A purine-sensitive pathway regulates multiple genes involved in axon regeneration in goldfish retinal ganglion cells.

In lower vertebrates, retinal ganglion cells (RGCs) can regenerate their axons and reestablish functional connections after optic nerve injury. We show here that in goldfish RGCs, the effects of several trophic factors converge on a purine-sensitive signaling mechanism that controls axonal outgrowth and the expression of multiple growth-associated proteins. In culture, goldfish RGCs regenerate their axons in response to two molecules secreted by optic nerve glia, axogenesis factor-1 (AF-1) and AF-2, along with ciliary neurotrophic factor. The purine analog 6-thioguanine (6-TG) blocked outgrowt

The purine-rich trinucleotide repeat sequences d(CAG)15 and d(GAC)15 form hairpins.

The structures of single-stranded (ss) oligonucleotides containing (CAG)15 [ss(CAG)15] or (GAC)15 [ss(GAC)15] were examined. At 10 degrees C, the electrophoretic mobilites of the two DNAs were similar to ss(CTG)15, a DNA that forms a hairpin containing base paired and/or stacked thymines. At 37 degrees C in 50 mM NaCl, single-strand-specific P1 nuclease cleaved the G33-G36 phosphodiesters of ss(GAC)15, and the G32-A34, G35-C36 phosphodiesters of ss(CAG)15 (where the loop apex of both DNAs = A34). Electrophoretic mobility melting profiles indicated that the melting temperature (Tm) of ss(CAG)15

Purine metabolite washout and platelet aggregation at reflow after tourniquet ischemia: effect of intravenous regional lidocaine.

Tourniquet ischemia will influence the biochemical milieu of tissue cells and affect the metabolism of purines in skeletal muscle distal to the occlusion. At reperfusion, generation of oxygen radicals by the hypoxanthine-xanthine oxidase system may ensue, influencing white blood cell and thrombocyte aggregation, causing damage to the endothelial cell barrier and inducing non-reflow type phenomena. Amide-type local anaesthetics are known to affect local vasotone, leukocyte adherence and platelet function but the influence of lidocaine on purine metabolite washout and platelet aggregation follow…

Characterization of the alterations in purine nucleotide metabolism in hypoxanthine-guanine phosphoribosyltransferase-deficient rat neuroma cell line.

A rat neuroma cell line (B103 4C), deficient of hypoxanthine-guanine phosphoribosyltransferase (HGPRT), was utilized as a model tissue in search for the biochemical basis of the Lesch-Nyhan syndrome (LNS). The HGPRT-deficient neurons exhibited the following properties: an almost complete absence of uptake of guanine and of hypoxanthine into intact cell nucleotides (0.92% and 0.69% of normal, respectively); a significant increase in the availability of 5'-phosphoribosyl-1-pyrophosphate; a three- to fourfold acceleration of the rate of de novo nucleotide synthesis; a normal excretion of xanthine

Bone marrow transplantation only partially restores purine metabolites to normal in adenosine deaminase-deficient patients.

To delineate the extent to which bone marrow transplantation provides "enzyme replacement therapy", we have determined metabolite concentrations in two patients with adenosine deaminase (ADA) deficiency treated with bone marrow transplants and rendered immunologically normal. 10 yr after engraftment of lymphoid cells, erythrocyte deoxy ATP was markedly decreased compared to the marked elevations of deoxy ATP observed in untreated patients, but was still significantly elevated (62 and 90 vs. normal of 6.0 +/- 6.0 nmol/ml packed erythrocytes). Similarly, deoxyadenosine and adenosine excretion we

Identification, characterization and localization of a (Ca2+ + Mg2+)-activated purine nucleoside triphosphate phosphohydrolase from calcifying cartilage.

