Generating mRNA Encoding Anti-HBV Designer Epigenome Modifiers.

Chronic hepatitis B virus (HBV) infection poses significant global health challenges, with an estimated 257.5 million individuals affected, leading to approximately 880,000 deaths annually from complications such as cirrhosis and hepatocellular carcinoma. Although antiviral therapies are available, they often fail to eliminate the stable viral reservoir of covalently closed circular DNA within infected cells. Recent advancements in epigenome editing offer a promising alternative to traditional mutagenic approaches, providing a strategy to silence HBV replication and transcription directly. Thi

Genomic characterization and pathogenicity of a porcine hemagglutinating encephalomyelitis virus strain isolated in China.

Porcine hemagglutinating encephalomyelitis virus (PHEV) is a member of the genus betacoronavirus within the family coronaviridae, which invades the central nervous system (CNS) via peripheral nervous system and causes encephalomyelitis or vomiting and wasting disease (VWD) in sucking piglets. Up to now, although few complete nucleotide sequences of PHEV have been reported, they are not annotated. This study aimed to illuminate genome characterization, phylogenesis and pathogenicity of the PHEV/2008 strain. The full length of the PHEV/2008 strain genome was 30,684 bp, with a G + C content of 37

A novel papillomavirus isolated from a nasal neoplasia in an Italian free-ranging chamois (Rupicapra r. rupicapra).

Most amniotes are the hosts of many, distantly related papillomaviruses (PVs). Infection by PVs can be asymptomatic, or lead instead to benign or malignant lesions. However, PVs infecting animals and associated with malignancies are still largely understudied. In the present study, we communicate the complete genome of a novel PV found in a nasal neoplasia of a free-ranging alpine chamois (Rupicapra r. rupicapra) in an Italian national park. Long-PCR and cloning approaches followed for Sanger sequencing were used to identify the first PV found in chamois. The genome of the novel virus - RrupPV

[Cytomegalovirus (CMV)].

Human cytomegalovirus (HCMV) is a ubiquitous beta human herpesvirus type 5. Compared to other human herpesviruses, HCMV is the largest, with a genome of approximately 235 kb containing approximately 250 ORFs with the potential to encode proteins. Usually, HCMV asymptomatically infects the host during childhood, and establishes life-long latency. The infection is life-threatening for infants and immunocompromised individuals, because of direct cytopathicity by viral replication, causing systemic organ injuries. Intrauterine infection occasionally causes microcephaly, sensorineural hearing loss

HTLV-1 genetic diversity of 52 complete sequences from 14 African countries reveals novel variants and a lack of typical P12/P8 and P30 accessory proteins in HTLV-1b, d, and f genotypes.

Central Africa is the largest region of human T-cell Leukaemia virus (HTLV-1) endemicity with several million people estimated to be infected. Based on the study of the LTR region, it is also the region with the highest HTLV-1 diversity, with the presence of genotypes a-b and d-g. However, complete genomic sequences are still lacking for Central African genotypes. Here, we report the first large collection of complete HTLV-1 sequences for genotypes b, d and f from Central Africa and neighbouring countries. We identified substantial diversity within the HTLV-1b genotype, including a newly defin

Evolutionary Patterns and Genotype-Specific Amino Acid Mutations of Tick-Borne Encephalitis Virus.

Tick-borne encephalitis virus (TBEV) is a significant tick-borne flavivirus responsible for severe human diseases. Here, we analyzed the genetic diversity and evolutionary dynamics of TBEV using 263 genome sequences from the NCBI database and identified key amino acid mutations. TBEV sequences were classified into five genotypes-Baikalian, European, Far-Eastern, Himalaya, and Siberian-showing ORF nucleotide similarity of 81.5% to 88.0% and amino acid similarity of 93.0% to 96.4%. Extensive recombination between genotypes was not observed. Entropy analyses revealed highly variable sites distrib

Ovine Herpesvirus 2 Glycoprotein B Complementation Restores Infectivity to a Bovine Herpesvirus 4 gB-Null Mutant.

