Nanoscale Metal-Organic Frameworks for Cancer Immunotherapy.

Cancer immunotherapy, particularly checkpoint blockade immunotherapy (CBI), has revolutionized the treatment of some cancers by reactivating the antitumor immunity of hosts with durable response and manageable toxicity. However, many cancer patients with low tumor antigen exposure and immunosuppressive tumor microenvironments do not respond to CBI. A variety of methods have been investigated to reverse immunosuppressive tumor microenvironments and turn "cold" tumors "hot" with the goal of extending the therapeutic benefits of CBI to a broader population of cancer patients. Immunostimulatory ad

Single-cell transcriptomics reveals ADRB1 as a key immune checkpoint regulating T cell exhaustion in lung cancer and its prognostic value in lung cancer.

BACKGROUND: Lung cancer, LUAD, and LUSC are both among the leading causes of cancer-related deaths. T cell exhaustion within the tumor microenvironment compromises the immune response and limits the benefits of immune checkpoint therapy. Adrenergic signaling primarily through ADRB1 induces T cell exhaustion and immune suppression. ADRB1 + T cells have not yet been studied regarding their role in lung cancer progression or prognosis. scRNA-seq analysis for tumor and adjacent nontumor tissues from patients with LUAD and LUSC. Dimensionality reduction and clustering with Scanpy, followed by ssGSE

Crosstalk on cell behavior in interactive cocultures of hMSCs with various oral cell types.

When prospectively applied for regenerative therapies, human bone-marrow-derived mesenchymal stem cells (hMSCs) interact with the locally residing host cells. With respect to the developmentally particular origin of oral cells, little is known about the putatively discriminative behavioral responses of hMSCs in interaction with various oral cell types, including human alveolar bone osteoblasts (hOAs), periodontal ligament fibroblasts (hPDLs), and gingival fibroblasts (hGFs). To assess the crosstalk between hMSCs and oral cells, interactive cocultures were established by combining well-characte

Tryptophan metabolism in tumor immune escape: mechanisms, cellular crosstalk, and therapeutic opportunities.

Tumor immune escape is increasingly recognized as an immunometabolic process shaped not only by immune checkpoints and suppressive cell populations, but also by nutrient competition and metabolic signaling within the tumor microenvironment. This nutrient-competitive environment is not limited to tryptophan depletion, but also involves glucose restriction, glutamine dependence, arginine metabolism, amino acid transporter competition, and impaired mitochondrial fitness of effector T cells. Among amino acid pathways, tryptophan metabolism has emerged as a central regulator of tumor-immune interac

Curcumin derivatives as photosensitizers in photodynamic therapy: photophysical properties and in vitro studies with prostate cancer cells.

Photodynamic therapy (PDT) is a minimally invasive approach to treat various forms of cancer, based on the ability of certain non-toxic molecules (photosensitizers) to generate reactive oxygen species (ROS) after excitation by light of a certain wavelength and eventually induce strong phototoxic reactions against malignant cells and other pathogens. Curcumin is one of the most extensively investigated phytochemicals with a wide range of therapeutic properties and has been shown to induce strong photocytotoxic effects in micromolar concentrations against a variety of cancer cell lines. Curcumin

Interleukin-37 overexpression enhances the therapeutic efficacy of adipose-derived mesenchymal stem cells in atopic dermatitis by activating autophagy and improving skin barrier dysfunction.

Adipose-derived mesenchymal stem cells (ADSCs) have emerged as a promising therapeutic option because of their immunomodulatory and tissue-repair capabilities in the treatment of Atopic dermatitis (AD). However, their efficacy is limited by cellular senescence and functional decline. Interleukin-37 (IL-37), an anti-inflammatory cytokine, has shown potential in mitigating inflammation and enhancing skin barrier function. This study investigated whether IL-37 overexpression in ADSCs could augment their therapeutic effects in AD and the underlying mechanism. Lentiviral-mediated IL-37 overexpressi

Stem Cell-based Therapeutic and Endogenous Regeneration Approaches for Congenital Tooth Agenesis.

