Loss of cytochrome oxidase in Saccharomyces cerevisiae during inhibition of mitochondrial protein synthesis by erythromycin and chloramphenicol.

There is a major reduction in respiratory competence, and inhibitionof growth, several hours after the addition of erythromycin or chloramphenicol to Saccharomyces cerevisiae growing in medium containing a non-fermentable carbon source. Spectrographic evidence is presented for a loss of cytochrome oxidase as a consequence of the antibiotic treatment. This loss is prevented by cyanide or oligomycin. When glucose is added, however, the loss occurs irrespective of the presence of the respiratory inhibitors. Cycloheximide does not affect respiratory competence or cause loss of cytochrome oxidase, …

Saccharomyces boulardii in patients with severe acute pancreatitis: a single center, open-label randomized controlled trial.

BACKGROUND: Nosocomial infections in patients with severe acute pancreatitis (SAP) are frequently driven by impaired intestinal barrier function, which facilitates bacterial translocation and contributes to adverse clinical outcomes. Saccharomyces boulardii (S. boulardii) can reconstitute gut microbiota composition. We investigated whether S. boulardii combined with enteral nutrition (EN) affects the microbiome and nosocomial infections in SAP. METHODS: This study is a single centre, open-label randomized controlled trial. We included 50 patients with SAP in a Chinese gastroenterology intensi…

Antioxidant and Anti-Aging Effects of Porphyra-334 Produced from Saccharomyces cerevisiae in Human Skin Models.

Porphyra-334 (PPR-334) is one of the species of mycosporine-like amino acids (MAAs), known as biological UV protection ingredients. In this study, we developed a large-scale purification process to extract PPR-334 from Saccharomyces cerevisiae and confirmed the previously identified efficacy of PPR-334, while also demonstrating its efficacy under UV-independent conditions. PPR-334 scavenged reactivity oxygen species (ROS) and increased catalase (CAT) gene expression in human epidermal keratinocyte cells (HEKa). In both HEKa and normal human dermal fibroblast cells (NHDF), PPR-334 suppressed th…

Ribosome-associated chaperones integrate co-translational mitochondrial protein import, stress signaling and proteostasis in Saccharomyces cerevisiae.

Mitochondrial biogenesis requires the coordinated synthesis, targeting, and import of nuclear-encoded mitochondrial precursor proteins. Although ribosome-associated chaperones support co-translational protein folding, their genetic contributions to mitochondrial protein import and cellular homeostasis remain incompletely defined. Here, we investigate the roles of the nascent polypeptide-associated complex (NAC) and the ribosome-associated Hsp70 system Ssb1/2 in Saccharomyces cerevisiae. We show that NAC and Ssb1/2 have distinct yet partially overlapping functions in the handling of mitochondri…

Saccharomyces cerevisiae and Bacillus subtilis show modest, node-dependent effects on insulin sensitivity in periparturient cows.

Periparturient immunometabolic adaptations yield small, node-dependent shifts in endotoxin proxies and insulin-sensitivity indices. Direct-fed microbials (DFM) are mechanistically plausible but often produce modest metabolic readouts. Test whether Saccharomyces cerevisiae (SC), Bacillus subtilis (BS), or SC + BS alter endotoxin (blood, milk) and metabolic families versus Control (Ctrl) across prespecified nodes, and evaluate prespecified equivalence where applicable. A 4-arm trial enrolled 20 Holsteins from -60 to + 21 d around calving (Ctrl, SC, BS, SC+BS). Blood and milk were sampled at mult…

Nano-formulated β-glucans from Saccharomyces cerevisiae enhance immunity and protection against coccidiosis in broiler chickens.

This study investigated the effects of a β-glucans nanoformulation (β-GN) derived from Saccharomyces (S.) cerevisiae on immune responses and protection against coccidiosis in broilers. For this purpose, S. cerevisiae was cultured from baker's yeast followed by enzymatic extraction of β-glucans with a yield rate of 36.67 ± 1.17%. The β-GN was prepared using the ionic gelation method and characterized by FTIR-spectroscopy, which confirmed the structural integrity of β-glucans. Toxicological safety of β-GN was assessed prior to the experimental trial. The β-GN was supplemented to the broiler diet…

Flow cytometric distinction of early apoptotic, late apoptotic and primary necrotic morphological markers in Saccharomyces cerevisiae.

Regulated cell death (RCD) is a fundamental biological process that ensures tissue homeostasis, mediates stress responses, and contributes to diverse pathological conditions. The budding yeast Saccharomyces cerevisiae has emerged as a valuable model organism for the study of RCD, providing evolutionary insights into conserved molecular pathways and enabling the systematic dissection of cell death mechanisms in a genetically tractable system. Upon a lethal stimulus, yeast populations may segregate into three major subtypes: primary necrotic cells with disrupted plasma membranes, early apoptotic…

Design of an oral vaccine against Salmonella pullorum using protein surface display on bioengineered Saccharomyces cerevisiae.

Salmonella enterica subsp. Salmonella Pullorum is a concerned pathogenic microorganism that poses a serious threat to the global poultry industry due to high mortality in chicken. Acquired immunity is considered the most effective means to prevent and control Salmonella pullorum. However, safe and effective vaccines are still lacking worldwide. In this study, Saccharomyces cerevisiae was used as a chassis to design an oral vaccine for pullorum by displaying Salmonella fimbriae and outer membrane proteins on its surface. The results demonstrated that the engineered yeast EY-01 exhibited strong …

Deciphering the Molecular Adapting Mechanism of Lactic Acid-Tolerant Saccharomyces cerevisiae Through Genomic and Transcriptomic Analysis.