Ovine herpesvirus 2 (OvHV-2) and bovine herpesvirus 4 (BoHV-4) are gamma herpesviruses that belong to the genera Macavirus and Rhadinovirus, respectively. As with all herpesviruses, both OvHV-2 and BoHV-4 express glycoprotein B (gB), which plays an essential role in the infection of host cells. In that context, it has been demonstrated that a BoHV-4 gB-null mutant is unable to infect host cells. In this study, we used homologous recombination to insert OvHV-2 ORF 8, encoding gB, into the BoHV-4 gB-null mutant genome, creating a chimeric BoHV-4 virus carrying and expressing OvHV-2 gB (BoHV-4∆gB

Multiple novel caliciviruses identified from stoats (Mustela erminea) in the United Kingdom.

The Caliciviridae family, comprising positive-sense RNA viruses, is characterised by its non-enveloped, small virions, broad host range, and notable tendency for host switching. These viruses are primarily associated with gastroenteric disease, though they can lead to haemorrhagic or respiratory infections. Our study employed a metagenomics analysis of faecal samples from stoats (Mustela erminea), identifying two novel calicivirus species, named stoat vesivirus and stoat valovirus. Stoat vesivirus was identified in three samples (ST008, ST006, ST004), exhibiting a genome wide nucleotide identi

Identification and characterization of interferon regulatory factor 1 from Lateolabrax japonicus involved in antiviral immune response against grouper nervous necrosis virus infection.

Interferon regulatory factors (IRFs) play a key role in mediating the host response against pathogen infection and other important biological processes. In the present study, an interferon regulation factor 1 gene was identified from Lateolabrax japonicus (designated LjIRF-1), the cDNA sequence of LjIRF-1 was 1394 bp long, and with an open reading frame (ORF) of 945 bp that encodes a peptide of 314 amino acids. Bioinformatics data showed that LjIRF-1 possesses a DNA-binding domain (DBD) and two low complexity regions, which shared 56-81% identity to other fish IRF-1s. The LjIRF-1 transcripts w

Norovirus GII.Pe Genotype: Tracking a Foodborne Outbreak on a Cruise Ship Through Molecular Epidemiology, Brazil, 2014.

Norovirus (NoV) is recognized as the most common cause of foodborne outbreaks. In 2014, an outbreak of acute gastroenteritis occurred on a cruise ship in Brazil, and NoV became the suspected etiology. Here we present the molecular identification of the NoV strains and the use of sequence analysis to determine modes of virus transmission. Food (cream cheese, tuna salad, grilled fish, orange mousse, and vegetables soup) and clinical samples were analyzed by ELISA, conventional RT-PCR, qRT-PCR, and sequencing. Genogroup GII NoV was identified by ELISA and conventional RT-PCR in fecal samples from

T-cell epitope mapping of ORF2 and ORF3 proteins of human hepatitis E virus.

Little data are available on cellular immune responses during infection with hepatitis E virus (HEV). We therefore mapped CD4 T-cell epitopes in open reading frame (ORF)2 and ORF3 proteins of HEV using lymphocyte proliferation assays and overlapping peptide libraries. Proliferation of peripheral blood mononuclear cells from 40 patients with acute hepatitis E and 21 healthy controls with recombinant HEV ORF2 protein or pools of overlapping HEV ORF2/ORF3 peptides was measured. HLA-DQB1 and HLA-DRB1 alleles were also determined. Mononuclear cells from patients with hepatitis E more often showed s

Differential expression of alternatively spliced pX mRNAs in HTLV-I-infected cell lines.

Human T cell leukemia/lymphotropic virus (HTLV) is a complex 9 kb human retrovirus with at least eight alternatively spliced mRNAs expressed from the 3' or pX region of the genome. These mRNAs allow for the expression of novel proteins from the previously recognized pX open reading frames I and II in addition to Tax, Rex and p21rex encoded from orf III and IV. These alternatively spliced messages have been detected using reverse-transcriptase polymerase chain reaction (RT/PCR) amplification in HTLV-I-transformed T cell lines as well as in peripheral blood mononuclear cells (PBMC) from infected

Expression of alternatively spliced human T-lymphotropic virus type I pX mRNA in infected cell lines and in primary uncultured cells from patients with adult T-cell leukemia/lymphoma and healthy carriers.

