The genetics of ankylosing spondylitis and axial spondyloarthritis.

Ankylosing spondylitis (AS) and spondyloarthritis are strongly genetically determined. The long-standing association with HLA-B27 is well described, although the mechanism by which that association induces AS remains uncertain. Recent developments include the description of HLA-B27 tag single nucleotide polymorphisms in European and Asian populations. An increasing number of non-MHC genetic associations have been reported, which provided amongst other things the first evidence of the involvement of the IL-23 pathway in AS. The association with ERAP1 is now known to be restricted to HLA-B27 pos

Integrating GWAS and transcriptomics to identify candidate genes associated with the crest trait in Yongshun crested duck.

The Yongshun crested duck (YCD) is a newly discovered ornamental duck breed characterized by a cluster of spherical feathers on the head. However, the genetic mechanisms underlying the crest trait in YCD remain unclear. In this study, we integrated whole-genome resequencing and transcriptomic analyses to identify candidate genes and regulatory pathways associated with the crest trait. A total of 175 YCD individuals (76 crested and 99 non-crested) were subjected to genome-wide association study (GWAS). Population genetic analyses revealed sufficient genetic recombination and no obvious populati

Multiple environmental test and genetic mapping jointly reveal genomic loci and associated genes underlying cotton Verticillium wilt resistance.

Based on GWAS, QTL mapping, BSA and RNA‒seq jointly reveal the genetic mechanisms underlying Verticillium wilt resistance in cotton and identify three candidate genes associated with this trait. Verticillium wilt, caused by the soil-borne vascular pathogen Verticillium dahliae, is a devastating disease that severely threatens global cotton production. However, the genetic basis of resistance to this disease remains poorly uncovered, hindering the advancement of genomics-assisted breeding programs for Verticillium wilt (VW) resistance. Here, we systematically evaluated VW resistance in a recomb

Biopsychosocial risk factors for Alzheimer's disease and related dementias in UK immigrants from the Middle East and North Africa (MENA).

The Middle East and North Africa (MENA) region represents the area of greatest projected growth in instances of Alzheimer's disease and related dementias (ADRDs) globally, yet, it remains virtually uncharacterized in health studies of aging and ADRDs. The UK Biobank is one of the largest and well characterized datasets of aging immigrants in the UK, offering an unprecedented opportunity to identify risk factors for ADRDs in individuals from MENA regions. Here we used the UK Biobank to compare sociodemographic, disease, lifestyle, genetic, and neuroimaging risk factors for ADRDs among UK immigr

Effects of COLQ Gene Missense Mutations on Growth and Meat Traits in Leizhou Black Goats.

As an indigenous goat breed unique to southern China, Leizhou Black Goats (LZBGs) are highly valued for their rapid growth, high reproductive performance, and superior meat quality. However, their offspring frequently exhibit symptoms of muscle atrophy and malnutrition, suggesting potential genetic defects underlying these adverse phenotypes. As a unique extracellular matrix component, collagen Q (COLQ) is specifically enriched within the synaptic basal lamina at vertebrate neuromuscular junctions (NMJs), where it anchors acetylcholinesterase (AChE) to facilitate efficient acetylcholine hydrol

Neither Diplectanum nor specific: a dramatic twist to the taxonomic framework of Diplectanum (Monogenea: Diplectanidae).

The taxonomy of the genus Diplectanum has been exclusively based on morphological features, with 28 nominal species parasitic on perciform fishes recognised. We used molecular data, to our knowledge for the first time, to evaluate the taxonomic framework of Diplectanum, infer the relationships amongst species attributed to this genus, re-assess the degree of host specificity and explore the population genetic structure of Diplectanum spp. parasitising Mediterranean sciaenids, which are potential target fish species for aquaculture diversification in the region. A minimum of 10 specimens of Dip

Interrogating the Genetic Determinants of Tourette's Syndrome and Other Tic Disorders Through Genome-Wide Association Studies.

OBJECTIVE: Tourette's syndrome is polygenic and highly heritable. Genome-wide association study (GWAS) approaches are useful for interrogating the genetic architecture and determinants of Tourette's syndrome and other tic disorders. The authors conducted a GWAS meta-analysis and probed aggregated Tourette's syndrome polygenic risk to test whether Tourette's and related tic disorders have an underlying shared genetic etiology and whether Tourette's polygenic risk scores correlate with worst-ever tic severity and may represent a potential predictor of disease severity. METHODS: GWAS meta-analys

Hereditary etiology of non-syndromic sensorineural hearing loss in the Republic of North Ossetia-Alania.

More than 50% of congenital hearing loss is hereditary, in which the majority form is non-syndromic. In this study we estimate the most prevalent pathogenic genetic changes in an Ossetian cohort of patients. This is useful for local public health officials to promote genetic counseling of affected families with regard to high allele frequencies of prevalent pathogenic variants and assortative mating in the community of people with hearing loss. In this study, genetic heterogeneity of hereditary non-syndromic sensorineural hearing loss (NSNHL) in a cohort of 109 patients and an assessment of th

Investigating Atopic Dermatitis Prevention With Omega-3 Supplementation in Different Ethnic Populations.

