Regulation of redox metabolism in the mouse oocyte and embryo.

Energy homeostasis of the oocyte is a crucial determinant of fertility. Following ovulation, the oocyte is exposed to the unique environment of the Fallopian tube, and this is reflected in a highly specialised biochemistry. The minute amounts of tissue available have made the physiological analysis of oocyte intermediary metabolism almost impossible. We have therefore used confocal imaging of mitochondrial and cytosolic redox state under a range of conditions to explore the oxidative metabolism of intermediary substrates. It has been known for some time that the early mouse embryo metabolises

Carbohydrate metabolism in erythrocytes of copper deficient rats.

Dietary copper deficiency is known to adversely affect the circulatory system of fructose-fed rats. Part of the problem may lie in the effect of copper deficiency on intermediary metabolism. To test this, weanling male Long-Evans rats were fed for 4 or 8 weeks on sucrose-based diets containing low or adequate copper content. Copper deficient rats had significantly lower plasma and tissue copper as well as lower plasma copper, zinc-superoxide dismutase activity. Copper deficient rats also had a significantly higher heart:body weight ratio when compared to pair-fed controls. Direct measurement o

Gas chromatography-mass spectrometric studies of canine urinary metabolism.

After the urine was treated with urease, lyophilized, and trimethylsilylated, it was examined for metabolic profiles in Dalmatian dogs and Shetland sheepdogs by gas chromatography-mass spectrometry (GC/MS), which simultaneously analyzes organic acids, amino acids, sugars, sugar alcohols, purine and pyrimidine bases, and nucleosides. The profiles were compared with those from human specimens. As clarified in past studies, Dalmatian dogs showed an extreme decrease in allantoin, which is the final product of purine metabolism in the canine of other species, and a marked detection of uric acid pea

Embryonic metabolism of foetal fuels in whole-embryo culture.

The whole-embryo culture technique has been extensively used to evaluate embryonic growth and development using morphological criteria. However, the system also provides an opportunity to assess the status of embryonic metabolism to determine biochemical maturation and alterations induced by xenobiotics or changes in substrate availability. For example, using (14)C-labelled substrates the rates of product formation may be determined for a variety of foetal fuels such as glucose and the ketone body, beta-hydroxybutyrate. During organogenesis glucose metabolism is characterized by high rates of

Age-dependent reprogramming of vascular metabolism compromises endothelial resilience to inflammation-induced endothelial dysfunction.

Endothelial dysfunction is a hallmark of vascular inflammation and aging. However, the role of cellular bioenergetics in the age-dependent resilience of endothelial function to inflammation has not yet been characterized. Therefore, the aim of this study was to determine whether IL-1β-induced inflammation differentially influences vascular metabolism and endothelial function in the isolated aorta of aged mice compared to young mice. Aorta from young (3-8 months) and old (22-28 months) C57BL/6 mice was isolated and subjected to ex vivo analysis of endothelial function using wire myography and v

Enzymatic Changes in Red Blood Cells of Diamond-Blackfan Anemia.

Diamond-Blackfan anemia is a congenital bone marrow failure syndrome characterized by red blood cell (RBC) aplasia with varied malformations in infants. Elevated activity of adenosine deaminase (ADA) has been considered as a useful biomarker of Diamond-Blackfan anemia, and ADA assay has been shown to be more sensitive than genetic diagnosis. Approximately, 80% of the examined patients showed elevated ADA activity, whereas genetic tests of ribosome subunit genes identified mutations in approximately 60% of the patients. We previously reported that reduced glutathione (GSH) levels in RBCs may se

Immunometabolism and HIV-1 pathogenesis: food for thought.

Antiretroviral therapies efficiently block HIV-1 replication but need to be maintained for life. Moreover, chronic inflammation is a hallmark of HIV-1 infection that persists despite treatment. There is, therefore, an urgent need to better understand the mechanisms driving HIV-1 pathogenesis and to identify new targets for therapeutic intervention. In the past few years, the decisive role of cellular metabolism in the fate and activity of immune cells has been uncovered, as well as its impact on the outcome of infectious diseases. Emerging evidence suggests that immunometabolism has a key role

Pyrimidine 5'-nucleotidase and oxidative damage in red blood cells transfused to beta-thalassemic children.

Pyrimidine 5' nucleotidase (P5'N) acquired deficiency has been found in several hematologic disorders including beta-thalassemia. Our previous studies suggested that the aldehydes produced during membrane lipid peroxidation could play a role in P5'N inactivation in thalassemia. To evaluate the effects of the thalassemic "environment" on transfused red blood cells, we tested P5'N, pyruvate kinase (PK), glucose 6-phosphate dehydrogenase (G-6PD) activity, creatine content, reduced glutathione (GSH) levels and the hexose monophosphate shunt (HMS) in the red cells of homozygous transfusion-dependen

Transaldolase haploinsufficiency in subjects with acetaminophen-induced liver failure.