Congenital tooth agenesis, including hypodontia and oligodontia, represents one of the most common developmental anomalies of the craniofacial complex and reflects failure of odontogenesis rather than postnatal tissue degeneration. The absence of teeth during growth affects mastication, speech, facial development and psychosocial well-being, while current management remains limited to prolonged temporization during childhood followed by definitive prosthetic rehabilitation after craniofacial maturation. No curative therapy is presently available. Recent advances in cell biology and development

Pirfenidone exacerbates Th2-driven vasculopathy in a mouse model of systemic sclerosis-associated interstitial lung disease.

BACKGROUND: Systemic sclerosis (SSc) is an autoimmune disease characterised by severe vasculopathy and fibrosis of various organs including the lung. Targeted treatment options for SSc-associated interstitial lung disease (SSc-ILD) are scarce. We assessed the effects of pirfenidone in a mouse model of SSc-ILD. METHODS: Pulmonary function, inflammation and collagen deposition in response to pirfenidone were assessed in Fra-2-overexpressing transgenic (Fra-2 TG) and bleomycin-treated mice. In Fra-2 TG mice, lung transcriptome was analysed after pirfenidone treatment. In vitro, pirfenidone effec

Intratumoral infiltrating lymphocytes correlate with improved survival in colorectal cancer patients: Independent of oncogenetic features.

BACKGROUND: The clinical relevance and prognostic value of tumor-infiltrating lymphocytes (TILs) as an interplay between malignant cells and immune function has been known for decades. On contrary, this potential may be different by T lymphocytes subsets endowed with a different function. Colorectal cancer (CRC) is a heterogeneous disease with different suggested prognostic biomarkers. So, this study was conducted to examine the prognostic value of CD8+ TILs on the survival rate of CRC as an independent factor of oncogenetic tumor features. METHODS: With respect to this, 281 formalin-fixed, p

Destructive inflammatory reaction after an autologous retinal pigment epithelium and choroid transplantation: no detection of an auto-immune response.

PURPOSE: Five patients who underwent uncomplicated retinal pigment epithelium (RPE)-choroid transplantation for neovascular age-related macular degeneration developed a destructive inflammatory reaction causing subretinal fluid accumulation and extensive RPE atrophy in the graft. We hypothesized that this inflammation could be caused by an auto-immune response against the graft, resulting in circulating auto-antibodies. The aim of our study was to examine a potential autoimmune origin, which would allow a more targeted therapy approach. METHODS: Five above-mentioned patients and four control

Glucose Induces DNMT1/IMPDH2-Dependent Metabolic Memory in Endothelial Cells Upon Reprograming Nucleotide Metabolism.

Type 2 diabetic (T2D) individuals are predisposed to enduring vascular complications despite therapeutic/lifestyle intervention due to 'metabolic memory', an epigenetic reprogramming in various cell/tissue types. The present study examined the potential role of DNMT isoforms in regulating glucose-induced metabolic memory and associated changes in endothelial metabolism leading to diabetic complications. The study involved micro/macro vascular endothelial cells (ECs), high-fat diet (HFD)-induced diabetic mouse models, and subjects with diabetic retinopathy (DR) at varying enforced levels of gly

Integrin α 1 β 1 Promotes Interstitial Fibrosis and Cyst Growth in a Mouse Model of Polycystic Kidney Disease.

KEY POINTS: Multiomic analysis of the Pkd1nl/nl mouse and human autosomal dominant polycystic kidney disease tissue identified upregulation of integrin- α 1. Global knockout of integrin subunit α 1 reduced kidney cyst size and preserved kidney function. Loss of integrin- α 1 in Pkd1nl/nl mice precluded interstitial fibrosis, which we suggest was due to reduced fibroblast cell proliferation. BACKGROUND: Cystogenesis and interstitial fibrosis are contributing factors to kidney failure in individuals with autosomal dominant polycystic kidney disease (ADPKD). The molecular and cellular mechanisms

Cerebrospinal Fluid Biomarkers of Myeloid and Glial Cell Activation Are Correlated With Multiple Sclerosis Lesional Inflammatory Activity.

Multiple sclerosis (MS)-related inflammation can be divided into lesional activity, mediated by immune cells migrating from the periphery to the central nervous system (CNS) and non-lesional activity, mediated by inflammation compartmentalized to CNS tissue. Lesional inflammatory activity, reflected by contrast-enhancing lesions (CELs) on the magnetic resonance imaging (MRI), is effectively inhibited by current disease modifying therapies (DMTs). While, the effect of DMTs on non-lesional inflammatory activity is currently unknown. Reliable and simultaneous measurements of both lesional and non

Safety reporting on implantation of autologous adipose tissue-derived stem cells with platelet-rich plasma into human articular joints.

