An essential step in the success of germ cell transplantation is the preparation of the recipient's testicular environment to increase the availability of stem cell niches. However, most methods for this purpose in birds face serious limitations such as partial germ cell depletion, high toxicity and mortality, or the need to use expensive technologies. Here, we validated a simple and practical technique of transferring quail testicular cells into chicken testes depleted of endogenous spermatozoa by fractioned chemotherapy (20 mg/kg/week busulfan for 5 weeks). This protocol resulted in a very l…

Neural crest mesenchyme (NCM) controls species-specific pattern in the craniofacial skeleton but how this cell population accomplishes such a complex task remains unclear. To elucidate mechanisms through which NCM directs skeletal development and evolution, we made chimeras from quail and duck embryos, which differ markedly in their craniofacial morphology and maturation rates. We show that quail NCM, when transplanted into duck, maintains its faster timetable for development and autonomously executes molecular and cellular programs for the induction, differentiation, and mineralization of bon…

Toxicity of cadmium in the young Japanese quail rapidly produced moderate growth depression, hypogonadism in the male, decreased bone ash, severe anemia, alterations of "indicator" tissue levels of several essential inorganic elements, and marked histological abnormalities of the duodenum, bone marrow, adrenal medulla, and esophageal mucus glands. Cadmium appeared to have direct effects on zinc and iron, particularly iron (III), by decreasing intestinal absorption of these elements. Small amounts of dietary ascorbic acid were protective against many of the adverse effects of cadmium. The young…

Aflatoxin B1 (AFB1 ) is a common feed contaminant that adversely affects bird performance and product quality. A total of 600 7-day-old quail chicks were randomly allotted to eight experimental groups in a completely randomized design with five replicate pens and 15 quails per pen. Experimental treatments including two levels of AFB1 (0 and 2.5 mg/kg) and 4 levels of Nigella sativa (NS) (0, 0.5, 1.0 and 1.5% of diet) were offered from 7 to 35 days of age to quail chicks. Although feeding of AFB1 impaired gain (G) and feed conversion ratio, dietary NS increased G (p < 0.05). Relative weight of …

The functional links of specific retinoid receptors to early developmental events in the avian embryo are not known. Before such studies are undertaken, knowledge is required of the spatiotemporal expression patterns of the receptor genes and their regulation by endogenous retinoic acid levels during the early stages of development. Here, we report the expression patterns of mRNAs for RARalpha, RARalpha2, RARbeta2, RARgamma, RARgamma2, RXRalpha, and RARgamma from neurulation to HH10 in the normal and vitamin A-deficient (VAD) quail embryo. The transcripts for all retinoid receptors are detecta…

Cells immunoreactive for the enzyme aromatase were localized in the forebrain of male zebra finches with the use of an immunocytochemistry procedure. Two polyclonal antibodies, one directed against human placental aromatase and the other directed against quail recombinant aromatase, revealed a heterogeneous distribution of the enzyme in the telencephalon, diencephalon, and mesencephalon. Staining was enhanced in some birds by the administration of the nonsteroidal aromatase inhibitor, R76713 racemic Vorozole) prior to the perfusion of the birds as previously described in Japanese quail. Large …

The proepicardial organ provides differentiated cell types to the myocardial wall and facilitates coronary development. Ingrowth of the coronary arteries into the aorta has recently been linked to apoptosis. This study was set up to examine the effect of an inhibition of epicardial outgrowth on apoptotic patterning and coronary development. Epicardial outgrowth was blocked at HH15-17 in quail embryos, which survived until HH25-35 (n=33). Embryos with complete inhibition of outgrowth did not survive after HH29. These embryos presented with thin compact myocardium, devoid of vessels. In embryos …

The addition of chicken egg yolk to semen extenders is thought to reduce the fertilizing potential of rooster spermatozoa--but not (or at least not as much) that of other avian species. The aim of the present study was to determine whether quail egg yolk, a novel extender additive, provides advantages over chicken egg yolk in the cryopreservation of rooster spermatozoa. Experiments were also performed to determine whether the harmful effect of egg yolk occurs during cryopreservation or during fertilization after artificial insemination. Heterospermic rooster semen samples were divided into ali…

We describe here the defects that arise in the central nervous system (CNS) of quail embryos when they develop in the absence of vitamin A. It has been assumed that because of the effects of excess vitamin A and its metabolites, particularly retinoic acid (RA), on the CNS they are involved in various aspects of CNS development. We show that this is indeed the case, because these deficient quail embryos have three defects in their CNS. First, the posterior hindbrain fails to develop because the cells fated to form this part of the CNS in the very early embryo die by apoptosis. Second, the neura…

