Seminal fluid promotes in vitro sperm-oviduct binding in the domestic cat (Felis catus).

From many endangered or threatened species which are expected to profit from assisted reproduction techniques, mainly epididymal sperm of dead or freshly castrated males are available. These sperm had contact to epididymal secretion products but not to seminal fluid components. Notably, products of accessory sex glands have been shown in domestic animals to condition sperm for fertilization, in particular by mediating sperm-oviduct interaction. We report for the first time that motile epididymal sperm from domestic cats are able to bind to fresh oviduct epithelial cell explants from preovulato

The effects of type I collagenase on the degelification of chimpanzee (Pan troglodytes) semen plug and sperm quality.

BACKGROUND: Semen from the chimpanzee species becomes a colloidal solid after ejaculation. The formation of this copulatory plug is believed to prevent additional spermatozoa of subsequent mating events from accessing the ova. However, this naturally preserved strategy hampers the processes for sperm preparation. In this study, we investigated whether collagenase can be used to degelify the semen plug and accelerate the semen liquefaction process in zoo captive chimpanzee species (Pan troglodytes). RESULTS: We showed that incubation of chimpanzee ejaculates with 0.1% type I collagenase effici

Epididymosomes: transfer of fertility-modulating proteins to the sperm surface.

A variety of glycosylphosphatidylinositol (GPI)-linked proteins are acquired on spermatozoa from epididymal luminal fluids (ELF) during sperm maturation. These proteins serve roles in immunoprotection and in key steps of fertilization such as capacitation, acrosomal exocytosis and sperm-egg interactions. Their acquisition on sperm cells is mediated both by membrane vesicles (epididymosomes, EP) which were first reported to dock on the sperm surface, and by lipid carriers which facilitate the transfer of proteins associated with the membrane-free fraction of ELF. While the nonvesicular fraction

Sperm cryopreservation in oncological patients: a 14-year follow-up study.

OBJECTIVE: Oncologic treatments can destroy spermatogenic dividing cells and cause azoospermia which could be irreversible. Sperm banking is the best option to preserve male fertility after these treatments. It is easy, inexpensive, and safe. To date, few clinical data are available about large series of cancer patients. Our objective was to determine the usefulness of these preventive sperm freezing protocols. DESIGN: Prospective study. SETTING: University-affiliated private fertility center. PATIENT(S): One hundred eighty-six cancer patients who banked sperm samples at our center before s

Hedgehog signalling in solitary motile cilia: detection and role of hedgehog factors in sperm functions and in vitro fertilization.

IN BRIEF: Systemic blockade of Hedgehog signalling reduces sperm motility without knowing whether this effect is direct or indirect on spermatozoa. This study demonstrates the presence of members of the Hedgehog signalling pathway in spermatozoa and the direct effects of activator or inhibitor of this pathway on sperm function. ABSTRACT: The Hedgehog (Hh) signalling pathway controls cellular functions important to organ development and homeostasis. While previously thought to be restricted to non-motile primary cilia, a study indicated that Hh modulates cAMP concentration and beating frequenc

Comparison of intrabursal transfer of spermatozoa, a new method for artificial insemination in mice, with intraoviductal transfer of spermatozoa.

PURPOSE: The objective of this paper was to compare the in vivo fertilizing abilities of fresh epididymal spermatozoa with a new method of artificial insemination in mice, so-called "intrabursal transfer of spermatozoa (ITS)," which requires transfer of spermatozoa into a space near the infundibulum between the ovary and ovarian bursa of superovulated females, and the previous method, so-called "intraoviductal transfer of spermatozoa (IOTS)," especially as regards sperm number and capacitation. METHODS: Spermatozoa freshly isolated from B6C3F1 males were injected into superovulated B6C3F1 fem

Efficient and repeatable in vitro fertilization in rabbits.

Rabbits constitute an interesting model to understand gamete interaction and test novel Artificial Reproductive Techniques, but in vitro fertilization (IVF) is particularly problematic in this species. We have conducted a series of experiments to develop a consistent IVF technique. Initially, we checked viability, acrosome integrity, capacitation and motility in ejaculated sperm purified by a density gradient and incubated at different times in three different media: Tyrode's Albumin Lactate Pyruvate (TALP), human tubal fluid (HTF), and Brackett and Oliphant (BO). Proteomic analysis of iTRAQ in melatonin-treated sheep epididymal epithelial cells.

During maturation, spermatozoa acquire motility and fertilizing capacity as they transit through the epididymis. Melatonin is a lipophilic hormone with multiple functions in regulating the fertility. Previous studies have shown that melatonin affected the capacitation or maturation of sperm in the epididymis. The aim of this study was to investigate the effects of melatonin on epididymal caput epithelial cells in sheep. In the study, we used iTRAQ labelling coupled with LC-MS/MS for quantitative identification of differentially expressed proteins in melatonin-treated sheep epididymal caput epi

Reproductive pathology and sperm physiology in acid sphingomyelinase-deficient mice.

