In vitro, ex vivo, and in silico studies of phytochemicals from the stem bark of Bischofia javanica.

The stem bark extract of Bischofia javanica, used in traditional Kenyan medicine afforded nine compounds named, santin (1), ermanin (2), rhamnocitrin (3), kumatakenin (4), abyssinone V-4'-methyl ether (5), β-sitosterol (6), betulinic acid (7), friedelin (8) and 1-methyl-1,10-phenanthrolin-1-ium (9). Structural elucidation was performed based on spectroscopic and spectrometric analyses, and the data were compared with those reported in the literature. The natural origin of 1-methyl-1,10-phenanthrolin-1-ium (9) in the plant kingdom is reported here for the first time. Ermanin (2) showed selectiv

Celastrol derivatives ameliorate arthritis in AIA rats via modulating calcium signaling.

BACKGROUND: Calcium (Ca2+) signaling is implicated in the pathogenesis of rheumatoid arthritis (RA). Our previous study found that celastrol (CEL), a natural product derived from herbal plant Tripterygium wilfordii Hook F, suppressed arthritic symptoms in AIA rats via modulating Ca2+ signaling pathway by inhibiting sarcoplasmic/endoplasmic reticulum Ca2+ ATPase (SERCA). However, its high toxicity and narrow therapeutic window severely limit its clinical application. Therefore, developing safer and effective CEL derivatives are of utmost importance. OBJECTIVE: This study aims to develop CEL de

The Effect of Myrtacine and Celastrol-Enriched Extract on Cutibacterium acnes Extracellular Vesicles.

BACKGROUND: Commensal bacteria interact with each other and their environment, secreting anti-microbial peptides, bacteriocins, and extracellular vesicles (EVs). Cutibacterium acnes (C. acnes), a commensal bacterium that plays a major role in acne, is classified into different phylotypes (IA1, IA2, IB, IC, II, and III), where overabundance of phylotype IA1 relative to other phylotypes is found in acne lesions. EVs secreted by C. acnes phylotype IA1 extracted from an acne lesion were previously found to induce a pro-inflammatory response in skin models. Here, we investigated the effects of Myrt

Same analytical method for both (bio)assay and zone isolation to identify/quantify bioactive compounds by quantitative nuclear magnetic resonance spectroscopy.

Differing sensitivity is the main obstacle for a direct combination of HPTLC with NMR spectroscopy. A sufficient amount of the isolated compound zone must be provided by HPTLC for subsequent offline NMR detection (HPTLC//NMR). To fill the gap, a straightforward procedure was developed using the same analytical HPTLC system for both bioprofiling and isolation of bioactive zones from multicomponent mixtures. The HPTLC-effect-directed analysis (EDA) revealed several bioactive compounds in five botanical extracts, i.e. Salvia officinalis, Thymus vulgaris and Origanum vulgare, all Lamiaceae, and pe

Celastrol inhibits prostaglandin E2-induced proliferation and osteogenic differentiation of fibroblasts isolated from ankylosing spondylitis hip tissues in vitro.

BACKGROUND: Heterotopic ossification on the enthesis, which develops after subsequent inflammation, is one of the most distinctive features in ankylosing spondylitis (AS). Prostaglandin E2 (PGE-2) serves as a key mediator of inflammation and bone remodeling in AS. Celastrol, a well-known Chinese medicinal herb isolated from Tripterygium wilfordii, is widely used in treating inflammatory diseases, including AS. It has been proven that it can inhibit lipopolysac-charide-induced expression of various inflammation mediators, such as PGE-2. However, the mechanism by which celastrol inhibits inflamm

Anti-Gastric Ulcer Effect of Betulinic Acid in Male Albino Rats.

Betulinic acid (BA) is a lupane-type triterpene that has been identified and isolated from various plant species used in ethnomedicine in various cultures across the world. This study was undertaken to elucidate the mechanisms underlying the anti-ulcer effect of Betulinic acid. The effect of BA on indomethacin-induced ulcer, gastric mucus secretion, gastric mucus cells count, basal and histamine-induced gastric acid secretion and levels of malondialdehyde formation were studied using dose of 0.5, 1.5, and 3.0 mg/kg. The results showed that BA reduced indomethacin-induced ulceration significant

HIV-1 maturation inhibitor bevirimat stabilizes the immature Gag lattice.

Maturation of nascent virions, a key step in retroviral replication, involves cleavage of the Gag polyprotein by the viral protease into its matrix (MA), capsid (CA), and nucleocapsid (NC) components and their subsequent reorganization. Bevirimat (BVM) defines a new class of antiviral drugs termed maturation inhibitors. BVM acts by blocking the final cleavage event in Gag processing, the separation of CA from its C-terminal spacer peptide 1 (SP1). Prior evidence suggests that BVM binds to Gag assembled in immature virions, preventing the protease from accessing the CA-SP1 cleavage site. To inv

Genomics of sterols biosynthesis in plants: Current status and future prospects.

Sterols produced by bacteria and all eukaryotic organisms are essential for membrane functionality and stability. They play a vital role in growth, development and in abiotic stress tolerance. They are involved in diverse responses to biotic and abiotic stresses that lead to providing resistance against multiple diseases. Additionally, sterols serve as defensive compounds against herbivorous insects and animals. Phytosterols derived from plants, improve human nutrition and health and cure different ailments. The biosynthetic pathways for sterols and triterpenes exhibit similarities until the s…

Reduction in Dressing Change Burden in Patients With Epidermolysis Bullosa-Impact of Oleogel-S10.

Epidermolysis bullosa (EB) is a severe genetic disorder marked by skin fragility and blistering from minimal trauma. Management relies on frequent and painful dressing changes. The EASE study (NCT03068780), the largest to date in EB, previously demonstrated accelerated wound healing and reduced wound burden for Oleogel-S10 (birch triterpenes) versus control gel. This post hoc analysis focused on dressing change frequency and related time and cost savings among patients with daily dressing changes at baseline (Oleogel-S10 n = 47, control gel n = 53). By Day 90, 35.6% of Oleogel-S10 patients req…

Bioactive Compounds Formulated in Phytosomes Administered as Complementary Therapy for Metabolic Disorders.

Metabolic disorders (MDs), including dyslipidemia, non-alcoholic fatty liver disease, diabetes mellitus, obesity and cardiovascular diseases are a significant threat to human health, despite the many therapies developed for their treatment. Different classes of bioactive compounds, such as polyphenols, flavonoids, alkaloids, and triterpenes have shown therapeutic potential in ameliorating various disorders. Most of these compounds present low bioavailability when administered orally, being rapidly metabolized in the digestive tract and liver which makes their metabolites less effective. Moreov…