Prenatal development of the secondary palate in duck was studied using light and electron microscopic, histochemical, autoradiographic and biochemical techniques. The secondary palate of duck develops between Days 7-12 of incubation (HH 25-37). During this period the palatal shelves grow horizontally toward one another but, unlike those in mammals and the alligator, they never fuse, and a physiological cleft persists between them. In contrast to both chick, where the MEE differentiates to orthokeratinisation, and quail, where the MEE becomes parakeratinised, the MEE of duck differentiates to a…

For the first time, the biological activity of the newly synthesized glycosides of vitamin D3 and of 1 alpha(OH)D3 has been studied in rats, chickens, and Japanese quails. Glucosidation did not reduce the activity of vitamin D3. In chickens, vitamin D3 glucoside has an activity of 90% to 95% as compared to the aglycon. In rats vitamin D3 glucoside is equally active when given by intravenous or oral route. Glycosides of 1 alpha (OH)D3, however, show a lower activity in rats, chickens, and quails. 1 alpha (OH)D3 glucoside has about 10% activity of the free 1 alpha (OH)D3, while 1 alpha (OH)D3 ce…

Calcium ion affects ion permeability and membrane potential among many other aspects of cell function. Initial effects of increasing extracellular calcium upon membrane potential were studied in a quail fibrosarcoma (QT35) where calcium had a dose dependent effect, and normal quail fibroblasts, where there was little effect. Comparisons were then made in six different human hepatocellular carcinomas (Tong, HepG2, Hep3B, PLC/PRF/5, Mahlavu, and HA22T) in response to smaller changes in concentration. There were insignificant changes in membrane potential in two cell lines and significant elevati…

The vitamin A deficient (VAD) quail embryo lacks active retinoids, fails to express normally GATA-4, and develops a nonlooping heart tube morphogenetic defect that is a model for congenital cardiomyopathy. VAD quail embryos, or chick embryos depleted specifically for GATA factors, show in addition abnormal foregut development, characterized by apoptosis of the endoderm cells associated with presumptive myocardium during the process of heart tube formation. Exogenous retinoic acid or transplantation of normal chick embryo anterior endoderm is sufficient to rescue apoptosis as well as GATA-4 exp…

Cryopreservation of domestic animal sperm has been widely used for artificial insemination (AI), and egg yolk is one of the most commonly used cryoprotectants during the freezing-thawing process. The objectives of this study were to compare the effectiveness of egg yolk from five avian species (domestic chicken, domestic duck, domestic goose, Japanese quail or domestic pigeon) and to optimize the concentration of egg yolk on the cryopreservation of bull sperm in terms of frozen-thawed sperm progressive motility and viability. The results were two-fold. First, they showed that pigeon egg yolk p…

Di(n-butyl) phthalate (DBP) is considered a substance of serious concern because of its reproductive toxicity and endocrine-disrupting properties. Exposure to DBP causes morphological and functional changes in the male reproductive system of birds and mammals. However, there are no detailed reports on the effects of DBP on the Sertoli cell and junctional complexes of the blood-testis barrier (BTB) in birds. The present study investigated dose-related ultrastructural changes in Sertoli cells and junctional complexes of the BTB in adult Japanese quail (Coturnix coturnix japonica) exposed to DBP …

The hypothalamo-pituitary-gonadal (HPG) axis plays a major role in coordinating the reproduction of fish and other vertebrates. Gonadotropin-releasing hormone (GnRH) is the primary stimulatory factor responsible for the hypothalamic control of gonadotropin secretion. In 2000, a previously unidentified hypothalamic neuropeptide was isolated from the brain of Japanese quail and termed gonadotropin-inhibitory hormone (GnIH) based on its ability to directly inhibit gonadotropin release from the cultured quail anterior pituitary gland. One year later, the cDNA sequence that encodes the quail GnIH p…

Physaloptera spp. are common nematodes found in the stomach and muscles of mammals, reptiles, amphibians, and birds. Physaloptera spp. have a complicated life cycle with multiple definitive hosts, arthropod intermediate hosts, aberrant infections, and possible second intermediate hosts or paratenic hosts. For example, Physaloptera sp. larvae have been found within the tissues of wild northern bobwhite quail (Colinus virginianus), and it is suspected that quail may serve as paratenic or secondary hosts of these parasites. However, because it is not known what role quail play in the life cycle o…
