Sperm binding glycoprotein is differentially present surrounding the lumen of isthmus and ampulla of the pig's oviduct.

In several mammals a sperm reservoir is formed at the isthmus of the Fallopian tube, providing viable, potentially fertile sperm for an extensive period. In pig (Sus scrofa) the spermadhesin AQN-1 seems to be involved in the establishment of the sperm reservoir. The pig oviductal protein, sperm binding glycoprotein (SBG), binds to sperm and exposes carbohydrate groups that can be recognized by AQN-1. In this study we obtain anti-SBG polyclonal antibodies and use them to localize SBG in the oviduct. Immunohistochemical analysis shows that SBG is present at the apical surface of isthmic and ampu…

Diagnosis and epidemiology of Trichinella infections in wildlife in The Netherlands.

Trichinella infections in foxes and wild boars were studied to determine the prevalence of infection in wildlife in the Netherlands. Muscles of 429 forelegs of foxes (Vulpes vulpes) and diaphragms of 11 wild boars (Sus scrofa) were artificially digested. Single larvae of Trichinella were identified at species level using random amplified polymorphic DNA (RAPD-PCR). In addition, an ELISA using ES antigen was used to test serum samples of 458 wild boars. The prevalence of Trichinella in foxes ranged from 3.9% in the the eastern part of the country, 13.1% in the central part of the country and 1.…

Integrative genotype-environment association and single-cell transcriptomic analyses identify candidate mechanisms of environmental adaptation in pigs.

As one of the earliest domesticated and most globally ubiquitous livestock, pigs (Sus scrofa) have undergone profound environmental adaptation while following human expansion across diverse bioclimatic zones. Despite their evolutionary significance, the genomic mechanisms enabling pigs to thrive in disparate ecological settings remain poorly understood. Here, we present a comprehensive global analysis using a dataset of 1,151 pig genome sequences representing 146 distinct populations. By integrating three complementary genotype-environment association (GEA) frameworks including Redundancy Anal…

Capacitation-Induced Zinc Ion Flux and Sperm Plasma Membrane Remodeling Predict Porcine In Vitro Fertilization Cleavage Success.

Semen evaluation in human and animal reproduction relies on sperm motility and morphology; however, these often fail to predict fertility. The domestic boar (Sus scrofa) serves as a biomedical model for male reproduction due to similarities in sperm size, capacitation dynamics, and acrosomal structure to humans in comparison to traditional rodent models. This study evaluated sperm capacitation biomarkers, particularly zinc signatures, to predict cleavage success after in vitro fertilization (IVF). Semen from 20 boars (3 replicates each) was analyzed at 0, 1 and 4 h post-in vitro capacitation (…

Domestication-Driven Expansion and Structural Convergence of the Porcine Antiviral Interferon Repertoire.

The porcine interferon (IFN) system is highly diversified, particularly within Type I subfamilies, yet its evolutionary trajectory across domestication and breed formation remains poorly characterized. We performed a comprehensive comparative genomic and structural analysis of 432 IFN sequences spanning all IFN types across 11 Sus scrofa breeds representing commercial, indigenous, and wild/outgroup lineages. Phylogenetic reconstruction, pairwise dN/dS selection pressure analysis, and AlphaFold2-based 3D structure prediction coupled with DALI structural similarity mapping were integrated to res…

Differential prevalence of PFAS, PCBs and pesticides in liver of hunted game.

Free foraging game animals are constantly exposed to environmental pollutants and can therefore serve as bioindicators for these substances. However, they can also pose a health risk to humans if polluted meat of wild game animals enters the food chain. We analysed 219 potentially critical pollutants in 164 liver samples of hunted wild boar (Sus scrofa) and roe deer (Capreolus capreolus) from north-eastern Germany. Overall, 24 pollutants were detected and 16 of them were detected quantitatively. PCBs, PFAS and rodenticides were more frequent and present in far higher concentrations in wild boa…

Comparison of Immunoprotective Efficacy of Six Antigenic Proteins of Pasteurella multocida Serotype a in KM Mice (Mus musculus).

Pasteurella multocida serotype A (P. multocida) is frequently associated with severe respiratory disease in swine (Sus scrofa), highlighting the need for effective preventive strategies. To identify protective antigens suitable for a subunit vaccine targeting porcine P. multocida infection, six recombinant proteins (rAspA, rLolA, rOmpP6, rOppA, rRps6, rSmpA) were expressed in a prokaryotic system, and their efficacy was evaluated in a Mus musculus (Kunming) mouse model. All proteins were purified using His-tag affinity chromatography, and SDS-PAGE analysis confirmed expression with bands at th…

Gastrointestinal parasites circulating in wild boars from central Italy and molecular characterization of Balantioides coli, Blastocystis sp. and Enterocytozoon bieneusi.

