Enthalpic barriers dominate the folding and unfolding of the human Cu, Zn superoxide dismutase monomer.

The rate-limiting step in the formation of the native dimeric state of human Cu, Zn superoxide dismutase (SOD1) is a very slow monomer folding reaction that governs the lifetime of its unfolded state. Mutations at dozens of sites in SOD1 are known to cause a fatal motor neuron disease, amyotrophic lateral sclerosis, and recent experiments implicate the unfolded state as a source of soluble oligomers and histologically observable aggregates thought to be responsible for toxicity. To determine the thermodynamic properties of the transition state ensemble (TSE) limiting the folding of this high-c

Alpha-crystallin can function as a molecular chaperone.

The alpha-crystallins (alpha A and alpha B) are major lens structural proteins of the vertebrate eye that are related to the small heat shock protein family. In addition, crystallins (especially alpha B) are found in many cells and organs outside the lens, and alpha B is overexpressed in several neurological disorders and in cell lines under stress conditions. Here I show that alpha-crystallin can function as a molecular chaperone. Stoichiometric amounts of alpha A and alpha B suppress thermally induced aggregation of various enzymes. In particular, alpha-crystallin is very efficient in suppre

Proteolysis targeting chimera technology: a novel strategy for treating diseases of the central nervous system.

Neurological diseases such as stroke, Alzheimer's disease, Parkinson's disease, and Huntington's disease are among the intractable diseases for which appropriate drugs and treatments are lacking. Proteolysis targeting chimera (PROTAC) technology is a novel strategy to solve this problem. PROTAC technology uses the ubiquitin-protease system to eliminate mutated, denatured, and harmful proteins in cells. It can be reused, and utilizes the protein destruction mechanism of the cells, thus making up for the deficiencies of traditional protein degradation methods. It can effectively target and degra

Cholera toxin B subunit-five-stranded α-helical coiled-coil fusion protein: "five-to-five" molecular chimera displays robust physicochemical stability.

To create a physicochemically stable cholera toxin (CT) B subunit (CTB), it was fused to the five-stranded α-helical coiled-coil domain of cartilage oligomeric matrix protein (COMP). The chimeric fusion protein (CTB-COMP) was expressed in Pichia pastoris, predominantly as a pentamer, and retained its affinity for the monosialoganglioside GM1, a natural receptor of CT. The fusion protein displayed thermostability, tolerating the boiling temperature of water for 10min, whereas unfused CTB readily dissociated to its monomers and lost its affinity for GM1. The fusion protein also displayed resista

Hsp70 from Cyprinion macrostomus macrostomus and Garra rufa obtuse: stability and stability-dependent activity.

Two fish species, Cyprinion macrostomus macrostomus and Garra rufa obtuse, tolerate adverse conditions in the Kangal hot springs and cope with multiple stressors such as food deprivation, extreme temperature, toxins, protein degradation, hypoxia, and microbial damage. These fish have evolved strategies to counteract the stressors including the induction of heat shock proteins (Hsps). Hsps play an essential role in maintaining cellular homeostasis, and one of the key proteins in the mechanism is Hsp70. Hsp70 itself is exposed to the same stressors as all other proteins, and, hence, the stabilit

HSP70 interacting protein prevents the accumulation of inclusions in polyglutamine disease.

Heat shock proteins (HSPs) are associated with the proteinaceous inclusions that characterise many neurodegenerative diseases. This suggests they may be associated with disease aetiology and/or represents an attempt to remove abnormal protein aggregates. In this study the adenoviral mediated over-expression of HSP70 interacting protein (HIP) alone was shown to significantly reduce inclusion formation in both an in vitro model of Spinal Bulbar Muscular Atrophy and a primary neuronal model of polyglutamine disease. Experiments to determine the mechanism of action showed that: denatured luciferas

Biochemical characterization of annexin B1 from Cysticercus cellulosae.

