To identify potential regulators of Hox gene expression in mice, we have screened for genes highly related to brahma (brm), an activator of homeotic gene expression in Drosophila. We have cloned a murine gene, brg1, which, like brm, encodes a member of the DEGH protein family, suggesting that brg1 may be a DNA-dependent ATPase or a helicase. brg1 also contains a bromodomain which may be involved in transactivation. Although the sequences of a number of mammalian genes similar to Drosophila brm have been reported, they are related to brm only within specific portions of the putative helicase re

Ehlers-Danlos syndrome is a heterogeneous disorder characterized by joint hypermobility, skin hyperextensibility, fragility, and other signs of connective tissue involvement. In addition to these, the type VI variant of the disease has some special characteristics such as kyphoscoliosis and ocular abnormalities. The biochemical abnormality in most patients with this autosomal recessively inherited type VI variant is a deficiency in the activity of lysyl hydroxylase (EC 1.14.11.4), the enzyme catalyzing the formation of hydroxylysine in collagens and other proteins with collagen-like amino acid

Hot season is a major constraint to production and reproduction in buffaloes. The present work aimed to investigate the effect of season on ovarian function, developmental competence, and the relative abundance of gene expression in buffalo oocytes. Three experiments were conducted. In experiment 1, pairs of buffalo ovaries were collected during cold season (CS, autumn and winter) and hot season (HS, spring and summer), and the number of antral follicles was recorded. Cumulus oocyte complexes (COCs) were aspirated and evaluated according to their morphology into four Grades. In experiment 2, G

Transcription progression is governed by multitasking regulators including SPT5, an evolutionarily conserved factor implicated in virtually all transcriptional steps from enhancer activation to termination. Here we utilize a rapid degradation system and reveal crucial functions of SPT5 in maintaining cellular and chromatin RNA polymerase II (Pol II) levels. Rapid SPT5 depletion causes a pronounced reduction of paused Pol II at promoters and enhancers, distinct from negative elongation factor (NELF) degradation resulting in short-distance paused Pol II redistribution. Most genes exhibit downreg

Genetic mutations in dozens of monogenic genes can lead to serious podocyte dysfunction, which is a major cause of steroid-resistant nephrotic syndrome (SRNS). The NUP160 gene is expressed in both human kidney and mouse kidney. However, whether knockdown of NUP160 impairs podocytes has not yet been established. Therefore, we knocked down NUP160 by targeted short hairpin RNA (shRNA) in conditionally immortalized mouse podocytes and observed the effect of NUP160 knockdown on the proliferation, apoptosis, autophagy and cell migration of podocytes. We also investigated the effect of NUP160 knockdo

Inflammasomes are cytoplasmic sensors that regulate the activity of caspase-1 and the secretion of interleukin-1β (IL-1β) or interleukin-18 (IL-18) in response to foreign molecules, including viral pathogens. They are considered to be an important link between the innate and adaptive immune responses. However, the mechanism by which inflammasome activation occurs during primary Epstein-Barr virus (EBV) infection remains unknown. Human B lymphocytes and epithelial cells are major targets of EBV, although it can also infect a variety of other cell types. In this study, we found that EBV could in

Hemangiomas of the ovary are rare with a majority described as individual reports of unusual clinical presentations or morphologic findings. Both the expected and unexpected pathologic features of these tumors in the ovary are not well detailed. Therefore, we collected the largest series of ovarian hemangiomas to comprehensively define their clinicopathologic associations and examine the significance of hormone receptors in their pathogenesis. In addition, a novel EWSR1-NFATC1 fusion has recently been described in a case of hemangioma of bone. To our knowledge, EWSR1 rearrangement has not been

Segmentation of the vertebrate hindbrain into multiple rhombomeres is essential for proper formation of the cerebellum, cranial nerves and cranial neural crest. Paralog group 1 (PG1) hox genes are expressed early in the caudal hindbrain and are required for rhombomere formation. Accordingly, loss of PG1 hox function disrupts development of caudal rhombomeres in model organisms and causes brainstem defects, associated with cognitive impairment, in humans. In spite of this important role for PG1 hox genes, transcriptional targets of PG1 proteins are not well characterized. Here we use ectopic ex

MARVELD1 (MARVEL domain-containing 1) is a member of MARVEL domain-containing proteins and located on human chromosome 10q24.2. MARVELD1 has no significant similarity with other members of MARVEL domain family at amino acid level. Gene expression arrays demonstrated that MARVELD1 is widely expressed in normal human tissues and is down-regulated in primary multiple tumors derived from ovary, vulva, uterus, cervix, breast, testis, kidney, bladder and liver. The down-regulation of MARVELD1 was further identified by real-time PCR and immunohistochemical staining in primary breast cancer. In additi