A purine nucleoside triphosphate phosphohydrolase (unspecified diphosphate phosphohydrolase, EC 3.6.1.15) was chromatographically separated from the bulk of alkaline phosphatase activity by gel filtration chromatography of butanol and EDTA extracts of fracture callus and bovine epiphyseal cartilage. The callus enzyme differed from alkaline phosphatase in a variety of characteristics. The purine nucleoside triphosphate phosphatase hydrolyzed a more specific group of substrates, required Ca2+ and Mg2+ for optimal activity, remained unaffected by a potent alkaline phosphatase inhibitor, and demon

SAR of carbon-linked, 2-substituted purines: synthesis and characterization of AP23451 as a novel bone-targeted inhibitor of Src tyrosine kinase with in vivo anti-resorptive activity.

Targeted disruption of the pp60(src) (Src) gene has implicated this tyrosine kinase in osteoclast-mediated bone resorption and as a therapeutic target for the treatment of osteoporosis and other bone-related diseases. Here, we describe structure activity relationships of a novel series of carbon-linked, 2-substituted purines that led to the identification of AP23451 as a potent inhibitor of Src tyrosine kinase with antiresorptive activity in vivo. AP23451 features the use of an arylphosphinylmethylphosphinic acid moiety which confers bone-targeting properties to the molecule, thereby increasin…

Persistence of methylated purines in the DNA of various rat fetal and maternal tissues and carcinogenesis in the offspring following a single transplacental dose of N-methyl-N-nitrosourea.

Formation and loss of methylated purines in DNA of various fetal and maternal tissues were measured up to 7 days following intravenous administration of N-[14C]methyl-N-nitrosourea to rats on the 21st day of gestation. Methylation products were detected in all tissues examined, the level in maternal liver being higher than in other tissues. The concentrations of 7-methylguanine and 3-methyladenine decreased faster in fetal than in corresponding maternal tissues, due to a higher rate of DNA synthesis in fetal tissues, as determined by incorporation of labelled thymidine. Removal of the promutag…

TIM3 Signaling in Effector T Cells Acts as an Immunometabolic Switch in the Purine Degradation Pathway to Suppress Intestinal Inflammation.

BACKGROUND & AIMS: Effector T lymphocytes have been found to play a key role in inflammatory bowel diseases (IBDs). However, the molecular pathways controlling their metabolism and functional activity remain poorly defined. We aimed to elucidate the role of the immune checkpoint receptor TIM3 for T-cell signaling in IBDs. METHODS: We combined experimental colitis models in TIM3-deficient mice with analyses of human IBDs and control samples. Blood and mucosal T cells were assessed using multicolor flow cytometry, cytokine profiling, and untargeted metabolomics. Single-cell sequencing data were

Synthesis of Cyclopropyl Carbocyclic Purine Nucleosides via Base-Promoted [2 + 1] Annulation of Alkenyl Sulfonium Salts.

We herein report a facile and efficient base-promoted annulation approach to construct cyclopropyl carbocyclic purine nucleoside analogues. Using α-purine-substituted acetones/acetophenones as challenging dinucleophiles, this reaction undergoes selective C/C nucleophilic attack to achieve [2 + 1] annulation with alkenyl sulfonium salts (up to 92% yield, 24 examples). This reaction employs safer and more readily available starting materials, proceeds under transition-metal-free and mild conditions, and possesses broad functional group tolerance, thus showing great potential for practical applic

Structure-activity relationship profiling of N-substituted 8-trifluoromethyl-9H-purin-6-amines as mitochondrial protonophores.

Mitochondrial dysfunction is a common cellular defect linked to the pathogenesis of diabetes, obesity, and metabolic dysfunction-associated fatty liver disease. Accordingly, there is increasing interest in treating these diseases with mitochondria-targeted agents, such as mitochondrial protonophores. Herein, we report a library of N-substituted 8-trifluoromethyl-9H-purin-6-amine derivatives derived from the potent protonophore BAM15. Our structure-activity relationship (SAR) study revealed that this scaffold has a wide range of tolerated substitutions that elicit sub-micromolar potency and imp

In Vitro and In Vivo Evaluation of Pediococcus acidilactici Pedio6-1 for Purine Metabolism Modulation in Diet-Induced Obese Mice.

