Actions of putative transmitters in the chicken vagus nerve/oesophagus and Remak nerve/rectum preparations.

1 Histamine (0.02-0.1 mug/ml) contracted the chicken rectum preparation. This effect was antagonized by mepyramine (0.01 mug/ml) but not by hyoscine (0.02 mug/ml).2 5-Hydroxytryptamine (0.05-0.25 mug/ml) relaxed the rectum preparation and at higher concentration produced a biphasic response. These responses were not antagonized by methysergide (0.01 mug/ml), and the relaxation was not antagonized by tetrodotoxin (0.1 mug/ml) or a combination of propranolol (0.05 mug/ml) and phentolamine (0.1 mug/ml).3 Neither mepyramine (0.1 mug/ml) nor methysergide (0.01 mug/ml) antagonized the contractions p

Reflex discharge patterns of cardiac vagal efferent fibres.

1. Unit activity was recorded from single and few fibre preparations in a cardiac branch of the right vagus nerve of the cat.2. Increases in blood pressure mediated solely by the right carotid sinus nerve produced bradycardia when all other nerves to the heart had been cut. Myelinated fibres in the cardiac branch of the right vagus nerve were reflexly activated by the same procedure.3. The fibres were silent when blood pressure was below 140-150 mm Hg. As the pressure began to rise, they discharged phasically with the cardiac cycle. At pressures greater than 180 mm Hg, the discharge was contin

Microglial reactivity in the hippocampal CA2 is associated with advanced neuronal α-synucleinopathy.

Lewy body diseases are thought to evolve by the spread of intraneuronal α-synuclein pathology. However, staging models of α-synuclein pathology in the human brain seldom evaluate regions with direct synaptic connectivity to model microglial processes in disease. Here, we address this gap by testing the hypothesis that, within the well-defined synaptic connectivity of the intrahippocampal circuit, neuronal α-synuclein pathology is associated with microglial reactivity. We selected a cohort of autopsy-confirmed Lewy body disease patients with hippocampal neuronal α-synuclein pathology and minima

A phase II study repurposing atomoxetine for neuroprotection in mild cognitive impairment.

The locus coeruleus is the initial site of Alzheimer's disease neuropathology, with hyperphosphorylated Tau appearing in early adulthood followed by neurodegeneration in dementia. Locus coeruleus dysfunction contributes to Alzheimer's pathobiology in experimental models, which can be rescued by increasing norepinephrine transmission. To test norepinephrine augmentation as a potential disease-modifying therapy, we performed a biomarker-driven phase II trial of atomoxetine, a clinically-approved norepinephrine transporter inhibitor, in subjects with mild cognitive impairment due to Alzheimer's d

A De Novo Arg649Trp Variant in GluA2 Impairs Its Assembly With GluA1: Associated With Epilepsy and Neurodevelopmental Disorders.

AMPA-type glutamate receptors (AMPARs) are the main mediators for fast excitatory synaptic transmission in the brain. Among AMPAR subunits, GluA2 is unique because of its role in forming Ca2+-impermeable heteromeric receptors with GluA1/A3/A4, a property dictated by an arginine residue (GluA2(R)) in the pore region. In contrast, GluA2(R)-lacking AMPARs are Ca2+-permeable. Through whole-exome sequencing (WES), we identified a de novo GRIA2 variant (c.1945A>T, p.Arg649Trp) in a patient with neurodevelopmental disorders (NDDs) and epilepsy. Heterologous expression in HEK293T cells revealed that h…

Cholesterol-dependent modulation of β-amyloid 1-40 peptide aggregation by membrane organization.

HYPOTHESIS: Cholesterol (chol) is a key lipid for nervous system development and function. While aging is associated with reduced brain chol, Alzheimer's disease (AD) is characterized by altered chol metabolism and asymmetric membrane distribution, particularly between neuronal body and synaptic compartments. The impact of these changes on β-amyloid (Aβ) aggregation, a central event in AD pathogenesis, remains unclear-especially under physiologically low Aβ concentrations. We hypothesize that Aβ nucleation in colloidal systems requires chol as an initial anchoring site, but that progression to

Transneuronal Transport of Ligand-Conjugated Upconversion Nanoparticles from the Peripheral to the Central Nervous System.

Despite significant advancements in neuro-nanotechnology, the movement of nanomaterials between peripheral and central neurons for applications such as cargo delivery, imaging, and tracking remains a formidable challenge. This limitation highlights the critical necessity for developing engineered materials that can facilitate efficient intra-axonal transport and trans-synaptic translocation. In this study, we report the design and synthesis of core-shell-shell upconversion nanoparticles (CSS-UCNPs) featuring a NaYF4:YbEr core sequentially coated with active NaYF4:YbNd and inert NaYF4 shells. Human Astrocytes Express Functional Protease-Activated Receptors that Increase Their Reactive Phenotype Under Certain Culture Conditions.

BACKGROUND: Protease-activated receptors are G protein-coupled receptors with a novel activation mechanism by which proteases cleave the N-terminus revealing a tethered ligand that binds to the second extracellular loop leading to receptor activation. We have previously shown that protease-activated receptor 2 (PAR2) activation impairs hippocampal synaptic transmission and is neuroprotective in in vitro excitotoxic assays, both of which are astrocyte-dependent. We have also shown that AC264613, a blood-brain barrier-permeable PAR2 activator, results in behavioural changes associated with depre…

Alcohol drinking sex-dependently regulates interleukin-1 pro-inflammatory signaling in the prefrontal cortex of mice and rhesus macaques.

Alcohol use disorder (AUD) causes executive dysfunction, leading to reduced treatment adherence, worse clinical outcomes, and relapse. Current medications do not address these symptoms. Targeting the neuroimmune interleukin-1 (IL-1) system has potential, but its mechanistic role in prefrontal cortex (PFC) dysfunction is unclear. Here we investigated how chronic alcohol consumption shifts IL-1 regulation of the rodent and non-human primate PFC. We found that alcohol drinking decreased reference memory and shifted the search strategy in mice of both sexes, and this deficit correlated with altere

RO4938581, a GABAA-α5 negative allosteric modulator rescued behavioral and EEG phenotypes of a mouse model of Dup15q syndrome.

We investigated the correlation between increased gene copy number of gamma aminobutyric acid type A (GABAA) receptor α5-containing subunits and electrophysiological and behavioral phenotypes in a mouse model of Dup15q syndrome (15q dup) and tested the hypothesis that selectively inhibiting the activity of GABAA-α5 receptors may have therapeutic effects. Dup15q syndrome is a rare neurodevelopmental disorder caused by copy number gains of the 15q11.2-q13.1 chromosomal region, which includes UBE3A and a cluster of three genes (GABRA5, GABRB3, and GABRG3) encoding GABAA receptor subunits, all of
