The reorganization of the motor system in primates after the loss of a limb.

As a result of an extensive search, we were able to obtain a limited number of macaque monkeys, New World squirrel monkeys, and prosimian galagos years after they had received a therapeutic amputation of a forelimb or hindlimb as a treatment for injury. Forelimb or hindlimb regions of primary motor cortex, M1, were stimulated in these primates with microelectrodes using amounts of current just sufficient to evoke a movement. Results were compared to those obtained from normal primates or from M1 contralateral to the intact limb in the same primates. In all cases, more sites in M1 contralateral…

Fibroblast growth factors 1 and 2 in the primate uterus.

Fibroblast growth factors (FGF) 1 and 2 are paracrine effectors of proliferation and angiogenesis in many tissues. To elucidate potential roles for these growth factors in uterine plasticity, we used in situ hybridization histochemistry to identify the cellular sources of FGF-1 and -2 production, and immunohistochemistry to identify the cellular and extracellular deposition sites of the peptides in the primate uterus. To evaluate the effects of estradiol on uterine FGFs, uteri from ovariectomized rhesus monkeys treated with estradiol- or vehicle-containing pellets were investigated. FGF-1 and

Cellular prion protein localization in rodent and primate brain.

The presence of an abnormal, protease-resistant form of the prion protein (PrP) is the hallmark of various forms of transmissible spongiform encephalopathies (TSE) which can affect a number of mammalian species, including humans. The normal, cellular form of this protein, PrPc, while abundant in brain is also present in many tissues and a number of species. In order to address the unresolved question of the precise localization of normal cerebral PrPc, we used a free-floating immunohistochemistry procedure to localize the protein at both the light and the electron microscopic levels in the bra

Nests, tree holes, and the evolution of primate life histories.

In contrast to the majority of primates, many prosimians, some New World monkeys, and the great apes rest in tree holes or self-constructed nests during their inactive periods. The goal of this comparative study was to examine possible functions of this interspecific variation. Information on resting behavior, maternal behavior, and basic life-history traits was gleaned from the literature and mapped onto a phylogenetic tree of primates for various comparative tests. Parsimony-based reconstructions revealed that only the use of nests or tree holes as shelters for young infants can be unequivoc…

Orthotopic heart transplantation in a transgenic pig-to-primate model.

BACKGROUND: Previous studies demonstrated that hearts from transgenic pigs expressing human decay-accelerating factor (hDAF) were not hyperacutely rejected when transplanted heterotopically into the abdomen of cynomolgus monkeys. This study examines orthotopic transplantation of hDAF transgenic pig hearts into baboon recipients. METHODS: Orthotopic xenogeneic heart transplantation was performed using piglets, transgenic for hDAF, as donors. Ten baboons were used as recipients and were immunosuppressed with a combination of cyclophosphamide, cyclosporine, and steroids. RESULTS: Five grafts fa

Activities of various compounds against murine and primate polyomaviruses.

Polyomavirus infections in humans are due to BK virus (BKV) and JC virus (JCV). Diseases associated with human polyomaviruses occur mostly in immunocompromised adults, e.g., progressive multifocal leukoencephalopathy (PML), caused by JCV, in AIDS patients and hemorrhagic cystitis and uretral stenosis, caused by BKV, in transplant recipients. No therapy is available for these diseases, which necessitates the development of chemical entities that are active against polyomaviruses. Several antiviral compounds were evaluated to determine their effects on the in vitro replication of mouse polyomavi

Homologizing primate facial displays: a critical review of methods.

The applicability of previously established criteria for identifying homologous behavioural traits to the ritualized facial displays of primates is discussed and illustrated with several examples. (1) Homologizing of facial displays is based on (a) their stereotypy, (b) their nature as compounds of individual facial actions, and--to a lesser extent--(c) the homology of the muscle substrate by which the displays are accomplished. (2) Sequences of gradually changing displays ('Merkmalsreihen') can be constructed by considering related extant species, different ontogenetic stages, and variation a…

Experimental fetal and transplacental Neospora infection in the nonhuman primate.

BACKGROUND: Neospora is a newly recognized Toxoplasma-like protozoan that causes spontaneous abortion and/or neonatal disease in a wide range of animals. The purpose of this study was to determine the susceptibility of primates to Neospora infection. EXPERIMENTAL DESIGN: In experiment 1, two rhesus macaque fetuses were inoculated in utero at gestational day 65 with 1 x 10(6) culture-derived Neospora tachyzoites. A control fetus was given uninfected vehicle. The fetuses were removed by hysterotomy between 13 and 22 days postinoculation. In experiment 2, two pregnant macaques were inoculated in…

Evolutionary origin of mutations in the primate cytochrome P450c21 gene.

