Loop-mediated isothermal amplification (LAMP) holds great promise for rapid nucleic acid detection. However, its application is hindered by … ity from 50 to 65 °C (optimal), and the coefficient of variation of Tt at the same concentration of nucleic acids is less than 5%. Our study optimizes the LAMP reaction using AuNRs, thereby reducing false positives and offering great potential for clinical diagnosis.

In recent years, misdiagnosis or delayed diagnosis of Chlamydia psittaci (C. psittaci) infections has led to frequent outbreaks of severe public health events, such as severe pneumonia and respiratory distress, drawing increasing attention. Rapid and simple detection methods are vital for early intervention to reduce severity and mortality. In this study, we designed highly specific RPA primers and crRNA (CRISPR RNA) based on the highly conserved CPSIT_0429 gene in the C. psittaci genome, and preliminarily established a nucleic acid detection method for C. psittaci using the RPA-CRISPR/Cas12a

CRISPR-based nucleic acid diagnostics are a promising class of point-of-care tools that could dramatically improve healthcare outcomes for millions worldwide. However, these diagnostics require nucleic acid preamplification, an additional step that complicates deployment to low resource settings. Here, we developed CATNAP (Cas trans-nuclease detection of amplified products), a method that integrates isothermal linear DNA amplification with Cas12a detection in a single reaction. CATNAP uses a nicking enzyme and DNA polymerase to continuously generate single-stranded DNA, activating Cas12a's tra

In line with the global research priorities on sexually transmitted infections (STIs), we developed a molecular point-of-care test (POCT) for the parasite Trichomonas vaginalis that causes the STI trichomoniasis. Trichomoniasis remains the most common curable nonviral STI. We report on the use of specific T. vaginalis single-stranded-tailed DNA primers in combination with recombinase polymerase amplification (RPA) in a protein-free nucleic acid lateral flow (NALF) device for use at the point of care. The use of aminated DNA probes eliminates the need for proteins (such as streptavidin or hapte

BACKGROUND: Accurate and prompt diagnosis of tuberculosis in children is challenging due to non-specific clinical presentation and the low bacillary load of samples. Low-complexity manual nucleic acid amplification tests (LC-mNAATs) such as loop-mediated isothermal amplification (TB-LAMP) are World Health Organization (WHO)-recommended rapid molecular diagnostic tests. Even in resource-limited settings, they have good diagnostic accuracy in adults.
OBJECTIVES: To determine the diagnostic accuracy of LC-mNAATs for the detection of pulmonary tuberculosis in children (< 10 years) with presumptiv

Molecular diagnostics is an increasingly important clinical tool, especially in routine sampling. We evaluated two non-invasive methods (oral swabs and mouthwashes) for sampling nucleic acids from the oral/pharyngeal area. We created a workflow from sample collection (n = 59) to RT-qPCR based analysis. The samples were further characterized in terms of their cellular composition as well as the purity, degradation and microbial content of the derived DNA/RNA. We determined the optimal housekeeping genes applicable for these types of samples. The cellular composition indicated that mouthwashes c…

Nucleic acid testing (NAT) has revolutionized diagnostics by providing precise, rapid, and scalable detection methods for diverse biological samples. These recent advancements satisfy the increasing demand for on-site diagnostics, yet sample preparation remains a significant bottleneck for achieving highly sensitive diagnostic assays. There is an unmet need for compatible, efficient, and lab-free sample preparation for point-of-care NAT. To address this, we developed a portable, lab-free, and battery-powered device for extracting nucleic acids. We explored using low centrifugal forces with exi…

Hepatitis B virus (HBV) infection is a major global health problem of widespread concern. Clinically, serological assays are the most widely used diagnostic tests for HBV infection, with the presence of HBsAg in the serum being indicative of acute and chronic hepatitis B infection. However, increased identification of HBV DNA positive but HBsAg negative cases has greatly promoted the use of molecular assays for more accurate HBV diagnosis. Over the past few decades, especially since the outbreak of COVID-19, significant advancements have been made in the techniques and devices for nucleic acid

Upstream sample preparation remains the bottleneck for point-of-need nucleic acid testing due to its complexity and time-consuming nature. Sample preparation involves extracting, purifying, and concentrating nucleic acids from various matrices. These processes are critical for ensuring the accuracy and sensitivity of downstream nucleic acid amplification and detection. However, current sample preparation methods are often laboratory-based, requiring specialized equipment, trained personnel, and several hours of processing time. As a result, sample preparation often limits the speed, portabilit…