BACKGROUND: Adipose tissue-derived stem cells (ADSCs), a type of mesenchymal stem cells (MSCs), have great potential as therapeutic agents in regenerative medicine. Numerous animal studies have documented the multipotency of ADSCs, showing their capabilities to differentiate into tissues such as muscle, bone, cartilage, and tendon. However, the safety of autologous ADSC injections into human joints is only beginning to be understood and the data are lacking. METHODS: Between 2009 and 2010, 91 patients were treated with autologous ADSCs with platelet-rich plasma (PRP) for various orthopedic co

Ascites-Derived Organoids to Depict Platinum Resistance in Gynaecological Serous Carcinomas.

Gynaecological serous carcinomas (GSCs) constitute a distinctive entity among female tumours characterised by a very poor prognosis. In addition to late-stage diagnosis and a high rate of recurrent disease associated with massive peritoneal carcinomatosis, the systematic acquisition of resistance to first-line chemotherapy based on platinum determines the unfavourable outcome of GSC patients. To explore the molecular mechanisms associated with platinum resistance, we generated patient-derived organoids (PDOs) from liquid biopsies of GSC patients. PDOs are emerging as a relevant preclinical mod

Elevated plasma IgG is associated with improved treatment response and survival in advanced non‑small cell lung cancer patients treated with anti‑PD‑1 therapy.

Immune checkpoint inhibitors (ICIs) targeting PD-1 improve outcomes in advanced non-small cell lung cancer (NSCLC), but responses are heterogeneous and tissue biomarkers are imperfect. Circulating immunoglobulins integrate B- and T-cell function and may reflect systemic immune competence. We investigated whether plasma immunoglobulin levels during ICI therapy are associated with clinical outcomes in stage IV NSCLC. In this single-center retrospective study, 55 patients received anti-PD-1-based regimens: anti-PD-1 monotherapy (n = 31), chemotherapy plus anti-PD-1 (n = 18), or ipilimumab plus ni

Targeting PIM kinases in cancer and inflammatory disorders: From mechanisms to clinical translation.

Proviral integration site for Moloney murine leukemia virus (PIM) kinase family (PIM1, PIM2, and PIM3) comprises constitutively active serine/threonine kinases that function as proto-oncogenic and immune-inflammatory signaling nodes. In cancer, PIM kinases promote tumor growth, therapeutic resistance, and immune microenvironment remodeling by phosphorylating substrates such as BCL2-associated agonist of cell death (BAD), Myc proto-oncogene protein (c-MYC), proline-rich AKT substrate of 40 kDa (PRAS40), and eukaryotic translation initiation factor 4E-binding protein 1 (4E-BP1)/eukaryotic transl

Breakthrough rhino-orbito-cerebral mucormycosis caused by Rhizopus arrhizus during posaconazole prophylaxis after allogeneic hematopoietic stem cell transplantation: a case report and focused literature review.

Rhino-orbito-cerebral mucormycosis (ROCM) is a rapidly progressive angioinvasive fungal infection associated with substantial mortality in profoundly immunocompromised patients. We report a fatal case of ROCM caused by Rhizopus arrhizus in a 60-year-old man with relapsed angioimmunoblastic T-cell lymphoma 67 days after haploidentical allogeneic hematopoietic stem cell transplantation. Medical records confirmed that oral posaconazole prophylaxis was continued after discharge from a previous hospitalization on July 21, and remained in use at the onset of sino-orbital symptoms. Posaconazole thera

Human epidermal growth factor receptor 2 (HER2) in salivary gland carcinomas.

BACKGROUND: HER2 is a target for antibody-based treatment of breast and gastric carcinoma which is highly successful in advanced disease as well as in the adjuvant setting. To determine the potential applicability of such therapies, salivary gland tumours were analysed for HER2 overexpression/amplification in this study. METHODS: A tissue microarray (TMA) was constructed from 994 carcinomas and 205 adenomas of the salivary gland. Slides were analysed for HER2 overexpression and gene amplification using FDA approved reagents for immunohistochemistry (IHC; Hercep-Test; Dako) and fluorescence in
