Submicromolar hydrogen peroxide disrupts the ability of Fur protein to control free-iron levels in Escherichia coli.

In aerobic environments, mutants of Escherichia coli that lack peroxidase and catalase activities (Hpx(-)) accumulate submicromolar concentrations of intracellular H(2)O(2). We observed that in defined medium these strains constitutively expressed members of the Fur regulon. Iron-import proteins, which Fur normally represses, were fully induced. H(2)O(2) may antagonize Fur function by oxidizing the Fur:Fe(2+) complex and inactivating its repressor function. This is a potential problem, as in iron-rich environments excessive iron uptake would endanger H(2)O(2)-stressed cells by accelerating hyd

Anthrax lethal toxin enhances cytokine-induced VCAM-1 expression on human endothelial cells.

Vascular endothelial dysfunction is thought to play a prominent role in systemic anthrax pathogenesis. We examined the effect of anthrax lethal toxin (LTx), a key virulence factor of Bacillus anthracis, on the expression of vascular cell adhesion molecule-1 (VCAM-1) on normal and cytokine-stimulated human lung microvascular endothelial cells. Confluent endothelial monolayers were treated with lethal factor (LF), protective antigen (PA), or both (LTx) in the presence or absence of tumor necrosis factor-alpha (TNFalpha). LTx enhanced cytokine-induced VCAM-1 expression and monocyte adhesion. LTx

RpoS is not central to the general stress response in Borrelia burgdorferi but does control expression of one or more essential virulence determinants.

Borrelia burgdorferi, the Lyme disease spirochete, undergoes dramatic changes in antigenic composition as it cycles between its arthropod and mammalian hosts. A growing body of evidence suggests that these changes reflect, at least in part, the need for spirochetes to adapt to the physiological stresses imposed by abrupt changes in environmental conditions and nutrient availability. In many microorganisms, global responses are mediated by master regulators such as alternative sigma factors, with Escherichia coli RpoS (sigmaS) serving as a prototype. The importance of this transcriptional activ

Ф. dksA is required for intercellular spread of Shigella flexneri via an RpoS-independent mechanism.

Pathogenesis of Shigella flexneri is dependent on the ability of the bacterium to invade and spread within epithelial cells. In this study, we identified dksA as a gene necessary for intercellular spread in, but not invasion of, cultured cells. The S. flexneri dksA mutant exhibited sensitivity to acid and oxidative stress, in part due to an effect of DksA on production of RpoS. However, an S. flexneri rpoS mutant formed plaques on tissue culture monolayers, thus excluding DksA regulation of RpoS as the mechanism responsible for the inability of the dksA mutant to spread intercellularly. The global activator GacA of Pseudomonas aeruginosa PAO positively controls the production of the autoinducer N-butyryl-homoserine lactone and the formation of the virulence factors pyocyanin, cyanide, and lipase.

The global activator GacA, a highly conserved response regulator in Gram-negative bacteria, is required for the production of exoenzymes and secondary metabolites in Pseudomonas spp. The gacA gene of Pseudomonas aeruginosa PAO1 was isolated and its role in cell-density-dependent gene expression was characterized. Mutational inactivation of gacA resulted in delayed and reduced formation of the cell-density signal N-butyryl-L-homoserine lactone (BHL), of the cognate transcriptional activator RhIR (VsmR), and of the transcriptional activator LasR, which is known to positively regulate RhIR expres

Distinct lines of chickens express different splenic cytokine profiles in response to Salmonella Enteritidis challenge.

Chicken meat and eggs contaminated with Salmonella result in economic losses in the poultry industry and potential human infection. Intestinal parasites have been shown to lead to a reduction in the utilization of nutrients and performance in poultry. This study provides insight into the immune responses used by hens of 3 genetically distinct chicken lines (broiler, Leghorn, and Fayoumi) in the presence and absence of Salmonella Enteritidis infection. Understanding the range of immune responses used by different lines in response to Salmonella Enteritidis may help the poultry industry genetica

Bioactive chitosan dressings functionalized with indole alkaloids suppress biofilm development and virulence traits in bacterial species relevant to wound infections.

