Molecular analysis of the human vitamin D binding protein (group specific component, Gc) in tuberous sclerosis complex (TSC).

Group specific component (Gc) is an abundant plasma protein whose functional role is not clearly established. Gc protein is synthesised in the liver and is known to bind vitamin D, vitamin D metabolites, and G actin; Gc protein is also implicated in macrophage activation. Several polymorphic electrophoretic variants of Gc protein are found in all human populations; the most common alleles are Gc-1f, Gc-1s, and Gc-2. In previous studies, Gc allele frequencies, determined using isoelectric focusing or immunofixation or both, were significantly different in patients with tuberous sclerosis comple

Association Between Serum 25-Hydroxyvitamin D and Blood Pressure in Young Adults.

BACKGROUND: Vitamin D has been associated with blood pressure across the life course in observational studies, although effect sizes are generally modest and findings are not fully consistent. We examined the association between serum 25-hydroxyvitamin D [25(OH)D] concentrations and multiple blood pressure indices in a community-based cohort of young adults. METHODS: We conducted a cross-sectional analysis within the Young Taiwanese Adults (YOTA) cohort, restricting the sample to adults aged 18-45 years with complete serum 25(OH)D and blood pressure data. Serum 25(OH)D was modeled as a contin

Characterization of bone marrow lesions in axial spondyloarthritis using quantitative T1 mapping MRI.

OBJECTIVE: Conventional magnetic resonance imaging (MRI) uses T1-weighted and short-tau inversion recovery (STIR) sequences to characterize bone marrow in axial spondyloarthritis. However, quantification is restricted to estimating the extent of lesions because signal intensities are highly variable both within individuals and across patients and MRI scanners. This study evaluates the performance of quantitative T1 mapping for distinguishing different types of bone marrow lesions of the sacroiliac joints. MATERIALS AND METHODS: In this prospective study, 62 patients underwent computed tomogra

Correlation between sinus rhythm deceleration zones and critical sites for localized reentrant atrial flutter: A retrospective multicenter analysis.

BACKGROUND: Atypical left atrial flutter (AFL) may be macroreentrant or spatially localized. The relationship between the critical isthmus (CI) for localized reentry with sinus rhythm (SR) conduction slowing has not been systematically examined. OBJECTIVE: To examine the correlation between CI sites for localized AFL (L-AFL) and deceleration zones (DZ) identified by isochronal late activation mapping (ILAM) during baseline rhythm. METHODS: Patients with localized AFL who underwent high-density activation mapping of both SR and AFL were retrospectively analyzed. L-AFL was defined as reentry r

Impaired recovery of cardiac output and mean arterial pressure after successful defibrillation in patients with low left ventricular ejection fraction.

BACKGROUND: Early defibrillation clearly improves survival from malignant arrhythmia. However, in some cases the cause of death will only be altered from arrhythmic to nonarrhythmic. We evaluated the impact of left ventricular ejection fraction (LVEF) on trend and recovery profile of beat-to-beat cardiac output (CO) and mean arterial blood pressure (MAP) after successful defibrillation. METHODS: We investigated 63 NYHA class I-III patients undergoing threshold testing in the course of insertion of an implantable cardioverter defibrillator (ICD) in monitored anaesthesia care. Isolation and mapping of a cosmid clone containing the human NAT2 gene.

The NAT2 gene encodes for a polymorphic arylalkylamine N-acetyltransferase and thus accounts for the human N-acetylation polymorphism. By a NAT2-specific primer set we have screened a human chromosome 8-specific cosmid library. A positive cosmid clone was mapped by fluorescence in situ hybridization to 8p22. The polymerase chain reaction followed by restriction analysis of the PCR product was used to identify allele 2 to be contained in the cosmid clone.

Abacavir inhibits but does not cause self-reactivity to HLA-B*57:01-restricted EBV specific T cell receptors.

Pre-existing pathogen-specific memory T cell responses can contribute to multiple adverse outcomes including autoimmunity and drug hypersensitivity. How the specificity of the T cell receptor (TCR) is subverted or seconded in many of these diseases remains unclear. Here, we apply abacavir hypersensitivity (AHS) as a model to address this question because the disease is linked to memory T cell responses and the HLA risk allele, HLA-B*57:01, and the initiating insult, abacavir, are known. To investigate the role of pathogen-specific TCR specificity in mediating AHS we performed a genome-wide scr

Serological investigation and genotyping of Mycobacterium avium subsp. paratuberculosis in sheep and goats in Inner Mongolia, China.

Paratuberculosis a contagious and chronic disease in domestic and wild ruminants, is caused by Mycobacterium avium subspecies paratuberculosis (MAP). Typical clinical signs include intractable diarrhea, progressive emaciation, proliferative enteropathy, and mesenteric lymphadenitis. Paratuberculosis is endemic to many parts of the world and responsible for considerable economic losses. In this study, different types of paratuberculosis and MAP in sheep and goats were investigated in Inner Mongolia, a northern province in China contiguous with two countries and eight other provinces. A total of

The Role of Navigated Transcranial Magnetic Stimulation Motor Mapping in Adjuvant Radiotherapy Planning in Patients With Supratentorial Brain Metastases.

Purpose: In radiotherapy (RT) of brain tumors, the primary motor cortex is not regularly considered in target volume delineation, although decline in motor function is possible due to radiation. Non-invasive identification of motor-eloquent brain areas is currently mostly restricted to functional magnetic resonance imaging (fMRI), which has shown to lack precision for this purpose. Navigated transcranial magnetic stimulation (nTMS) is a novel tool to identify motor-eloquent brain areas. This study aims to integrate nTMS motor maps in RT planning and evaluates the influence on dosage modulation

Association between time to target mean arterial pressure and 90-day mortality in septic shock: a post hoc analysis of the OPTPRESS trial.

