Simulated microgravity produces attenuated baroreflex-mediated pressor, chronotropic, and inotropic responses in mice.

Whether myocardial contractile impairment contributes to orthostatic intolerance (OI) is controversial. Accordingly, we used transient bilateral carotid occlusion (TBCO) to compare the in vivo pressor, chronotropic, and inotropic responses (parts 1 and 2) to open-loop selective carotid baroreceptor unloading in anesthetized mice. In part 3, in vitro myocyte responses to isoproterenol in mice exposed to hindlimb unweighting (HLU) for approximately 2 wk were determined. Heart rate (HR) and mean arterial pressure (MAP) responses to TBCO were measured. In control mice, TBCO increased HR (15 +/- 2

Gravity related research with fishes--perspectives in regard to the upcoming International Space Station, ISS.

During the entire evolution of life on Earth, the development of all organisms took place under constant gravity conditions, against which they achieved specific countermeasures for compensation and adaptation. On this background, it is still an open question to which extent altered gravity such as hypergravity (centrifuge) or microgravity (spaceflight) affects the normal individual development, either on the systemic level of the whole organism or on the level of individual organs or even single cells. The present review provides information on these questions, comprising gravistimulated effe

Skeletal adaptation to external loads optimizes mechanical properties: fact or fiction.

The skeleton adapts to a changing mechanical environment but the widely held concept that bone cells are programmed to respond to local mechanical loads to produce an optimal mechanical structure is not consistent with the high frequency of bone fractures. Instead, the author suggests that other important functions of bone compete with mechanical adaptation to determine structure. As a consequence of competing demands, bone architecture never achieves an optimal mechanical structure.

Restoration of peak vascular conductance after simulated microgravity by maximal exercise.

We sought to determine if (i) peak vascular conductance of the calf was reduced following prolonged exposure to simulated microgravity, and (ii) if maximal cycle ergometry performed at the end of microgravity exposure stimulated a restoration of peak calf vascular conductance. To do this, peak vascular conductance of the calf was recorded following ischaemic plantar flexion exercise to fatigue in seven men after 16 days of head-down tilt (HDT) under two conditions: (i) after one bout of maximal supine cycle ergometry completed 24 h prior to performance of ischaemic plantar flexion exercise, an

Development of gravity-sensing organs in altered gravity.

Experiments are described in which the development of the gravity-sensing organs was studied in newt larvae reared in microgravity on the IML-2 mission and in Aplysia embryos and larvae reared on a centrifuge at 1 to 5 g. In Aplysia embryos, the statolith (single dense mass on which gravity and linear acceleration act) was reduced in size in a graded fashion at increasing g. In early post-metamorphic Aplysia or even in isolated statocysts from such animals, the number of statoconia produced is reduced at high g. Newt larvae launched before any of the otoconia were formed and reared for 15 days

Space vestibulo-neuroscience in the new century.

In the new century there will probably be more long spaceflights and fewer short spaceflights. There will probably be several flights to Mars, flights that (if chemical rockets are used) will involve one year to get there, a second year on Mars, and a third year to return. The three-year Mars scenario will probably include rotation of the spacecraft to provide artificial gravity. If nuclear thermal rockets are used, a manned exploration of Mars could be accomplished in less than one year. Vestibular research will be directed towards ensuring that astronauts, upon landing on Mars, will be able

Experiment on "Discovery" STS 51-C: aggregation of red cells and thrombocytes in heart disease, hyperlipidaemia and other conditions.

The aim of this experiment was to study aggregation of red cells in the blood of patients with ischaemic heart disease, diabetes, hyperlipidaemia, and (silent) cancer, and in two normal donors. Reconstituted blood using IgG was also used. The instrument, the automated slit-capillary photo-viscometer (100 kg weight) was set on the middeck of the Space Shuttle. An analogous instrument was at the Kennedy Space Center. Blood was obtained from donors, anticoagulated, and adjusted to haematocrit of 30% using native plasma. Experiments took place at 25 degrees C, during which blood was forced to flow

Manifesto of space medicine: the next dermatologic frontier.

As the fields of astronomy, cosmology, and space travel move rapidly forward, so must space medicine. The manned space program and medical knowledge and support have developed in tandem. Dermatology will play a fundamental role in survival during space flight. This paper reviews past, present, and future accomplishments of the space program as they relate to medicine and characterizes some of dermatology's multiple roles in the future. It further explores the immunologic alterations noted during space flight and the attendant implications for health and well-being both in flight and on return

Chemogenetic inhibition of the carotid bodies blunts hind-limb suspension microgravity-induced muscle alterations in rats.

Microgravity is known to promote muscle loss and impair physical performance. The carotid body (CB) chemoreceptors are sensitive to several stimuli and have been associated with peripheral vascular control and deterioration in exercise performance. Accordingly, it is plausible that the CB chemoreflex drive may modify the microgravity-induced muscle changes and, consequently, exercise performance. Thus, it is reasonable to propose that a microgravity environment can alter the CB chemoreflex drive, affecting exercise performance. Hence, we aimed to determine the effects of simulated microgravity

Comparing effects of microgravity and amyotrophic lateral sclerosis in the mouse ventral lumbar spinal cord.

