Optimization of autologous adipose tissue transplantation: The impact of patient-specific and intraoperative factors on cellular viability before and after cryopreservation.

BACKGROUND: Autologous adipose tissue transplantation is a versatile surgical procedure used for aesthetic, reconstructive, and regenerative purposes. Cryopreservation of adipose tissue collected during liposuction reduces the need for repeated fat collection, minimizing patient discomfort while preserving volumetric and regenerative properties. There are currently no studies in the literature correlating cellular viability, before and after thawing, with patient-specific variables and collection techniques. This study investigates patient-specific factors affecting cellular viability before a

Autofluorescence Quenching in Decellularized Plant Scaffolds for Tissue Engineering.

PURPOSE: Autofluorescence in plant-derived scaffolds interferes with fluorescence imaging by overlapping with commonly used fluorophores such as Hoechst and FITC. This limits the ability to visualize cell behavior and scaffold integration in tissue engineering applications. This study evaluated whether copper sulfate, ammonium chloride, or sodium borohydride can reduce autofluorescence in decellularized plant scaffolds without compromising mechanical integrity or cell viability. METHODS: The effectiveness of the three quenching agents was evaluated in decellularized leatherleaf viburnum, spin

Center experience is associated with greater survival following donation after circulatory death heart transplantation.

BACKGROUND: Higher center volume has been associated with improved outcomes in solid organ transplantation. However, the impact of center experience on donation after circulatory death (DCD) heart transplantation outcomes remains unclear. This study evaluates the association between cumulative DCD center experience and DCD post-transplant survival. METHODS: The United Network for Organ Sharing registry was queried for adult recipients who underwent DCD heart transplantation from January 1, 2019 to December 31, 2023. Recipients were stratified by cumulative DCD center experience into low (≤10)

ZIF-67-anchored halloysite nanotubes infused in chitosan hydrogel for enhanced bone tissue engineering.

Despite the intrinsic regenerative capacity of bone, there remains a clinical need for advanced biomaterials that can simultaneously accelerate osteogenesis and promote vascularization. To address this, a chitosan (CS)-based hydrogel film was developed by incorporating zeolitic imidazolate framework-67 (ZIF-67) anchored functionalized halloysite nanotubes (fHNTs) using a simple, low-cost, solution casting technique. Structural analyses using X-ray diffraction (XRD) and Fourier transform infrared spectroscopy (FTIR) validated the successful synthesis of the CS/ZIF-67/fHNT hydrogel films. Moreov

Kidney transplantation using a pediatric en bloc graft after normothermic regional perfusion and donation after circulatory death procurement.

Kidney transplantation is the ultimate treatment for patients with end stage renal disease. However, with the expanding gap between organ supply and demand, utilization of organs from extended-criteria donors is an important strategy to address this problem. The adoption of normothermic regional perfusion (NRP) after donation after circulatory death has been shown to increase organ utilization and posttransplant outcomes. In this case report, we present the first reported case of a successful kidney transplantation and 1-year follow-up using an en bloc graft from a 2-month-old infant who under

Toward improved auricle reconstruction: The role of FDM 3D printing with PCL and TPU materials.

Microtia, along with trauma, represents one of the main causes of external ear malformation. Different clinical techniques were developed for the reconstruction of the auricle, but they all have some drawbacks. This work is focused on the development of an innovative 3D porous scaffold, printed by Fused Deposition Modelling (FDM) and based on laser-scanned images of the healthy contralateral ear of the patient. The scaffold was printed using polycaprolactone (PCL) and thermoplastic polyether urethane (TPU) to mimic the components of the cartilage and adipose tissue, respectively. After the opt

Biofabrication and evaluation of 3D printed and cast PCL / collagen-alginate hydrogel tubular scaffolds for urethral tissue engineering.

