumption where the terminal event depends on the status but not the timing of the intermediate event. To bridge the research gap, we propose a new definition of counterfactual hazard that relaxes the Markov assumption by considering the entire history of the intermediate event. We derive an identification formula that involves an integral with respect to the probability density function

Competing risk model with a nonparametric form of relative risks.

Competing risks, referring to multiple mutually exclusive cause-specific events in survival data, are commonly seen in medical and engineering studies. In the presence of covariates, the Cox proportional hazards model is the most common approach to modeling covariate effects as the relative risk component for each cause-specific hazard function. However, the linear form of covariate effects in the existing models can be too restrictive to satisfy in practice. In this work, we propose a Cox regression model for competing risk survival data where the relative risk of each cause-specific hazard t

A storage model with random release rate for modeling exposure to food contaminants.

This paper presents the study of a continuous-time piecewise-deterministic Markov process for describing the temporal evolution of exposure to a given food contaminant. The quantity X of food contaminant present in the body evolves through its accumulation after repeated dietary intakes on the one hand, and the pharmacokinetics behavior of the chemical on the other hand. In the dynamic modeling considered here, the accumulation phenomenon is modeled by a simple marked point process with positive i.i.d. marks, and elimination in between intakes occurs at a random linear rate theta X, randomness

Comparison of machine learning and logistic regression as predictive models for adverse maternal and neonatal outcomes of preeclampsia: A retrospective study.

INTRODUCTION: Preeclampsia, one of the leading causes of maternal and fetal morbidity and mortality, demands accurate predictive models for the lack of effective treatment. Predictive models based on machine learning algorithms demonstrate promising potential, while there is a controversial discussion about whether machine learning methods should be recommended preferably, compared to traditional statistical models. METHODS: We employed both logistic regression and six machine learning methods as binary predictive models for a dataset containing 733 women diagnosed with preeclampsia. Particip

Computing assumption-lean bounds for per-protocol effects.

BACKGROUND: A common approach used to estimate per-protocol effects, or the effect of perfect adherence to a defined protocol on a specified outcome, is to artificially censor individuals when they deviate from the protocol. As adherence is not under control of the investigator, selection bias is a threat to the validity of per-protocol analyses. METHODS: Here, we describe nonparametric bounds for the per-protocol effect. Importantly, these bounds describe the range of all possible per-protocol effects concordant with the observed data under the sole additional assumption-beyond those of a st

SAM-HC: a Bayesian nonparametric construction of hybrid control for randomized clinical trials using external data.

In situations where high-quality external data are available or when it is challenging to recruit participants to the control arm of a randomized and controlled clinical trial (eg rare or pediatric diseases), it is desirable to borrow information from external data to augment the control arm. However, a main challenge in borrowing information from external data is to accommodate potential heterogeneous subpopulations across the external and trial data. We apply a Bayesian nonparametric model called the Shared Atoms Model (SAM) to identify overlapping and unique subpopulations across datasets,

Negatives about positivity and consistency as conditions for causal inference.

Positivity and causal consistency are sometimes presented as general validity requirements. Positivity, however, is not a general requirement for causal inference and does not address the more demanding practical needs for adequate numbers of observations. Common statistical methods in health and medical research depend on large-sample approximations; yet, approximation accuracy is rarely discussed in research reports, even in cases where it has clearly failed. Prioritization by generality and practical importance shifts the emphasis to approximation accuracy over positivity, with the latter b

A Simple Diagnostic for the Positivity Assumption for Continuous Exposures.

The positivity or experimental treatment assignment assumption is a fundamental requirement in causal analyses, invoked to ensure that identifiability holds without extrapolating beyond what the observed data can reveal. Positivity is well understood in the context of binary and categorical treatments, and has been thoroughly discussed-from how the assumption can be assessed to approaches that may be used when the assumption is suspected not to hold. Positivity extends to the context of continuous exposures, such as doses, however it has been given very little formal consideration. In this man

Stratification-based instrumental variable analysis framework for nonlinear effect analysis.

Nonlinear causal effects are prevalent in many research scenarios involving continuous exposures, and instrumental variables (IVs) can be employed to investigate such effects, particularly in the presence of unmeasured confounders. However, common IV methods for nonlinear effect analysis, such as IV regression or the control-function method, have inherent limitations, leading to either low statistical power or potentially misleading conclusions. In this work, we propose an alternative IV framework for nonlinear effect analysis, which has recently emerged in genetic epidemiology and addresses m

Flexible and efficient estimation of causal effects with error-prone exposures: a control variates approach for measurement error.

