Site-directed mutagenesis of the ATM promoter: consequences for response to proliferation and ionizing radiation.

Although ATM, the protein defective in ataxia-telangiectasia (A-T), is activated primarily by radiation, there is also evidence that expression of the protein can be regulated by both radiation and growth factors. Computer analysis of the ATM promoter proximal 700-bp sequence reveals a number of potentially important cis-regulatory sequences. Using nucleotide substitutions to delete putative functional elements in the promoter of ATM, we examined the importance of some of these sites for both the basal and the radiation-induced activity of the promoter. In lymphoblastoid cells, most of the mut

Cytogenetic monitoring of hospital workers occupationally exposed to ionizing radiation using the micronucleus centromere assay.

A cytogenetic study was performed in lymphocytes of hospital workers occupationally exposed to X- and gamma-rays using the micronucleus centromere assay. A comparison of the data for the exposed group and an age-matched group of non-exposed hospital workers showed a significant (P < 0.05) increase in centromere-positive micronuclei for the radiation workers, while no effect on centromere-negative micronuclei was present. The observed systematic increase in micronucleus frequency with age was mainly due to increased chromosome loss, reflected in the centromere-positivity of the micronuclei. The

Identification of potential mRNA biomarkers in peripheral blood lymphocytes for human exposure to ionizing radiation.

Since early in the Atomic Age, biological indicators of radiation exposure have been sought, but currently available methods are not entirely satisfactory. Using cDNA microarray hybridization to discover new potential biomarkers, we have identified genes expressed at increased levels in human peripheral blood lymphocytes after ex vivo irradiation. We recently used this technique to identify a large set of ionizing radiation-responsive genes in a human cell line (Oncogene 18, 3666-3672, 1999). The present set of radiation markers in peripheral blood lymphocytes was identified 24 h after treatme

Exposure to ionizing radiation in children undergoing Amplatzer device placement to close atrial septal defects.

To evaluate exposure to ionizing radiation during Amplatzer device occlusion, a prospective study was performed to measure surface entrance radiation dose by thermoluminescent dosimetry (TLD). Between June 1998 and April 1999, dosimetry was carried out on 12 patients with Amplatzer device occlusion of atrial septal defects (n = 10) or Fontan fenestration (n =) and 12 age-matched patients who underwent diagnostic catherization. TLD chips were placed at the posterior (PA) and right lateral (LA) chest wall as well as the thyroid (TH) and gonadal (GN) regions. The Amplatzer group had a median age

[DNA-dependent protein kinase: a major protein involved in the cellular response to ionizing radiation].

DNA-dependent protein kinase (DNA-PK) is a DNA-activated nuclear serine/threonine protein kinase. DNA-PK consists of a regulatory sub-unit, the heterodimeric Ku protein (composed of a 70- and a 86-kDa subunit) which binds DNA ends and targets the catalytic sub-unit, DNA-PKcs to DNA strand breaks. DNA-PK plays a major role in the repair of double-strand breaks induced in DNA after exposure to ionizing radiation as shown by the extreme radiosensitivity of cells with mutations in Ku86, Ku70 or DNA-PKcs genes. Cells deficient in DNA-PK activity also exhibit hypersensitivity to genotoxic drugs such

Oxidative damage of mitochondrial and nuclear DNA induced by ionizing radiation in human hepatoblastoma cells.

PURPOSE: Since reactive oxygen species (ROS) act as mediators of radiation-induced cellular damage, the aim of our studies was to determine the effects of ionizing radiation on the regulation of hepatocellular reduced glutathione (GSH), survival and integrity of nuclear and mitochondrial DNA (mtDNA) in human hepatoblastoma cells (Hep G2) depleted of GSH prior to radiation. METHODS AND MATERIALS: GSH, oxidized glutathione (GSSG), and generation of ROS were determined in irradiated (50-500 cGy) Hep G2 cells. Clonogenic survival, nuclear DNA fragmentation, and integrity of mtDNA were assessed in

[Lipoxygenase activity and 15-hete content of rat spleen lymphocytes after exposure to ionizing radiation].