Atopic dermatitis (AD) is a chronic inflammatory skin disease affecting approximately 5% of adults and 13% of children worldwide. Individuals with skin of color (SOC) are disproportionately affected and often experience more severe disease manifestations, including pigmentary changes and lichenification. Despite these disparities, SOC populations remain underrepresented in AD research, limiting the development of targeted prevention strategies. Emerging evidence suggests that omega-3 polyunsaturated fatty acids may play a protective role in AD through anti-inflammatory effects and support of t

The relationship between evolvability and plasticity under experimental warming across species' elevational ranges.

Phenotypic variation among individuals of a population arises from genetic and environmental effects. These may be correlated if they have similar underlying developmental mechanisms or selection histories. We tested empirically for such links over elevation gradients, on which species typically encounter predictable niche limits, range limits, and limits to adaptive evolution. We exposed plants from the elevational core and range edges of three montane and three alpine Brassicaceae species to experimental warming and measured key ecological traits that are divergent over elevation. Plasticity

Genome-wide association study in the Taiwan Biobank identifies four novel genes for human height: NABP2, RASA2, RNF41 and SLC39A5.

Numerous genome-wide association studies (GWASs) have been conducted for the identification of genetic variants involved with human height. The vast majority of these studies, however, have been conducted in populations of European ancestry. Here, we report the first GWAS of adult height in the Taiwan Biobank using a discovery sample of 14 571 individuals and an independent replication sample of 20 506 individuals. From our analysis, we generalize to the Taiwanese population genome-wide significant associations with height and 18 previously identified genes in European and non-Taiwanese East A

Cross-Species Insights Into Genomic Adaptations to Hypoxia.

Over millions of years, vertebrate species populated vast environments spanning the globe. Among the most challenging habitats encountered were those with limited availability of oxygen, yet many animal and human populations inhabit and perform life cycle functions and/or daily activities in varying degrees of hypoxia today. Of particular interest are species that inhabit high-altitude niches, which experience chronic hypobaric hypoxia throughout their lives. Physiological and molecular aspects of adaptation to hypoxia have long been the focus of high-altitude populations and, within the past

A New Risk Polymorphism rs10403848 of CARD8 Significantly Associated with Psoriasis Vulgaris in Northeastern China.

Caspase recruitment domain family member 8 (CARD8) is an adaptor molecule that negatively regulates nuclear factor-κB (NF-κB) activation, interleukin (IL)-1β secretion, and apoptosis. These play important roles in the pathogenesis of psoriasis. Genetic variants of CARD8 have been associated with an increased risk of several inflammatory diseases and psoriasis in Europe. However, nothing is known about the association of the polymorphisms of CARD8 and psoriasis vulgaris (PsV) in the Han population of northeastern China. To investigate the potential association between them, we designed a case-c

GWAS identifies two novel colorectal cancer loci at 16q24.1 and 20q13.12.

Colorectal cancer (CRC) is the fourth leading cause of cancer mortality worldwide. Genome-wide association studies (GWAS) identified more than 50 CRC loci. However, most of the previous studies were conducted in European population, and host genetic factors among Japanese population are largely remained to be identified. To identify novel loci in the Japanese population, here, we performed a large-scale GWAS using 6692 cases and 27 178 controls followed by a replication analysis using more than 11 000 case-control samples. We found the significant association of 10 loci (P < 5 × 10-8), includi

Association study to evaluate FoxO1 and FoxO3 gene in CHD in Han Chinese.

BACKGROUND: Coronary heart disease (CHD) is one of the leading causes of mortality and morbidity in China. Genetic factors that predispose individuals to CHD are unclear. In the present study, we aimed to determine whether the variation of FoxOs, a novel genetic factor associated with longevity, was associated with CHD in Han Chinese populations. METHODS: 1271 CHD patients and 1287 age-and sex-matched controls from Beijing and Harbin were included. We selected four tagging single nucleotide polymorphisms (SNPs) of FoxO1 (rs2755209, rs2721072, rs4325427 and rs17592371) and two tagging SNPs of

Transport proteins of the ABC systems superfamily and their role in drug action and resistance in nematodes.

The completion of a number of nematode genomes has provided significant information on ABC systems in these organisms. Nematodes have more ABC systems genes and greater diversity than do mammalian species. Class 1 and class 2 ABC systems, more commonly known as ABC transporters, are present. As in other organisms, nematode ABC systems are characterized by a highly conserved ATP-binding domain (ABC_2) and a less conserved transmembrane domain (ABC_TM1/TM1F). Studies of drug resistance in nematodes have suggested that ABC transporters are part of the resistance mechanism. Evidence in support of