AIMS: Chronic wound infections are frequently associated with biofilm-forming pathogens that impair healing and reduce the effectiveness of antimicrobial therapies. This study evaluated the antimicrobial, antibiofilm, synergistic, and antivirulence activities of two indole alkaloids, 3-(1H-indol-3-yl)propanoic acid (C1) and 1H-indole-2-carboxylic acid (C2), and investigated their incorporation into chitosan-based biodressings for infected wound management. METHODS AND RESULTS: Antimicrobial activity was evaluated by agar diffusion, minimum inhibitory concentration (MIC), minimum bactericidal

TLR4 signalling: the key to controlling EV71 replication and inflammatory response.

Hand, foot, and mouth disease (HFMD) is a common infectious disease caused by enterovirus 71 (EV71) that frequently affects children, leading to severe infections in some cases. In general, when infection occurs, the body upregulates inflammatory responses to eliminate pathogenic microorganisms to protect the host from infection. However, EV71 may inhibit host's innate immunity to promote virus infection. At present, it is not fully understood how EV71 hijack the host cells for its own replication. Toll-like receptor 4 (TLR4), a natural immune receptor, historically associated with bacterial e

Perturbation of maternal gut microbiota in mice during a critical perinatal window influences early neurobehavioral outcomes in offspring.

The gut microbiota is increasingly recognized as a key environmental factor that shapes host development and physiology, including neural circuits formation and function. Concurrently, there has been growing concern that early-life antibiotic exposure may alter brain developmental trajectories, increasing the risk for neurodevelopmental disorders such as autism spectrum disorder (ASD). Here, we assessed whether perturbation of the maternal gut microbiota in mice during a narrow critical perinatal window (last week of pregnancy and first three postnatal days), induced by exposure to a commonly

Neutrophil Survival Signaling During Francisella tularensis Infection.

Neutrophils are the most abundant and shortest-lived leukocytes in humans and tight regulation of neutrophil turnover via constitutive apoptosis is essential for control of infection and resolution of inflammation. Accordingly, aberrant neutrophil turnover is hallmark of many disease states. We have shown in previous work that the intracellular bacterial pathogen Francisella tularensis markedly prolongs human neutrophil lifespan. This is achieved, in part, by changes in neutrophil gene expression. Still unknown is the contribution of major neutrophil pro-survival signaling cascades to this pro

A nucleotide-binding leucine-rich repeat gene OsCNL1 contributes to strain-specific resistance to bacterial blight in rice.

Bacterial blight (BB), caused by Xanthomonas oryzae pv. oryzae (Xoo), remains a major threat to rice production, and continued discovery and functional validation of resistance genes remain important for durable disease management. Here, we functionally characterized OsCNL1 (LOC_Os11g37759), a rice coiled-coil (CC)-type nucleotide-binding leucine-rich repeat (NLR) gene associated with BB resistance in the resistant line B5N494. OsCNL1 encodes a 968-amino-acid CC type NLR protein and showed pathogen-responsive expression after PXO99A inoculation. CRISPR/Cas9-mediated disruption of OsCNL1 in B5N

Ascophyllum nodosum based plant biostimulant shapes the bacterial community in the rhizosphere of corn.

BACKGROUND: Plant biostimulants are an emerging class of agricultural inputs known to enhance plant growth and improve their tolerance to abiotic stress. While biostimulants are widely used, their mechanisms of action remain poorly understood. This study investigates the effects of an Ascophyllum nodosum-based biostimulant (ANE) on the rhizosphere bacterial communities of corn (Zea mays). RESULTS: Root exudates from ANE root-treated plants promoted the swarming motility of Pseudomonas protegens (CHA0), a plant growth-promoting rhizobacterium. Gene expression analysis showed that root exudates

Epigenetic Mechanisms in Aging: Extrinsic Factors and Gut Microbiome.

BACKGROUND/OBJECTIVES: Aging is a natural physiological process involving biological and genetic pathways. Growing evidence suggests that alterations in the epigenome during aging result in transcriptional changes, which play a significant role in the onset of age-related diseases, including cancer, cardiovascular disease, diabetes, and neurodegenerative disorders. For this reason, the epigenetic alterations in aging and age-related diseases have been reviewed, and the major extrinsic factors influencing these epigenetic alterations have been identified. In addition, the role of the gut microb

Neisseria cinerea Expresses a Functional Factor H Binding Protein Which Is Recognized by Immune Responses Elicited by Meningococcal Vaccines.

