Mixed-field agglutination observed in column agglutination testing is not always associated with the A3 subgroup.

Mixed-field agglutination (MFA) can be observed in forward typing of samples from A3 individuals with serologic ABO typing methods. The results of column agglutination testing (CAT) and tube agglutination testing using different antibody clones can be discordant. In this report, we reveal our experience using polymerase chain reaction-sequence-based typing (PCR-SBT) of ABO exon 7 to clarify serologic method discordance of A subgroup blood typing in Northern Thai donors. A total of 21 group A blood donors with either MFA or weak agglutination on routine ABO CAT were recalled. CAT was repeated w

Screening for Celiac Disease in Irritable Bowel Syndrome: An Updated Systematic Review and Meta-analysis.

OBJECTIVES: Celiac disease (CD) and irritable bowel syndrome (IBS) share similar symptoms, leading to confusion between the two and diagnostic delay. International guidelines recommend screening individuals with IBS for CD, via serological testing. However, studies published recently have cast doubt on the utility of this. We updated a previous meta-analysis examining this issue. METHODS: MEDLINE, EMBASE, and EMBASE Classic were searched through to May 2016. Eligible studies recruited adults with IBS according to symptom-based criteria, physician's opinion, or questionnaire data. Tests for CD

Serology for trachoma surveillance after cessation of mass drug administration.

BACKGROUND: Trachoma, caused by Chlamydia trachomatis (Ct), is the leading infectious cause of blindness worldwide. Yearly azithromycin mass … ough our use of catalytic modelling overcomes some of these limitations. CONCLUSIONS/SIGNIFICANCE: Serologic tests support NAAT findings of very low to zero prevalence of ocular Ct in this community and have potential to provide objective measures of transmission and useful surveillance tools for trachoma elimination programs.

Cerebral Schistosoma japonicum infection confirmed by histopathology and PCR: Diagnostic and therapeutic challenges in multiple parasitic coinfections.

Cerebral schistosomiasis, a rare manifestation of schistosomiasis, is challenging to diagnose due to non-specificity in the clinical and radiologic findings and cross-reactivity in serologic testing with other helminths. Coinfection with multiple parasites further complicates diagnosis and treatment. A previously healthy 32-year-old Filipino man presented with a seizure. Brain magnetic resonance imaging (MRI) revealed multiple clustered enhancing nodules with surrounding vasogenic edema in the left frontal lobe. Serologic testing was positive for anti-Schistosoma japonicum, Strongyloides, and

Challenges in Diagnosing and Managing Hepatitis E in Transplant Recipients: A Case Report.

Hepatitis E virus (HEV) is a common but often underdiagnosed cause of hepatitis in both the general population and transplant patients. Diagnosis is usually made by detecting IgM anti-HEV antibodies, with immunosuppressed individuals being at higher risk for chronic infection. We present the case of a 56-year-old man who developed asymptomatic liver cytolysis 4 months after a kidney transplant, with no significant physical findings or relevant epidemiological history. Abdominal ultrasound revealed hepatic steatosis. Drug-induced liver injury was suspected, prompting the discontinuation of pote

Dynamics of Borrelial Antibodies in European Lyme Borreliosis Patients with Erythema Migrans.

Background/Objectives: The diagnosis of typical erythema migrans (EM) is clinical, and serological testing for Borrelia-specific antibodies is not required. However, characterization of the immune response in patients with a reliable clinical diagnosis may improve the interpretation of serological findings in early Lyme borreliosis (LB) without characteristic skin lesions. Methods: Serum IgM and IgG antibodies to Borrelia burgdorferi sensu lato were evaluated in 1356 Slovenian adults with EM at baseline and at a follow-up visit approximately two months after treatment initiation, using a chemi

Diagnostic outcomes after gluten challenge in adult patients with unconfirmed coeliac disease already on a gluten-free diet: A 20-year retrospective cohort study.

