A Monte Carlo risk assessment model was developed to estimate the public health risk of importing prime cuts of beef infested with Taenia saginata to Norway from an endemic area in southern Africa. The model predicted that 21 (lower 5% = 1, upper 95% = 56) viable cysts would be present in domestic prime cuts during 1996 and 1997, with 8 (0 to 21) of them being ingested without sufficient heat treatment to kill the parasite. These cysts were expected to cause 2 (0 to 7) human infections. Corresponding figures for the imported prime cuts were 1,260 (99 to 2,900) viable cysts, 462 (37 to 1,065) i

A dynamic simulation model was used to assess the long-term effect of importing beef from an area with a high prevalence of Taenia saginata among cattle. The input of the model was from a Monte Carlo simulation model. The model predicted substantial increases in the prevalence of T. saginata in domestic cattle and in the incidence and prevalence of infections in humans that would last for more than one decade even if importation of infected beef was stopped after 2 years. The results were strongly influenced by various assumptions about proportion of human carriers infected abroad. Only in a s

One of the key events in the HIV-1 life cycle is reverse transcription (RT), during which single-stranded viral RNA (ssRNA) is converted into double-stranded DNA (dsDNA). This process occurs inside the mature HIV-1 capsid and, once it reaches a critical threshold, drives capsid rupture. This uncoating is essential for infection because it releases viral genetic material into the host cell nucleus. Despite its importance, many mechanistic details of this process remain to be fully understood. To address this gap, we develop a multiscale computational method for simulating RT inside the capsid,

INTRODUCTION: Proton therapy is an advanced radiotherapy technique that reduces radiation exposure to healthy tissues while maintaining tumor control. However, out-of-field radiation dose remains a concern due to their potential contribution to secondary cancer risks, particularly in pediatric patients and in the fetus of pregnant patients. Accurate assessment of out-of-field dose is challenging due to the mixed radiation field composed of neutrons, protons, electrons, photons, and alphas with a wide energy range.
METHODS: This study investigates how in-phantom measurements with lithium fluor

BACKGROUND: In-beam positron emission tomography (PET) integrates dedicated detectors into proton therapy systems, enabling real-time acquisition of proton-induced positron-emitting activity. By pre-delivering a subset of single-energy proton spots as probes, in-beam PET has the potential to support online proton range verification and may inform subsequent spot delivery. However, the limited acquisition time and low yields of positron emitters result in noisy reconstructed images, and the open geometry of dual-panel PET further introduces stretching artifacts.
PURPOSE: To address these issue

BACKGROUND: Health care systems face growing fiscal pressure while AI reaches clinical parity in several domains. UK National Health Service expenditure rose by 52%, while Australia's health expenditure grew by 29% between 2019 and 2023. Yet large-scale cost savings from AI remain limited, largely because implementation constraints continue to outweigh technical capability.
OBJECTIVE: This study develops a Bayesian budget-impact framework to estimate AI-driven gross cost savings in radiology, workflow optimization, and workforce optimization in the United Kingdom and Australia, explicitly acc

Mediation analysis is a powerful tool for exploring the causal relationships between exposures and outcomes that are mediated by intermediate variables. In this paper, we propose a flexible Bayesian mediation analysis framework to accommodate zero-inflated mediators, compatible with a wide range of outcome distributions. This novel technique employs Bayesian models for both the mediator and the outcome, utilizing Markov Chain Monte Carlo algorithms for parameter estimation. While addressing the challenges posed by an excess of zeros, we further decompose the mediation effects into components i

BACKGROUND: Accurate and non-invasive detection of tumor cells remains a major challenge in biomedical engineering and clinical diagnostics. Traditional imaging methods often face limitations in resolution, accessibility, or invasiveness. We propose a computational framework combining Bayesian inference with the Virtual Element Method (VEM) to address the inverse problem of tumor characterization using surface temperature measurements.
METHODS: The forward thermal response of biological tissues was modeled using Pennes' bioheat equation, with skin surface temperature distributions as measurab

Intrinsically disordered proteins (IDPs) and regions (IDRs) challenge structural characterization due to their dynamic conformational ensembles. Existing computational approaches for modeling these ensembles are often computationally expensive, inflexible, or inaccessible to non-experts, particularly for multi-domain or multi-chain proteins. We present Ensemblify, an open-source, user-friendly Python package for generating and analyzing conformational ensembles of IDPs/IDRs. Ensemblify uses a Monte Carlo algorithm coupled with neighbor-aware sampling of dihedral angles from curated or user-def

