Causal interplay between trigeminal neuralgia and systemic inflammatory markers: A bidirectional Mendelian randomization study.

Trigeminal neuralgia (TN) is a debilitating neuropathic pain disorder with profound impacts on quality of life. Although alterations in circulating inflammatory proteins have been reported, observational findings remain inconsistent and are vulnerable to confounding and reverse causation. Whether these immune changes are causal or merely downstream consequences of TN remains uncertain, underscoring the need for robust causal inference methods. We conducted a bidirectional 2-sample Mendelian randomization (MR) analysis to investigate the causal relationships between circulating inflammatory pro

Loci Associated With Susceptibility to Biliary Atresia-A Genome-Wide Association Study in Taiwan.

BACKGROUND: Biliary atresia (BA) is the leading cause of pediatric liver transplantation worldwide. However, the genetic background remains unclear in most patients. In this genome-wide association study (GWAS), we aimed to identify genetic loci that contribute to susceptibility to BA. METHODS: We recruited 189 BA patients (85 males and 104 females) and 558 non-BA controls (275 males and 283 females) for this GWAS analysis using the Axiom Genome-Wide TWB 2.0 Array with 686,463 single-nucleotide polymorphisms (SNPs). The MassARRAY Sequenom analysis was then applied to validate the identified t

Genome-Wide Association Reveals Signalling-Linked Infection Tolerance in Hibernating Bats.

Hibernation profoundly alters host-pathogen dynamics by suppressing metabolism and immune function, posing unique challenges for infection control. In this study, we examined how genomic variation modulates infection and physiological traits in temperate bats during hibernation. We combined infection screening, haematology, blood biochemistry, and whole-genome sequencing across five vespertilionid species, identifying over 170,000 single nucleotide variants (SNVs) and assessing their associations with 23 health-related variables. Using the phylogenetically informed treeWAS framework, we detect

Population-Specific Mutational Spectrum of Autosomal Recessive Nonsyndromic Hearing Loss in Croatian Roma: Implications for Clinical Genetics.

BACKGROUND/OBJECTIVES: Hearing impairment is a highly prevalent sensory disorder resulting from a variety of causes. A high proportion of autosomal recessive non-syndromic hearing impairment is linked to the GJB2 (OMIM 121011) gene which encodes for a gap junction protein, connexin-26. Alterations of genes that encode for connexins can lead to changes in cell ion content and cause hearing impairment. METHODS: GJB2 gene polymorphisms (c.71G>A, p.Trp24*rs104894396; c.457G>A, p.Val153Ile, rs111033186; c.380G>A, p.Arg127His, rs111033196; c.109G>A, p.Val37Ile, rs72474224; and c.269T>C, p.Leu90Pro,

Phenome-wide screening of the putative causal determinants of bipolar affective disorder using genetic data.

Bipolar affective disorder (BPAD) is a chronic psychiatric condition with high heritability (60-85%) and significant phenotypic variability, complicating its etiological understanding. Genome-wide association studies (GWAS) have revealed genetic overlaps with diverse traits, suggesting pleiotropy. This study aimed to identify shared genetic factors and infer causal relationships using large-scale GWAS data.BPAD GWAS summary statistics were obtained from the UK Biobank via the Complex Trait Genetics Virtual Lab (CTG-VL) platform. Genetic correlations with 1,504 traits were estimated using linka

Candidate Stress-Specific microRNA Expression in Pediatric Thrombosis and Healthy Children: A Comparative Analysis of Serum miR-34a-5p in Relation to Stress Levels and Inflammatory Genetic Traits.

BACKGROUND: Pediatric thrombosis (PT) is a rare clinical entity manifesting from neonatal life to adolescence with possibly severe clinical complications and increasing prevalence in the last decade. Its pathogenesis has been attributed to a multitude of factors, all of which share stress and inflammation as a common denominator. Although stress-induced inflammation, marked by the upregulation of proinflammatory interleukins, such as IL-1β, creates a prothrombotic environment, its relation with thrombosis remains incompletely defined, especially in the context of PT, which differs substantiall

ZDHHC5 may regulate the function of NKT cells and the immune response in Meniere's disease.

Meniere's disease (MD) is a heterogeneous, rare inner ear disorder characterized by recurrent vertigo, fluctuating sensorineural hearing loss, tinnitus, and aural fullness. Its molecular mechanisms remain unclear due to significant clinical and immunological heterogeneity. In this study, we integrated multi‑omics genetic data to systematically screen for palmitoylation regulatory genes associated with MD risk. We first intersected 31 core palmitoylation genes with blood cis‑eQTL datasets to identify candidate expression‑related genes, then performed two‑sample Mendelian randomization (MR) to e

Widespread benzimidazole resistance and β-tub-1 F200Y SNP in gastrointestinal nematodes of sheep across Portugal.

Anthelmintic resistance (AR) to benzimidazoles (BZs) in gastrointestinal nematodes (GINs) of sheep was assessed across 35 farms in northern (Trás-os-Montes, TM), north-central (Beira Interior, BI), central (Estremadura, ES), and southern (Alentejo, AL) Portugal. Pooled fecal samples were tested for AR by egg hatch tests (EHTs) with albendazole (ABZ) and thiabendazole (TBZ). Effective concentrations inhibiting 50% egg hatch (EC50) were calculated, with EC50 > 0.13 µg/mL (ABZ) and > 0.10 µg/mL (TBZ) indicating AR. ABZ resistance was detected on 69% of farms, while TBZ, never administered, showed

Causal association between blood immune cells and chronic hepatitis B: Univariable and multivariable Mendelian randomization.

