The influences of hypothyroidism on behavioral measures of the taste function in male and female Long-Evans rats were determined. Experimental rats' preferences for and ability to detect NaCl, HCl, sucrose, and quinine sulfate were examined before, during, and after 9 weeks of maintenance on 0.1% propylthiouracil (PTU), an agent that produces marked hypothyroidism, with similar determinations made for control animals. Despite significant decreases in PTU-treated rats' serum triiodothyronine (T3) and thyroxin (T4), there were no changes in sensitivity or responsivity to the target tastants. How…

Receptive fields (RFs) of 36 taste (the 22 parabrachio-thalamic relay (P-T) and 14 non-P-T) and 23 mechanoreceptive neurons (7 P-T and 16 non-P-T) were located in the oral cavity of rats. All of the taste and most of the mechanoreceptive units examined had an RF on the ipsilateral side of the tongue or palate, but some mechanoreceptive P-T and non-P-T units had RFs bilaterally. When the RFs of taste neurons were examined with the most effective of the four basic taste (the best stimulus) and non-best stimuli, no difference was noticed in the location of RFs between the P-T and non-P-T neurons.…

BACKGROUND: Cephalic phase responses (CPRs) are anticipatory and conditioned responses elicited by the stimulation of food-related cues in the oral and nasal cavities. By initiating autonomic and endocrine reactions before absorption of nutrients, CPRs can blunt acute-postprandial hyperglycemia. Taste-related information is critical for triggering CPRs, but the neural mechanisms underlying taste-driven CPRs remain incompletely elucidated.
HIGHLIGHT: Current evidence supports a model in which taste-driven CPRs arise from coordinated processing across peripheral taste receptors, the rostral com…

An animal's ability to taste is critical for nutrient selection and rejection of potentially toxic substances. Factors such as age, diet, environment, and internal state can also influence food-related behavioral outcomes. One such factor-exposure to taste cues during early development-remains poorly understood. Here, we evaluate how larval taste input affects adult feeding preference in Drosophila melanogaster. We find that larval dietary exposure to papaverine, a bitter alkaloid, attenuates feeding avoidance of papaverine in adult flies. This behavioral modification appears compound specific…

BACKGROUND: Adrenal insufficiency often presents with nonspecific symptoms, impeding early diagnosis. Taste and smell disturbances are rarely recognized as presenting features.
CASE PRESENTATION: We report the case of a 54-year-old man with a 3-month history of persistent taste and smell disturbances accompanied by fatigue, loss of appetite, weight loss, and diffuse body pain, including thigh stiffness. Endocrine evaluation with stimulation tests revealed markedly low levels of cortisol and adrenocorticotropic hormone, leading to a diagnosis of secondary adrenal insufficiency. After glucocort…

Taste disorders and malnutrition are both age-related conditions that, individually, may have negative consequences but which are also probably associated with each other, making the consequences even worse. This scoping review was performed to systematically map and synthesize the existing evidence on the association between taste disorders and malnutrition in older adults. Four electronic literature databases (PubMed, Embase, CINAHL, and Web of Science) were searched. Title and abstract screening as well as full-text screening was performed by 2 authors independently. Studies of older adults…

V. volvacea is an easy-to-cultivate short-cycle edible mushroom that is rich in protein and high-quality peptides. The V. volvacea peptides, hydrolyzed by alcalase, showed an umami response value of 15.51 ± 0.11 and a salty response value of 4.64 ± 0.27 after the Maillard reaction at 120 °C via electronic tongue. The fluorescence intensity initially increased and subsequently decreased as reaction temperatures rose, and a substantial formation of browning products was observed beyond 120 °C, resulting in a diminished taste perception. High temperatures enhanced the degradation of Maillard inte…

This study investigated the impact of cold water with increasing temperature (CI) and boiling water with constant temperature (BC) steaming on the metabolites and taste of Eriocheir sinensis. Sensory evaluation and electronic tongue analysis indicated that CI group enhanced ovaries umami, whereas BC group increased muscle umami (p < 0.05). Widely targeted metabolomics showed impacts on amino acid and energy metabolism. CI group significantly increased total amount of free amino acids and nucleotides. Enzyme activity experiments confirmed that cold water with increasing temperature augmented ad…

Exposure to environmental enrichment (EE) has demonstrated a remarkable ability to modulate cognitive processes. These effects have been associated with brain-derived neurotrophic factor (BDNF), a protein recognized for regulating synaptic plasticity in the adult brain. Previous research from our group revealed that exposure to EE prior to the acquisition of a strong form of conditioned taste aversion (CTA) weakens the aversive response by restoring BDNF levels in the insular cortex (IC), a region in the temporal lobe involved in multimodal sensory integration and learning and memory processes…