During the solid-state brewing process of traditional Chinese Baijiu, lactic acid is the most abundant organic acid, which inhibits the growth and metabolism of Saccharomyces cerevisiae. To reveal the lactic acid tolerance mechanism of S. cerevisiae, the growth, metabolic performance, and antioxidant enzyme activity of S. cerevisiae NCUF309.5-44 and S. cerevisiae NCUF309.5 were measured under 4% (v/v) lactic acid stress. Additionally, whole-genome re-sequencing and transcriptomic analyses were performed to identify genetic variations and differentially expressed genes between the two strains u…

Activated carbon facilitates Saccharomyces cerevisiae colonization in an electron-donor self-sustaining chain elongation fermentation system.

Chain elongation is a promising technology for the resourceful utilization of organic waste and wastewater. Glucose and starch were used as model substrates to demonstrate that exogenous electron donors are essential for chain elongation. Glucose and starch affects caproate synthesis in the chain elongation lacking of electron donor producing microorganisms, while glucose and ethanol significantly influence the succession of microbial community. Activated carbon enhanced the colonization of Saccharomyces cerevisiae and strengthened the chain elongation process, with the relative abundance of S…

Enhancing ethanol tolerance and metabolic activity of Saccharomyces cerevisiae during Rice fermentation by fixed-frequency ultrasound.

Ultrasound pretreatment shows potential for enhancing microbial fermentation efficiency, but its mechanochemical impact on S. cerevisiae during rice fermentation, particularly regarding ethanol tolerance and metabolic activity, requires systematic investigation. Therefore, the multi-dimensional influence of ultrasound on S. cerevisiae was studied from the aspects of biomass dynamics under ethanol stress, membrane biophysics, metabolic markers, and oxidative stress response. Following the ultrasound treatment (logarithmic metaphase, 28 kHz, and 45 W/L), the growth rate of S. cerevisiae was fast

Chromosome III Aneuploidy Enhances Ethanol Tolerance in Industrial Saccharomyces cerevisiae by Increasing the TUP1 Copy Number.

Ethanol stress poses a considerable challenge for Saccharomyces cerevisiae during fermentation. Strains carrying an extra copy of chromosome III exhibit enhanced ethanol tolerance. Here, we investigated the underlying mechanisms of this tolerance, focusing on gene dosage effects and differential gene expression under ethanol stress. We compared the gene expression profiles of a strain with three copies of chromosome III and its derivative with two copies, exposed to 6% and 10% ethanol. Our analysis identified TUP1, located on chromosome III, as a key regulator of the ethanol stress response. D…

Iron, manganese, and copper complexes protect saccharomyces cerevisiae from methyl methanesulfonate-induced genotoxicity and oxidative stress.

Methyl methanesulfonate (MMS) is a well-known classical alkylating agent that induces DNA damage, mutagenesis, and cell dysfunction. Alternatives to mitigate the damage caused by MMS are essential for understanding DNA repair mechanisms and developing approaches to reduce mutagenic and cytotoxic effects of alkylating agents. This study explores the potential of synthetic antioxidants containing iron (1), manganese (2), or copper (3) to protect Saccharomyces cerevisiae from MMS-induced damage. The wild type strain, BY4741, and its isogenic DNA-repair mutants rad9Δ and rad54Δ strains were used t…

Interactions of Saccharomyces cerevisiae and Lactiplantibacillus plantarum Isolated from Light-Flavor Jiupei at Various Fermentation Temperatures.

Chinese Baijiu is a famous fermented alcoholic beverage in China. Interactions between key microorganisms, i.e., Saccharomyces cerevisiae and Lactiplantibacillus plantarum, have recently been reported at specific temperatures. However, empirical evidence of their interactions at various temperatures during fermentation is lacking. The results of this study demonstrated that S. cerevisiae significantly suppressed the viability and lactic acid yield of L. plantarum when they were cocultured above 15 °C. On the other hand, L. plantarum had no pronounced effect on the growth and ethanol yield of S…

Yap1-mediated Flr1 expression reveals crosstalk between oxidative stress signaling and caffeine resistance in Saccharomyces cerevisiae.

Caffeine, a methylxanthine derivative, affects various physiological conditions such as cell growth, proliferation, and energy metabolism. A genome-wide screening for genes required for caffeine resistance in Schizosaccharomyces pombe revealed several candidates, including Pap1 and downstream target genes involved in caffeine efflux. We found that Yap1, a budding yeast AP-1 homolog required for oxidative stress response, has a caffeine tolerance function. Although the Yap1 mutant is not sensitive to caffeine, overexpression of Yap1 renders cells resistant to high concentrations of caffeine. Ca…

Saccharomyces boulardii modulates oxidative stress and renin angiotensin system attenuating diabetes-induced liver injury in mice.

Type 1 diabetes (T1DM) is a chronic disease characterized by hyperglycemia due to a deficiency in endogenous insulin production, resulting from pancreatic beta cell death. Persistent hyperglycemia leads to enhanced oxidative stress and liver injury. Several studies have evaluated the anti-diabetic and protective effects of probiotic strains in animal models. In the present study, we investigated, through histopathological and biochemical analyses, the effects of eight weeks of administration of Saccharomyces boulardii (S. boulardii) yeast on the liver of streptozotocin (STZ) induced diabetic C…