The CYP21 gene codes for the enzyme cytochrome P450c21 (21-hydroxylase), which is critically involved in the synthesis of glucocorticoids and mineralocorticoids. Standard human haplotypes contain two copies of CYP21--a functional gene and a pseudogene. Inactivation of the functional gene leads to congenital adrenal hyperplasia (CAH). The pseudogene has three main defects: an 8-bp deletion in exon 3, a T insertion in exon 7, and a stop codon in exon 8. To determine the origin of these defects and to shed light on the evolution of the CYP21 gene, we sequenced relevant segments of 10 primate CYP2

Effect of cyclosporine on the development of cerebral vasospasm in a primate model.

The authors studied the effect of the immunosuppressive drug cyclosporine (CS) on the development of cerebral vasospasm after subarachnoid hemorrhage (SAH) using a primate model of vasospasm. Eighteen monkeys were randomly divided into three groups: a sham-operated control group, an SAH group, and a CS-treated group. To induce SAH, the right side of the circle of Willis was dissected free of the arachnoid and an autologous blood clot was placed around the arteries. In the CS group, CS (5 mg/kg/day) was administered intramuscularly for 7 days after the induction of SAH. The vessel caliber was e

Surgical manipulation of primate cerebral arteries in established vasospasm.

It is generally believed that surgery in the face of angiographic vasospasm is dangerous due to an increased incidence of postoperative cerebral ischemia. One theory is that arterial narrowing is exacerbated by surgical manipulation of vasospastic vessels during aneurysm dissection and clipping. This theory was tested in a primate model of cerebral vasospasm and the results reported. Six monkeys underwent baseline cerebral angiography, followed by induction of subarachnoid hemorrhage (SAH) on both sides of the circle of Willis. An equal amount of fresh autologous blood clot was placed around e

Reinnervation of primate ciliary muscle following ciliary ganglionectomy.

Ciliary ganglionectomy and/or postganglionic ciliary neurectomy in the cynomolgus monkey was followed by supersensitivity to intramuscular (i.m.) pilocarpine and lack of response to topical eserine and to electrical stimulation of the Edinger-Westphal nucleus. Normal responsiveness to pilocarpine and eserine returned in most instances after about 6 months. An accommodative response to stimulus of the Edinger-Westphal nucleus was also present and, as in control eyes, could be blocked by hexamethonium. Collectively, these findings demonstrate that parasympathetic innervation to the ciliary muscl

The morphology and distribution of Merkel cells in primate gingival mucosa.

The morphology and distribution of Merkel cells in primate gingival mucosa have been studied by correlated light and electron microscopic techniques. The gingival mucosa is composed of a stratified squamous epithelium with a dense underlying connective tissue stroma. The epithelium inter-digitates with the underlying connective tissue forming long interconnected rete ridges. Merkel cells and their associated axons are abundant in gingival mucosa where they are located, either individually or in clusters, at the base of epithelial rete ridges. These cells have an identical morphology to Merkel

Acute hyperammonemia in the young primate: physiologic and neuropathologic correlates.

Infusion-induced acute (less than or equal to 24 h) hyperammonemia to concentrations up to five times normal (0.19 +/- 0.03 versus 0.90 +/- 0.08 mM) was studied in eleven 6-9-month-old Macaca mulatta. The young primates developed a progressive reduction of consciousness that correlated in severity directly with the elevation of blood ammonia concentration. Hyperventilation, electroencephalographic slowing, occasional seizure activity, and, eventually, apneustic breathing also occurred. Intracranial pressure rose from 76 +/- 7 to 167 +/- 12 mmH2O. Arterial oxygen and blood pressure remained wit…

High efficiency DNA-mediated transformation of primate cells.

Tissue culture cells from several mammalian species, including three primate lines, were transfected with recombinant vectors carrying Escherichia coli xanthine-guanine phosphoribosyltransferase or Tn5 aminoglycoside phosphotransferase dominant selectable markers. Human HeLa and SV40-transformed xeroderma pigmentosum cells exhibited stable transformation frequencies of at least 10(-3) (0.1 percent). CV-1, an African green monkey kidney cell line, could be stably transformed with the exceptionally high frequency of 6 X 10(-2) (6 percent).

The primate locus coeruleus and effects of clonidine on opiate withdrawal.

Studies of nonhuman primates suggested that the noradrenergic locus coeruleus (LC) may be involved as part of the neural substrate for a brain alarm function which includes attentiveness, arousal, anxiety, fear, terror, and the physiological correlates of these states. The studies compared the effects of electrical activation of tiny electrodes in the locus coeruleus with the effects of other agents and conditions which increased or decreased LC activity. The results suggested that the activation of the LC system is prevented by endogenous morphine-like substances and by opiates and that the o…

The pathoparasitology of the alimentary tract and pancreas of nonhuman primates: a review.

A review of the literature concerning the gross and histologic lesions associated with protozoal and metazoal parasitism in the alimentary tract and pancreas of nonhuman primates is presented. In addition, the natural history, morphology, life cycle, methods for diagnosis, and potential for zoonotic disease are reviewed briefly for each parasite discussed. The parasite species reviewed in detail are those most common or most likely to produce lesions in the alimentary tract and pancreas of the nonhuman primate host. All parasites, both pathogenic and nonpathogenic, in each major group (protozo…
