Hemoglobin solution as a plasma expander.

Stroma-free homoglobin solution has received extensive experimental trials in dogs and nonhuman primates. It has been shown to have no coagulant activity and to have no deleterious effect on renal function. Also pathological examination of autopsy and biopsy specimens fails to demonstrate any kidney damage after infusion of large amounts of stroma-free hemoglobin solution. In vivo data would also indicate that the solution can both carry oxygen and liberate it into tissues despite the fact that its in vitro oxygen dissociation curve is shifted to the left. Animals can be maintained with a 3% h…

Genomic structural variations distinguishing archaic and modern humans are associated with neurodevelopment.

Understanding the divergence in neurodevelopment between extinct archaic hominins and modern humans is a central goal in evolutionary biology. Structural variants (SVs) are key genomic drivers of phenotypic change. Here, we identified a set of SVs present in archaic humans by screening fixed SV events from diverse non-human primate (NHP) lineages, termed archaic hominin structural variants (AHSVs). AHSVs are significantly enriched in genes governing brain development and function, many of which are implicated in neuropsychiatric disorders. We found that several AHSVs overlap with brain cis-reg

Sensitivity to permanent non-functionality in Goffin's cockatoos (Cacatua goffiniana).

While the ability to recognize that something is irreparably broken is important for human physical and social cognition, its underpinnings remain largely unexplored in non-human animals. Recognizing the permanent loss of function is highly relevant to foraging on perishable resources and to interacting with breakable objects such as wooden tools. Nevertheless, learning how to respond to such permanent non-functionality has broader implications, as it has been proposed to represent an important prerequisite for the development of a minimal concept of death. The Goffin's cockatoo, an opportunis

Ferrets and genetically modified ferrets as model organisms in biomedical research: a review.

Domestic ferrets have served as experimental animals for over a century, offering a balance between practicality and biological relevance while bridging the gap between rodent and non-human primate models. Their anatomical and physiological similarities to humans, particularly in the respiratory and nervous systems, make ferrets an invaluable model for biomedical research across various disciplines. Recent advances in genome-editing technologies have enabled the efficient creation of genetically modified (GM) ferrets, significantly expanding their applications in basic and translational resear

PRMT5 inhibition promotes cross-species spermatogonia expansion and suppresses differentiation.

Spermatogonial stem cells (SSCs) hold great promise for treating male infertility, but their clinical translation is impeded by the lack of optimal conditions to maintain their undifferentiated state in vitro. In this study, we focused on epigenetic regulators upregulated during differentiation as potential targets. Through a small-molecule screen targeting such conserved regulators, we found that PRMT5 inhibition suppressed mouse SSC differentiation and enhanced their proliferation in a GDNF-deficient, differentiation-prone microenvironment in vitro. Using SSC transplantation assays, we confi

Inferential planning in the frontal cortex.

How the brain plans and maintains sequences of future actions remains a central question in systems neuroscience. Studies in the frontal cortex revealed that multiple elements of a sequence are represented simultaneously in separable neural subspaces, challenging classical sequential planning models. Here, we show that these representations emerge naturally under inferential planning, in which sequential actions are inferred from sensory evidence and goals. Using a hierarchical generative model, we reproduce key neural phenomena observed in the primate frontal cortex, including the simultaneou

Role of Oxytocin in Obesity.

PURPOSE OF REVIEW: This review will summarize the role of the hormone, oxytocin (OT), in the control of energy balance and glucose homeostasis in pre-clinical and clinical models and the role of sex in contributing to these effects. RECENT FINDINGS: OT reduces intake of chow and highly palatable foods, including sucrose, high fat diet, and chocolate cookies, suggesting that it suppresses homeostatic and hedonic feeding. Chronic administration reduces body weight in diet-induced obese (DIO) rodent models. These effects occur through a selective reduction of fat mass while sparing lean mass. Wh

Animal models of chronic graft-versus-host disease with oral involvement: A review.