Microgravity (MG) exposure and motor neuron diseases, such as amyotrophic lateral sclerosis (ALS), lead to motor deficits, including muscle atrophy and loss of neuronal activity. Abnormalities in motor neurons and muscles caused by MG exposure can be recovered by subsequent ground exercise. In contrast, the degeneration that occurs in ALS is irreversible. A common phenotype between MG exposure and ALS pathology is motor system abnormality, but the causes may be different. In this study, to elucidate the motor system that is affected by each condition, we investigated the effects of MG and the

Microgravity-induced ocular changes are related to body weight.

On Earth, tissue weight generates compressive forces that press on body structures and act on the walls of vessels throughout the body. In microgravity, tissues no longer have weight, and tissue compressive forces are lost, suggesting that individuals who weigh more may show greater effects from microgravity exposure. One unique effect of long-duration microgravity exposure is spaceflight-associated neuroocular syndrome (SANS), which can present with globe flattening, choroidal folds, optic disk edema, and a hyperopic visual shift. To determine whether weight or other anthropometric measures a

Comparative analysis of anti-polyglutamine Fab crystals grown on Earth and in microgravity.

Huntington's disease is one of nine neurodegenerative diseases caused by a polyglutamine (polyQ)-repeat expansion. An anti-polyQ antigen-binding fragment, MW1 Fab, was crystallized both on Earth and on the International Space Station, a microgravity environment where convection is limited. Once the crystals returned to Earth, the number, size and morphology of all crystals were recorded, and X-ray data were collected from representative crystals. The results generally agreed with previous microgravity crystallization studies. On average, microgravity-grown crystals were 20% larger than control

Preflight screening techniques for centrifuge-simulated suborbital spaceflight.

INTRODUCTION: Historically, space has been the venue of the healthy individual. With the advent of commercial spaceflight, we face the novel prospect of routinely exposing spaceflight participants (SPFs) with multiple comorbidities to the space environment. Preflight screening procedures must be developed to identify those individuals at increased risk during flight. We examined the responses of volunteers to centrifuge accelerations mimicking commercial suborbital spaceflight profiles to evaluate how potential SFPs might tolerate such forces. We evaluated our screening process for medical app

Simulated stand tests and centrifuge training to prevent orthostatic intolerance on Earth, moon, and Mars.

One proposed method to overcome postflight orthostatic intolerance is for astronauts to undergo inflight centrifugation. Cardiovascular responses were compared between centrifuge and gravitational conditions using a seven-compartment cardiovascular model. Vascular resistance, heart rate, and stroke volume values were adopted from literature, while compartmental volumes and compliances were derived from impedance plethysmography of subjects (n=8) riding on a centrifuge. Three different models were developed to represent the typical male subject who completed a 10-min postflight stand test ("mal

Analysis of posture and eye movement responses to Coriolis stimulation under 1 G and microgravity conditions.

To detect the effect of microgravity on vestibular responses, we conducted Coriolis stimulation experiments at 1 G and μ G. Five men with vision occluded were asked to tilt their head forward while rotating at 100 degrees/sec. Postural changes were recorded by a 3D linear accelerometer, and the distance of upper body movement was derived from recordings of linear acceleration. Eye movements were recorded by a CCD camera. For a second period after commencing head tilt, the upper body moved 10 cm in the direction of inertia input at 1 G, but it moved to the opposite direction at μ G, i.e., 4 cm

Development and validation of a predictive bone fracture risk model for astronauts.

There are still many unknowns in the physiological response of human beings to space, but compelling evidence indicates that accelerated bone loss will be a consequence of long-duration spaceflight. Lacking phenomenological data on fracture risk in space, we have developed a predictive tool based on biomechanical and bone loading models at any gravitational level of interest. The tool is a statistical model that forecasts fracture risk, bounds the associated uncertainties, and performs sensitivity analysis. In this paper, we focused on events that represent severe consequences for an explorati

Melanoma growth and tumorigenicity in models of microgravity.

INTRODUCTION: Spaceflight involves numerous biological stressors that could affect long-term cancer incidence and tumor behavior. Ground-based models of microgravity can be used to investigate in vitro and in vivo tumor growth as a preparation for later work in space. The incidence of tumor growth and carcinogenesis in microgravity is as yet unknown. Hence, we investigated the effects of modeled microgravity on tumor growth and tumorigenicity using ground-based in vitro and in vivo models. METHODS: Murine B16-F10 melanoma cells were cultured in a tissue culture flask (FL) and in a rotating-wa

G-factor as a tool in basic research: mechanisms of orthostatic tolerance.

The inability to tolerate upright standing posture due to development of orthostatic hypotension is a clinical problem experienced by more than 500,000 people in the United States. In addition, orthostatic intolerance is an operational problem since as many as 25 to 64 percent of the crew members from U.S. space shuttle flights have been reported to experience presyncopal incidents during postflight stand tests. Last year alone, more than 10,000 cases of unexplained syncope were reported in active duty personnel in all U.S. military services. Many clinical investigations have focused on measur

Comparison of treatment strategies for Space Motion Sickness.

Treatment strategies for Space Motion Sickness (SMS) were compared using the results of postflight oral debriefings. Standardized questionnaires were administered to all crewmembers immediately following Space Shuttle flights by NASA flight surgeons. Cases of SMS were graded as mild, moderate, or severe based on published criteria, and medication effectiveness was judged based on subjective reports of symptom relief. Since October 1989, medication effectiveness is reported inflight through Private Medical Conferences with the crew. A symptom matrix was analyzed for 19 crewmembers treated with
