Flow cytometric analysis of platelets from children with the Wiskott-Aldrich syndrome reveals defects in platelet development, activation and structure.

The pathophysiology of platelet dysfunction in the Wiskott-Aldrich immune deficiency syndrome (WAS) remains unclear. Using flow cytometry, we have characterized the functional properties of platelets from 10 children with WAS. Patients with WAS had thrombocytopenia, small platelets, increased platelet-associated IgG and reduced platelet-dense granule content. Levels of reticulated 'young' platelets were normal in the WAS patients. Although the mean numbers of platelet glycoprotein (GP) Ib, GPIIbIIIa and GPIV molecules per platelet appeared lower in WAS patients than in healthy controls, analys

Dual effects of oleic acid on Ca2+ mobilization and protein phosphorylation in human platelets in presence or absence of platelet activating factor.

This laboratory demonstrated earlier that oleic acid inhibited platelet activating factor (PAF)-induced aggregation and serotonin release of … that elevation of adenylate cyclase activity was not associated with free fatty acid inhibition of platelet activation. Interestingly, both PAF and oleic acid added separately to human platelets induced protein-tyrosine phosphorylation, but oleic acid did not cause any inhibition of PAF-induced protein-tyrosine phosphorylation.(ABSTRACT TRUNCATED AT 400 WORDS)

The paradox of platelet activation and impaired function: platelet-von Willebrand factor interactions, and the etiology of thrombotic and hemorrhagic manifestations in essential thrombocythemia and polycythemia vera.

Patients with essential thrombocythemia (ET) and polycythemia vera (PV), complicated by microvascular ischemic or thrombotic events, have shortened platelet survival, increased beta-thromboglobulin, platelet factor 4, and thrombomodulin levels, and increased urinary thromboxane B2 excretion. These are all reversible by inhibition of platelet cyclooxygenase 1 with aspirin, and are therefore indicative of platelet activation and platelet-mediated thrombotic processes. The thrombotic tendency persists as long as platelet counts are above the upper limit of normal (400 x 10 (9)/L). Despite strong …

Sociodemographic and Clinical Correlates of Markers of Immune Activation, Exhaustion and Platelet Activation among HIV-Infected Patients Initiating Antiretroviral Therapy in Dar es Salaam, Tanzania.

Chronic inflammation and persistent immune activation (IA) during HIV infection are associated with non-AIDS complications. We investigated sociodemographic and clinical characteristics influencing IA and exhaustion (IE), and platelet activation (PA) in newly diagnosed people living with HIV (PLHIV) and identified modifiable factors for early interventions. We analyzed baseline blood samples from 365 PLHIV participating in a trial investigating the effect of aspirin on IA, IE, and PA. We assessed levels of markers of monocyte activation (soluble CD14), platelet activation (soluble P-selectin),…

Serum activity of platelet-activating factor acetylhydrolase is a potential clinical marker for leptospirosis pulmonary hemorrhage.

Pulmonary hemorrhage has been recognized as a major, often lethal, manifestation of severe leptospirosis albeit the pathogenesis remains unclear. The Leptospira interrogans virulent serogroup Icterohaemorrhagiae serovar Lai encodes a protein (LA2144), which exhibited the platelet-activating factor acetylhydrolase (PAF-AH) activity in vitro similar to that of human serum with respect to its substrate affinity and specificity and thus designated L-PAF-AH. On the other hand, the primary amino acid sequence of L-PAF-AH is homologous to the alpha1-subunit of the bovine brain PAF-AH isoform I. The L

Serum platelet-activating factor acetylhydrolase (PAF-AH) activity in patients with hyperbilirubinemic hepatobiliary disease.

Platelet-activating factor acetylhydrolase (PAF-AH) is an enzyme that catalyzes hydrolysis of PAF and is important for regulation of serum PAF concentration. Twenty-seven patients with hyperbililubinemic hepatobiliary diseases (eight patients with liver cirrhosis, five with acute hepatitis and 14 with obstructive cholestasis) were studied. Serum PAF-AH activity was elevated in hyperbilirubinemic condition associated with liver cirrhosis, obstructive cholestasis or acute hepatitis. There was a significant correlation of serum PAF-AH activity with serum cholesterol (r=0.891, P=0.014 in liver cir

Platelet-activating factor enhances urokinase-type plasminogen activator gene expression in corneal epithelium.

PURPOSE: To determine whether platelet-activating factor (PAF), a lipid mediator that is accumulated in the cornea after alkali burn, induces the gene expression of urokinase-type plasminogen activator (uPA) in the corneal epithelium. Possible signaling mechanisms of uPA gene induction by PAF also were examined. METHODS: Rabbit corneas were cultured with or without PAF. One hour before stimulation, PAF antagonists or other modulators were added to PAF. In some experiments, the corneas were permeabilized to introduce guanosine triphosphate analogs into the corneal epithelial cells. Corneal epi

Systemic Platelet Activation and Respiratory Exacerbations, Pulmonary Symptoms, and Mortality among Current and Former Smokers in SPIROMICS.

