Baseline Tumor Features and Systemic Immune Dynamics Underlying Efficacy in MSS Metastatic Colorectal Cancer Treated with Regorafenib, Ipilimumab, and Nivolumab.

Microsatellite-stable metastatic colorectal cancer (MSS mCRC) remains resistant to conventional immunotherapies. In a phase I trial, we observed encouraging efficacy of the combination of regorafenib, ipilimumab, and nivolumab (RIN), with a 27.6% overall response rate and a median overall survival of 20 months. The most pronounced benefits were observed in patients without liver metastases. To uncover immunologic mechanisms underlying response and resistance, we performed correlative studies of the tumor microenvironment (TME) and systemic immune features. Tumor biopsies from 8 patients and pe

Lenvatinib plus transarterial chemoembolization and PD-1 inhibitors as conversion therapies for unresectable intermediate-advanced hepatocellular carcinoma: a phase 2 trial and exploratory biomolecular study.

Conversion therapy remains an uncommon strategy for managing unresectable hepatocellular carcinoma (uHCC) due to limited evidence supporting its efficacy. To address this gap, we initiated a prospective phase 2 multicenter trial (NCT04997850) comparing the LEN-TAP regimen, combining lenvatinib, transarterial chemoembolization (TACE), and PD-1 inhibitors, against TACE alone in uHCC patients. The study's primary outcome was salvage liver resection (SLR) rate; secondary measures included objective response rate (ORR), overall survival (OS), event-free survival (EFS), recurrence-free survival (RFS

ABHD17C regulates the efficacy of lenvatinib in suppressing hepatocellular carcinoma.

INTRODUCTION: Lenvatinib is a first-line therapy for hepatocellular carcinoma (HCC), but its clinical efficacy is limited by drug resistance. ABHD17C, a depalmitoylation enzyme involved in HCC progression, has not been investigated in lenvatinib response. This study aimed to determine whether ABHD17C regulates the anti-tumor efficacy of lenvatinib in HCC. METHODS: Published single-cell RNA sequencing (scRNA-seq) data were analyzed to characterize ABHD17C expression in the HCC tumor microenvironment. Functional assays were performed in HCC cell lines to evaluate the effects of lenvatinib and A

Hepatocellular carcinoma organoids enable drug discovery and combination therapy across heterogeneous tumors.

Hepatocellular carcinoma (HCC) remains a leading cause of cancer mortality, with limited therapeutic options and poor clinical outcomes. Tumor heterogeneity is a defining feature of HCC and a major barrier to effective treatment. To address this, we established a biobank of patient-derived HCC organoids generated from diagnostic biopsies, capturing the diversity of clinical features across disease stages. Using a high-throughput phenotypic screening platform, we tested more than 1,600 compounds and identified multiple agents with strong antitumor activity, including candidates suitable for dru

Matched pair suitability of 89Zr and 177Lu-labeled L804-LLP2A theranostics for targeting VLA-4 in melanoma.

UNLABELLED: There is a need for chelators that are versatile for complexing multiple radiometals. We previously demonstrated that the macrocyclic 1,2-HOPO chelator, L804, has utility for complexing 177Lu(III) for radiopharmaceutical therapy and 89Zr(IV) for PET imaging with antibody-based agents. Here, we investigated whether 89Zr-radiolabelled peptidomimetic LLP2A could function as a diagnostic surrogate for [177Lu]Lu-LLP2A therapy in the targeting of VLA-4 overexpressed in melanoma, with and without an albumin-binding moiety (pIBA) for biological half-life extension. METHODS: L804-LLP2A (1)

Construction of ultra-stable magnetic imine-cyclization covalent organic frameworks for efficient extraction of phenylurea herbicides in plant beverages.

Imine-linked covalent organic frameworks (COFs) are considered as promising adsorbents to remove the water contaminants. However, the reversibility and hydrolyzation of imine bond may generate limited stability in long-term aqueous environment-based applications, and the improved stability of imine-linked COFs is highly desired. In this work, a carboxyl-quinoline linked COFs (CQ-COF) was synthesized by the cyclization of imine linkage via one-step modification. CQ-COF demonstrated high crystallinity, excellent stability and large surface area. Further integrating with magnetic nanoparticles en

Late-line options for patients with metastatic colorectal cancer: a review and evidence-based algorithm.

Over the past few years, several novel systemic treatments have emerged for patients with treatment-refractory metastatic colorectal cancer, thus making selection of the most effective later-line therapy a challenge for medical oncologists. Over the past decade, regorafenib and trifluridine-tipiracil were the only available drugs and often provided limited clinical benefit compared to best supportive care. Results from subsequent practice-changing trials opened several novel therapeutic avenues, both for unselected patients (such as trifluridine-tipiracil plus bevacizumab or fruquintinib) and
