Fat Grafting in Radiation-Induced Soft-Tissue Injury: A Narrative Review of the Clinical Evidence and Implications for Future Studies.

Radiation-induced changes in skin and soft tissue result in significant cosmetic and functional impairment with subsequent decrease in quality of life. Fat grafting has emerged as a therapy for radiation-induced soft-tissue injury, and this narrative review aims to evaluate the current clinical evidence regarding its efficacy. A review was conducted to examine the current clinical evidence of fat grafting as a therapy for radiation-induced injury to the skin and soft tissue and to outline the clinical outcomes that can be used to more consistently quantify chronic radiation-induced injury in f

Sox9-expressing cells promote regeneration after radiation-induced lung injury via the PI3K/AKT pathway.

BACKGROUND: Radiation-induced lung injury (RILI) is considered one of the most common complications of thoracic radiation. Recent studies have focused on stem cell properties to obtain ideal therapeutic effects, and Sox9 has been reported to be involved in stem cell induction and differentiation. However, whether Sox9-expressing cells play a role in radiation repair and regeneration remains unknown. METHODS: We successfully obtained Sox9CreER, RosatdTomato and RosaDTA mice and identified Sox9-expressing cells through lineage tracing. Then, we evaluated the effects of the ablation of Sox9-expr

Gut Microbiota-Derived l-Histidine/Imidazole Propionate Axis Fights against the Radiation-Induced Cardiopulmonary Injury.

Radiation-induced cardiopulmonary injuries are the most common and intractable side effects that are entwined with radiotherapy for thorax cancers. However, the therapeutic options for such complications have yielded disappointing results in clinical applications. Here, we reported that gut microbiota-derived l-Histidine and its secondary metabolite imidazole propionate (ImP) fought against radiation-induced cardiopulmonary injury in an entiric flora-dependent manner in mouse models. Local chest irradiation decreased the level of l-Histidine in fecal pellets, which was increased following feca

Bone marrow stromal cell therapy improves survival after radiation injury but does not restore endogenous hematopoiesis.

The only available option to treat radiation-induced hematopoietic syndrome is allogeneic hematopoietic cell transplantation, a therapy unavailable to many patients undergoing treatment for malignancy, which would also be infeasible in a radiological disaster. Stromal cells serve as critical components of the hematopoietic stem cell niche and are thought to protect hematopoietic cells under stress. Prior studies that have transplanted mesenchymal stromal cells (MSCs) without co-administration of a hematopoietic graft have shown underwhelming rescue of endogenous hematopoiesis and have delivere

Persea americana Mill. crude extract exhibits antinociceptive effect on UVB radiation-induced skin injury in mice.

Persea americana, popularly known as avocado, has been empirically used as analgesic and anti-inflammatory including in the skin disorder treatment. Species of the genus Persea also show a photoprotective effect against UVB radiation. We investigated the antinociceptive and anti-inflammatory effects from a topical formulation containing the P. americana leaf extract in a UVB irradiation-induced burn model in mice and performed a gel-formulation stability study. The antinociceptive and anti-inflammatory effect was evaluated through mechanical allodynia, paw oedema, and inflammatory cell infiltr

Nanoencapsulation of coenzyme Q10 and vitamin E acetate protects against UVB radiation-induced skin injury in mice.

This study aimed to investigate the feasibility of producing semisolid formulations based on nanocapsule suspensions containing the association of the coenzyme Q10 and vitamin E acetate by adding gellan gum (2%) to the suspensions. Furthermore, we studied their application as an alternative for the treatment of inflammation induced by ultraviolet B (UVB) radiation. For this, an animal model of injury induced by UVB-radiation was employed. All semisolids presented pH close to 5.5, drug content above 95% and mean diameter on the nanometric range, after redispersion in water. Besides, the semisol

Transcriptional corepressor MTG16 regulates small intestinal crypt proliferation and crypt regeneration after radiation-induced injury.

Myeloid translocation genes (MTGs) are transcriptional corepressors implicated in development, malignancy, differentiation, and stem cell function. While MTG16 loss renders mice sensitive to chemical colitis, the role of MTG16 in the small intestine is unknown. Histological examination revealed that Mtg16(-/-) mice have increased enterocyte proliferation and goblet cell deficiency. After exposure to radiation, Mtg16(-/-) mice exhibited increased crypt viability and decreased apoptosis compared with wild-type (WT) mice. Flow cytometric and immunofluorescence analysis of intestinal epithelial ce

Radiation-induced lung injury and inflammation in mice: role of inducible nitric oxide synthase and surfactant protein D.

