Combination of lactoferrin-based microparticles and carboanhydrase II inhibitor demonstrates enhanced inhibition effect on Ewing sarcoma cells.

Ewing sarcoma (ES) is a highly aggressive pediatric malignancy with limited treatment options and frequent development of drug resistance. In this case, novel drug delivery systems may overcome tumor resistance and improve therapeutic efficacy. We developed lactoferrin-chondroitin sulfate microparticles (Lf-ChS MPs) that can be loaded with the carbonic anhydrase II inhibitor OX72. Their physicochemical properties were characterized by AFM, FTIR, zeta potential, DSC/TGA, and drug release assays. In vitro cytotoxicity was evaluated in ES cell lines (A673, ES36, T69, and doxorubicin-resistant A67

Neoadjuvant chemotherapy-enabled tumor conversion and surgical resection in pediatric primary pulmonary Ewing sarcoma: a case report.

BACKGROUND: Ewing sarcoma (EWS) is a highly aggressive malignant tumor that primarily affects the skeletal system in children and adolescents. Primary pulmonary Ewing sarcoma (PPEWS) is extremely rare, particularly in children, with very few cases reported in the literature. CASE PRESENTATION: This report describes a 10-year-old girl who was admitted with an intermittent cough lasting over 7 months. Chest computed tomography (CT) revealed a mass in the right lower lung lobe. Bronchoscopic biopsy demonstrated a highly malignant undifferentiated small round cell tumor, with immunohistochemistry

Distinct epidemiology and treatment outcomes between skeletal and extraskeletal Ewing sarcoma in Japan: a population-based study.

BACKGROUND: Ewing sarcoma arises at both skeletal (SES) and extraskeletal (EES) sites; however, whether the anatomical origin influences outcomes in current practice remains uncertain. This study compared clinical characteristics, treatment, and survival rates of patients with SES and EES in a large nationwide Japanese cohort. METHODS: We analyzed patients diagnosed with Ewing's sarcoma between 2016 and 2019 in Japan, classified as having SES or EES, using a population-based cancer registry. Demographics, stage, treatment, hospital characteristics, and overall survival (OAS) were evaluated us

Imaging of ewing sarcoma: an updated analysis including presenting features, prognostic imaging biomarkers, and treatment response assessment.

Ewing sarcoma is a highly aggressive small round cell sarcoma primarily affecting children and adolescents. Imaging plays a central role from diagnosis to staging, treatment response assessment, and follow-up. This review synthesizes current evidence across the various imaging modalities involved at each stage of patient management, including conventional radiography, CT, MRI, and nuclear imaging, emphasizing their complementary roles. Radiographs and CT delineate bone destruction patterns, cortical breaches, and periosteal reactions, while MRI provides superior visualization of intramedullary

Pathologic and Molecular Diagnosis of Ewing Sarcoma: A Multicenter Analysis From the Latin American Cooperative Group Trial.

INTRODUCTION: Ewing Sarcoma (ES) is a small, round, blue cell tumor (SRBCT) characterized by a chromosomal translocation between chromosomes 11 and 22 in ~85% of cases, alongside immunohistochemical (IHC) expression of the surface glycoprotein CD99. Despite advancements in molecular diagnostics, low-income countries continue to face challenges in tumor classification and identification of fusion partners. METHODS: This study retrospectively analyzed pathology reports from 396 patients enrolled in the Latin American Cooperative Group (GALOP) trial, with data collected until December 2021. CD99

A Rare Case of Ewing Sarcoma Presenting in the Adult Bladder-Treatments and Need for More Discussion.

Ewing sarcoma (ES) is a rare, aggressive malignancy seldom found in the urinary bladder. We report a 33-year-old male diagnosed with extra-skeletal ES of the bladder following symptoms initially attributed to prostatitis. Despite multimodal therapy-chemotherapy, radiation, and partial cystectomy-he developed early widespread metastases and died within a year. Pathology revealed positive surgical margins and limited treatment response. This case highlights the diagnostic and therapeutic complexity of bladder ES, the importance of surgical margin status, and the patient's autonomy in surgical de

Metastatic-Site Radiation Therapy for Ewing Sarcoma and Rhabdomyosarcoma: Consensus Guidelines From the National Pediatric Cancer Foundation.