Annexin B1 from Cysticercus cellulosae has recently been identified using immunological screening in an attempt to find novel antigens for vaccine development against cysticercosis. The protein possesses anticoagulant activity and carries significant therapeutic potential due to its thrombus-targeting and thrombolytic properties. We investigated the biochemical properties of annexin B1 using liposome and heparin Sepharose copelleting assays, as well as CD spectroscopy. The calcium-dependent binding to acidic phospholipid membranes is reminiscent of other mammalian annexins with a clear prefere

Asymmetric mutations in the tetrameric R67 dihydrofolate reductase reveal high tolerance to active-site substitutions.

Type II R67 dihydrofolate reductase (DHFR) is a bacterial plasmid-encoded enzyme that is intrinsically resistant to the widely-administered antibiotic trimethoprim. R67 DHFR is genetically and structurally unrelated to E. coli chromosomal DHFR and has an unusual architecture, in that four identical protomers form a single symmetrical active site tunnel that allows only one substrate binding/catalytic event at any given time. As a result, substitution of an active-site residue has as many as four distinct consequences on catalysis, constituting an atypical model of enzyme evolution. Although we

Mutation causing self-aggregation in human gammaC-crystallin leading to congenital cataract.

PURPOSE: Many forms of congenital hereditary cataract are associated with mutations in the crystallin genes. The authors focus attention on congenital lamellar cataract, which is associated with the R168W mutation in gammaC-crystallin, and congenital zonular pulverulent cataract, which is associated with a 5-bp insertion in the gammaC-crystallin gene. METHODS: To understand the molecular phenotypes-i.e., the functional defects that have occurred in the mutant gammaC-crystallin molecule in two cases described-the authors cloned, expressed, isolated, and compared the solution state structural f

Infrequent cavity-forming fluctuations in HPr from Staphylococcus carnosus revealed by pressure- and temperature-dependent tyrosine ring flips.

Infrequent structural fluctuations of a globular protein is seldom detected and studied in detail. One tyrosine ring of HPr from Staphylococcus carnosus, an 88-residue phosphocarrier protein with no disulfide bonds, undergoes a very slow ring flip, the pressure and temperature dependence of which is studied in detail using the on-line cell high-pressure nuclear magnetic resonance technique in the pressure range from 3 MPa to 200 MPa and in the temperature range from 257 K to 313 K. The ring of Tyr6 is buried sandwiched between a beta-sheet and alpha-helices (the water-accessible area is less t

Viral safety of solvent-detergent treated blood products.

Laboratory research commencing in 1982 led to licensing in the United States in 1985 of a solvent/detergent (SD)-treated anti-haemophilic factor (AHF) concentrate. Licensing was based on (a) studies demonstrating the inactivation of several marker viruses [vesicular stomatitis virus (VSV), Sindbis virus, Sendai virus], human immunodeficiency virus (HIV), hepatitis B virus (HBV), and non-A, non-B hepatitis virus [NANBHV; now known to be principally hepatitis C virus (HCV)] added to AHF just before treatment, (b) the realization that the principal viruses of concern in a transfusion setting (e.g

Stabilization of Enzymes via Covalent Tethering to Appropriately Tuned Random Copolymer Brushes.

Immobilizing enzymes can often cause their inactivation due to enzyme-support interactions and changes in the enzyme's microenvironment. As such, the design of novel supports remains largely empirical, and a rational approach may avoid time-consuming and tedious experiments. This chapter describes a method for stabilizing enzymes via immobilization on heterogeneous polymer brush supports. By varying the composition of the brush, the enzyme-brush interface may be finely tuned to prevent enzyme unfolding and rescue denatured enzymes via a chaperone-like mechanism. The design of polymer brush sup

Western blot analysis for the detection of serum antibodies recognizing linear Aquaporin-4 epitopes in patients with Neuromyelitis Optica.

The current study presents a western blot assay showing good sensitivity (81%) and specificity (97%) for detecting serum antibodies to Aquaporin-4 (AQP4) in patients with Neuromyelitis Optica (NMO). By using western blot, we were able, for the first time, to distinguish serum immunoreactivity against either of the two AQP4 isoforms, showing that antibodies recognizing the linear AQP4-M1 isoform are specific for NMO. Moreover, the high sensitivity and specificity of the western blot assay in detecting serum anti-AQP4 antibodies in NMO patients suggest that such auto-antibodies may be of linear

The reconstitution of unfolded myoglobin with hemin dicyanide is not accelerated by fly-casting.