The urethra is a fibromuscular tube that transports urine from the bladder to the exterior of the body. Congenital disorders and urethral wall strictures caused by traumatic injuries, infections, iatrogenic injuries, and tumor removals can impair urethral function. This research aimed to create a 3D printed scaffold of the urethra using casting and 3D printing techniques, and then the constructed scaffolds were characterized. The final scaffolds consisted of a 3D-printed mesh made of poly-ε-caprolactone (PCL) that was cast with a hybrid hydrogel of collagen-alginate and crosslinked. The swelli

A 4D printed nanoengineered super bioactive hydrogel scaffold with programmable deformation for potential bifurcated vascular channel construction.

Four-dimensional (4D) printing of hydrogels enabled the fabrication of complex scaffold geometries out of static parts. Although current 4D fabrication strategies are promising for creating vascular parts such as tubes, developing branched networks or tubular junctions is still challenging. Here, for the first time, a 4D printing approach is employed to fabricate T-shaped perfusable bifurcation using an extrusion-based multi-material 3D printing process. An alginate/methylcellulose-based dual-component hydrogel system (with defined swelling behavior) is nanoengineered with carbonized alginate

Lung Transplantation From Controlled and Uncontrolled Donation After Circulatory Death (DCD) Donors With Long Ischemic Times Managed by Simple Normothermic Ventilation and Ex-Vivo Lung Perfusion Assessment.

OBJECTIVES: Reduction of LDL-cholesterol (LDLc) is essential to decrease the cardiovascular mortality in rheumatoid arthritis (RA). Between 2005 and 2010, French recommendations for dyslipidaemia defined the LDLc target based on the number of cardiovascular risk factors. In 2006, it was recommended to consider LDLc objectives with RA being counted as an additional cardiovascular risk factor. Our objective was to assess lipid target achievement between 2006 and 2010 in a cohort of patients with recent-onset RA. METHODS: 814 patients were included between 2002 and 2005 in a French cohort of pat

Comparison of Biliary Complications Rates After Brain Death, Donation After Circulatory Death, and Living-Donor Liver Transplantation: A Single-Center Cohort Study.

Donation-after-circulatory-death (DCD), donation-after-brain-death (DBD), and living-donation (LD) are the three possible options for liver transplantation (LT), each with unique benefits and complication rates. We aimed to compare DCD-, DBD-, and LD-LT-specific graft survival and biliary complications (BC). We collected data on 138 DCD-, 3,027 DBD- and 318 LD-LTs adult recipients from a single center and analyzed patient/graft survival. BC (leak and anastomotic/non-anastomotic stricture (AS/NAS)) were analyzed in a subset of 414 patients. One-/five-year graft survival were 88.6%/70.0% for DCD

Evolving utilization of donation after circulatory death livers in liver transplantation: The day of DCD has come.

Compared to donation after brain death (DBD), livers procured for transplantation from donation after circulatory death (DCD) donors experience more ischemia-reperfusion injury and higher rates of ischemic cholangiopathy due to the period of warm ischemic time (WIT) following withdrawal of life support. As a result, utilization of DCD livers for liver transplant (LT) has generally been limited to short WITs and younger aged donor grafts, causing many recovered DCD organs to be discarded without consideration for transplant. This study assesses how DCD liver utilization and outcomes have change

Donor Gamma-Glutamyl Transferase Is Associated With Liver Allograft Discard and Failure.

INTRODUCTION: The disparity between the number of individuals on the wait list and available liver allografts creates the need for a system that maximizes donor liver utilization and predicts graft failure. RESEARCH QUESTION: This study aimed to determine the relationship between donor Gamma-Glutamyl Transferase (GGT), liver discard, and graft failure. DESIGN: Through multivariate analysis from 53 966 deceased liver donors, we adjusted for donor clinical and demographic characteristics and compared donor GGT with allograft discard. We compared donor GGT ranges with graft failure and analyzed

Enhanced dual network hydrogels consisting of thiolated chitosan and silk fibroin for cartilage tissue engineering.