Exposure measurement error is a ubiquitous but often overlooked challenge in causal inference with observational data. Existing methods accounting for exposure measurement error largely rely on restrictive parametric assumptions, while emerging data-adaptive estimation approaches allow for less restrictive assumptions but at the cost of flexibility, as they are typically tailored toward rigidly defined statistical quantities. There remains a critical need for assumption-lean estimation methods that are both flexible and possess desirable theoretical properties across a variety of study designs

Sensitivity analysis for studies transporting prediction models.

We consider estimation of measures of model performance in a target population when covariate and outcome data are available from a source population and covariate data, but not outcome data, are available from the target population. In this setting, identification of measures of model performance is possible under an untestable assumption that the outcome and population (source or target) are independent conditional on covariates. In practice, this assumption is uncertain and, in some cases, controversial. Therefore, sensitivity analysis may be useful for examining the impact of assumption vi

Joint mirror procedure: controlling false discovery rate for identifying simultaneous signals.

In many applications, the process of identifying a specific feature of interest often involves testing multiple hypotheses for their joint statistical significance. Examples include mediation analysis, which simultaneously examines the existence of the exposure-mediator and the mediator-outcome effects, and replicability analysis, aiming to identify simultaneous signals that exhibit statistical significance across multiple independent studies. In this work, we present a new approach called the joint mirror (JM) procedure that effectively detects such features while maintaining false discovery

Application of hypothetical strategies in acute pain.

Since the publication of ICH E9 (R1), "Addendum to statistical principles for clinical trials: on choosing appropriate estimands and defining sensitivity analyses in clinical trials," there has been a lot of debate about the hypothetical strategy for handling intercurrent events. Arguments against the hypothetical strategy are twofold: (1) the clinical question has limited clinical/regulatory interest; (2) the estimation may need strong statistical assumptions. In this article, we provide an example of a hypothetical strategy handling use of rescue medications in the acute pain setting. We arg

How to analyze continuous and discrete repeated measures in small-sample cross-over trials?

To optimize the use of data from a small number of subjects in rare disease trials, an at first sight advantageous design is the repeated measures cross-over design. However, it is unclear how these within-treatment period and within-subject clustered data are best analyzed in small-sample trials. In a real-data simulation study based upon a recent epidermolysis bullosa simplex trial using this design, we compare non-parametric marginal models, generalized pairwise comparison models, GEE-type models and parametric model averaging for both repeated binary and count data. The recommendation of w

Infinite hidden Markov models for multiple multivariate time series with missing data.

Exposure to air pollution is associated with increased morbidity and mortality. Recent technological advancements permit the collection of time-resolved personal exposure data. Such data are often incomplete with missing observations and exposures below the limit of detection, which limit their use in health effects studies. In this paper, we develop an infinite hidden Markov model for multiple asynchronous multivariate time series with missing data. Our model is designed to include covariates that can inform transitions among hidden states. We implement beam sampling, a combination of slice s

Emergence of criticality in the transportation passenger flow: scaling and renormalization in the Seoul bus system.

Social systems have recently attracted much attention, with attempts to understand social behavior with the aid of statistical mechanics applied to complex systems. Collective properties of such systems emerge from couplings between components, for example, individual persons, transportation nodes such as airports or subway stations, and administrative districts. Among various collective properties, criticality is known as a characteristic property of a complex system, which helps the systems to respond flexibly to external perturbations. This work considers the criticality of the urban transp

The estimation of direct and indirect causal effects in the presence of misclassified binary mediator.

Mediation analysis serves to quantify the effect of an exposure on an outcome mediated by a certain intermediate and to quantify the extent to which the effect is direct. When the mediator is misclassified, the validity of mediation analysis can be severely undermined. The contribution of the present work is to study the effects of non-differential misclassification of a binary mediator in the estimation of direct and indirect causal effects when the outcome is either continuous or binary and exposure-mediator interaction can be present, and to allow the correction of misclassification. Benchmarking road safety: lessons to learn from a data envelopment analysis.

Road safety performance indicators (SPI) have recently been proposed as a useful instrument in comparing countries on the performance of different risk aspects of their road safety system. In this respect, SPIs should be actionable, i.e. they should provide clear directions for policymakers about what action is needed and which priorities should be set in order to improve a country's road safety level in the most efficient way. This paper aims at contributing to this issue by proposing a computational model based on data envelopment analysis (DEA). Based on the model output, the good and bad a