Current evidence underscores the pivotal role of the immune system in the pathogenesis of chronic hepatitis B (CHB). Nevertheless, the precise causal nexus between immune cells and CHB remains enigmatic, with numerous unresolved queries. Leveraging data from the latest summary-level genome-wide association studies, we examined immune cell genetics in 3757 individuals from Sardinia. CHB data predominantly originated from the Biobank Japan project, encompassing 1394 cases and 211,059 controls. Our investigation entailed a comprehensive univariable Mendelian randomization (MR) analysis to dissect

Machine learning-driven discovery of host genetic factors for paratuberculosis in goats within the one health framework.

Paratuberculosis, caused by Mycobacterium avium subsp. paratuberculosis (MAP), remains a persistent One Health concern due to its slow clinical course, environmental resilience, wide circulation in ruminant systems, and unresolved zoonotic implications. To characterise MAP exposure across Türkiye’s goat populations, we conducted a nationwide genomic survey encompassing seven breeds from 36 farms in 11 provinces. High-density SNP genotyping combined with mutual-information–based feature preselection retained informative, non-redundant loci capturing both linear and nonlinear components of disea

Circulating metabolites associated with protection against Mycobacterium tuberculosis infection and inhibition of mycobacterial growth.

A significant proportion of individuals who are heavily exposed to infectious tuberculosis patients do not acquire Mycobacterium tuberculosis (Mtb) infection, as detected by an interferon gamma release assay (IGRA). We examined circulating metabolite profiles and metabolic genotypes in 199 heavily exposed IGRA-negative tuberculosis household contacts in Indonesia. Based on differentially abundant metabolites, activity of several pathways including arachidonic acid, arginine and proline, glutathione, and tryptophan metabolism, correlated with a negative IGRA at three months. SNPs near PRODH (in

Whole-genome sequencing and machine learning reveal candidate genomic features associated with Listeria monocytogenes persistence in two ice cream facilities.

Listeria monocytogenes is a major foodborne pathogen responsible for listeriosis. Ice cream has emerged as an important vehicle for L. monocytogenes transmission, yet genomic data describing strain persistence in ice cream production environments in China remain limited. In this study, we conducted whole-genome sequencing (WGS) of 57 L. monocytogenes isolates recovered from two ice cream production facilities in China between 2016 and 2018 to characterize population structure, persistence, and associated risk factors. Genomic typing revealed dominance of lineage II serotype 1/2a sequence type

Emergence of distinct Burkholderia pseudomallei lineages in Brazil: Phenotypic, genotypic, and phylogenetic agreement.

Burkholderia pseudomallei is a Gram-negative facultative intracellular bacterium that causes melioidosis, a neglected tropical disease that spreads beyond traditional endemic regions. This study examined B. pseudomallei isolates obtained from five melioidosis cases in Piauí, Brazil, using phenotypic, molecular, and genomic methods. Isolates were identified as B. pseudomallei using VITEK 2 system and MALDI-TOF mass spectrometry. Conventional PCR targeting the orf11, burk17, and burk475 gene regions confirmed species-level identification, with sequences showing 98.78-100% identity to GenBank B.

Identifying candidate genetic variants for egg number by analyzing over 1,000 fully sequenced layers.

BACKGROUND: Egg production over a long laying cycle until 700 days of age is preferred for modern layer chicken breeding. It is influenced by the onset of laying, stability during the peak period, and persistence at late laying stages. Conventional single-single nucleotide polymorphism (SNP) association analyses have identified additive loci, but few studies have explored dominance effects or integrated multiomics data to investigate the genetic basis of egg production traits from the onset to 700 days of age. A full diallel cross of 1,004 chickens was·subjected to whole-genome sequencing. Tra

Association of FOXP3 rs3761548 With Cancer: Systematic Review and Two Approaches of X-chromosome Genotypic Meta-analysis.

BACKGROUND/AIM: Cancer development involves complex interactions between immune mechanisms and the tumor microenvironment, with regulatory T cells (Tregs) being implicated in suppressing anti-tumor immunity. The X-linked gene Forkhead Box P3 (FOXP3), which regulates Tregs' function, and its promoter variant rs3761548 C>A have been widely studied for their role in tumorigenesis. This meta-analysis aims to re-evaluate the association between rs3761548 and cancer risk using two statistical approaches to account for sex-based genotypic differences and X-chromosome statistical challenges. Integration of genome and transcriptome reveals core genes and allele-specific expression genetic variants associated with immune differentiation of taurine ancestry dairy cattle from indicine ancestry cattle.

BACKGROUND: Disease is a key factor to shorten productive lifespan of dairy cattle. Shifting from classical 'treating existing diseases' to 'preventing diseases before they occur' is a challenge in high-producing dairy cattle. Immunity plays a crucial role in defending against disease. Substantial evidence indicates that the immune function of taurine ancestry cattle and indicine ancestry cattle are distinct. Therefore, this study aims: (1) to identify core immune genes (CIG) and allele-specific expression (ASE) variants by comparing taurine ancestry dairy cattle with indicine ancestry cattle;