Thiolated chitosan (CS-NAC) was synthesized and the selected CS-NAC was used together with silk fibroin (SF) to produce dual network CS-NAC/SF hydrogels. The CS-NAC/SF solutions with formulated compositions were able to form hydrogels at physiological temperature and pH. Rheological measurements showed that elastic modulus of some CS-NAC/SF gels could reach around 3 kPa or higher and was much higher than their respective viscous modulus, indicating that they behaved like strong gels. Deformation measurements verified that CS-NAC/SF gels had well-defined elasticity. The optimized CS-NAC/SF gels

Generation of a 3D Liver Model Comprising Human Extracellular Matrix in an Alginate/Gelatin-Based Bioink by Extrusion Bioprinting for Infection and Transduction Studies.

Bioprinting is a novel technology that may help to overcome limitations associated with two-dimensional (2D) cell cultures and animal experiments, as it allows the production of three-dimensional (3D) tissue models composed of human cells. The present study describes the optimization of a bioink composed of alginate, gelatin and human extracellular matrix (hECM) to print human HepaRG liver cells with a pneumatic extrusion printer. The resulting tissue model was tested for its suitability for the study of transduction by an adeno-associated virus (AAV) vector and infection with human adenovirus

Use of Distal Medial Sural Vessels as Recipient Vessels in Free Tissue Transfer.

Background The medial sural vessels have been used extensively as donor vessels in free tissue transfer; however, there is sparse literature detailing their use as recipient vessels. The few existing reports involve small numbers of patients and describe operative approaches that are unnecessarily cumbersome. We explored our experience in free tissue transfer to the medial sural vessels and share a simplified operative approach to this recipient site. Methods A retrospective review was performed of all patients who underwent free tissue transfers to the medial sural vessels between December 20

Corticosteroids in the management of brain-dead potential organ donors: a systematic review.

Summary Current guidelines recommend the administration of hormonal combination therapy including immunosuppressive doses of corticosteroids to donors with low left ventricular ejection fractions and to consider hormonal therapy administration to all donors. However, these recommendations are largely based on observational data. The aim of this systematic review (SR) was to assess the clinical efficacy and safety of corticosteroids in brain-dead potential organ donors. MEDLINE and EMBASE were searched from the earliest accessible date up to March 2013 with a qualified librarian. Studies compar

The influence of 3% hydrogen peroxide on the survival rate of hair grafts when used as an antiseptic solution for surgical wound care: experience with five patients.

BACKGROUND: A solution of 3% hydrogen peroxide is one of the most widely used antiseptics to decrease bacterial colonization. OBJECTIVE: We performed this study to evaluate influence of 3% hydrogen peroxide on the survival rate of hair grafts. METHODS: We designed two templates on both frontoparietal recesses of five patients with male pattern alopecia. Follicular units containing single hairs were obtained from occiput. We grafted approximately 50 follicular units on each template. The gauze with normal saline was applied on the left template and the gauze with 3% hydrogen peroxide on the r

Optimizing a canine survival model of orthotopic lung transplantation.

INTRODUCTION: While acute models of orthotopic lung transplantation have been described in dogs, the technical considerations of developing a survival model in this species have not been elaborated. Herein, we describe optimization of a canine survival model of orthotopic lung transplantation. METHODS: Protocols of orthotopic left lung transplantation and single lung ventilation were established in acute experiments (n=9). Four dogs, serving as controls, received autologous, orthotopic lung transplants. Allogeneic transplants were performed in 16 DLA-identical and 16 DLA-mismatched unrelated

Size-reduced lung transplantation: an advanced operative strategy to alleviate donor organ shortage.

BACKGROUND: The increasing need for donor lungs, especially for small and pediatric recipients, has not been matched by an adequate supply. This disparity has stimulated the development of new operative techniques, which allow downsizing of larger lungs for use in smaller recipients, thus potentially expanding the donor pool. This approach has recently gained more widespread use, especially for highly urgent recipients; however, is still not considered a standard procedure. PATIENTS AND METHODS: All primary size-reduced lung transplants performed from January 2001 to December 2003 were retros
