Few molecular studies have identified the current status of cystic echinococcosis in Egypt. The present study aimed to ascertain the genotype(s) of Echinococcus granulosus responsible for human hydatidosis in different Egyptian governorates (regions). Animal isolates were collected from 40 camels, 5 pigs and 44 sheep. 27 human isolates were included in the present study. Specific PCR was performed and followed by DNA sequencing for mitochondrial 12S ribosomal RNA gene and BLAST analysis. The sheep cysts were not hydatid cysts. G6 genotype (camel starin) predominates in human, camel and pig isol

The risk of human hookworm infections from land application of wastewater matrices could be high in regions with high hookworm prevalence. A rapid, sensitive and specific hookworm detection method from wastewater matrices is required in order to assess human health risks. Currently available methods used to identify hookworm ova to the species level are time consuming and lack accuracy. In this study, a real-time PCR method was developed for the rapid, sensitive and specific detection of canine hookworm (Ancylostoma caninum) ova from wastewater matrices. A. caninum was chosen because of its mo

Indonesian thin-tail (ITT) sheep can resist infection with Fasciola gigantica but not F. hepatica and presents an ideal model to investigate the mechanisms of liver fluke resistance in a natural host. This study examines the local and systemic immune responses of sheep during Fasciola infection and demonstrates that different anatomical tissues display distinct cytokine profiles consistent with liver fluke migration. The study also reveals a significant difference in the cytokine and antibody profiles of ITT sheep infected with F. gigantica compared with F. hepatica, with a higher ratio of IL-

Bladder carcinoma accounts for 26% of reported human malignancies in Egypt, and has been strongly associated with urinary schistosomiasis. Nevertheless, the immediate role of schistosomal egg proteins in bladder carcinogenesis is unexplored. We investigated the effects of crude soluble egg antigens (SEA) of Schistosoma hematobium on urothelial cell proliferation. The proliferation of bovine endothelial Endo, human urothelial J82 and smooth muscle SMC cell lines was assessed by low-density growth assays. SEA induced proliferation of both J82 and Endo cells in a dose-dependent fashion, but not S

Between April and June 2009, 1,075 feral rainbow trout from 10 different lakes involved with aquaculture activities in Los Lagos Region, Chile, were inspected for Diphyllobothrium species. All viscera and muscles of the fish were examined using stereomicroscopy; pyloric cecae and stomachs infected with plerocercoids were checked by histology and scanning electron microscopy. Plerocercoids of Diphyllobothrium dendriticum were confirmed by PCR and sequencing of COI and 18S rRNA + ITS1 + 5.8S rRNA + ITS2 genes for the first time in Chile. Overall prevalence of plerocercoids of D. dendriticum was

The methylation of lysine 4 of Histone H3 (H3K4me) is an important component of epigenetic regulation. H3K4 methylation is a consequence of transcriptional activity, but also has been shown to contribute to "epigenetic memory"; i.e., it can provide a heritable landmark of previous transcriptional activity that may help promote or maintain such activity in subsequent cell descendants or lineages. A number of multi-protein complexes that control the addition of H3K4me have been described in several organisms. These Set1/MLL or COMPASS complexes often share a common subset of conserved proteins,

Ribosomal protein S9 (RPS9) is an essential functional gene that participates in DNA repair and developmental regulations. A sequence homolog of RPS9 has been found to be upregulated in the protoscoleces (PSCs) of Echinococcus granulosus treated with artemisinin. However, E. granulosus RPS9 (EgRPS9) has not been identified before. In the present study, the 657-base pair (bp) cDNA encoding EgRPS9 was cloned. Amino acid sequence analysis showed that EgRPS9 was similar to the RSP9 proteins from Schistosoma japonicum (SjRPS9, 86%) and Schistosoma mansoni (SmRPS9, 79%). Phylogenetic tree analysis s

In onchocerciasis patients and in O. volvulus-exposed individuals without signs of onchocericiasis, T- and B-cell responses to two recombinantly expressed O. volvulus enzymes were analysed and compared to responses to total protein extract of adult parasites. The cytosolic enzymes Cu/Zn superoxide dismutase 1 (OvSOD1) and glutathione S-transferase 2 (OvGST2) represent 2 detoxifying molecules which may play an important role in parasite defense against host-induced oxidative stress. The T-cell response to the two recombinant proteins was analysed by investigating the cytokine responses of perip

The Trematoda family Didymozoidae comprises over 250 species that typically use marine fishes as their definitive hosts. Species identification of adult didymozoids based solely on morphology is often problematic owing to the difficulty of observing their detailed internal organs. Although molecular analyses are generally essential for trematode taxonomy, reference sequences (e.g. 28S rDNA, ITS, and COI) for morphologically identified adult didymozoids remain scarce. In this study, adult didymozoids were collected from five marine fish species in Japan between 2018 and 2025. Morphological obse

A new entomopathogenic nematode, Steinernema hattingi n. sp., was isolated during a survey in Mpumalanga Province of South Africa. Morphological and morphometric traits place the species in the glaseri-group, characterised by large infective juveniles (IJs) with average body length exceeding 1000 μm. The IJs of S. hattingi n. sp. have a mean body length of 1059 μm, lateral fields composed of eight equally spaced ridges (nine incisures) at mid-body (formula 2, 6, 8, 4, 2), a relatively short tail (c ratio 16 on average) with a hyaline region occupying approximately 45% of its length, and are re

