Tempol Mitigates Cisplatin-Induced Retinopathy by Modulating ER Stress, Enhancing Autophagy, and Preserving Neurotrophic Support.

The-state-of-the-art literature is eagerly searching for protective strategies of chemotherapy-induced retinal neurotoxicity, driven by oxidative stress, ER stress, mitochondrial dysfunction, and autophagy impairment. We investigated the neuroprotective effects of tempol, a stable nitroxide antioxidant, capable of crossing biological barriers and mitigating cisplatin-induced cytomolecular insult. Forty-eight Wistar rats were divided into control, cisplatin-treated, tempol-treated, and CIS + TEM-cotreated groups. Retinal tissues were analyzed through histological, immunohistochemical, and biomo

Platelets and diseases: signal transduction and advances in targeted therapy.

Platelets are essential anucleate blood cells that play pivotal roles in hemostasis, tissue repair, and immune modulation. Originating from megakaryocytes in the bone marrow, platelets are small in size but possess a highly specialized structure that enables them to execute a wide range of physiological functions. The platelet cytoplasm is enriched with functional proteins, organelles, and granules that facilitate their activation and participation in tissue repair processes. Platelet membranes are densely populated with a variety of receptors, which, upon activation, initiate complex intracel

Management strategies for fluoride toxicity and the critical role of epigenetic therapy.

Fluoride is an essential trace element primarily known for its role in preventing dental caries; however, excessive exposure can lead to adverse health effects. Emerging research suggests possible links between fluorosis and neurodevelopmental disorders, including reduced cognitive function and increased prevalence of ADHD. Its impact on liver health such as hepatotoxicity and hormone-related infertility also raises concern. Moreover, high fluoride exposure has been associated with various toxicities, including skeletal fluorosis, reproductive toxicity, and neurotoxicity, potentially driven by

The role of VCAM1 in post-stroke cognitive impairment: Molecular mechanisms and therapeutic potential.

Ischemic stroke constitutes a critical medical condition that can lead to post-stroke cognitive impairment (PSCI), yet the underlying molecular mechanisms remain incompletely understood. This study investigated the role of vascular cell adhesion molecule 1 (VCAM1) in cognitive dysfunction following ischemic stroke and evaluated its therapeutic potential. In a rat model of middle cerebral artery occlusion (MCAO), VCAM1 knockdown resulted in improved motor function, enhanced spatial memory, and reduced infarct size. Mechanistically, silencing VCAM1 was found to have mitigated blood-brain barrier

Dry Eye Disease and Psychiatric Disorders: Neuroimmune Mechanisms and Therapeutic Perspectives.

Dry eye disease (DED) is a highly prevalent multifactorial disorder of the ocular surface that extends beyond local tear film pathology to involve systemic immune, neuroendocrine, and neurosensory mechanisms. Increasing evidence reveals a strong and bidirectional association between DED and psychiatric disorders, particularly depression, anxiety, post-traumatic stress disorder (PTSD), and sleep disturbances. This review synthesises the current knowledge on shared molecular, neuroimmune, and neuropathic pathways that underlie this comorbidity. Key mechanisms include hypothalamic-pituitary-adren

Characterization of intestinal microbiota in patients with psoriasis combined with metabolic syndrome.

The co-morbidity of psoriasis (PsO) and metabolic syndrome (MetS) is prevalent. While the mechanisms causing PsO in MetS remain unknown, it is understood that gut microbiota plays a critical role in maintaining immune and metabolic balance. However, the specific mechanisms and whether biomarkers are involved in the pathophysiological process of PSO in MetS patients have yet to be elucidated. This study involved 29 patients with PSO and MetS (PM) and 29 with MetS (M). It analyzed their clinical parameters, microbial composition, function, diagnostic, and predictive performance. The results unve

Nebulized and intravenous enzyme replacement therapy in mice with mucopolysaccharidosis type II.

Mucopolysaccharidosis Type II is a hereditary lysosomal storage disease characterized by deficiency in the enzyme iduronate 2-sulfatase (IDS). IDS is critical in the breakdown of sulfated glycosaminoglycans and its deficiency leads to an accumulation of these compounds across various tissue types resulting in multisystemic dysfunction. Intravenous administration of recombinant IDS (idursulfase) substantially improves patients' quality and length of life. However, recombinant IDS delivered intravenously is sequestered in the liver and respiratory failure remains as the leading cause of death fo

Nucleotide-Binding Oligomerization Domain 2 in Signaling, Immunity, and Mycobacterial Infection.

INTRODUCTION: Nucleotide-binding oligomerization domain 2 (NOD2) functions primarily as a cytoplasmic pattern recognition receptor (PRR) that detects muramyl dipeptide (MDP), a conserved bacterial cell wall component, thereby playing a pivotal role in pathogen surveillance. However, emerging evidence reveals that NOD2 exerts broad immunomodulatory effects beyond its canonical role as a PRR, though its effects can be contradictory, depending on genetic background, immunological microenvironment, and disease context. In this review, we integrate recent advances in understanding NOD2's functional

The role of gut microbiota in autoimmune disease progression and therapy: a comprehensive synthesis.

Autoimmune diseases arise from the immune system's dysregulated attack on the body's own tissues, influenced by a complex interplay of genetics, environment, and the microbiome. This comprehensive review and meta-analysis examines the dynamic relationship between gut microbiota and autoimmune diseases, highlighting their role in disease onset, progression, and potential therapeutic interventions. Emerging evidence underscores the bidirectional interactions between microbiota and immune pathways, particularly through mechanisms like mucosal immune modulation and regulatory T-cell activity.

Exploring the metabolic patterns and response mechanisms of bile acids during fasting: A study with poultry as an example.

Fasting is beneficial to alleviate fatty liver, lose weight and improve reproductive function. However, previous studies have shown that, during fasting, disorders of bile acid metabolism were strongly associated with intestinal inflammation. The physiological and biochemical parameters and gene expression of multiple tissues of chickens at every critical time node were measured by ELISA and qPCR. In addition, association analysis was performed based on liver transcriptome sequencing and cecum metabolome data. At the cellular level, the regulatory effects of cecal metabolites on host bile acid

Bromodomain-Containing Protein 9 Regulates Signaling Pathways and Reprograms the Epigenome in Immortalized Human Uterine Fibroid Cells.

Bromodomain-containing proteins (BRDs) are involved in many biological processes, most notably epigenetic regulation of transcription, and BRD dysfunction has been linked to many diseases, including tumorigenesis. However, the role of BRDs in the pathogenesis of uterine fibroids (UFs) is entirely unknown. The present study aimed to determine the expression pattern of BRD9 in UFs and matched myometrium and further assess the impact of a BRD9 inhibitor on UF phenotype and epigenetic/epitranscriptomic changes. Our studies demonstrated that the levels of BRD9 were significantly upregulated in UFs