Transaldolase (TAL) is an enzyme in the pentose phosphate pathway (PPP) that generates NADPH for protection against oxidative stress. While deficiency of other PPP enzymes, such as transketolase (TKT), are incompatible with mammalian cell survival, mice lacking TAL are viable and develop progressive liver disease attributed to oxidative stress. Mice with homozygous or heterozygous TAL deficiency are predisposed to cirrhosis, hepatocellular carcinoma (HCC) and acetaminophen (APAP)-induced liver failure. Both mice and humans with complete TAL deficiency accumulate sedoheptulose 7-phosphate (S7P)

The Redox Role of G6PD in Cell Growth, Cell Death, and Cancer.

The generation of reducing equivalent NADPH via glucose-6-phosphate dehydrogenase (G6PD) is critical for the maintenance of redox homeostasis and reductive biosynthesis in cells. NADPH also plays key roles in cellular processes mediated by redox signaling. Insufficient G6PD activity predisposes cells to growth retardation and demise. Severely lacking G6PD impairs embryonic development and delays organismal growth. Altered G6PD activity is associated with pathophysiology, such as autophagy, insulin resistance, infection, inflammation, as well as diabetes and hypertension. Aberrant activation of

Hexose uptake and transport in polymorphonuclear leukocytes from patients with glycogen storage disease Ib.

Neutrophil functions and glucose metabolism are known to be impaired in glycogen storage disease (GSD) Ib patients. The uptake of nonmetabolizing glucose analogues into polymorphonuclear leukocytes (PMN) of GSD Ib patients was studied. 2-Deoxyglucose (2-DOG) and 3-O-methylglucose are transported across the cell membrane by facilitated diffusion mediated by the glucose transporter. Because 2-DOG is phosphorylated within the cell, its uptake rate reflects hexose transport as long as phosphorylation is not rate-limiting. These conditions prevail only at low 2-DOG concentrations. Transport of 5 mi

Biodegradation of oxidized lignite by Pseudomonas sp. TX1: Degradation products and metabolomic characterization.

Microbial degradation offers a green pathway for resource recovery and clean conversion of low-rank coal. However, the mechanisms governing the extracellular depolymerization of coal macromolecules and intracellular metabolic regulation remain poorly understood. This study integrated solid-phase structural characterization, liquid-phase product analysis, and untargeted metabolomics to investigate lignite biodegradation by an indigenous isolate, Pseudomonas sp. TX1. During TX1 treatment of oxidized lignite, the culture supernatant reached an absorbance at 450 nm (A450) of 2.67 after 10 days, in

NUAK isoforms promote glucose and glutamine metabolic reprogramming driven colon cancer progression and migration through regulation of AMPK/mTOR signaling.

Colorectal cancer (CRC) cells undergo significant alterations in glucose and glutamine metabolism to support rapid growth and migration. However, the molecular mechanisms underlying these metabolic changes remain incompletely understood. Although NUAK1 and NUAK2, members of the NUAK kinase family have been implicated in CRC tumorigenesis, their specific roles in metabolic reprogramming that drives CRC progression and metastasis have not been elucidated. In this study, we demonstrate that NUAK1 and NUAK2 are overexpressed in human CRC tissues and that their expression is associated with increas

Comparative lifespan trajectories of brain energy metabolism in human and macaque.

The brain's high energy demands are met by a suite of metabolic pathways that selectively appropriate glucose across development, yet how this metabolic strategy changes across the lifespan and whether it is conserved across species remains incompletely understood. We previously mapped five core energy metabolism pathways across the human cortex and lifespan, revealing a fundamental dichotomy between anabolic and energy-producing pathway expression: the pentose phosphate pathway, which contributes to biomass, peaks prenatally, while glycolysis, the tricarboxylic acid cycle, and oxidative phosp

Plant-Derived Modulators of Tumor Metabolism as Novel, Efficacious, and Low-Toxicity Therapeutic Agents for Cancer Treatment.

Metabolic reprogramming is a core hallmark of malignancy, enabling tumor cells to sustain rapid proliferation, evade immune elimination, and develop resistance to therapy. Although a wide range of plant-derived phytochemicals exhibit anticancer activity with comparatively low toxicity, their capacity to disrupt specific metabolic dependencies exploited by tumors has not been comprehensively synthesized. This review brings together current mechanistic evidence showing how major phytochemical classes, including polyphenols, terpenes and terpenoids, glucosinolates, and alkaloids, interfere with p

Temporal Pattern and Metabolic Mechanisms of Ecosystem Carbon Fluxes Under Decadal-Long Nitrogen Enrichment in an Alpine Grassland.

Due to widespread nitrogen limitation, reactive nitrogen input is expected to stimulate vegetation carbon fixation, thus potentially offsetting the decomposition-induced soil carbon losses. However, this projection is largely based on short-term measurements. The crucial question is whether such responses persist over time. Combining a decade-long nitrogen-addition experiment in an alpine grassland with biochemical and plant metabolomics analyses, we showed that a decade of nitrogen addition elicited only a transient increase in net ecosystem productivity (NEP; balance between ecosystem carbon
