Increased alcohol drinking and seizure susceptibility in female humanized ApoE4 knockin rats.

Epidemiological analyses suggest that the Ɛ4 allele of apolipoprotein E (ApoE) genes may influence the effects of alcohol on cognitive and executive function and dementia risk compared to the Ɛ3 allele. Here, we investigated this question in female rats given that women are more vulnerable than men to the Ɛ4 genotype effects on various diseases. Experiment 1 examined the effects of alcohol drinking on performance in a Barnes maze and an operant strategy set-shifting (OSS) task during abstinence in wildtype (WT) and homozygous ApoE4 knock-in (E4) rats. Experiment 2 repeated the behavioral asses

Unifying network model links recency and central tendency biases in working memory.

The central tendency bias, or contraction bias, is a phenomenon where the judgment of the magnitude of items held in working memory appears to be biased toward the average of past observations. It is assumed to be an optimal strategy by the brain and commonly thought of as an expression of the brain's ability to learn the statistical structure of sensory input. On the other hand, recency biases such as serial dependence are also commonly observed and are thought to reflect the content of working memory. Recent results from an auditory delayed comparison task in rats suggest that both biases ma

Visuospatial working memory capacity moderates the relationship between anxiety and OCD related checking behaviors.

BACKGROUND: Compulsive checking behavior is the most prevalent compulsive behavior in patients with obsessive-compulsive disorder (OCD). While some studies have shown that anxiety and executive function influence compulsive checking behavior, the relationship between these constructs is inconclusive. Hence, we sought to explore the interplay between executive function, anxiety and compulsive checking behavior. MATERIALS AND METHODS: 47 healthy participants (HC) and 51 patients with OCD participated in the study. Symptoms and emotional states were assessed using the Yale-Brown Obsessive Compul

Impulsivity is related to overhasty risk learning in attention-deficit/hyperactivity disorder: A computational psychiatric approach.

Attention-deficit/hyperactivity disorder (ADHD) is often accompanied by excessive risky behavior, and an impulsive trait has been proposed to be associated with risk-taking. However, the aspect of the cognitive process that impulsivity influences is not well understood. We hypothesized that impulsivity could be related to an overhasty shifting of beliefs during risk learning, thereby resulting in enhanced risk-taking behavior. In this study, we tested our hypothesis using the Bayesian modeling approach and predicted overhasty learning by a data-driven approach. We used an openly available task

Impaired Cognitive Function after Perineuronal Net Degradation in the Medial Prefrontal Cortex.

Perineuronal nets (PNNs) are highly organized components of the extracellular matrix that surround a subset of mature neurons in the CNS. These structures play a critical role in regulating neuronal plasticity, particularly during neurodevelopment. Consistent with this role, their presence is associated with functional and structural stability of the neurons they ensheath. A loss of PNNs in the prefrontal cortex (PFC) has been suggested to contribute to cognitive impairment in disorders such as schizophrenia. However, the direct consequences of PNN loss in medial PFC (mPFC) on cognition has no

Sex Differences in μ-Opioid Receptor Regulation of the Rat Locus Coeruleus and Their Cognitive Consequences.

Stress-related neuropsychiatric pathologies are more prevalent in females compared with males. An important component of the stress response is activation of the locus coeruleus (LC)-norepinephrine system. Because LC activation is tempered by endogenous opioid release during stress, the magnitude of opioid regulation of the LC could determine stress vulnerability. Here we report convergent evidence for decreased μ-opioid receptor (MOR) function in the female rat LC. The selective MOR agonist, DAMGO (10 pg), completely inhibited LC discharge of male but not female rats and DAMGO (30 pg) produce

The left parietal cortex and motor attention.

The posterior parietal cortex, particularly in the right hemisphere, is crucially important for covert orienting; lesions impair the ability to disengage the focus of covert orienting attention from one potential saccade target to another (Posner, M. I. et al., Journal of Neuroscience, 1984, 4, 1863-1874). We have developed a task where precues allow subjects to covertly prepare subsequent cued hand movements, as opposed to an orienting or eye movement. We refer to this process as motor attention to distinguish it from orienting attention. Nine subjects with lesions that included the left pari

Is impaired set-shifting a feature of "pure" anorexia nervosa? Investigating the role of depression in set-shifting ability in anorexia nervosa and unipolar depression.

Impaired set-shifting has been reported in patients with anorexia nervosa (AN) and in patients with affective disorders, including major depression. Due to the prevalent comorbidity of major depression in AN, this study aimed to examine the role of depression in set-shifting ability. Fifteen patients with AN without a current comorbid depression, 20 patients with unipolar depression (UD) and 35 healthy control participants were assessed using the Trail Making Test (TMT), the Wisconsin Card Sorting Test (WCST) and a Parametric Go/No-Go Test (PGNG). Set-shifting ability was intact in patients wi

Contributions of cortical neuron firing patterns, synaptic connectivity, and plasticity to task performance.