BACKGROUND: In uncertain cases of coeliac disease (CD), gluten challenge (GC) may be necessary to confirm or exclude the diagnosis. However, data on diagnostic outcomes after GC are limited. AIMS: We aimed to evaluate outcomes after GC in patients with unconfirmed CD who had already started a gluten-free diet (GFD), and identify predictors of a confirmed diagnosis. METHODS: Patients with unconfirmed CD already on a GFD, who underwent GC and subsequent testing with endomysial antibodies (EmA) and duodenal biopsy between 06/2000-06/2021 were included. Clinical data, prior test results, and fin

Screening, Prevalence and Management of Chronic Viral Hepatitis C in Mental Health Setting: Towards the Eradication of A Forgotten Reservoir.

The rate of HCV infection is higher in people with psychiatric conditions than in the general population. However, access to somatic care for populations with mental or dual diagnoses remains insufficient. These circumstances could constitute a barrier to the eradication of HCV infection. A retrospective study was performed in the public mental health institution of Ville Evrard. Screening for HCV infection should be systematically proposed to patients attending the institution. On-site assessment of liver disease severity and treatment with direct-acting antivirals prescribed by physicians no

Pepsinogen II in gastritis and Helicobacter pylori infection.

BACKGROUND AND AIM: In the gastric mucosa, pepsinogen II (PgII) is produced/secreted by glands in the mucus-secreting antral and cardia compartments, but also by the chief cells and the oxyntic glands. Increasing PgII serum levels are associated with the whole spectrum of gastric inflammatory diseases, including gastritis induced by Helicobacter pylori (H. pylori). This review critically addresses the clinical value of PgII serology for assessing gastric mucosal inflammation, and as a marker of H. pylori status, in both H. pylori-positive patients and after eradication therapy. RESULTS: A sea

Validation of a Harmonized Enzyme-Linked-Lectin-Assay (ELLA-NI) Based Neuraminidase Inhibition Assay Standard Operating Procedure (SOP) for Quantification of N1 Influenza Antibodies and the Use of a Calibrator to Improve the Reproducibility of the ELLA-NI With Reverse Genetics Viral and Recombinant Neuraminidase Antigens: A FLUCOP Collaborative Study.

Current vaccination strategies against influenza focus on generating an antibody response against the viral haemagglutination surface protein, however there is increasing interest in neuraminidase (NA) as a target for vaccine development. A critical tool for development of vaccines that target NA or include an NA component is available validated serology assays for quantifying anti-NA antibodies. Additionally serology assays have a critical role in defining correlates of protection in vaccine development and licensure. Standardisation of these assays is important for consistent and accurate re

Celiac disease: Understandings in diagnostic, nutritional, and medicinal aspects.

Celiac disease (CD) is characterized by clinical polymorphism, with classic, asymptomatic or oligosymptomatic, and extra-intestinal forms, which may lead to diagnostic delay and exposure to serious complications. CD is a multidisciplinary health concern involving general medicine, pediatric, and adult gastroenterology, among other disciplines. Immunology and pathology laboratories have a fundamental role in diagnosing and monitoring CD. The diagnosis consists of serological testing based on IgA anti-transglutaminase (TG2) antibodies combined with IgA quantification to rule out IgA deficiency,

Immune responses to SARS-CoV-2 in three children of parents with symptomatic COVID-19.

Compared to adults, children with severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) have predominantly mild or asymptomatic infections, but the underlying immunological differences remain unclear. Here, we describe clinical features, virology, longitudinal cellular, and cytokine immune profile, SARS-CoV-2-specific serology and salivary antibody responses in a family of two parents with PCR-confirmed symptomatic SARS-CoV-2 infection and their three children, who tested repeatedly SARS-CoV-2 PCR negative. Cellular immune profiles and cytokine responses of all children are similar to t

Contagious epididymitis due to Brucella ovis: relationship between sexual function, serology and bacterial shedding in semen.

BACKGROUND: Contagious Epididymitis (CE) due to Brucella ovis (B. ovis) is a contagious disease that impairs rams' fertility due to epididymis, testicle and accessory sexual gland alterations. An increased incidence of CE has been observed in South Eastern France ("PACA" region) since the Rev.1 vaccination against B. melitensis has been stopped in 2008. The objective of this study was to evaluate the relationship between the infection by B. ovis and the sexual function of rams. Two-hundred eighteen sexually-mature rams, from 11 seropositive flocks, were submitted to a clinical examination of t
