Serum Levels of miR-223-3p and miR-223-5p in Prostate Diseases.

INTRODUCTION: Prostate Cancer (PCa) is the second most common cancer in males and the fifth in cancer-associated mortality. Although the Prostate-Specific Antigen (PSA) test is widely used in PCa screenings, it has significant limitations in the differential diagnosis of PCa. Therefore, studies on developing new biomarkers for PCa diagnosis are ongoing. MiRNAs are good candidate biomarkers for the diagnosis of cancers, including prostate cancer, as they can be easily detected from circulation. OBJECTIVE: In this study, it is aimed to determine the diagnostic value of serum levels of miR-223-3

Neglected Tropical Diseases: The Potential Application of microRNAs for Monitoring NTDs in the Real World.

Neglected Tropical Diseases (NTDs) are a common health problem and require an efficient campaign to be eradicated from tropical countries. Almost a million people die of NTDs every year in the world, and almost forty percent of the patients are under 20 years. Mass Drug Administration (MDA) is an effective tool for eradication of this health condition. However, a monitoring system is required to evaluate treatment-response and early detection of the re-emerging NTD. The relevance of current tests depends on good quality of the specimen. Thus, new molecular methods with high sensitivity and spe

Update on the Regulation of HIPK1, HIPK2 and HIPK3 Protein Kinases by microRNAs.

UNLABELLED: The Homeodomain-Interacting Protein Kinases (HIPKs) HIPK1, HIPK2 and HIPK3 are Ser/Thr kinases which interact with homeobox proteins and other transcription factors, acting as transcriptional coactivators or corepressors. HIPKs contribute to regulate several biological processes, such as signal transduction, apoptosis, embryonic development, DNA-damage response, and cellular proliferation, in response to various extracellular stimuli. Recently it has emerged that, in addition to their role in cancer, fibrosis and diabetes, HIPKs may also be involved in other human diseases, includi

Prognostic and Therapeutic Implications of MicroRNA in Malignant Pleural Mesothelioma.

Malignant Pleural Mesothelioma (MPM) is an aggressive disease characterized by a dismal prognosis, mainly due to late diagnosis. To date, there are very few treatment options available and the refractoriness to the majority of therapeutic strategies, leading to consider MPM a relevant problem in public health. Therefore, the identification of novel prognostic markers and alternative therapeutic strategies remain a top priority. Several efforts have been made in this direction and to date a number of studies have investigated the role of microRNA as biomarkers in MPM, identifying the potential

miR-29c-3p is an Effective Biomarker of Abdominal Aortic Aneurysm in Patients Undergoing Elective Surgery.

BACKGROUND: Abdominal aortic aneurysm (AAA) is usually asymptomatic and mostly diagnosed incidentally. Aortic dilatation progresses along with the wall weakening and eventually leads to a life-threatening rupture. Currently, there are no effective screening biomarkers for AAA. MicroRNAs, a class of small noncoding RNA molecules, offer great potential as biomarkers because of their stability in the bloodstream and expression profile specific for different diseases. OBJECTIVE: To assess the circulating miR-29c-3p as a potential biomarker of AAA and to elucidate the biological functions of miR-2

Delayed Cell Cycle Progression in STHdh(Q111)/Hdh(Q111) Cells, a Cell Model for Huntington's Disease Mediated by microRNA-19a, microRNA-146a and microRNA-432.

Several indirect evidences are available to indicate that abnormalities in cell cycle may contribute to pathogenesis of Huntington's disease (HD). Here, we show that the cell cycle progression in STsdh(Q111)/Hdh(Q111)cells, a cell model of HD, is delayed in S and G2-M phases compared to control STHdhQ7/HdhQ7cells. Expression of 17 genes, like PCNA and CHEK1, was increased in STHdh(Q111)/Hdh(Q111)cells. Increased expressions of PCNA, CHEK1 and CCNA2, and an enhanced phosphorylation of Rb1 were observed in primary cortical neurons expressing mutant N-terminal huntingtin (HTT), R6/2 mice and STHd

Involvement of microRNA181a in differentiation and cell cycle arrest induced by a plant-derived antioxidant carnosic acid and vitamin D analog doxercalciferol in human leukemia cells.

1,25-dihydroxyvitamin D3 (1,25D) has been shown to influence differentiation, cell proliferation and cell death in cultured leukemia cells. However, its clinical use is limited by its hypercalcemic effects. An analog of 1,25D, doxercalciferol (1-D2), has anti-tumor activity, with markedly reduced calcemic effects, which makes it a potential agent for clinical treatment of AML. Previous studies suggested that the combination of 1,25D with other agents, such as plant-derived antioxidants, can have additive or synergistic anti-cancer activities in leukemia cells. Here we report that 1-D2 induced

The miR-29 Family in Virus-Associated Cancers: Mechanisms, Biomarkers, and Therapeutic Opportunities.

