Exploring the recurrence and metastasis of breast invasive ductal carcinoma based on machine learning and survival analysis.

OBJECTIVE: Invasive ductal carcinoma (IDC), the predominant histopathological subtype comprising about 80% of breast malignancies, continues to pose a significant clinical challenge due to frequent recurrence. Existing relapse prediction models remain limited in accuracy and generalizability. This study aimed to construct and validate machine learning-based models for predicting 5-year (short- to medium term) recurrence and metastasis risk in IDC, based on recurrence-free survival (RFS) analysis. METHODS: A total of 640 IDC cases diagnosed between January 2017 and December 2019 were enrolled,

SRC as a Prognostic and Immunomodulatory Biomarker in Acute Myeloid Leukemia: A Multi-Omics Study.

The bone marrow tumor microenvironment (TME) is critical for acute myeloid leukemia (AML) progression, immune evasion, and treatment resistance. SRC, a non-receptor tyrosine kinase involved in multiple oncogenic pathways, has not been systematically characterized in AML in relation to prognosis and immune regulation. We integrated bulk transcriptomic and single-cell RNA-sequencing datasets from TCGA, BeatAML, and GEO. Immune-related targets were identified using xCell-based immune scoring and weighted gene co-expression network analysis (WGCNA), followed by protein-protein interaction analysis

Integrative bulk and single-cell transcriptomic analysis identify an ac4C-related signature in lung adenocarcinoma.

BACKGROUND: N4-acetylcytidine (ac4C) RNA modification is a critical epitranscriptomic regulator of cancer progression, yet its specific biological functions and regulatory patterns in lung adenocarcinoma (LUAD) remain poorly understood. This study aims to characterize the clinical relevance and potential regulatory patterns of ac4C-related features in the LUAD tumor microenvironment (TME). METHODS: We integrated bulk RNA-seq data from TCGA and single-cell RNA-seq (scRNA-seq) data from GEO. Weighted gene co-expression network analysis (WGCNA) identified prognosis-related modules. A robust six-

Machine learning-driven transcriptomic and single-cell profiling of programed cell death patterns in colon cancer.

ObjectiveColon cancer ranks among the most prevalent malignancies globally. Despite advances in therapy, patients' prognosis remains poor, particularly in advanced stages. Programed cell death (PCD), including over 20 patterns, plays a pivotal role in colon cancer progression. However, a systematic analysis of the PCD regulatory network in colon cancer is lacking.MethodsWe comprehensively analyzed various PCD patterns in colon cancer using bulk transcriptomic and single-cell transcriptomic data from GEO and TCGA databases. Multiple machine learning algorithms were used to identify Key PCD patt

Prediction of isometric motor tasks and effort levels based on high-density EMG in patients with incomplete spinal cord injury.

OBJECTIVE: The development of modern assistive and rehabilitation devices requires reliable and easy-to-use methods to extract neural information for control of devices. Group-specific pattern recognition identifiers are influenced by inter-subject variability. Based on high-density EMG (HD-EMG) maps, our research group has already shown that inter-subject muscle activation patterns exist in a population of healthy subjects. The aim of this paper is to analyze muscle activation patterns associated with four tasks (flexion/extension of the elbow, and supination/pronation of the forearm) at thre

Brain regions with abnormal network properties in severe epilepsy of Lennox-Gastaut phenotype: Multivariate analysis of task-free fMRI.

OBJECTIVE: Lennox-Gastaut syndrome, and the similar but less tightly defined Lennox-Gastaut phenotype, describe patients with severe epilepsy, generalized epileptic discharges, and variable intellectual disability. Our previous functional neuroimaging studies suggest that abnormal diffuse association network activity underlies the epileptic discharges of this clinical phenotype. Herein we use a data-driven multivariate approach to determine the spatial changes in local and global networks of patients with severe epilepsy of the Lennox-Gastaut phenotype. METHODS: We studied 9 adult patients an

Three-dimensional virtual histology of the human hippocampus based on phase-contrast computed tomography.

We have studied the three-dimensional (3D) cytoarchitecture of the human hippocampus in neuropathologically healthy and Alzheimer's disease (AD) individuals, based on phase-contrast X-ray computed tomography of postmortem human tissue punch biopsies. In view of recent findings suggesting a nuclear origin of AD, we target in particular the nuclear structure of the dentate gyrus (DG) granule cells. Tissue samples of 20 individuals were scanned and evaluated using a highly automated approach of measurement and analysis, combining multiscale recordings, optimized phase retrieval, segmentation by m

Research on Pathological Voice Recognition Based on XGBoost.

With the continuous growth of human social communication, the number of people suffering from voice disorders is also increasing. Due to the objective and non-invasive advantages of acoustic detection methods for pathological voice, the use of speech signal analysis for pathological voice recognition has become a research hotspot. This article first selected 101 continuous vowels /a/ from the German SVD database as the research object. Secondly, using wavelet packet technology for time-frequency analysis, four nonlinear dynamic parameters, namely approximate entropy, sample entropy, fuzzy entr

Discovery of a DNA methylation episignature for Weiss-Kruszka syndrome.