Reactive nitrogen species (RNS) generated after exposure to radiation have been implicated in lung injury. Surfactant protein D (SP-D) is a pulmonary collectin that suppresses inducible nitric oxide synthase (iNOS)-mediated RNS production. Herein, we analyzed the role of iNOS and SP-D in radiation-induced lung injury. Exposure of wild-type (WT) mice to γ-radiation (8 Gy) caused acute lung injury and inflammation, as measured by increases in bronchoalveolar lavage (BAL) protein and cell content at 24 h. Radiation also caused alterations in SP-D structure at 24 h and 4 weeks post exposure. These

Inhibition of p38 MAPK attenuates ionizing radiation-induced hematopoietic cell senescence and residual bone marrow injury.

Exposure to a moderate or high total-body dose of radiation induces not only acute bone marrow suppression but also residual (or long-term) bone marrow injury. The induction of residual bone marrow injury is primarily attributed to the induction of hematopoietic cell senescence by ionizing radiation. However, the mechanisms underlying radiation-induced hematopoietic cell senescence are not known and thus were investigated in the present study. Using a well-established long-term bone marrow cell culture system, we found that radiation induced hematopoietic cell senescence at least in part via a

Diffusion-weighted imaging in the follow-up of treated high-grade gliomas: tumor recurrence versus radiation injury.

BACKGROUND AND PURPOSE: Diffusion-weighted (DW) MR imaging is a means to characterize and differentiate morphologic features, including edema, necrosis, and tumor tissue, by measuring differences in apparent diffusion coefficient (ADC). We hypothesized that DW imaging has the potential to differentiate recurrent or progressive tumor growth from treatment-induced damage to brain parenchyma in high-grade gliomas after radiation therapy. METHODS: We retrospectively reviewed follow-up conventional and DW MR images obtained starting 1 month after completion of radiation treatment with or without c

[(ADP-ribose)n-transferase activity of enterocyte nuclei during radiation injury of small intestine mucosa in rats].

The (ADP-ribose)n transferase activity of enterocyte nuclei of small intestine is studied in rats in normal and under radiation injury of intestine mucosa. It is shown that kinetics of [14C]-NAD incorporation into the acid-insoluble fraction of enterocyte nuclei is of a two-step character when the medium contains 1-15 microM of NAD. The X-ray irradiation of animals (a dose of 0.21 cells per kg) evokes changes in the (ADP-ribose)n transferase activity in nuclei of enterocytes isolated 1-72 h after irradiation. The irradiation results in distortion of the two-step kinetics of (ADP-ribose) synthe

Stimuli-responsive hydrogels for radiation-induced skin injury: from passive barriers to autonomous drug delivery systems.

Radiation-induced skin injury (RISI) affects over 95% of radiotherapy patients. Current clinical management remains confined to passive supportive care, lacking mechanistic precision for RISI's unique pathophysiology. This review adopts a function-centric perspective, classifying hydrogel systems across three generations: first-generation passive moisture barriers; second-generation bioactive platforms incorporating antioxidants, growth factors, stem cells and exosomes; and third-generation stimuli-responsive systems integrating autonomous drug release, self-healing capabilities and biosensor

Chlorella-derived extracellular vesicle-based nanogels suppress cGAS-STING for treatment of radiation-induced lung injury.

Radiation-induced lung injury (RILI) represents a critical complication associated with radiation exposure, requiring targeted treatment strategies in clinical practice. Here, we pioneered the extraction of extracellular vesicles (EVs) from Chlorella and innovatively developed a multifunctional nanogel platform (RU.521-EVs NPs) by combining them with the cGAS inhibitor RU.521. The Chlorella-derived EVs possess a naturally high abundance of antioxidant enzymes (e.g., SOD2, CAT) and anti-inflammatory lipids (e.g., linolenic acid). RU.521-EVs NPs could synergistically inhibit excessive activation

Deciphering Radiation-Induced Lung Injury: From Therapeutic Challenges to Macrophage-Centered Molecular Mechanisms and Innovative Interventions.

Radiation-induced lung injury (RILI) is a life-threatening complication of thoracic radiation therapy for malignancies. It poses 2 challenges in clinical management: the progression from acute radiation pneumonitis to irreversible pulmonary fibrosis and the limitations of current therapies, such as glucocorticoids and antifibrotic drugs, because of efficacy constraints and adverse effects. Recent studies have revealed that pulmonary macrophages form a spatiotemporally regulated inflammatory-fibrotic coupling network through phenotypic switching. Specifically, alveolar macrophages exhibit M1 pr