Eurytrema spp. flukes are responsible for economic losses in ruminant production, leading to hyperplasia and hypertrophy of the bile ducts, periductal fibrosis and loss of pancreatic parenchyma. These flukes affect ruminants, small carnivores and occasionally humans. The aim of this study was to perform a precise identification by morphometry and molecular markers in Eurytrema flukes from Argentina, also clarify their phylogenetic relationships. Eurytrema flukes were collected from the pancreatic ducts of breed cattle examined during necropsies in Montecarlo, Misiones province, Argentina. Prec

A bottlenose dolphin, Tursiops sp., stranded on the coast of South Carolina, USA was found to be heavily infected in its intestine by tapeworms, which we identified molecularly. Sequencing of portions of the mitochondrial cytochrome c oxidase I (COI) and nuclear large subunit ribosomal RNA (28S rRNA) genes showed the cestodes to be Diphyllobothrium stemmacephalum, commonly known as a broad tapeworm. Infections of marine mammals by Diphyllobothrium have been previously reported in the Northwestern Atlantic Ocean, but only to genus level. Infection by tapeworms may be rare in dolphins in South C

Schistosoma is the causative agent of schistosomiasis, a common infectious disease distributed worldwide. Our previous phosphoproteomic analysis suggested that glycogen synthase kinase 3 (GSK3), a conserved protein kinase in eukaryotes, is likely involved in protein phosphorylation of Schistosoma japonicum. Here, we aimed to identify the interacting partners of S. japonicum GSK3β (SjGSK3β) and to evaluate its role in parasite survival. Toward these ends, we determined the transcription levels of SjGSK3β at different developmental stages and identified its interacting partners of SjGSK3β by scr

Canine dirofilariasis is common in Brazil, but molecular diagnosis is rare even though molecular studies increase our knowledge about molecular epidemiology and circulating genotypes from helminths worldwide. This study aims to estimate the prevalence of infection with a modified Knott's test and to perform molecular characterization of Dirofilaria immitis (Leidy, 1856) Railliet and Henry, 1911, in dogs from endemic areas of Maricá and Niterói municipalities, Rio de Janeiro State, Brazil. Molecular characterization was performed in 33 blood samples from dogs positive for microfilariae and 4 ad

We have undertaken a detailed analysis of the biotransformation of five of the most therapeutically important benzimidazole anthelmintics - albendazole (ABZ), mebendazole (MBZ), thiabendazole (TBZ), oxfendazole (OxBZ) and fenbendazole (FBZ) - in Caenorhabditis elegans and the ruminant parasite Haemonchus contortus. Drug metabolites were detected by LC-MS/MS analysis in supernatants of C. elegans cultures with a hexose conjugate, most likely glucose, dominating for all five drugs. This work adds to a growing body of evidence that glucose conjugation is a major pathway of xenobiotic metabolism i

BACKGROUND: Schistosomiasis traditionally has been diagnosed by detecting eggs in stool or urine. However, the sensitivity of these examinations is limited, especially in travelers with a low worm burden. Serologic tests have a greater sensitivity, but their results remain positive regardless of treatment and thus cannot be used for follow-up of patients. We hypothesized that detection of worm microRNAs (miRNAs) in serum can overcome the drawbacks of the existing diagnostic methods.
METHODS AND RESULTS: Twenty-six returning travelers with schistosomiasis (based on positive results of serologi

Parasites of deep-water fishes are less known in comparison to the parasites of fishes living in the demersal and epipelagic ocean zones. In the present research note we report the occurrence of larval trypanorhynch cestodes in a rare deep-water fish, the deep-water brotula, Cataetyx laticeps. Based on the 28S rDNA (region D1-D3) sequence homology and the phylogenetic analysis, the larval cestodes are putatively assigned to the genus Grillotia. It is suggested that the definitive host of this trypanorhynch is a batoid.

Cichlidocestus n. gen. is proposed to accommodate 2 new species of proteocephalidean cestodes, Cichlidocestus gillesi n. sp. from Cichlasoma amazonarum in Peru (type species) and Cichlidocestus janikae n. sp. from Hypsophrys nicaraguensis (all Perciformes: Cichlidae) in Costa Rica. The new genus is unique among all but 1 proteocephalidean genera in the position of the ovary that occupies the middle and posterior thirds of the median region of proglottids (vs. the ovary in the posterior third of proglottids near their posterior margin in all but 1 remaining taxa). In addition, Cichlidocestus is

The aim of this paper was to evaluate the genetic relationship between two isolates of Opisthorchis (O. pedicellata sp. 1, O. pedicellata sp. 2) from two siluroid fish hosts, based on three nuclear DNA (18S, internal transcribed spacer 1 (ITS1) and ITS2). Molecular analysis revealed that both isolates of 18S sequences showed low intraspecific variability (1.6%), while this was 1.9% for ITS1 and 2.6% for ITS2. The secondary structure of the ITS2 region exhibited a remarkable four-helix model, with helices 1 and 4 being relatively short, three U-U mismatches in helix 2 and the longest helix (3)