Chronic graft-versus-host disease (cGVHD) remains a significant complication following allogeneic hematopoietic stem cell transplantation, with the oral cavity being one of the most frequently affected sites. Oral cGVHD manifests mucosal inflammation, salivary gland dysfunction, and fibrosis, all of which substantially impair patients' quality of life. To date, research efforts have predominantly focused on clinical investigations, with relatively limited exploration of animal models for in vivo studies, despite animal models playing a pivotal role in deciphering the pathobiology of cGVHD and

Single-cut gene therapy in a one-step generated rhesus monkey model of Duchenne muscular dystrophy.

Progress in Duchenne muscular dystrophy (DMD) treatment is hindered by the lack of animal models that closely replicate human pathology and enable the evaluation of therapy efficacy and safety based on these models. To address this need, we optimize the generation of nonhuman primate DMD models, reducing the development time from 6 to 7 years to under 1 year, enabling the rapid generation of DMD monkey models. These models closely mimic human DMD pathology and motor dysfunction, making them suitable for testing gene therapies. Using these models, we develop a single-cut gene therapy strategy t

The flexible, the stereotyped and the in-between: putting together the combinatory tool use origins hypothesis.

Tool use research has long made the distinction between tool using that is considered learned and flexible, and that which appears to be instinctive and stereotyped. However, animals with an inherited tool use specialisation can exhibit flexibility, while tool use that is spontaneously innovated can be limited in its expression and facilitated by predispositions for ecological specialisations. Furthermore, recent evidence does not support the proposed division of flexible tool use along primate-bird taxonomic lines. Instead, we hypothesise that tool use is a more complementary phenomenon than

Notes on the threadworm Strongyloides fuelleborni (Nematoda: Strongyloididae) in vervet monkeys (Chlorocebus pygerythrus) and zoonotic strongyloidiasis in southern Malawi.

We sought to ascertain whether zoonotic strongyloidiasis occurred in vervet monkeys (Chlorocebus pygerythrus), a non-human primate (NHP) species becoming increasingly common in Southern Malawi. Faecal collection took place in four locations: Nyala Park, a private wildlife reserve adjacent to a sugarcane plantation in Chikwawa District, and three public locations, each near tourist lodges in Mangochi District. Our sampling took place during July 2023, when 32 faecal samples were inspected with parasitological methods. After faecal culture, threadworm larvae were noted in both districts that wer

Biochemical Characterisation of the Short Isoform of Histone N-Terminal Acetyltransferase NAA40.

N-alpha-acetyltransferase 40 (NAA40) is an evolutionarily conserved N-terminal acetyltransferase (NAT) linked to oncogenesis and chemoresistance. A recent study reported the generation of a second, shorter NAA40 isoform (NAA40S) through alternative translation, which we proceeded to further characterise. Notably, recombinant NAA40S had a greater in vitro enzymatic activity and affinity towards its histone H2A/H4 substrates compared to full-length NAA40 (NAA40L). Within cells, NAA40S was enzymatically active, based on its ability to suppress the H2A/H4S1Ph antagonistic mark in CRISPR-generated

Convolutional neural networks develop major organizational principles of early visual cortex when enhanced with retinal sampling.

Primate visual cortex exhibits key organizational principles: cortical magnification, eccentricity-dependent receptive field size and spatial frequency tuning as well as radial bias. We provide compelling evidence that these principles arise from the interplay of the non-uniform distribution of retinal ganglion cells, and a quasi-uniform convergence rate from the retina to the cortex. We show that convolutional neural networks outfitted with a retinal sampling layer, which resamples images according to retinal ganglion cell density, develop these organizational principles. Surprisingly, our re

Alpha-gal syndrome (AGS) in a glaucoma suspect with narrow iridocorneal angles.