RATIONALE: Platelet activation is elevated in chronic obstructive pulmonary disease (COPD) and associated with self-reported respiratory symptoms. Observational studies link the antiplatelet drug aspirin to lower exacerbation rates and fewer symptoms. However, it is unknown if platelet activation predicts incident respiratory exacerbations or mortality. OBJECTIVE: Is systemic platelet activation prospectively associated with respiratory exacerbations and mortality among ever-smokers with or at risk for COPD? METHODS: We measured two systemic platelet activation biomarkers, urinary 11-dehydro…

Illicit drug N-ethylpentylone potentiates platelet activation through 5-hydroxytryptamine2A receptor-mediated mitogen-activated protein kinase signaling.

BACKGROUND: Several illicit drugs have been reported to be associated with ischemic stroke and myocardial infarction. However, their underlying mechanisms remain poorly investigated. OBJECTIVES: In this study, we focused on N-ethylpentylone (NEP), a synthetic cathinone with potent hallucinogenic properties, and explored its role in platelet activation. METHODS: Platelet function assays and flow cytometry were used to evaluate the effects of NEP on platelet aggregation, spreading, clot retraction, and integrin αIIbβ3 activation. The role of 5-hydroxytryptamine2A receptor (5-HT2AR) in NEP-medi…

Col003 attenuates sepsis-associated coagulation dysfunction in mice in association with reduced platelet activation and NET formation.

BACKGROUND: Coagulation dysfunction is a major contributor to the increased mortality associated with sepsis (Levi et al., 2013; Gando et al., 2019 [1,2]), in which platelet activation, neutrophil activation, and neutrophil extracellular trap (NET) formation play important roles. Recent studies have suggested that HSP47 is involved in venous thromboembolism and platelet/neutrophil activation (Thienel et al., 2023a [3]); however, whether HSP47 contributes to platelet-mediated NET formation and coagulation dysfunction during sepsis remains unclear. OBJECTIVES: This study aimed to investigate th…

Biomimetic platelet membrane-camouflaged nanomicelles for synergistic hypoxia alleviation and immune activation to potentiate immunotherapy in TNBC.

Triple-negative breast cancer (TNBC) remains poor response to immunotherapy, largely attributable to the hypoxic tumor microenvironment that drives functional exhaustion of effector T cells and natural killer (NK) cells. To counter this, we engineered platelet-membrane-biomimetic nanomicelles (PM@(M/B)NM) for the co-delivery of metformin and BMS-1 with acid-responsive release profiles. The biomimetic nanocarriers retained characteristic platelet membrane proteins, effectively evading macrophage phagocytosis, prolonging systemic circulation, and enhancing active tumor accumulation. Platelet activation and mitophagy induction by thymic stromal lymphopoietin (TSLP) is associated with thrombosis in Kawasaki disease.

Kawasaki disease (KD) is an acute vasculitis of early childhood with unknown cause, which can be complicated by coronary artery lesions resulting in thrombosis. Fu and coworkers identify thymic stromal lymphopoietin (TSLP) to be increased in KD patients with thrombosis. They demonstrate that TSLP can activate platelets, induce mitophagy and trigger thrombus formation in vitro, suggesting that TSLP may have potential as a therapeutic target against KD-associated thrombosis. Commentary on: Fu L, et al. TSLP induces platelet mitophagy and promotes thrombosis in Kawasaki disease. Br J Haematol. 20

The binding of autotaxin to integrins mediates hyperhomocysteinemia-potentiated platelet activation and thrombosis in mice and humans.

Hyperhomocysteinemia (HHcy) is associated with an exaggerated platelet thrombotic response at sites of vascular injury. In this study, human medical examination showed that elevated human plasma Hcy levels correlated positively with enhanced blood coagulation and platelet activity, suggesting that humans with HHcy are more prone to thrombus formation at the sites of vascular injury. Accordingly, we observed accelerated platelet activation, primary hemostasis, and thrombus formation in apolipoprotein E-deficient (ApoE-/-) mice with acute or chronic HHcy. Upon homocysteine (Hcy) administration i…

Triggering receptor expressed on myeloid cells (TREM) like transcript-1 (TLT-1) reveals platelet activation in preeclampsia.

Triggering receptor expressed on myeloid cells (TREM) like transcript-1 (TLT-1) is a membrane protein receptor found in α-granules of megakaryocytes and platelets. Upon platelet activation TLT-1 is rapidly relocated to the surface of platelets. In plasma, a soluble form of TLT-1 (sTLT-1) is present. Plasma levels of sTLT-1 are significantly elevated in thrombotic diseases. In the present study, we investigated to whether TLT-1 reflects platelet activation in pregnant women with preeclampsia. We studied 30 preeclamptic patients who were matched with 30 normotensive pregnant women and 30 non-pre…