The miR-29 family (miR-29a, miR-29b, and miR-29c) demonstrates context-depend-ent roles in cancers associated with oncogenic viruses (HPV, HBV/HCV, EBV, and HTLV-1), which collectively contribute to 15-20% of human malignancies. This comprehensive review ex-amines evidence that miR-29 primarily functions as a tumor suppressor by targeting DNMT3A, PTEN, and MCL-1, thereby regulating proliferation, apoptosis, and metastasis, though it exhibits paradoxical oncogenic activity in specific contexts, such as HBV-related Hepatocellular Carci-noma (HCC). Clinical data reveal virus-specific expression p

MicroRNAs in Cystic Fibrosis: Dual Role in Pathogenesis and Therapeutic Potential.

Cystic fibrosis (CF) is a severe inherited disorder caused by dysfunction of the cystic fibrosis transmembrane conductance regulator (CFTR), an epithelial ion channel essential for maintaining airway surface hydration and effective mucociliary clearance. Although over a thousand CFTR mutations have been described, their functional consequences vary widely, with some genotypes retaining partial CFTR activity and presenting as residual-function phenotypes. Despite CFTR modulator therapies substantially improved clinical outcomes in many patients, their efficacy remains incomplete and strongly de

LncRNAs at the Crossroads of Breast Cancer Metastasis: Emerging Mechanisms and Therapeutic Prospects.

Breast cancer metastasis is one of the leading causes of cancer-related mortality in women, which occurs through an intricate molecular network and cellular events. Over time, lncRNAs have emerged as an important regulator of metastatic progression. These RNAs affect important processes including EMT, ECM remodelling, angiogenesis, immune evasion, and can-cer stemness. LncRNAs regulate this process by acting as molecular decoys, scaffolds, guides, or competing endogenous RNAs, thereby controlling gene expression at both transcriptional and post-transcriptional levels. Various lncRNAs, includin

Do Circulating miRNAs Exhibit Distinct Patterns in Coronary Artery Disease Patients?

INTRODUCTION: Coronary artery disease (CAD) remains a leading cause of morbidity and mortality worldwide. Identifying reliable biomarkers for early diagnosis and understanding the underlying molecular mechanisms are critical for improving patient outcomes. Circulating microRNAs (miRNAs) have emerged as promising non-invasive biomarkers due to their stability in blood and involvement in various pathophysiological processes. This pilot case-control study aims to perform small RNA sequencing using next-generation sequencing in the plasma of Indian CAD patients to identify differentially expressed

miR-139 is a Tumor Suppressor microRNA with a Guanine-rich Precursor Terminal Loop.

INTRODUCTION/OBJECTIVE: MicroRNAs (miRNAs) are key post-transcriptional regulators of gene expression that modulate cancer-related processes, including cell growth, proliferation, differentiation, and invasion. While numerous oncogenic (oncomiRs) and tumor suppressor (TS) miRNAs have been characterized, structural features distinguishing these groups are not fully understood. This study investigates whether guanine (G) enrichment in precursor miRNA (pre-miRNA) terminal loop (TL) regions across the human miRNome is associated with tumor-suppressive activity in lung cancer. METHODS: We analyzed

In Silico Analysis of Potential miRNAs-EGFR Binding Complex for EGFR-TKI Resistance Lung Cancer Cells.

INTRODUCTION: Overexpression of Epidermal Growth Factor Receptor (EGFR) plays a critical role in the progression and high mortality of Non-Small Cell Lung Cancer (NSCLC). Although EGFR tyrosine kinase inhibitors (EGFR-TKIs) demonstrate promising efficacy as first-line therapy, acquired resistance remains a major clinical challenge. Increasing evidence suggests that microRNAs (miRNAs) regulate EGFR signaling pathways and may influence therapeutic response and the development of resistance. METHODS: This study aimed to identify EGFR-targeting microRNAs (miRNAs) and evaluate their binding stabil

mRNA Vaccines: Development, Mechanisms and Advances in Delivery Systems.

For more than over two decades, mRNA vaccines have been successfully administered and continues to be a promising platform for preventive and therapeutic uses. A key advantage of mRNA-based therapies over DNA-based approaches is that the mRNA molecule only needs to reach the cytoplasm for translation, bypassing the need for nuclear entry. Unlike pre-designed peptide vaccines, which may be restricted to specific MHC haplotypes, mRNA vaccines encode full-length antigens, allowing the host's cells to process and present a diverse array of epitopes suitable for a wide range of MHC haplotypes withi