Although human T-cell lymphotropic virus type I (HTLV-I) is the etiologic agent of adult T-cell leukemia/lymphoma (ATL), the role of viral gene expression in the progression to and maintenance of the leukemic state in vivo is unclear because of the inability of most previous studies to readily detect HTLV-I RNA in infected individuals. By using the reverse transcriptase-polymerase chain reaction, we detected spliced messages for the HTLV-I pX regulatory genes in primary uncultured cells from ATL patients and healthy asymptomatic carriers. In addition to the expected doubly spliced pX message,

Isolation and characterization of a highly pathogenic Orthoflavivirus associated with ascites syndrome in Chinese soft-shelled turtles (Pelodiscus sinensis).

Since 2016, mass mortality events characterized primarily by severe abdominal swelling and ascites have emerged in Chinese soft-shelled turtle (Pelodiscus sinensis) farms in China, leading to substantial economic losses. However, the cause of the disease has remained elusive, as no bacteria or parasites were detected in the diseased turtles. Tissue homogenate filtrate was used to infect Vero, GCO, TSK, TSL, and C6/36 cells, all of which produced cytopathic effects (CPE). Spherical, suspected viral particles with diameters approximately 30 nanometers (nm) were observed in the kidney tissue of d

Generation of Interferon-γ-Expressing Single-Cycle Viral Hemorrhagic Septicemia Virus (rVHSV-A-IFNγ-ΔG) and Potential as a Vaccine in Olive Flounder (Paralichthys olivaceus).

Single-cycle viruses generated by the deletion of gene(s) essential for viral replication in the genome can be a way to enhance the safety of attenuated virus-based vaccines because single-cycle viruses can infect cells only once and cannot produce infective viral particles. In the present study, not only to guarantee safety as a prophylactic vaccine but also to enhance the protective efficacy, a single-cycle viral hemorrhagic septicemia virus (VHSV) lacking the G gene and containing olive flounder (Paralichthys olivaceus) interferon-γ (IFNγ) ORF between N and P genes of the viral genome (rVHS

Interferon regulatory factor 6 gene from Oreochromis niloticus is involved in resistance against tilapia lake virus.

Tilapia lake virus (TiLV) is an emerging virus that seriously threatens the tilapia industries worldwide. Interferon regulatory factors (IRFs) are the crucial mediators regulating the response of interferon (IFN) to combat invading viruses. Here, a IRF6 homologs from tilapia (Oreochromis niloticus) were characterized and analyzed. The full length of OnIRF6 mRNA contains a 249 bp 5'untranslated region (UTR), an open reading frame (ORF) of 1473 bp and a 3' UTR of 1209 bp. The OnIRF6 typically shared the conserved domains and phylogenetic relationship with IRF homologs of other species and showed

Description of the first isolates of guinea fowl corona and picornaviruses obtained from a case of guinea fowl fulminating enteritis.

Guinea fowl fulminating enteritis has been reported in France since the 1970s. In 2014, a coronavirus was identified and appeared as a possible viral pathogen involved in the disease. In the present study, intestinal content from a guinea fowl involved in a new case of the disease in 2017 was analysed by deep sequencing, revealing the presence of a guinea fowl coronavirus (GfCoV) and a picornavirus (GfPic). Serial passage assays into the intra-amniotic cavity of 13-day-old specific pathogen-free chicken eggs and 20-day-old conventional guinea fowl eggs were attempted. In chicken eggs, isolatio

Inhibition of interferon I induction by non-structural protein NSs of Puumala virus and other vole-associated orthohantaviruses: phenotypic plasticity of the protein and potential functional domains.

The orthohantavirus Puumala virus (PUUV), which is transmitted by bank voles (Clethrionomys glareolus), and other vole-borne hantaviruses contain in their small (S) genome segment two overlapping open reading frames, coding for the nucleocapsid protein and the non-structural protein NSs, a putative type I interferon (IFN-I) antagonist. To investigate the role of NSs of PUUV and other orthohantaviruses, the expression pattern of recombinant NSs constructs and their ability to inhibit human IFN-I promoter activity were investigated.