PURPOSE: Despite the urgent need for improved outcomes in patients with metastatic Ewing sarcoma (EWS) and rhabdomyosarcoma (RMS), it is unknown how to best approach metastatic-site radiation therapy for these patients and whether such treatment provides a significant oncologic benefit that outweighs the toxicities. METHODS AND MATERIALS: We gathered a panel of pediatric radiation oncologists from academic hospitals to identify and discuss current controversies regarding the role of radiation in the management of metastatic EWS and RMS. The panel reviewed existing clinical data and ongoing tr

Shifting from a Biological-Agnostic Approach to a Molecular-Driven Strategy in Rare Cancers: Ewing Sarcoma Archetype.

Sarcomas of the thoracic cavity are rare entities that predominantly affect children and young adults. They can be very heterogeneous encompassing several different histological entities. Ewing Sarcoma (ES) can potentially arise from every bone, soft tissue, or visceral site in the body. However, it represents an extremely rare finding when it affects the thoracic cavity. It represents the second most frequent type of thoracic sarcoma, after chondrosarcoma. ES arises more frequently in sites that differ from the thoracic cavity, but it displays the same biological features and behavior of extr

Outcome for Neoadjuvant Treatment of Parotid Gland Adamantinoma-Like Ewing Sarcoma: Case Report and Review of Literatures.

Adamantinoma-like Ewing sarcoma typically shows t(11;22) EWSR1::FLI1 translocation and complex epithelial differentiation. It poses a diagnostic challenge, especially in the head and neck region, due to its under-recognition and significant histologic overlap with other malignancies. Neoadjuvant and adjuvant treatment information on head and neck Adamantinoma-like Ewing sarcoma is limited. Herein, we report a case of a 78-year-old female with Adamantinoma-like Ewing sarcoma of the parotid gland, including the imaging findings and clinical response to neoadjuvant therapy followed by surgery. Cabozantinib in patients with advanced Ewing sarcoma or osteosarcoma (CABONE): a multicentre, single-arm, phase 2 trial.

BACKGROUND: Patients with Ewing sarcoma or osteosarcoma have a median overall survival of less than 12 months after diagnosis, and a standard treatment strategy has not yet been established. Pharmacological inhibition of MET signalling and aberrant angiogenesis has shown promising results in several preclinical models of Ewing sarcoma and osteosarcoma. We aimed to investigate the activity of cabozantinib, an inhibitor of MET and VEGFR2, in patients with advanced Ewing sarcoma and osteosarcoma. METHODS: We did a multicentre, single-arm, two-stage, phase 2 trial in patients with advanced Ewing

Cutting Edge: The RNA-Binding Protein Ewing Sarcoma Is a Novel Modulator of Lymphotoxin β Receptor Signaling.

Lymphotoxin β receptor (LTβR) signaling is crucial for lymphoid tissue organogenesis and immune homeostasis. To identify novel regulatory mechanisms for signaling, we implemented a two-step screen that uses coexpression analysis of human fibroblasts undergoing LTβR stimulation and affinity-purification mass spectrometry for the LTβR signaling protein TNFR-associated factor 3 (TRAF3). We identify Ewing sarcoma (EWS) protein as a novel LTβR signaling component that associates with TRAF3 but not with TNFR-associated factor 2 (TRAF2). The EWS:TRAF3 complex forms under unligated conditions that are

Pregnancy-Associated Plasma Protein-A (PAPP-A) in Ewing Sarcoma: Role in Tumor Growth and Immune Evasion.

BACKGROUND: Ewing sarcoma (EWS) manifests one of the lowest somatic mutation rates of any cancer, leading to a scarcity of druggable mutations and neoantigens. Immunotherapeutics targeting differentially expressed cell surface antigens could provide therapeutic benefit for such tumors. Pregnancy-associated plasma protein A (PAPP-A) is a cell membrane-associated proteinase produced by the placenta that promotes fetal growth by inducing insulinlike growth factor (IGF) signaling. METHODS: By comparing RNA expression of cell surface proteins in EWS (n = 120) versus normal tissues (n = 42), we com