BACKGROUND: In septic shock resuscitation, maintaining a mean arterial pressure (MAP) of at least 65 mmHg is recommended; however, the clinical relevance of the time required to achieve target MAP remains unclear. We evaluated the association between time to target MAP (TTT-MAP) and clinical outcomes in septic shock. METHODS: This post hoc secondary analysis used data from the OPTPRESS randomized controlled trial. TTT-MAP was defined as the time from randomization to first attainment of the randomized-arm lower-bound MAP target (80 mmHg in the high-target arm; 65 mmHg in the low-target arm) a

Effect of Aerobic Exercise with Blood Flow Restriction on Postexercise Hypotension in Young Adults: The Role of Histamine Receptors.

We tested hypothesis that aerobic exercise with blood flow restriction (BFR) induced postexercise hypotension (PEH), and the reduction in blood pressure (BP) was due to peripheral vasodilation via the histamine receptors. Ten male subjects participated in this study. The subjects were randomly assigned to walk for 10 min at 6.4 km/h, 0% grade with or without BFR after taking histamine receptor blockade. Following exercise, BP was measured at 10 min interval for 60 min. Heart rate (HR), stroke volume (SV), cardiac output (CO), mean arterial pressure (MAP), and total peripheral resistance (TPR)

Blood pressure indices for predicting incident cardiovascular disease: A 13-year follow-up study in Japanese population.

BACKGROUND: Several blood pressure (BP) indices have been associated with incident cardiovascular disease (CVD); however, evidence comparing their long-term prognostic value in Asian populations is limited. We investigated the association between multiple BP indices and CVD risk over a 13-year follow-up period in a large Japanese population. METHODS: Data from a health check-up program conducted by the Panasonic Corporation covering 166 operational sites from 2008 to 2021, including 163,956 participants not receiving anti-hypertensive drugs, were analyzed. The primary outcome was the incidenc

Association of mean arterial pressure with non-alcoholic fatty liver disease: results from the NAGALA study.

OBJECTIVE: Several recent reports have suggested the use of mean arterial blood pressure (MAP) to assess/predict the risk of developing atherosclerosis, chronic kidney disease, diabetes, metabolic syndrome, and poor prognosis in a variety of cardiovascular and cerebrovascular diseases. The current study aimed to investigate the association of MAP with non-alcoholic fatty liver disease (NAFLD) and to explore the differences in this association across populations. METHODS: This study used data from the NAGALA study from 1994 to 2016. MAP was calculated as 1/3 systolic blood pressure (SBP) + 2/3

Therapeutic management of Mycobacterium avium subspecies paratuberculosis infection with complete resolution of symptoms and disease in a patient with advanced inflammatory bowel syndrome.

BACKGROUND: A 26-year-old male had a history of frequent bowel movements, mushy stool with mucus and loss of 25 kg body weight in 6 months was diagnosed as a case of inflammatory bowel disease (IBD). The patient did not respond to routine and standard treatment for IBD. His condition was steadily deteriorating, and he was in a very precarious state when he reported to us. METHODS: Upon laboratory investigation by using IS900 specific PCR [which is specific for Mycobacterium avium subspecies paratuberculosis (MAP)], the blood and stool samples were found negative. However, the presence of low

The Ikaros gene encodes a family of lymphocyte-restricted zinc finger DNA binding proteins, highly conserved in human and mouse.

The Ikaros gene is an essential regulator in the development and homeostasis of the mouse lymphopoietic system. To study the role of the Ikaros gene in the human lymphopoietic system, we cloned and characterized human Ikaros cDNAs. In the human, as in the mouse, differential splicing of Ikaros primary transcripts generates a family of lymphoid-restricted zinc finger DNA binding proteins, highly conserved in sequence composition and relative expression to the mouse homologues. Expression of Ikaros isoforms is highly restricted to the lymphopoietic system and is particularly enriched in maturing

Localization of deletion breakpoints in radiation-induced mutants of the hprt gene in hamster cells.

DNA was analysed from a large set of hamster hprt gene mutants, some induced by ionising radiations and others occurring naturally, to identify those with large alterations in part of the gene. DNA from these mutants was restricted further with different endonucleases and probed to establish the patterns of restriction fragments remaining. Of 15 mutants characterized, one showed a duplication of part of the 5' end of the gene, and the remainder showed deletions of various sizes. It was possible to approximately locate the breakpoints of the deletions by comparison of fragment patterns to a rec

Cerebellar corticonuclear projection in the cat. The vermis of the anterior and posterior lobes.

The projection of the vermis of the anterior and posterior lobes of the cerebellum to the fastigial nucleus was studied. It was attempted to obtain lesions restricted to the lobules and this was achieved for the posterior lobe only. The extent of lesions and of additional encroachments were determined by mapping degenerated descending cortical fibers in serial sections. Evidence was obtained that the projection of the vermis is restricted to the fastigial nucleus, without a contribution to the interposed nuclei, in the posterior lobe. Indirect evidence supports the existence of a similar arran

From research method to community resource: Co-developing a body mapping toolkit for peer support with Long Covid patients.

The paper describes the development of a Body Mapping Toolkit for Long Covid Patients, co-created in collaboration with the patient-led organisation Long Covid Support. We discuss how the use of body mapping, a participatory and arts-based research method, can support a more holistic and embodied understanding of Long Covid attentive to the way illness experiences are shaped by patients' social, cultural, and economic contexts. We further demonstrate how, through collaboration with patient organisations, body mapping might be extended beyond this research application to create spaces for peer