Neuronal responses during behavior are diverse, ranging from highly reliable 'classical' responses to irregular 'non-classically responsive' firing. While a continuum of response properties is observed across neural systems, little is known about the synaptic origins and contributions of diverse responses to network function, perception, and behavior. To capture the heterogeneous responses measured from auditory cortex of rodents performing a frequency recognition task, we use a novel task-performing spiking recurrent neural network incorporating spike-timing-dependent plasticity. Reliable and

Use of prism adaptation in children with unilateral brain lesion: Is it feasible?

INTRODUCTION: Unilateral visuospatial deficits have been observed in children with brain damage. While the effectiveness of prism adaptation for treating unilateral neglect in adult stroke patients has been demonstrated previously, the usefulness of prism adaptation in a pediatric population is still unknown. The present study aims at evaluating the feasibility of prism adaptation in children with unilateral brain lesion and comparing the validity of a game procedure designed for child-friendly paediatric intervention, with the ecological task used for prism adaptation in adult patients. Dopamine receptors regulate preference between high-effort and high-risk rewards.

RATIONALE: Optimal decision-making necessitates evaluation of multiple rewards that are each offset by distinct costs, such as high effort requirement or high risk of failure. The neurotransmitter dopamine is fundamental toward these cost-benefit analyses, and D1-like and D2-like dopamine receptors differently modulate the reward-discounting effects of both effort and risk. However, measuring the role of dopamine in regulating decision-making between options associated with distinct costs exceeds the scope of traditional rodent economic decision-making paradigms. OBJECTIVES: We developed the

Effects of inpatient treatment of adolescents with anorexia nervosa are associated with body representation: a monocentric pilot study.

This monocentric pilot study investigates the relationship between nutritional treatment and body representation distortion in adolescents with Anorexia Nervosa (AN) using the Rubber Hand Illusion (RHI) task. A total of 28 adolescents participated, including 14 patients with restrictive AN and 14 healthy controls. The RHI and Robot Hand Illusion (RoHI) tasks were conducted, with assessments taken at admission and discharge for the AN group. Sense of Ownership (SO) and Sense of Agency (SA) rated via 7-item Likert; proprioceptive drift was millimetre shift in perceived hand position after stimul

Patterns of GP and nurse independent prescriber prescriptions for antibiotics dispensed in the community in England: a retrospective analysis.

BACKGROUND: Physician-nurse task shifting, a process of delegation whereby tasks are moved to other specialized healthcare workers, is used in primary care in many countries to improve access, efficiency and quality of care. One such task is the prescription of medicines. OBJECTIVES: To identify nurse independent prescriber (NIP) and GP numbers in England, the proportions and types of NIP and GP antibiotic prescriptions dispensed in the community, and the impact of COVID-19 on the volume, rate and types of antibiotic prescriptions dispensed. METHODS: Descriptive population-based retrospectiv…

Improving power in functional magnetic resonance imaging by moving beyond cluster-level inference.

Inference in neuroimaging typically occurs at the level of focal brain areas or circuits. Yet, increasingly, well-powered studies paint a much richer picture of broad-scale effects distributed throughout the brain, suggesting that many focal reports may only reflect the tip of the iceberg of underlying effects. How focal versus broad-scale perspectives influence the inferences we make has not yet been comprehensively evaluated using real data. Here, we compare sensitivity and specificity across procedures representing multiple levels of inference using an empirical benchmarking procedure that

Group cognitive remediation therapy for children and adolescents in intensive day-patient treatment for anorexia nervosa: a feasibility study.

BACKGROUND: Cognitive Remediation Therapy (CRT) is an adjunct treatment targeting set-shifting (SS) and weak central coherence (CC) thought to play a role in maintaining anorexia nervosa (AN). This study aimed to test the feasibility of brief group CRT for young people with AN in a day-patient setting using neuropsychological assessment of SS and CC. It also explored the benefit for young people with Autism Spectrum Disorder (ASD) features. METHODS: Twenty female day-patients (12-18 years) with AN or Atypical AN completed neuropsychological assessment of SS (using the Wisconsin Card Sorting T

Magnetoencephalographic (MEG) brain activity during a mental flexibility task suggests some shared neurobiology in children with neurodevelopmental disorders.

BACKGROUND: Children with neurodevelopmental disorders (NDDs) exhibit a shared phenotype that involves executive dysfunctions including impairments in mental flexibility (MF). It is of interest to understand if this phenotype stems from some shared neurobiology. METHODS: To investigate this possibility, we used magnetoencephalography (MEG) neuroimaging to compare brain activity in children (n = 88; 8-15 years) with autism spectrum disorders (ASD), attention deficit hyperactivity disorder (ADHD) and obsessive-compulsive disorder (OCD), as they completed a set-shifting/mental flexibility task.

Whole body organization during a symmetric bimanual pick up task in overweight and obese children.

Information on the effects of obesity on the biomechanics of whole body movement control in children is limited. The purpose of the current study is to test the hypothesis that during a simple pick up task, overweight and obese children will organize their whole body movements differently than those in normal weight children. Twelve children who were overweight or obese (5-13 years old) and twelve age matched normal weight children participated in the study. Children picked up an empty box to waist height at a self-selected pace while kinematic and kinetic data were recorded and analyzed using