Weiss-Kruszka syndrome (WSKA; OMIM 618619) is a rare autosomal dominant neurodevelopmental disorder caused by haploinsufficiency of ZNF462, a zinc-finger transcription factor involved in chromatin regulation and early embryonic development. WSKA is characterized by developmental delay, hypotonia, craniofacial dysmorphic features (around 8) and variable congenital anomalies. Genome-wide DNAm profiling was performed on peripheral blood DNA from 9 WSKA cases with (likely) pathogenic ZNF462 variants and matched controls to look for differential methylation. Analysis using the EpiSign™ pipeline ide

Multi-omics clustering combined with multiple machine learning to identify epigenetic features in low-grade glioma patients.

BACKGROUND: Epigenetic heterogeneity has been demonstrated in a wide range of cancers including leukemia, colorectal cancer, and glioma; in the case of low-grade glioma (LGG), its association with tumor malignant progression and immune microenvironmental regulation has been less reported. This study aimed to further explore the application of epigenetics in LGG. METHODS: Epigenetic subtypes were identified based on consensus clustering of multi-omics data, and prognostic models were constructed based on 101 machine learning combinations. Biological differences among different risk groups were

Nonsubsampled rotated complex wavelet transform (NSRCxWT) for medical image fusion related to clinical aspects in neurocysticercosis.

Neurocysticercosis (NCC) is a parasite infection caused by the tapeworm Taenia solium in its larvae stage which affects the central nervous system of the human body (a definite host). It results in the formation of multiple lesions in the brain at different locations during its various stages. During diagnosis of such symptomatic patients, these lesions can be better visualized using a feature based fusion of Computed Tomography (CT) and Magnetic Resonance Imaging (MRI). This paper presents a novel approach to Multimodality Medical Image Fusion (MMIF) used for the analysis of the lesions for t

Machine learning identifies a distinct microbiota signature in immune checkpoint inhibitor colitis compared with inflammatory bowel disease.

BACKGROUND: Immune checkpoint inhibitors (ICIs) have revolutionized oncology by enhancing antitumor immune responses. However, their use is frequently associated with immune-mediated adverse events, including colitis (ICIs-colitis). This condition shares clinical and histological features with inflammatory bowel diseases (IBD), such as Crohn's disease (CD) and ulcerative colitis (UC). Emerging evidence highlights the gut microbiota's role in both ICIs-colitis and IBD pathogenesis. This study aimed to determine whether a distinct microbiota profile, assessed via machine learning, could differen

Automated detection of schizophrenia using nonlinear signal processing methods.

Examination of the brain's condition with the Electroencephalogram (EEG) can be helpful to predict abnormality and cerebral activities. The purpose of this study was to develop an Automated Diagnostic Tool (ADT) to investigate and classify the EEG signal patterns into normal and schizophrenia classes. The ADT implements a sequence of events, such as EEG series splitting, non-linear features mining, t-test assisted feature selection, classification and validation. The proposed ADT is employed to evaluate a 19-channel EEG signal collected from normal and schizophrenia class volunteers. A dataset

Pathway activation patterns in diffuse large B-cell lymphomas.

Deregulation of cell signaling pathways controlling cell growth and cell survival is a common feature of all cancers. Although a core repertoire of oncogenic mechanisms is widely conserved between various malignancies, the constellation of pathway activities can vary even in patients with the same malignant disease. Modern molecularly targeted cancer drugs intervene in cell signaling compensating for pathway deregulation. Hence characterizing tumors with respect to pathway activation will become crucial for treatment decisions. Here we have used semi-supervised machine learning methodology to

Depth-Sensitive Optical Sensing for Non-Invasive Measurement of Human Muscle Activity.

Human muscle anatomy consists of multiple layers, each contributing to movement through complex patterns of activation. Conventional non-invasive sensing techniques, such as surface electromyography (sEMG) and mechanomyography (MMG), primarily capture aggregate muscle activity and provide limited depth-dependent information. As different movements may involve distinct combinations of superficial and deeper muscles, access to depth-dependent information could improve the discrimination of motion patterns that are difficult to distinguish using surface measurements alone. To address this limitat

Diagnostic host gene signature for distinguishing enteric fever from other febrile diseases.

Misdiagnosis of enteric fever is a major global health problem, resulting in patient mismanagement, antimicrobial misuse and inaccurate disease burden estimates. Applying a machine learning algorithm to host gene expression profiles, we identified a diagnostic signature, which could distinguish culture-confirmed enteric fever cases from other febrile illnesses (area under receiver operating characteristic curve > 95%). Applying this signature to a culture-negative suspected enteric fever cohort in Nepal identified a further 12.6% as likely true cases. Our analysis highlights the power of data-
