[Caerulein-induced contraction of the ileal longitudinal smooth muscle isolated from various animal species].

An effect of caerulein was studied on a contractile response of the ileal longitudinal smooth muscle isolated from seven animal species, mon … g. TTX inhibited the contractions in all the ilea. As the contraction was inhibited by atropine and scopolamine in dog ileum but not in rabbit one, the contraction may be due to an excitation of the cholinergic neuron or an excitation of non-cholinergic excitatory neuron, respectively. On the other hand, it is supposed that the contraction in guinea-pig ileum …

Hippocampal criticality tracks cognitive demand and shifts with memory impairment.

Dynamical systems exhibit transitions between ordered and disordered states, and a "critical state" occurs when the system lies at the borde … urons in freely behaving mice during rest, a novel object recognition (NOR) task, and NOR following scopolamine administration that severely impairs spatial memory. We find that while hippocampal networks exhibit some features of a near-critical system at rest, network activity shifts significantly closer to a critical state during NOR, and away from critical …

Turmeric nanovesicles loaded with vitamins C and E attenuate oxidative stress and improve cognitive outcomes in rats with memory deficits.

AIM: Oxidative stress (OS) contributes to Alzheimer's disease (AD) pathology. Vitamins C and E target OS, but their efficacies are limited by poor intracellular delivery. Encapsulation in nanovesicles (NVs) may improve targeted delivery for AD treatment. METHODS: Forty-five rats were randomly assigned to nine groups (n = 5 per group): normal control, scopolamine control, vitamin C, vitamin E, vitamin C + E, turmeric NVs (TNV), TNV-C, TNV-E, and donepezil. Treatment was administered intraperitoneally (IP) for one week before induction of the AD-like model using daily IP administration of scopo…

Semi-Synthesis and Biological Evaluation of Phyllanthin Derivatives as Potential Neuroprotective Agents.

Oxidative stress is responsible for the functional loss and sensory impairment of neurons, which leads to progressive loss of brain and spinal cord cells and contributes significantly to several neurological disorders, thus eventually resulting in brain atrophy and death. This study involves the isolation of lignans, including phyllanthin, hypophyllanthin, and nirtetraline, from Phyllanthus amarus. Furthermore, the phyllanhtin was semi-synthetically modified to give phyllanthin oxadiazole derivatives and was evaluated for neuroprotective activity in scopolamine-injured neuroblastoma-2a (N2A) c…

Preventive Effect of Probiotic Leuconostoc mesenteroides H40 Against Cognitive Disorder by Anti-Inflammatory, Synaptic Plasticity Regulation, and Antioxidant Effects.

Live Leuconostoc mesenteroides H40 has been reported to have probiotic properties; however, the effect of its live probiotic form on cognitive ability has not been reported. This study investigated modulatory effects of the probiotic L. mesenteroides H40 in an ICR mouse model (male) of cognitive disorders. Cognitive disorders were induced in mice by the addition of scopolamine (1 mg/kg/day) with donepezil (2 mg/kg/day) as a medicinal control. L. mesenteroides H40 significantly attenuated scopolamine-induced cognitive disorder in the novel object recognition and Y-maze tests in a concentration-…

M1 muscarinic receptor is a key target of neuroprotection, neuroregeneration and memory recovery by i-Extract from Withania somnifera.

Memory loss is one of the most tragic symptoms of Alzheimer's disease. Our laboratory has recently demonstrated that 'i-Extract' of Ashwagandha (Withania somnifera) restores memory loss in scopolamine (SC)-induced mice. The prime target of i-Extract is obscure. We hypothesize that i-Extract may primarily target muscarinic subtype acetylcholine receptors that regulate memory processes. The present study elucidates key target(s) of i-Extract via cellular, biochemical, and molecular techniques in a relevant amnesia mouse model and primary hippocampal neuronal cultures. Wild type Swiss albino mice…

Phytochemical analysis, antioxidant, antistress, and nootropic activities of aqueous and methanolic seed extracts of ladies finger (Abelmoschus esculentus L.) in mice.

Abelmoschus esculentus L. (ladies finger, okra) is a well-known tropical vegetable, widely planted from Africa to Asia and from South Europe to America. In the present study, we investigated the in vitro antioxidant capacity and in vivo protective effect of the aqueous and methanolic seed extracts of Abelmoschus esculentus against scopolamine-induced cognitive impairment using passive avoidance task and acute restraining stress-induced behavioural and biochemical changes using elevated plus maze (EPM) and forced swimming test (FST) in mice. Our results demonstrated that the pretreatment of mic…

An NGF mimetic, MIM-D3, stimulates conjunctival cell glycoconjugate secretion and demonstrates therapeutic efficacy in a rat model of dry eye.

The aim of this study was to evaluate the efficacy of MIM-D3, a small molecule nerve growth factor (NGF) peptidomimetic, as a therapeutic agent in rats with scopolamine induced dry eye. NGF plays an important role in ocular surface maintenance and corneal wound healing and was recently shown to have mucin secretagogue activity in conjunctival cells. We investigated whether MIM-D3 increased glycoconjugate secretion in conjunctival cells in vitro and in rat tear fluids in vivo. Primary rat conjunctival cell cultures were treated with increasing concentrations of MIM-D3 and evaluated for glycocon…

The selective 5-HT6 receptor antagonist Ro4368554 restores memory performance in cholinergic and serotonergic models of memory deficiency in the rat.