PURPOSE: Alpha-gal syndrome (AGS) is an allergy to non-primate mammalian carbohydrate (galactose-alpha-1,3-galactose) which may cause anaphylaxis. Allergic patients must avoid ophthalmic drugs containing animal-derived ingredients. OBSERVATIONS: We report a 59-year-old non-Hispanic white woman who was referred for a glaucoma evaluation. She had been diagnosed with AGS after a tick bite in 2017. Ophthalmic exam revealed potentially occludable, narrow iridocorneal angles and laser iridotomy was recommended. Prior to performing the iridotomy, we investigated the ophthalmic medications required f

Programming cytomegalovirus as an HIV vaccine.

The initial development of cytomegalovirus (CMV) as a vaccine vector for HIV/simian immunodeficiency virus (SIV) was predicated on its potential to pre-position high-frequency, effector-differentiated, CD8+ T cells in tissues for immediate immune interception of nascent primary infection. This goal was achieved and also led to the unexpected discoveries that non-human primate (NHP) CMVs can be programmed to differentially elicit CD8+ T cell responses that recognize viral peptides via classical MHC-Ia, and/or MHC-II, and/or MHC-E, and that MHC-E-restricted CD8+ T cell responses can uniquely med

Novel Case Presentation of Abulia After Lone Star Tick Bite As Evidenced by Raised Titers of Alpha-Gal Specific IgM Immunoglobulin and a Possibility of Alpha-Gal Driven Hypothalamic Dysfunction As the Pathomechanism.

Galactose-α-1,3-galactose, an oligosaccharide epitope better acknowledged as α-Gal, is present in non-primate mammal meat, tick bites, microorganisms, and vaccines as a glycoprotein or glycolipid moiety. This can manifest hyperimmune reactions as it enters the human body, known as α-Gal syndrome (AGS). Complement-targeted therapies in kidney transplantation-insights from preclinical studies.

Aberrant activation of the complement system contributes to solid-organ graft dysfunction and failure. In kidney transplantation, the complement system is implicated in the pathogenesis of antibody- and cell-mediated rejection, ischemia-reperfusion injury, and vascular injury. This has led to the evaluation of select complement inhibitors (e.g., C1 and C5 inhibitors) in clinical trials with mixed results. However, the complement system is highly complex: it is composed of more than 50 fluid-phase and surface-bound elements, including several complement-activated receptors-all potential therape

Functional Insulinomas in a Rhesus Macaque (Macaca mulatta).

We report a rare case of functional insulinomas in a 16.7-year-old female Rhesus macaque (Macaca mulatta) that was presented with neuroglycopenic signs to the breeding colony hospital at the Tulane National Primate Research Center. At initial and follow-up examinations, the animal was consistently hypoglycaemic and was clinically maintained with additional fruits, other high-sugar food items and dextrose supplementation. Occasional episodes of seizure and collapse resolved quickly on administration of high-sugar food items. At necropsy, the uncinate process of the pancreas had a 2.2 cm diamete

FATAL FASCIOLOIDES MAGNA IN A LESSER SPOT-NOSED GUENON (CERCOPITHECUS PETAURISTA).

A 4-yr-old male intact lesser spot-nosed guenon (Cercopithecus petaurista), housed at a North American zoological facility, presented with acute lethargy, inappetence, and mild neurologic signs. Physical examination revealed hemorrhagic pleural effusion in the right hemithorax. This guenon's condition improved over several days but then deteriorated, and the guenon presented with lethargy and weakness. A hemorrhagic pleural effusion was identified within the left hemithorax. The guenon developed respiratory and cardiac arrest while anesthetized. Gross examination revealed tract formation in th

The posterior parietal cortex contributes to visuomotor processing for saccades in blindsight macaques.

Patients with damage to the primary visual cortex (V1) lose visual awareness, yet retain the ability to perform visuomotor tasks, which is called "blindsight." To understand the neural mechanisms underlying this residual visuomotor function, we studied a non-human primate model of blindsight with a unilateral lesion of V1 using various oculomotor tasks. Functional brain imaging by positron emission tomography showed a significant change after V1 lesion in saccade-related visuomotor activity in the intraparietal sulcus area in the ipsi- and contralesional posterior parietal cortex. Single unit