The bcl-2 oncogene blocks programmed cell death (apoptosis). Epstein-Barr virus (EBV) can immortalize B lymphocytes into continuously growing lymphoblastoid cell lines (LCL) by the coordinate expression of at least 9 latent genes (EBV nuclear antigen [EBNA] 1-6, latent membrane protein [LMP], and terminal proteins [TP] 1 and 2). We analyzed transcription and expression of bcl-2 and latent EBV genes in Burkitt's lymphoma (BL) cell lines with a germinal center phenotype (group I) as well as activated BL cell lines (group III) and LCLs. We found high expression of bcl-2 as well as the full spectr

This study aims to investigate how emodin, an active component of Polygonum multiflorum, regulates macrophage polarization through the HDAC4/NF-κB/CXCL12 pathway and impacts the progression of hepatocellular carcinoma (HCC). The inhibitory effect of emodin on M2 polarization was assessed by collecting conditioned cell culture medium from HepG2 cells and treating macrophages with emodin. Network pharmacology and other experiments were used to identify HDAC4 as a key target, and this was further validated through molecular docking, surface plasmon resonance (SPR), and Western blot analysis. For decades, apoptosis has reigned supreme in cell death therapeutics, yet its clinical limitations in resistant cancers and degenerative diseases have unveiled the critical role of non-apoptotic regulated cell death (RCD) pathways, including ferroptosis, necroptosis, pyroptosis, and parthanatos. This review examines chromatin as a key regulatory layer influencing these pathways through dynamic histone modifications, DNA methylation, non-coding RNAs, and 3D genome architecture. We dissect how chromatin landscapes integrate metabolic, oxidative, and inflammatory signals in a cell-type- and line

A vital question in transcription regulation is how and to what extent transcription of different DNA species, including viral DNA, episomal DNA and endogenous retroelements, is differentially regulated to transcription of host genes. RPRD2 has previously been characterized as an HIV restriction factor that blocks reverse transcription. However, RPRD2 has also been characterized as a regulator of global transcription of host genes. Therefore, we hypothesized that RPRD2 may also regulate nascent transcription from HIV provirus and endogenous retroelements. First, we used a combination of experi

Epilepsy is a common chronic neurological disorder. Approximately one-third of patients respond poorly to existing anti-seizure medications. There is an urgent need for novel therapeutic strategies targeting the fundamental disease processes. Ferroptosis is an iron-dependent form of regulated cell death characterized by the accumulation of lipid peroxides. In recent years, its role in the pathological mechanisms of epilepsy has gained increasing attention. This review systematically elaborates the core molecular mechanisms of ferroptosis. These include dysregulated iron metabolism, failure of

N6-methyladenosine (m6A) methylation is the most common intramolecular modification in eukaryotic mRNA; its dynamic regulation depends on "writers" (methyltransferases: METTL3/METTL14/WTAP/VIRMA), "erasers" (demethylases: FTO/ALKBH5), and "readers" (binding proteins: YTHDF/YTHDC/IGF2BP families), thereby regulating RNA splicing, nuclear export, translation, and degradation. In rheumatoid arthritis (RA), this epigenetic network is severely disrupted: abnormal expression of writers leads to post-transcriptional activation of pro-inflammatory genes, while an imbalance in erasers compromises the s

OBJECTIVE: Cases of diffuse large B-cell lymphoma (DLBCL) of the uterus co-expressing CD5 are exceedingly rare and diagnostically challenging due to its nonspecific clinical and radiological features, which often mimic other uterine malignancies. This study retrospectively analyzes the cytopathological and histopathological characteristics of this entity in uterine cavity drainage fluid to facilitate its recognition.
METHODS: We analyzed a case of uterine CD5-positive DLBCL by collecting clinical data, imaging findings, cytology from uterine drainage fluid, and histology from endometrial cure

Non-syndromic orofacial clefts (nsOFCs), such as cleft lip, cleft lip with or without cleft palate (CL/P), and cleft palate only, are among the most common craniofacial malformations in humans. We identified a range of rare and de novo structural variants (SVs) associated with nsOFCs in a cohort of children of African and Asian ancestry from the Gabriella Miller Kids First Data Resource Center. Most of these novel candidate SVs are located in non-coding regions. More specifically, we characterized SVs associated with nsOFCs across the DOT1L (disruptor of telomeric silencing 1-like) genomic loc

Arabidopsis thaliana possesses hitherto unknown Occludin domain containing ELL ortholog identified as CARP9. The AtCARP9 protein similar to ELL orthologs across species, interacts with both the Arabidopsis EAF orthologs. Transcription is one of the early steps during which the expression of most protein-coding genes is controlled in cells. Accumulating evidence suggests that elongation of transcription is another dynamic step, which is regulated by 'transcription elongation factors'. ELL (eleven nineteen lysine rich leukemia) belongs to a family of conserved transcription elongation factors, w
