Reports of environmental and human health impacts of per- and polyfluoroalkyl substances (PFAS) have greatly increased in the peer-reviewed literature. The goals of the present review are to assess the state of the science regarding toxicological effects of PFAS and to develop strategies for advancing knowledge on the health effects of this large family of chemicals. Currently, much of the toxicity data available for PFAS are for a handful of chemicals, primarily legacy PFAS such as perfluorooctanoic acid and perfluorooctane sulfonate. Epidemiological studies have revealed associations between

New psychoactive substances, especially synthetic cannabinoids (SC), are gaining increasing relevance in postmortem forensic toxicology. Particularly, the interpretation of analytical results is challenging, as usually, no toxicokinetic (TK) data concerning distribution in organs and tissues are available. Thus, a controlled pig TK study allowing for examination of organ and tissue distribution of SC was performed. For this purpose, 12 pigs received a single pulmonary dose of 200 µg/kg body weight each of 4-ethylnaphthalene-1-yl-(1-pentylindole-3-yl)methanone (JWH-210), 2-(4-methoxyphenyl)-1-(

The UV filter 2-ethylhexyl salicylate (EHS) is used in sunscreens and other personal care products worldwide and has been found in a variety of environmental media. We aimed to provide human toxicokinetic data on EHS as a tool for risk assessment. For that purpose, we investigated metabolism and urinary metabolite excretion after a single oral EHS dose (57.4-75.5 μg/(kg body weight)) in three male volunteers. In a suspect screening, we tentatively identified seven EHS metabolites. Three EHS specific metabolites were quantitatively investigated: 2-ethyl-5-hydroxyhexyl 2-hydroxybenzoate (5OH-EHS

A comprehensive weight-of-the-evidence evaluation of 2,4-dichlorophenoxyacetic acid (2,4-D) was conducted for potential interactions with the estrogen, androgen and thyroid pathways and with steroidogenesis. This assessment was based on an extensive database of high quality in vitro, in vivo ecotoxicological and in vivo mammalian toxicological studies. Epidemiological studies were also considered. Toxicokinetic data provided the basis for determining rational cutoffs above which exposures were considered irrelevant to humans based on exceeding thresholds for saturation of renal clearance (TSRC

A database of embryo-fetal developmental toxicity (EFDT) studies of 379 pharmaceutical compounds in rat and rabbit was analyzed for species differences based on toxicokinetic parameters of area under the curve (AUC) and maximum concentration (Cmax) at the developmental lowest adverse effect level (dLOAEL). For the vast majority of cases (83% based on AUC of n = 283), dLOAELs in rats and rabbits were within the same order of magnitude (less than 10-fold different) when compared based on available data on AUC and Cmax exposures. For 13.5% of the compounds the rabbit was more sensitive and for 3.

Epoxiconazole, a triazole-based fungicide, was tested in toxicokinetic, prenatal and pre-postnatal toxicity studies in guinea pigs, following oral (gavage) administration at several dose levels (high dose: 90 mg/kg body weight per day). Maternal toxicity was evidenced by slightly increased abortion rates and by histopathological changes in adrenal glands, suggesting maternal stress. No compound-related increase in the incidence of malformations or variations was observed in the prenatal study. In the pre-postnatal study, epoxiconazole did not adversely affect gestation length, parturition, or

Studies about formaldehyde (FA) published since the guideline of 0.1 mg/m(3) by the World Health Organization (WHO) in 2010 have been evaluated; critical effects were eye and nasal (portal-of-entry) irritation. Also, it was considered to prevent long-term effects, including all types of cancer. The majority of the recent toxicokinetic studies showed no exposure-dependent FA-DNA adducts outside the portal-of-entry area and FA-DNA adducts at distant sites were due to endogenously generated FA. The no-observed-adverse-effect level for sensory irritation was 0.5 ppm and recently reconfirmed in hyp

SCH 59884 is an IV prodrug of SCH 56592, the broad-spectrum azole antifungal agent that is active both orally and intravenously in animal models of infection. SCH 56592 is in phase III clinical trials for the treatment of serious systemic fungal infections. SCH 59884 is a carboxylate ester of SCH 56592 with gamma-butyric acid phosphate. Following IV administration of SCH 59884, the compound is rapidly dephosphorylated to SCH 207962 which is then hydrolyzed to SCH 56592. A high-performance liquid chromatographic (HPLC) method was developed for the simultaneous determination of SCH 59884, SCH 20

Scorpion stings are very frequent in Tunisia; yet a method for evaluating envenoming severity and consequently victim treatment, has never been adequately established nor has its efficiency been properly evaluated. Indeed, a management of envenomed patients requires the optimization of envenoming antivenom immunotherapy. This task requires either, an accurate evaluation of toxicokinetic parameters of scorpion venoms in absence and in presence of antivenom, using animals as models, and the establishment of a quantitative relationship between human blood scorption venom levels, envenoming severi

