Autophagic degradation of glycogen in skeletal muscles of the newborn rat.

Large amounts of glycogen accumulate in rat skeletal muscle fibers during the late fetal stages and are mobilized in the first postnatal days. This glycogen depletion is relatively slow in the immature leg muscles, in which extensive deposits are still found 24 hr after birth and, to some extent, persist until the 3rd day. In the more differentiated psoas muscle and especially in the diaphragm, the glycogen stores are completely mobilized already during the early hours. Section of the sciatic nerve 3 days before birth or within the first 2 hr after delivery does not affect glycogen depletion i

The Role of Skeletal Muscle Mitochondria in NLRP3 Inflammasome Signaling.

Skeletal muscle mitochondria possess the ability to autoregulate their health and functioning by the orchestration of mitochondrial quality control (MQC) pathways. This plasticity allows them to adapt to various stimuli, such as exercise. However, under pathological conditions, mitochondria can become dysfunctional, generating damage-associated molecular patterns (DAMPs), such as reactive oxygen species (ROS) and oxidized mitochondrial DNA (mtDNA). These DAMPs can launch an innate immune response, with consequences of widespread inflammation and atrophy. Integral to this is the NLRP3 inflammas

Current Progress in the Role of Ferroptosis in Skeletal Muscle Atrophy.

Atrophy of skeletal muscles, caused by multiple factors, including ageing, disuse, trauma and systemic diseases, is a major pathological condition that affects physical performance and systemic health by the loss of muscle function; in effect, weakening the whole body and leading to disabling conditions such as sarcopenia, frailty and increased fall risk. First reported in 2012 by Dixon et al., ferroptosis is a newly identified, distinct type of regulated cell death characterised by unique biochemical and morphological features and differs from classic apoptosis and necrosis. One hallmark feat

Dysregulation of the ubiquitin-proteasome system in aging skeletal muscle.

Protein homeostasis (proteostasis) is essential for maintaining skeletal muscle integrity, and its disruption is a central feature of aging related sarcopenia. The ubiquitin-proteasome system (UPS) is the primary pathway responsible for selective protein degradation in muscle. However, its regulation during physiological aging remains incompletely understood. Most studies have focused on muscle-specific E3 ubiquitin ligases, particularly MuRF1 and MAFbx/atrogin-1, which are widely used as molecular markers of muscle atrophy. However, changes in E3 ligase expression do not consistently correspo

Cloning and characterization of intermediate Homer1E of human skeletal muscle.

HOMER proteins are scaffolding proteins critically involved in intracellular signaling, calcium homeostasis, receptor trafficking and synaptic plasticity. Three human HOMER genes (HOMER1, HOMER2, HOMER3) are expressed in both neurons and skeletal muscle fibers. HOMER1 long isoforms multimerize via their carboxy-terminal coiled-coil domain, forming signaling clusters with postsynaptic density proteins. Despite growing evidence of HOMER1 relevance in skeletal muscle physiology, molecular data on human muscle remain scarce. This study investigates the expression of alternatively spliced HOMER1 tr

The impact of hyperparathyroidism on skeletal muscle pathophysiology and physical function.

Primary hyperparathyroidism (PHPT), increasingly diagnosed in its asymptomatic form, is associated with clinically significant neuromuscular dysfunction. Growing evidence indicates that skeletal muscle is a direct target of parathyroid hormone (PTH), with chronic PTH excess impairing mitochondrial bioenergetics, promoting proteolysis, and altering muscle-bone-adipose endocrine crosstalk. Experimental studies confirm PTH receptor type 1 (PTHR1) expression in muscle fibers and satellite cells, while transcriptomic analyses of PHPT muscle reveal dysregulation of calcium signaling and oxidative me

Redox Control of Skeletal Muscle Function and Adaptations to Exercise.

Since the first clear observation that skeletal muscle contained increased amounts of free radical species in the early 1980s, there has been increasing interest in how such species are formed and what functions they might have in skeletal muscle. Superoxide and nitric oxide are formed in increased amounts in skeletal muscle during contractile activity and these species together with the important derivative, hydrogen peroxide, are now recognised to play an important role in the physiological responses of muscle to contractile activity or exercise. In this chapter the potential sites for gener

Alcohol-Mediated Skeletal Muscle Adaptations and Their Impact on Comorbidities.

At-risk alcohol use has significant adverse effects on multiple tissue and organ systems, including the skeletal muscle. The pathophysiological mechanisms underlying alcohol-induced dysfunctional skeletal muscle (SKM) mass are multifactorial, involving decreased protein synthesis, increased protein degradation, impaired glucose homeostasis, bioenergetic dysregulation, aberrant extracellular matrix remodeling, impaired satellite cell function, circadian rhythm disruption, and epigenomic adaptations. This review provides a brief overview of these major alcohol-induced mechanisms of SKM dysfuncti

The role of microRNAs in dexamethasone-induced skeletal muscle atrophy.

Muscle atrophy is characterized by a decrease in muscle mass, strength, and activity. Recently, it was determined that microRNAs (miRNAs) can regulate muscle atrophy and that dexamethasone (Dex), an allergy and autoimmune disorder treatment that can induce muscle atrophy. Therefore, this study was designed to identify miRNAs expressed in Dex-induced muscle atrophy in mice using small RNA sequencing. A total of 820 miRNAs were identified, with 58 miRNAs expressed explicitly in atrophic muscles. Dex-induced muscle atrophy miRNAs clustered separately from the differential miRNAs in aging, disuse,

Aging in the Skeletal Muscle Revealed by Molecular Immunohistochemical Imaging.

The skeletal muscle is a complex organ mainly composed of multinucleated fibres responsible for contractile activity, but it also contains postnatal myogenic stem cells (i.e., satellite cells), connective cells and nervous cells. The skeletal muscle is severely affected by aging, undergoing a progressive reduction in muscle mass, strength and endurance in a condition known as sarcopenia. The mechanisms underlying sarcopenia still need to be completely clarified, but they are undoubtedly multifactorial, involving all cell types constituting the skeletal muscle. Immunohistochemistry has widely b

Role of Peptides in Skeletal Muscle Wasting: A Scoping Review.

BACKGROUND: Systemic muscle wasting is a prevalent condition that predicts adverse health outcomes in aging and disease. Despite its clinical relevance, the development of predictive biomarkers and effective pharmacological therapies remains limited. Peptides have recently gained attention for their diverse bioactive functions, positioning them as promising biomarkers and therapeutic agents for muscle wasting. METHODS: This scoping review systematically identifies studies examining the direct association between well-defined peptides and clinical components of muscle wasting: muscle mass, str

MBNL splicing factors regulate the microtranscriptome of skeletal muscles.

Muscleblind like splicing regulators (MBNLs) govern various RNA-processing steps, including alternative splicing, polyadenylation, RNA stability and mRNA intracellular localization. In myotonic dystrophy type 1 (DM1), the most common muscular dystrophy in adults, MBNLs are sequestered on toxic RNA containing expanded CUG repeats, which leads to disruption of MBNL-regulated processes and disease features of DM1. Herein, we show the significance of MBNLs in regulating microtranscriptome dynamics during the postnatal development of skeletal muscles and in microRNA (miRNA) misregulation observed i