Types A and B Niemann-Pick disease (NPD) are lysosomal storage disorders resulting from the deficient activity of acid sphingomyelinase (ASM). In this manuscript we report the pathobiology of male gonadal tissue and sperm in a knockout mouse model of NPD and demonstrate the importance of ASM for normal sperm maturation and function. Characteristic lipid-filled vacuoles were evident in light micrographs of testis' seminiferous tubules and epithelial cells lining the epididymis of -/- mice. Electron micrographs extended these findings and revealed storage vesicles within Sertoli cells of the sem

Oxidative stress is one of the few defined causes of male infertility affecting at least one third of patients attending infertility clinics … tion because their biology is heavily dependent on redox regulation. ROS are important mediators of sperm capacitation, stimulating the generation of cAMP and prostaglandins, inhibiting protein phosphatases and encouraging removal of cholesterol from the plasma membrane. Furthermore, during fertilization, the ability of ROS to activate metalloproteinases facilitates …

ProAKAP4 as a potential biomarker of fertility and sperm freezability in males of different species - A review.

In recent years, special attention has been paid to proAKAP4, the precursor of A-kinase anchoring protein 4 (AKAP4), which plays a crucial role in the structure and function of the sperm flagellum. This testis-specific protein forms the fibrous sheath and local signal transduction required for proper sperm capacitation and motility. Highly conserved across species, proAKAP4 levels are strongly correlated with fertility and sperm survivability post-thaw. This review summarizes the biological roles of proAKAP4 and its potential as a semen quality biomarker in various mammalian species, including…

Mutations in the prostate specific antigen (PSA/KLK3) correlate with male infertility.

Prostate specific antigen (PSA/KLK3) is known to be the chief executor of the fragmentation of semenogelins, dissolution of semen coagulum, thereby releasing sperm for active motility. Recent research has found that semenogelins also play significant roles in sperm fertility by affecting hyaluronidase activity, capacitation and motility, thereby making PSA important for sperm fertility beyond simple semen liquefaction. PSA level in semen has been shown to correlate with sperm motility, suggesting that PSA level/activity can affect fertility. However, no study investigating the genetic variatio

Mammalian glutathione peroxidases control acquisition and maintenance of spermatozoa integrity.

In mammals, posttesticular epididymal sperm maturation is considered an essential step in the transformation of immature testicular gametes to mature spermatozoa capable of fertilization. Reactive oxygen species (ROS) have been shown to be key actors in this maturation process, and it is now clear that ROS are central for sperm physiology in processes such as sperm maturation and capacitation. However, during epididymal maturation and storage and until the onset of fertilization, oxidative damage is a threat spermatozoa must face more than any other cells. Spermatozoa were found to be extremel

Binding of arylsulfatase A to mouse sperm inhibits gamete interaction and induces the acrosome reaction.

We have shown previously that male germ cell-specific sulfoglycolipid, sulfogalactosylglycerolipid (SGG), is involved in sperm-zona pellucida binding, and that SGG and its desulfating enzyme, arylsulfatase A (AS-A), coexist in the same sperm head area. However, AS-A exists at a markedly low level in sperm as compared to SGG (i.e., 1/400 of SGG molar concentration). In the present study, we investigated whether perturbation of this molar ratio would interfere with sperm-egg interaction. We demonstrated that purified AS-A bound to the mouse sperm surface through its high affinity with SGG. When

Properties of human epididymal sperm obtained from an alloplastic spermatocele: motility assessment and penetration of zona-free hamster oocytes in the presence and absence of caffeine.

Sperm were collected over a period of months from a human alloplastic spermatocele implanted at the corpus/caudal epididymal junction and were evaluated for their maturity, motility, and ability to capacitate and acrosome react, as assessed by the hamster zona-free oocyte sperm penetration assay (SPA). A mean of 11 X 10(6) sperm were obtained with each aspiration, with 34% to 40% being mature, normal forms. Motility was poor; 15% +/- 5% showing nonprogressive movement. SPA results were 22% +/- 3% oocyte penetration. Addition of 7.5 mM caffeine markedly enhanced motility and improved the SPA re

Human spermatozoa and temperature: the elephant in the room.

Men are notable for low sperm production, relative to that of other large mammals, and often inferior morphology and motility of their spermatozoa. The extent to which temperature plays a role in this picture has been a moot point. However, animal experiments suggest that an increased scrotal temperature of approximately +4°C brought by inguinal clothing has a negative impact on the germinal epithelium and on the epididymis in man. In two animal species with inguinal testes, their transposition to the abdomen, raising the testis temperature by a modest approximate 1.5°C brought reduced sperm p