The growing abundance and geographic spread of wild boar (Sus scrofa) populations in Europe have heightened concerns about their contribution to the transmission of parasites of zoonotic and veterinary importance. This study investigated the occurrence and zoonotic potential of gastrointestinal parasites in 46 wild boars sampled during the 2023 hunting season in Italy. Copromicroscopic analyses revealed that 47.8% of animals were positive for at least one parasite taxon, with frequent coinfections. Thirteen taxa were identified by microscopy, including protozoans (Entamoeba coli, Iodamoeba but…

Streptococcus suis in free-ranging wild boars from Southern Brazil: A preliminary survey.

Streptococcus suis (S. suis) is a pathogen associated with pigs and human meningitis. Previous studies indicate wild boars (Sus scrofa) as a S. suis reservoir; however, in Brazil this has not yet been assessed in free-ranging wild boars. Therefore, this study aimed to identify the frequency of S. suis in wild boar samples from South Brazil. For that, convenient lung and lymph node samples collected during 2013, 2014, 2015, and 2019 of free-ranging wild boars from Rio Grande do Sul, South Brazil were evaluated. The detection of S. suis was based on the presence of gdh (glutamate dehydrogenase) …

Embryonic signals mediate extracellular vesicle biogenesis and trafficking at the embryo-maternal interface.

BACKGROUND: Extracellular vesicles (EVs) are membrane-coated nanoparticles secreted by almost all cell types in living organisms. EVs, as paracrine mediators, are involved in intercellular communication, immune response, and several reproductive events, including the maintenance of pregnancy. Using a domestic animal model (Sus scrofa) with an epitheliochorial, superficial type of placentation, we focused on EV biogenesis pathway at the embryo-maternal interface, when the embryonic signaling occurs for maternal recognition and the maintenance of pregnancy. RESULTS: Transmission electron micros…

Adaptation of the Porcine Pituitary Transcriptome, Spliceosome and Editome during Early Pregnancy.

The physiological mechanisms of the porcine reproduction are relatively well-known. However, transcriptomic changes and the mechanisms accompanying transcription and translation processes in various reproductive organs, as well as their dependence on hormonal status, are still poorly understood. The aim of this study was to gain a principal understanding of alterations within the transcriptome, spliceosome and editome occurring in the pituitary of the domestic pig (Sus scrofa domestica L.), which controls basic physiological processes in the reproductive system. In this investigation, we perfo…

Feral Swine as Indirect Indicators of Environmental Anthrax Contamination and Potential Mechanical Vectors of Infectious Spores.

Anthrax is a disease that affects livestock, wildlife, and humans worldwide; however, its relative impacts on these populations remain underappreciated. Feral swine (Sus scrofa) are relatively resistant to developing anthrax, and past serosurveys have alluded to their utility as sentinels, yet empirical data to support this are lacking. Moreover, whether feral swine may assist in the dissemination of infectious spores is unknown. To address these knowledge gaps, we intranasally inoculated 15 feral swine with varying quantities of Bacillus anthracis Sterne 34F2 spores and measured the seroconve…

Early steps of embryo implantation are regulated by exchange of extracellular vesicles between the embryo and the endometrium.

In early pregnancy, as the embryo arrives in the uterus, intensive communication between the embryo and uterus begins. Hundreds of molecules are known to be involved, but despite numerous findings, full understanding of the complexity of the embryo-maternal dialog remains elusive. Recently, extracellular vesicles, nanoparticles able to transfer functionally active cargo between cells, have emerged as important players in cell-cell communication, and as such, they have gained great attention over the past decade also in reproductive biology. Here, we use a domestic animal model (Sus scrofa) wit…

Formaldehyde exposure induces differentiation of regulatory T cells via the NFAT-mediated T cell receptor signalling pathway in Yucatan minipigs.

The use of minipigs (Sus scrofa) as a platform for toxicological and pharmacological research is well established. In the present study, we investigated the effect of formaldehyde (FA) exposure on helper T cell-mediated splenic immune responses in Yucatan minipigs. The minipigs were exposed to different inhaled concentrations of FA (0, 2.16, 4.62, or 10.48 mg/m3) for a period of 2 weeks. Immune responses elicited by exposure to FA were determined by assessing physiological parameters, mRNA expression, and cytokine production. Additionally, the distribution of helper T cells and regulatory T (T…