This study explores how the kinetics of a coupled folding/binding reaction depend on the initial conformation of the protein. Stopped-flow spectroscopy is used to monitor the reaction of apo-myoglobin (aMb) with hemin dicyanide at pH 7.2. Different initial aMb conformations are tested. In the case of acid-denatured aMb, the observed kinetics are consistent with a "fly-casting" scenario [Shoemaker et al., Proc. Natl. Acad. Sci. USA 97 (2000) 8868-8873]. However, the formation of a compact complex proceeds more rapidly in the case of prefolded aMb. This finding is opposite to what would be expec

A nondenaturing solid phase pharmaceutical carrier comprised of surface-modified nanocrystalline materials.

Numerous carrier systems have been developed for the controlled delivery of biologically active molecules such as drugs and diagnostic agents. The biophysical interactions between the biologically active molecules and their carriers, however, may denature the former and lead to reduced biological activity. In this study, a model nondenaturing carrier comprised of a nanocrystalline (10 -7 m) tin oxide core and a surface-charge-reducing organic bonding layer (GF292) was synthesized. A subsequently bound protein (human transferrin) showed significant retained conformation by immunoelectron micros

Tailoring chickpea protein functionality for food applications through supercritical CO2 processing.

This study evaluates the use of supercritical CO2 (SCCO2) as a dual-purpose method for defatting and modulating the functional properties of chickpea protein. Two processing routes were investigated: 1) post-extraction SCCO2 modification of protein obtained from full-fat flour (CP series), and 2) flour defatting followed by protein extract modification with SCCO2 at different pressures (DFCP series). CP0 and DFCP0 refer to proteins from untreated and SCCO2-defatted flours, respectively; CP75-800 and DFCP75-800 underwent additional SCCO2 treatments at indicated pressures. SCCO2 defatting at 850

Cross-linking studies of the protein topography of rat liver microsomes.

A cleavable cross-linking reagent, dithiobis(succinimidyl propionate), DSP, was used to study the topography of the proteins in the endoplasmic reticulum membrane of rat liver. Reaction of untreated (control), phenobarbital- or 3-methylcholanthrene-induced microsomes with 0.5 mM DSP for 30 min at 0 degrees C resulted in the cross-linking of a protein with a molecular weight of about 52 000 to form an apparent dimer. In phenobarbital microsomes, a smaller amount of a 52 000-dalton protein also appeared in a dimer in the absence of DSP if N-ethylmaleimide was not included during homogenization.

High hydrostatic pressure modulation of whey protein-malvidin derivative complexes: Mechanisms, structural changes, and stability.

This study investigated how high hydrostatic pressure (HHP) modulates the structural and functional properties of whey protein isolate (WPI) complexes with Malvidin-3-O-6-[acrylic acid-(2-hydroxy,4-carboxy-cyclohexanol) ester]-guaiacol (MV3ACEC). HHP at 350 MPa maximized the binding ratio (78.41 ± 0.94%) and thermal stability (denaturation temperature: 123.76 °C) of WPI-MV3ACEC, while reducing particle size (237 nm) and enhancing system stability (zeta potential: 12.7 mV). Structural analyses revealed HHP-induced conformational changes in WPI-MV3ACEC, including decreased α-helix and increased

Modulation of walnut meal protein functionalities through low-temperature subcritical defatting: Insights into aggregation behavior and structural changes.

This study compared walnut meal protein (WMP) obtained from walnut meal defatted using low-temperature subcritical butane extraction (SBE-WMP) with that obtained from walnut meal produced by traditional high-temperature pressing (HP-WMP) to elucidate the effects of defatting methods on the structural and functional properties of WMP. Compared with HP-WMP, SBE-WMP exhibited a more ordered molecular conformation, with reduced oxidation and aggregation. These structural characteristics were accompanied by a smaller mean particle size (7.25 vs. 147.50 μm), higher solubility (66.93% vs. 51.88%) and