Neisseria meningitidis is a major cause of bacterial meningitis and sepsis worldwide. Capsular polysaccharide vaccines are available against meningococcal serogroups A, C, W, and Y. More recently two protein-based vaccines, Bexsero and Trumenba, against meningococcal serogroup B strains have been licensed; both vaccines contain meningococcal factor H binding protein (fHbp). fHbp is a surface-exposed lipoprotein that binds the negative complement regulator complement factor H (CFH), thereby inhibiting the alternative pathway of complement activation. Recent analysis of available genomes has ind

Interaction effects of Micropterus salmoides and Edwardsiella piscicida under ammonia nitrogen and nitrite stress.

This study investigated the interactive effects of ammonia nitrogen (AN) and nitrite stress on Micropterus salmoides during infection with Edwardsiella piscicida (E. piscicida). The results indicated that combined stress of 2 mg/L nitrite initially induced a short-term immune enhancement in the host. This enhancement was characterized by the activation of genes associated with antigen recognition (CD40, TLR-2, IgM, and IFN-γ), innate immunity genes (MsP-2 and HSP70), inflammatory factors (IL-8 and TNF-α), and autophagy (VAMP8 and STX17) in the gills, liver, and head kidney. However, this immun

The RpfF Acyl-ACP thioesterase of major rice pathogen Xanthomonas oryzae pv. oryzae is essential for maintenance of membrane function and virulence.

In Xanthomonas, RpfF is a bifunctional enzyme having both enoyl-ACP dehydratase and acyl-ACP thioesterase activities catalyzed by a single active site. While the hydratase activity is essential for producing quorum-sensing signals of the diffusible signal factor (DSF) family, the physiological role of the thioesterase activity remained unclear. Singh and coworkers have reported that deletion of the gene encoding RpfF (a ∆rpfF strain) of the rice pathogen X. oryzae pv. oryzae (Xoo) resulted in loss of membrane integrity (Singh et al, 2022). We report that the loss in membrane integrity is due t

HMOX1-mediated ferroptosis participates in gangrenous mastitis in dairy goats.

Gangrenous mastitis is a severe disease caused by bacterial infection. It can lead to sepsis and even death in affected goats, posing a significant threat to the development of the dairy goat industry. The pathogenesis of this disease is not fully understood. In this study, a Staphylococcus aureus isolate obtained from a goat with gangrenous mastitis was used to establish in vivo and in vitro models. Using omics sequencing and detection of iron, MDA, GSH, and ROS levels, along with RT-qPCR and western blot, the occurrence of ferroptosis was assessed. Furthermore, by employing ferrostatin-1 (Fe

Arabidopsis mlo3 mutant plants exhibit spontaneous callose deposition and signs of early leaf senescence.

Arabidopsis thaliana mlo3 mutant plants are not affected in pathogen infection phenotypes but-reminiscent of mlo2 mutant plants-exhibit spontaneous callose deposition and signs of early leaf senescence. The family of Mildew resistance Locus O (MLO) proteins is best known for its profound effect on the outcome of powdery mildew infections: when the appropriate MLO protein is absent, the plant is fully resistant to otherwise virulent powdery mildew fungi. However, most members of the MLO protein family remain functionally unexplored. Here, we investigate Arabidopsis thaliana MLO3, the closest re

Molecular characterization and functional activity of CXCL8_L3 in large yellow croaker Larimichthys crocea.

CXCL8, also called interleukin-8, is a typical CXC chemokine that plays a key role in promoting inflammation. Phylogenetically, fish CXCL8 chemokines can be divided into three subgroups, CXCL8_L1, CXCL8_L2, and CXCL8_L3, of which CXCL8_L3 is a new subgroup. The CXCL8_L3 gene sequences have been reported in many fish species, but their function remains unknown. Here, a CXCL8_L3 (LycCXCL8_L3) gene was cloned from large yellow croaker Larimichthys crocea. Its open reading frame (ORF) was 309 nucleotides long, encoding a peptide of 102 amino acids. The deduced LycCXCL8_L3 protein contains an 18-aa
