Afferent and efferent innervation of the rat esophagus. A tracing study with horseradish peroxidase and nuclear yellow.

The technique of retrograde labeling of nerve cells with HRP and nuclear yellow as well as transganglionic anterograde HRP-tracing of sensory projections into the CNS were used to establish the motor and sensory innervation pattern of two parts of the rat esophagus: the cervical and the abdominal segment. For comparison, also the innervation of the anterior wall of the stomach was studied. Application of HRP to the cervical part of the esophagus resulted in bilateral labeling of neurons in the nucleus ambiguus exclusively, while application of the tracer to the abdominal part was followed by l

Distribution of glycoconjugates in normal human skin using biotinyl lectins and avidin-horseradish peroxidase.

Lectin binding patterns in normal human skin were studied using five different biotinyl lectins and avidin-horseradish peroxidase. The staining pattern was specific for each lectin. In the epidermis, peanut agglutinin (PNA) and soybean agglutinin (SBA) preferentially stained the cell membranes of keratinocytes in the spinous and granular cell layers, indicating changes in the saccharide residues during keratinocyte differentiation. In the secretory segment of an eccrine sweat gland, the superficial cells gave a strong granular staining with Ricinus communis agglutinin (RCA). Dolichos biflorus

Mechanism of action of thioureylene antithyroid drugs in the rat: possible inactivation of thyroid peroxidase by propylthiouracil.

We have previously shown that the thioureylene antithyroid drugs 6-propyl-2-thiouracil (PTU) and 1-methyl-2-mercaptoimidazole (MMI) can inactivate thyroid peroxidase (TPO) in a model iodination system containing relatively high concentrations of iodide. The purpose of the present study was to determine whether these drugs may also inactivate TPO in vivo in rats. Assays for total TPO activity after injection of PTU or MMI did not prove to be a valid approach. As TPO inactivation might be expected to result in a relatively prolonged inhibition of enzyme activity, most of our experiments involved

Enzyme-linked immunoassay in the diagnosis of leptospirosis in domestic animals using peroxidase-conjugated protein-A.

The ELISA test for detection of antibodies to Leptospirosis in domestic animals was performed using Staphylococcal protein-A coupled to peroxidase in place of antisera to IgG. Genus- and type-specific antigens were extracted with SDS technique from four pathogenic serotypes and two non-pathogenic ones, and they were identified with the aid of ELISA using specific rabbit antisera. Micro-agglutination (MA) and ELISA were compared using a total of 48 positive swine sera and a 100% agreement was obtained, since with sera from 16 dogs clinically suspected of Leptospirosis the ELISA resulted highly

Neural pathways to lower urinary tract identified by retrograde axonal transport of horseradish peroxidase.

A new technique for tracing nerve fibers supplying specific organs by retrograde axonal transport of the enzyme horseradish peroxidase (HRP) was used to determine the innervation of various segments of the lower urinary tract. HRP was injected at the level of the bladder neck, postprostatic urethral segment, pelvic nerve, pudendal nerve, and pelvic floor musculature to determine precise nerve supply to each site, as well as possible overlap. The technique determines the exact spinal level of innervation, as well as the number of nuclei directly related to the motor neuron axons supplying inner

[Study of the connections of brain stem respiratory nuclei by the technic of retrograde transport of horseradish peroxidase].

The method of horseradish peroxidase retrograde axonal transport was used to study the connections between neurons of respiratory nuclei and reticular formation of medulla oblongata and descending fibre pathways from these structures in the spinal cord of cats. It is shown that neurons of the nucleus ambiguus and nucleus retroambiguus (the ventral respiratory group) are connected with the ventrolateral part of nucleus tractus solitarius (the dorsal respiratory group) and from direct bilateral connections with each other and with the medial part of medulla oblongata. A wide developed system of

Blood glutathione-peroxidase levels in skin diseases: effect of selenium and vitamin E treatment.

Blood glutathione-peroxidase (GSH-Px) was determined in 61 healthy subjects and 506 patients with various skin disorders. Depressed levels were observed in patients with psoriasis, eczema, atopic dermatitis, vasculitis, mycosis fungoides and dermatitis herpetiformis. Low values of GSH-Px were also found in some patients with pemphigoid, acne conglobata, polymyositis, rheumatoid arthritis, scleroderma and systemic lupus erythematodes. Vegetarian diet, malnutrition and alcohol abuse could possibly account for the low values in some patients. Fifty patients with low GSH-Px levels were treated wit

Erythrocyte superoxide dismutase, catalase and glutathione peroxidase in glucose-6-phosphate dehydrogenase deficiency.

Glucose-6-phosphate dehydrogenase (G-6-PD) deficient erythrocytes are particularly sensitive to oxidant stress. In order to evaluate if these cells are protected against oxidant damage, we assayed the antioxidant enzymes superoxide dismutase (SOD), catalase and glutathione peroxidase (GSH-Px) in erythrocytes of G-6-PD deficient (hemizygous and heterozygous) subjects. Normal levels of antioxidant enzymes were found in all subjects examined both with positive and negative histories of haemolytic crisis after fava bean or drug ingestion. In contrast, high levels of catalase and GSH-Px were found

Preparation of protein A-peroxidase monoconjugate using a heterobifunctional reagent, and its use in enzyme immunoassays.