Antagonists at serotonin type 6 (5-HT(6)) receptors show activity in models of learning and memory. Although the underlying mechanism(s) are not well understood, these effects may involve an increase in acetylcholine (ACh) levels. The present study sought to characterize the cognitive-enhancing effects of the 5-HT(6) antagonist Ro4368554 (3-benzenesulfonyl-7-(4-methyl-piperazin-1-yl)1H-indole) in a rat object recognition task employing a cholinergic (scopolamine pretreatment) and a serotonergic- (tryptophan (TRP) depletion) deficient model, and compared its pattern of action with that of the a…

Chronic haloperidol produces a time- and dose-related slowing of lick rhythm in rats: implications for rodent models of tardive dyskinesia and neuroleptic-induced parkinsonism.

In order to characterize the development of orolingual motor effects of chronic haloperidol treatment in rats, this typical neuroleptic was administered for 102 days while daily measurements of tongue movement dynamics (peak force, lick rhythm, number of licks) during water licking were recorded. After chronic haloperidol dosing (vehicle, 0.06. 0.12, 0.24 mg/kg for four separate groups) for 32 days and continuing every second or third day of the chronic dosing period, the effects of cholinergic (scopolamine: 0.05-0.20 mg/kg; trihexyphenidyl: 0.15-1.0 mg/kg) or serotonergic (ritanserin: 0.5-4.0…

Correlation of brain levels of 9-amino-1,2,3,4-tetrahydroacridine (THA) with neurochemical and behavioral changes.

9-Amino-1,2,3,4-tetrahydroacridine (THA) has been reported to cause improvement in patients with senile dementia of the Alzheimer's type. We have examined some effects of THA in vitro and in vivo to define its mechanism of action. In vitro, THA inhibits acetylcholinesterase (AChE) (IC50 = 223 nM) and blocks [3H]AFDX-116 (M2) and [3H]telenzepine (M1) binding (IC50 s of 1.5 and 9.1 microM respectively). In vivo levels of THA were 10-fold higher in brain than plasma following 3.2 mg/kg i.p., a dose which was found to be active in reversing amnesia induced by scopolamine assessed in T-maze tests i…

Methylscopolamine and conditioned location avoidance.

On alternate days, rats were confined to one side of a shuttlebox following IP administration of saline and to the other following the peripherally-acting muscarinic antagonist, methylscopolamine (1.2 mg/kg). They later avoided the side associated with the drug effect. By duplicating an earlier finding with centrally- and peripherally-acting scopolamine, this result identified aversive peripheral actions of the two drugs as mainly responsible for the effects observed.

[Effectiveness of the preparation Gavinton in preventing motion sickness].

The effectiveness of the Hungarian drug kavinton used to prevent motion sickness was assessed. During the study 8 motion sickness susceptible test subjects were kept in a chamber rotating at a rate of 6 rpm for 5 hours. The effectiveness of the drug taken regularly during the exposure was compared with that of scopolamine and placebo taken as a single dose. The results obtained are suggestive of a positive effect of kavinton as an antimotion drug.

Uropharmacology: part VI. Parasympathetic depressants.

A discussion of the various parasympathetic depressants is presented. Belladonna alkaloids include atropine, the prototype, and scopolamine. Synthetic drugs include quaternary ammonium compounds with antimuscarinic activity such as methantheline, propantheline, and other drugs such as isopropamide, pipenzolate methylbromide, and diphemanil methylsulfate. A miscellaneous class of drugs such as hemicholinium, valethamate, and oxybutynin chloride also possesses parasympathetic depressant activity. The pharmacologic properties and clinical usage of these drugs are discussed.

[The adequacy of a new method for assessing the vestibular protective effect of biologically active substances].

The effects of some neuropsychotropic agents on changes in pain sensitivity in albino male rats under the action of complex accelerations were investigated. It was found that administration of drugs with the antimotion-sickness activity--scopolamine, phenamine, phenibut prevented the development of analgesia with the opiate component, the use of low-effective and non-effective with respect to suppression of vestibulo-vegetative disturbances agents--diazepam, ephedrine, haloperidol failed to exert such an effect.

Anticholinergic behavioral effect of phencyclidine.

Phencyclidine (PCP) impaired spatial alternation performance in rats. This effects was mimicked by antimuscarinic anticholinergics (scopolamine, atropine) and PCP derivatives (ketamine, cyclohexamine) but not by a variety of other agents. Muscarinic cholinergic agonists antagonized PCP. Impairment of spatial alteration performance by PCP appears to be mediated, at least in part, by and anticholinergic action; this is the first instance of a behavioral effect of PCP that can be largely attributed to a specific mechanism.

The effects of some drugs on the caloric induced nystagmus.

A striking difference between the two anticholinergic drugs was observed. Atropine only modified the eye speed of the slow phase (mes), whereas scopolamine modified both parameters measured, mes and duration. Amphetamine and caffeine prolonged the duration, leaving the mes unchanged. All antiemetic drugs inhibited both parameters. It is concluded that the effect of a drug on the caloric reaction gives insufficient information in the preclinical evaluation of vestibulo-suppressive drugs.

Inhibiting cholinergic signalling in the cerebellar interpositus nucleus impairs motor behaviour.

The role of neuromodulators in the cerebellum is not well understood. In particular, the behavioural significance of the cholinergic system in the cerebellum is unknown. To investigate the importance of cerebellar cholinergic signalling in behaviour, we infused acetylcholine receptor antagonists, scopolamine and mecamylamine, bilaterally into the rat cerebellum (centred on interpositus nucleus) and observed the motor effects through a battery of behavioural tests. These tests included unrewarded behaviour during open field exploration and a horizontal ladder walking task and reward-based beam …