Replenishment of taste cells in taste buds throughout life requires innervation. Recently, we provided evidence that R-spondin is sufficient to promote taste cell generation and restore taste buds in a nerve transection model. Yet, the necessity of gustatory-neuron-supplied R-spondin 2 (RSPO2) in taste tissue homeostasis has remained unresolved. We used genetic approaches to investigate this. In a strain that carries an Rspo2 hypomorphic allele, the number of taste buds was significantly reduced in these mice, compared to wild-type mice, in both anterior and posterior tongue. Specific ablation…

The sense of taste generally shows diminishing sensitivity to prolonged sweet stimuli, referred to as sweet adaptation. Yet, its mechanistic landscape remains incomplete. Here, we report that glia-like type I cells provide a distinct mode of sweet adaptation via intercellular crosstalk with chemosensory type II cells. Using the microfluidic-based intravital tongue imaging system, we found that sweet adaptation is facilitated along the synaptic transduction from type II cells to gustatory afferent nerves, while type I cells display temporally delayed and prolonged activities. We identified that…

Prevention and treatment protocols for taste changes observed during hematopoietic cell transplantation (HCT) are not well-established. The purpose of this study was to assess the efficacy of photobiomodulation (PBM) in relieving taste changes and preventing lingual papillae atrophy. HCT patients received PBM (n = 42) on the tongue dorsum using an InGaAIP laser (660 nm, 100 mW, 1.1 W/cm2, 8.8 J/cm2). During the HCT conditioning (T0), severe neutropenia (T1), and after neutrophil engraftment (T2), taste acuity for sweet, bitter, sour, and salty solutions, and clinical appearance of lingual papi…

BACKGROUND: The loss of smell and taste are some of the chronic complications that have impacted and continue to impact patients who suffered a COVID-19 infection. Impairment of smell and taste have significant impact on patients' lives. Current treatment modalities are very limited.
CASE REPORT: We present a 40-year-old patient who completely lost smell and taste following a COVID-19 infection. The patient was treated with bilateral stellate ganglion blocks (SGBs) where each side was blocked at separate visits. At the 6-month follow-up, taste and smell both improved by 90%.
CONCLUSIONS: SGB…

Taste buds on the tongue contain taste receptor cells (TRCs) that detect sweet, sour, salty, umami and bitter stimuli. Like non-taste lingual epithelium, TRCs are renewed from basal keratinocytes, many of which express the transcription factor SOX2. Genetic lineage tracing has shown that SOX2+ lingual progenitors give rise to both taste and non-taste lingual epithelium in the posterior circumvallate taste papilla (CVP) of mice. However, SOX2 is variably expressed among CVP epithelial cells, suggesting that their progenitor potential may vary. Using transcriptome analysis and organoid technolog…

So far, few studies have examined the effect of salt taste receptors genetic variation on dietary intake in the Iranian population. We aimed to evaluate associations between single nucleotide polymorphisms (SNPs) in salt taste receptors' genes with dietary salt intake and blood pressure. A cross-sectional study was carried out among 116 randomly selected healthy adults aged ≥ 18 in Isfahan, Iran. Participants underwent sodium intake determination by 24-h urine collection, as well as dietary assessment by semi-quantitative food frequency questionnaire and blood pressure measurement. Whole blood…

Western diet (WD), characterized by a high intake of fats and sugary drinks, is a risk factor for male reproductive impairment. However, the molecular mechanisms underlying this remain unclear. Taste receptor type 1 member 3 (TAS1R3), activated by ligands of WD, is highly expressed in extra-oral tissues, particularly in the testes. Here, we investigated to determine the effects of WD intake on male reproduction and whether TAS1R3 mediates WD-induced impairment in male reproduction. Male C57BL/6 J wild-type (WT) and Tas1r3 knockout (KO) mice were fed either a normal diet and plain water (ND) or…

Taste receptor cells are taste bud epithelial cells that are dependent upon the innervating nerve for continuous renewal and are maintained by resident tissue stem/progenitor cells. Transection of the innervating nerve causes degeneration of taste buds and taste receptor cells. However, a subset of the taste receptor cells is maintained without nerve contact after glossopharyngeal nerve transection in the circumvallate papilla in adult mice. Here, we revealed that injury caused by glossopharyngeal nerve transection triggers the remaining differentiated K8-positive taste receptor cells to dedif…