Gut lactic acid bacteria are emerging as potential targets for modulating host purine metabolism and alleviating hyperuricemia-related disorders. This study aimed to isolate purine-degrading lactic acid bacterial strains from porcine intestine and systematically evaluate their probiotic potential through both in vitro and in vivo approaches. A strain designated Pedio6-1 was isolated and identified as Pediococcus acidilactici based on 16S rRNA sequencing. In vitro assays demonstrated that P. acidilactici Pedio6-1 exhibited nearly complete adenine clearance (approaching 100%) and a total purine

Integrative multi-omics profiling implicates purine metabolism and the PI3K-Akt pathway in radiation-induced lung injury.

BACKGROUND: Thoracic radiotherapy commonly causes radiation-induced lung injury (RILI); however, its mechanisms remain poorly defined, and no FDA-approved therapies are available. METHODS: To identify key pathways and regulators, we analyzed metabolic and transcriptomic alterations in a murine model of RILI. Following a 15 Gy thoracic irradiation, lung tissues were assessed by histopathology (H&E and Masson staining), LC-MS-based metabolomics, and transcriptomics. RESULTS: Irradiated lungs showed erythrocyte exudation, inflammatory infiltration, fibrosis, and epithelial damage, whereas contr

Paralogous guanine deaminases acquired from bacteria by horizontal gene transfer promote purine homeostasis in Caenorhabditis elegans.

Disruptions in purine metabolism contribute to a range of human diseases, from rare genetic disorders such as Lesch-Nyhan syndrome and xanthinuria to common conditions including gout and cancer. To better understand the metabolic networks that regulate purine homeostasis, we developed a Caenorhabditis elegans model of xanthine dehydrogenase ( xdh-1 ) deficiency. Remarkably, xdh-1 mutant animals form rare xanthine stones, recapitulating a hallmark of human xanthinuria. To uncover genetic regulators of purine homeostasis, we performed a forward genetic screen for mutations that exacerbate xanthi

Benefit of Salbutamol for the Treatment of Neuromuscular Junction Dysfunction in Patients With Purine-Rich Element Binding Protein A (PURA) Syndrome.

Purine-rich element-binding protein A (PURA) syndrome is a rare neurodevelopmental disorder caused by pathogenic variants in the PURA gene and is characterized by neonatal hypotonia, feeding difficulty, respiratory dysregulation, and severe developmental impairment. The PURA gene has recently been identified as a cause of congenital myasthenic syndrome. We report two unrelated infants with genetically confirmed PURA syndrome who were treated with neuromuscular junction-directed therapy and reviewed three additional published cases. All five patients had neonatal hypotonia and apnea or respirat

Adenosine Deaminase-Mediated Purine Dysfunction Leads to DNA Repair Inhibition and Senescence in Sporadic Amyotrophic Lateral Sclerosis.

Amyotrophic lateral sclerosis (ALS) is a neurodegenerative disease characterised by the death of motor neurons leading to paralysis and death generally 3-5 years post-symptom onset. ALS is a cell- and non-cell-autonomous disease, with glia such as astrocytes influencing disease pathology and progression. Our laboratory has previously identified purine metabolism dysfunction in induced neural progenitor cell-derived astrocytes (iAstrocytes) from sporadic ALS (SALS) cases, driven by loss of the enzyme adenosine deaminase (ADA). Here, we have demonstrated that loss of ADA, along with changes to e

Pathway coessentiality mapping reveals complex II is required for de novo purine biosynthesis in acute myeloid leukaemia.

Understanding how cellular pathways interact is crucial for treating complex diseases like cancer. Individual gene-gene interaction studies have provided valuable insights, but may miss pathways working together. Here we develop a multi-gene approach to pathway mapping which reveals that acute myeloid leukaemia (AML) depends on an unexpected link between complex II and purine metabolism. Through stable-isotope metabolomic tracing, we show that complex II directly supports de novo purine biosynthesis and that exogenous purines rescue AML cells from complex II inhibition. The mechanism involves …