Protein A-peroxidase monoconjugate was prepared in solution using a heterobifunctional reagent, N-succinimidyl-3-(2-pyridyldithio)-propionate. The yield of the monoconjugate was much higher than that obtained with current methods. An immunoassay method was developed in which protein A-peroxidase monoconjugate served as a universal tool. Protein A-peroxidase monoconjugate was taken up by IgG molecules immobilized on an excess of solid phase antigen. The ability of free antigen to inhibit the binding of antibody, measured as inhibition of conjugate up take, served as the basis for quantification

Uptake sites of horseradish peroxidase after injection into peritoneal structures: defining some pitfalls.

Following injection of horseradish peroxidase (HRP) either into the jejunal wall or the peritoneal cavity, neurons in the dorsal motor nucleus of the vagus nerve, celiac, nodose and spinal ganglia, and ventral and lateral horns of the spinal cord from the mid-thoracic to lumbar segments were labeled. When HRP was injected into the wall of the exteriorized gut, neurons of the spinal cord were not labeled. Furthermore, there was a significant decrease in the number of labeled neurons in the dorsal motor nucleus and ganglia examined. These results indicate that HRP injected into the intestinal wa

A kinetic study on the diffusion-coupled reaction of a basic horseradish peroxidase adsorbed on the carboxymethylcellulose membrane.

The rate of reactions of a basic horseradish peroxidase adsorbed on the carboxymethylcellulose membrane was measured spectrophotometrically by monitoring the state of the enzyme. For the reactions with hydrogen peroxide and cyanide dissolved in media the diffusion process was rate limiting both in the static and flowing media, and kinetic features of the enzyme were masked. The rate of CO recombination of the ferrous enzyme was measured by flash photolysis in the static media containing varying amounts of CO. Apparently, the initial rate of CO recombination was no more affected by the diffusio

Projection from the vestibular nuclei to the inferior olive in the cat: an autoradiographic and horseradish peroxidase study.

Injections of horseradish peroxidase (HRP) in the inferior olive of 13 cats resulted in the labeling of neurons in the medial (MVN) and descending vestibular nuclei (DVN) as well as in subgroups g, p.h., s.v., x and z. The contribution of groups p.h. and z. could not be definitively established. Injections of triated leucine in the vestibular nuclei and adjacent reticular formation in 4 cats indicated that the previously identified vestibular nuclei and subgroups project bilaterally to the dorsomedial cell column and ipsilaterally to subnucleus beta of the inferior olive. In view of the known

Resonance Raman study on yeast cytochrome c peroxidase. Effect of coordination and axial ligands.

Resonance Raman spectra are reported for native ferric cytochrome c peroxidase, its cyanide and fluoride compounds, those of the ferrous enzyme and its cyanide and carbonyl compounds, and the spectrum of the hydrogen peroxide compound, compound I. Band frequencies of ferric horseradish peroxidase isoenzyme C2 and its derivatives are also given. Comparison of the frequencies of the bands around 1400, 1500, 1560-1580, and 1610-1640 cm-1 with those of other hemoproteins and heme model compounds showed that in ferric highspin compounds in particular the bands are not only spin and oxidation sensit

[Absorption and magnetic circular dichroism spectra of nonequilibrium states of hemoproteins. III. Complexes of peroxidase].

Absorption and magnetic curcular dichroism spectra of nonequilibrium states of peroxidase and its complexes with F-, N3-, CN- produced by reduction of oxidased forms of proteins by thermalysed electrons at 77 degrees K were studied. Mixtures of high spin and low spin ferroforms were found in nonequilibrium states of peroxidase and complexes with F- and N3-, the content of the high spin ferroform increasing as follows: N3- complex less than peroxidase less than fluorine complex. Only low spin ferroforms was found after low temperature reduction of the cyanide complex. The existence of the low s

Isoelectric enzyme patterns of leukocyte peroxidase in normal controls and patients with neuronal ceroid-lipofuscinoses.

Using macro- and microanalytical isoelectric focussing techniques for separation of the soluble leukocytic peroxidase (hydrogen donor: p-phenylenediamine) we found 4 main isoenzyme components with pI at 9.6 (9.0), 7.6 (7.5), 6.2 (6.2) and 4.2 (4.7), which exhibited additional heterogeneities. The isoenzymes of the membrane-bound peroxidase displayed a similar pattern. The isoelectric subfractions of peroxidase in controls and patients with late-infantile (Jansky-Bielschowsky), juvenile (Spielmeyer-Sjögren) and adult (Kufs) types of neuronal ceroid-lipofuscinosis did not reveal any significant

Root growth restraint can be an acclimatory response to low pH and is associated with reduced cell mortality: a possible role of class III peroxidases and NADPH oxidases.

Low pH (<5.0) can significantly decrease root growth but whether this is a direct effect of H(+) or an active plant response is examined her … e that root growth inhibition from low pH can be part of an active plant response, and suggest that peroxidases may have a critical early role in reducing loss of cell viability and in the observed root growth constraint.