Objective.Scattered coincidences are a major source of quantitative bias in positron emission tomography (PET) and must be compensated during reconstruction using an estimate of scattered coincidences per line-of-response and time-of-flight bin. Such estimates are typically obtained from simulators with simple cylindrical scanner models that omit detector physics. Incorporating detector sensitivities for scatter is challenging, as scattered coincidences have less constrained properties (e.g. incidence angles) than true coincidences.Approach.We integrated a 5D single-photon detection probabilit

Intraoperative electron radiation therapy (IOERT) is commonly delivered without intraoperative imaging, limiting patient-specific planning and requiring fast, reliable treatment calculations in the operating room. As a result, monitor unit (MU) calculations are often performed using spreadsheets and static look-up tables derived from measurements in water. We present Kali MC, an open source, Python-based software developed to streamline IOERT absorbed dose visualization, MU calculation and report generation for treatments delivered with the Liac HWL mobile accelerator. MOTIVATION: Cell-cell interactions (CCIs) are fundamental to multicellular organisms and play crucial roles in diverse biological processes and disease mechanisms. Understanding CCIs is vital for deciphering disease pathogenesis and developing therapeutic strategies. Although numerous computational methods have been developed to infer CCIs from complex biological data, most existing approaches rely primarily on single-gene expression levels and ligand-receptor databases, often failing to capture the nuanced network-wide changes characteristic of disease states.
RESULT: We propose MetaCCI, a n

PURPOSE: In radiotherapy, the primary goal is to eliminate cancer cells while minimizing damage to healthy cells. The low-energy electrons generated by radiation beams interact with deoxyribonucleic acid (DNA), causing double-strand breaks that are challenging to repair, ultimately leading to tumor cell death, since the radiation provokes a clustered damage and the tumor cells have defective or dysregulated DNA repair pathways.
METHODS: We performed Monte Carlo simulations using standard GEANT4 classes, employing an atomistic model of DNA composed of 1122 base pairs, with each atom represente

Cyclic peptides have emerged as a compelling class of bioactive scaffolds, but de novo design of target-binding cyclic peptides from protein structures remains challenging. Here, we present HighMorph, an interaction-guided framework that combines protein-protein interaction information with artificial intelligence for rational cyclic peptide design. HighMorph integrates Monte Carlo tree search with a Transformer-based policy-value network to efficiently explore cyclic peptide sequence space, while incorporating explicit atomic-level hydrogen bond constraints extracted from reference protein-pr

Spectral shaping is a computed tomography (CT) dose optimization technique that adjusts source voltage and filtration to reduce patient radiation exposure without compromising image quality. Traditionally, radiation dose has been assessed using the computed tomography dose index (CTDI). Emerging dosimetric approaches aim to enable patient-specific evaluations by estimating organ absorbed doses, providing a more accurate representation of the biological impact. This study investigates spectral shaping for an extremity photon-counting detector (PCD) CT, through organ absorbed dose estimation. Mo

BACKGROUND: Estimating effective dose (E) for CT is often done with dose-length product (DLP) and effective dose conversion factors, a.k.a. k-factors. Previous studies have primarily focused on k-factor values derived from certain fixed-sized phantoms but have not extensively explored all the contributing factors and associated variations in E.
PURPOSE: In this study, we investigated the contributing factors and variations of E using k-factors as surrogates in adult and pediatric CT exams and provided empirical formula to estimate a comprehensive new set of k-factors.
METHODS: Effective dose

Monte Carlo simulations were conducted to evaluate the dose distribution characteristics and motion robustness of fixed-angle versus arc irradiation techniques in boron neutron capture therapy (BNCT), using a head phantom. For fixed-angle irradiation, an anterior-posterior (AP) beam was oriented towards the facial plane. In contrast, the arc technique involved a continuous 180° rotation, from the left to the right, around the central axis traversing the face. The target was located in the nasal cavity region. Results demonstrated the dosimetric superiority of the arc technique, which increased

This paper established a Susceptible-Infected-Recovered-Dead (SIRD) model to fit and predict the COVID-19 epidemic in China. On the premise of passing the Markov Chain Monte Carlo (MCMC) robustness check, we counterfactually assessed the effectiveness of partial unlocking and vaccination policies. Our key findings are as follows: (1) The SIRD model predicted that China could fully contain COVID-19 in July, 2020, which is consistent with reality; (2) If the strict social distancing policies are relaxed before the epidemic ends, it will lead to a second rebound. Even worse, the rebound increases