Pretreatment of rat spleen lymphocytes with 0.5 mM A23187 had no influence on the prostaglandins E2 and F2 alpha content, but was followed by the increasing of both 15-HETE and leukotriene B4 levels. Lypoxygenase activity of lymphocytes towards exogenous substrate linoleic acid was increased within 3-12 h after 1 Gy irradiation and decreased below the control level at 24 h. The changes of lypoxygenase activity correlate with that of 15-HETE content. Additional incubation of the cells, obtained at 3 h after irradiation, was followed by the intensification of the chromatin internucleosomal fragm

Radiation protection against early and late effects of ionizing irradiation by the prostaglandin inhibitor indomethacin.

Protective effects of indomethacin, a prototype prostaglandin-inhibiting agent, against early and late sequelae of radiation injury (after X-rays or gamma rays) in mice were investigated. The following tissues or organs were examined: hematopoietic tissue, esophagus, jejunum, colon, lung, hair follicles, and tissues involved in the development of radiation-induced leg contractures. In addition, the effect of indomethacin was tested against radiation-induced carcinogenesis. In all experiments, the radiation was delivered as a single dose. Indomethacin led to significant protection of hematopoie

Progressive behavioral changes in rats after exposure to low levels of ionizing radiation in utero.

The deleterious effects of ionizing radiation on the developing brain may be not only prolonged but progressive. Fetuses were exposed to 0.75 Gy of ionizing radiation on gestational day 15 through whole body exposure of the pregnant rat. Three behavioral tests (gait analysis, continuous corridor activity and photographic analysis of sequences of behavioral acts) were performed at 1 and 3 months, postnatally. Body weight and thickness of the cerebral cortex of irradiated rats were 10-15 percent below controls throughout the period of study. Behavior in all tests was more affected at 3 months th

A case-control study of congenital malformations and occupational exposure to low-level ionizing radiation.

In a case-control study, the authors investigated the association of parental occupational exposure to low-level external whole-body penetrating ionizing radiation and risk of congenital malformations in their offspring. Cases and controls were ascertained from births in two counties in southeastern Washington State, where the Hanford Site has been a major employer. A unique feature of this study was the linking of quantitative individual measurement of external whole-body penetrating ionizing radiation exposure of employees at the Hanford Site, using personal dosimeters, and the disease outco

The effects of hyperthermia and ionizing radiation in normal and ataxia telangiectasia human fibroblast lines.

The effects of 45 degrees C hyperthermia and gamma radiation have been studied in three normal human fibroblast lines (GM38, GM730, WI38) and compared to the effects in two lines derived from patients with the hereditary disease ataxia telangiectasia (AT3BI, AT5BI). All lines, both normal and gamma-sensitive AT, showed a similar resistance to killing by heat alone, suggesting that the defect responsible for the increased radiation sensitivity in AT lines does not confer increased heat sensitivity. Shouldered survival curves were obtained in each case indicating the ability to accumulate sublet

[Dynamics of morphological changes in the spinal cord following exposure to non-ionizing microwave radiation].

The structure of different portions (cervical, thoracic, lumbar, sacral and coccygeal) of the spinal cord were studied in the experiments on 50 animals with the use of the Nissl, Zimmermann, Cajal and other methods on days 1, 10, 20 and 30 after exposure to non-ionizing microwave radiation (NMI). Single exposure to NMI (wave length 12.6 cm, intensity 400-500 mW/cm2) for one hour (cats) or four hours (dogs) produces a severe distress of glial neurones and cells, which is marked by the appearance of dystrophic processes along the entire spinal cord. The disease progresses, leading to abnormaliti

"Ionizing Radiation Is Not in My Toolbox": Ethical Collaboration for Emerging Psychiatric Indications in Radiosurgery.

Recent advances in stereotactic radiosurgery have expanded consideration of non-malignant and functional indications, including emerging applications in psychiatric illness. In response to the review by Ehret et al. we offer a multidisciplinary perspective-co-authored by radiation oncology and psychiatry-on the ethical and educational implications of this evolving frontier. While psychiatric radiosurgery remains investigational, its irreversible neural effects and identity-relevant outcomes raise distinct considerations related to informed consent, decisional capacity, and long-term monitoring