Some environmental and physiological factors that affect the toxicokinetic behaviors of organic solvents were examined using a physiologically based pharmacokinetic model developed for trichloroethylene (TRI). The conclusions are as follows: 1) Body fat content substantially affects the kinetic behavior of TRI: the TRI concentration in blood and the urinary excretion rate of its metabolites are higher in slim men than in obese men during exposure, but these parameters eventually become higher in obese men. 2) There is a sex difference in the pharmacokinetic profiles of TRI. Although retention

Heavy metals that contaminate soils and water usually exist in various oxidation states and form a number of compounds with different physical and chemical characteristics. These differences are often reflected in dramatic variation in toxicokinetic and biologic properties. Such variation in properties, critical in determining intrinsic toxicity, often causes a great deal of uncertainty in analyses of public health risks at sites where metal exposure is evaluated. In the Superfund program, such uncertainties may substantially undermine attempts to characterize potential impacts to populations

The widespread use of pesticides in agriculture also results in the exposure of non-target organisms. These non-target organisms can differ widely in their sensitivity to pesticides. The sensitivity of organisms to a compound is typically assessed using standardized ecotoxicological tests that rely on fixed-time metrics such as four-day LC50 values. These metrics ignore the time-dependency of toxicity and provide little mechanistic understanding of the observed differences in species sensitivity to toxic substances. To quantify potential differences in the development of toxicity over time, se

Honey bees are important pollinators of wild and cultivated plants. They are therefore a key species in the environmental risk assessment of plant protection products in the European Union. As empirical assessments of effects on honey bees on field and landscape level are challenging, mechanistic modelling offers a powerful complement. Such models can predict exposure and effects in the field and are therefore increasingly accepted within the regulatory framework. The BEEHAVEecotox model is a model which links exposure to colony effects in a mechanistic manner. It uses standard regulatory data

Micro(nano)plastics (MNPs) are emerging environmental contaminants, yet pharmaceuticals and medical procedures represent a distinct, direct exposure pathway that bypasses primary physiological barriers. This access, via intravenous administration, implants, or injections, is hypothesized to alter toxicokinetic profiles compared to environmental ingestion, representing a unique but unquantified risk. This review synthesizes current knowledge on MNPs introduced via pharmaceutical and clinical routes through a systematic analysis of 27 core studies identified from Web of Science, PubMed, Scopus,

Per- and polyfluoroalkyl substances (PFAS) continue to be one of the most persistent global contaminants and are increasingly recognized as leading metabolic- and hepatic-dysfunction mediators. Despite extensive investigation of PFAS toxicity, a critical gap in the identification and integration of toxicokinetic drivers of hepatic bioaccumulation with mechanistic pathways driving mitochondrial and nuclear receptor-related injury, more specifically, with respect to alternative PFAS strategies, still remains. Legacy PFAS, including PFOA and PFOS, accumulate in the liver and disturb mitochondrial

Dysosma versipellis (DV), despite its broad-spectrum antitumor potential, faces clinical limitations due to cholestatic hepatotoxicity, prompting investigation into its mechanisms and environmental health implications. Using a toxicological evidence chain (TEC) approach in Sprague–Dawley rats administered 1.2 g/kg DV ethanol extract for 3 days, integrated analyses revealed characteristic cholestatic injury, including significantly reduced liver weight (P < 0.01), elevated serum biomarkers (ALT, AST, TBA; P < 0.01), and bile duct hyperplasia. Toxicokinetic profiling identified five hepatotoxic

Following its introduction as an alternative to perfluorooctanoic acid, hexafluoropropylene oxide dimer acid (HFPO-DA) has been extensively detected in various environmental matrices. Despite this prevalence, limited information is available regarding its hepatotoxicity biomarkers. In this study, toxicokinetic simulations indicated that under repeated treatment, HFPO-DA in mice serum reached a steady state by the 4th day. To assess its subacute hepatic effects and identify potential biomarkers, mice were administered HFPO-DA orally at doses of 0, 0.1, 0.5, 2.5, 12.5, or 62.5 mg/kg/d for 7 d. R

BACKGROUND: The occurrence of severe liver injury by the herbal medicine Polygoni Multiflori Radix (PMR) has drawn significant attention. The fact that processing attenuates PMR-induced hepatotoxicity has been well accepted, but the mechanisms are still ambiguous.
PURPOSE: This study aimed to illuminate the mechanism of processing-based attenuation of PMR hepatotoxicity.
METHODS: The contents of emodin-8-O-β-d-glucoside (EG) and emodin (EMD) in raw and processed PMR were quantified. The difference in toxicokinetic behaviors of EG and EMD was determined in vivo, and the disposition properties

Although microsampling of blood is recommended to promote the 3Rs in toxicokinetic (TK) evaluation, there are few reports applying microsampling in actual toxicity evaluation. Here, we assessed the effects of microsampling on toxicological evaluation of methapyrilene hydrochloride, a hepatotoxic substance. Female SD rats received methapyrilene hydrochloride orally at dose levels of 0 (vehicle), 10, and 30 mg/kg BW, once daily for 4 weeks. Each dose level included a microsampling group and a non-microsampling group (n = 5). In the microsampling groups, blood sampling (50 µL/time point) was perf
