Left Ventricular Assist Device Flow Pattern Analysis Using a Novel Model Incorporating Left Ventricular Pulsatility.

Our current understanding of flow through the circuit of left ventricular assist device (LVAD), left ventricle and ascending aorta remains incompletely understood. Computational fluid dynamics, which allow for analysis of flow in the cardiovascular system, have been used for this purpose, although current simulation models have failed to fully incorporate the interplay between the pulsatile left ventricle and continuous-flow generated by the LVAD. Flow-through the LVAD is dependent on the interaction between device and patient-specific factors with suboptimal flow patterns evoking increased ri

Contextual dimensions of pediatric tuberculosis imaging: radiation exposure, access, and system capacity in high- and low-resource settings.

BACKGROUND: Pediatric tuberculosis diagnosis relies heavily on imaging, yet access, equipment standards, and dose monitoring differ widely across health systems. Evidence describing how these contextual factors influence imaging use and radiation exposure in children remains scarce. OBJECTIVE: To describe pediatric tuberculosis imaging practices and estimated radiation doses across two distinct resource settings, Spain (hospital-based, high-resource) and Mozambique (primary care-based, low-resource), to inform strategies for safe, equitable, and context-appropriate imaging. METHODS AND MATER

The characteristics of upper lobe-origin hemoptysis: analysis based on 31 patients.

BACKGROUND: Hemoptysis is associated with a variety of respiratory system diseases. Bronchial artery embolization (BAE) is effective for this condition, and preoperative computed tomography angiography (CTA) is helpful for the implementation of BAE. Some non-life-threatening cases have intermittent and recurrent episodes after conservative treatment, and no obvious abnormalities were found during tests and examinations, and they were defined as upper lobe-origin hemoptysis (ULOH), with concerns of embolism. This study aimed to describe the clinical characteristics of a distinct subtype of hemo

Hemodynamic comparisons of different shunt positions and geometrical model simplification strategies in the simulation of transjugular intrahepatic portosystemic shunt (TIPS).

Transjugular intrahepatic portosystemic shunt (TIPS) is a widely used surgery for portal hypertensive patients, whose potential postoperative complications are closely related to the hemodynamic condition of the portal venous system. The selection of shunt position in the surgery may affect the postoperative hemodynamics; however, it is difficult for clinical studies to investigate the influence. Therefore, this study aims to employ the computational model simulating TIPS to compare the hemodynamic differences resulting from different shunt positions, and also to investigate the influences of

Efficiency of Heterogenous Functional Connectomes Explains Variance in Callous-Unemotional Traits After Computational Lesioning of Cortical Midline and Salience Regions.

Introduction: Callous-unemotional (CU) traits are a youth antisocial phenotype hypothesized to be a result of differences in the integration of multiple brain systems. However, mechanistic insights into these brain systems are a continued challenge. Where prior work describes activation and connectivity, new mechanistic insights into the brain's functional connectome can be derived by removing nodes and quantifying changes in network properties (hereafter referred to as computational lesioning) to characterize connectome resilience and vulnerability. Methods: Here, we study the resilience of c

Computational Modeling of Macrophage Iron Sequestration during Host Defense against Aspergillus.

Iron is essential to the virulence of Aspergillus species, and restricting iron availability is a critical mechanism of antimicrobial host defense. Macrophages recruited to the site of infection are at the crux of this process, employing multiple intersecting mechanisms to orchestrate iron sequestration from pathogens. To gain an integrated understanding of how this is achieved in aspergillosis, we generated a transcriptomic time series of the response of human monocyte-derived macrophages to Aspergillus and used this and the available literature to construct a mechanistic computational model

Diversity of potassium channels in neuronal dendrites.

Complex computations in the nervous system begin with electrical signals generated in single neurons. Such signals include action potentials mediated by the opening of voltage-dependent ion channels, and synaptic potentials arising from neurotransmitter receptor activation. The amplitude, waveform, and propagation of action potentials and synaptic potentials influence cellular signaling in profound ways, and are largely determined by activities of ion channels in the cell membrane. The location and properties of ion channels therefore play critical roles in shaping electrical signaling in the

Central Nervous System Biodistribution and Pharmacokinetics of Radiolabeled Tofersen in Rodents, Nonhuman Primates, and Humans.

Antisense oligonucleotides (ASOs) are an important therapeutic modality across several therapeutic areas, offering currently available and potential future treatment options for patients. ASO pharmacokinetics, biodistribution, and regional brain uptake are not fully characterized, particularly in humans. Here, we report preclinical studies and the first-in-human imaging trial measuring the biodistribution of [99mTc]Tc-MAG3-tofersen. The tracer was designed to be a proxy for tofersen (Qalsody; Biogen), an ASO approved for the treatment of amyotrophic lateral sclerosis in adults who have a varia

Development of a treatment planning system for superficial x-ray radiotherapy using Monte Carlo database.

Objective. Currently, superficial x-ray radiotherapy does not take advantage of modern treatment planning technologies. To address the problem, a treatment planning system for superficial x-ray radiotherapy has been developed using a database generated through Monte Carlo simulations. The system, called SXRTDose, can be used to optimize irradiation strategies by adjusting energy, filtration, and applicator aiming to deliver a planned dose to the target volume while minimizing radiation risk to surrounding normal tissues.Approach. TOPAS Monte Carlo code was used for establishing the dosimetric

Comparing a BCI communication system in a patient with Multiple System Atrophy, with an animal model.

Paralysis affects many people worldwide, and the people affected often suffer from impaired communication. We developed a microelectrode-based Brain-Computer Interface (BCI) for enabling communication in patients affected by paralysis, and implanted it in a patient with Multiple System Atrophy (MSA), a neurodegenerative disease that causes widespread neural symptoms including paralysis. To verify the effectiveness of the BCI system, it was also tested by implanting it in a non-human primate (NHP). Data from the human and NHP were used to train binary classifiers two different types of machine

Modeling the interplay of sex hormones in cardiac hypertrophic signaling.

Biological sex plays a crucial role in the outcomes of cardiac health and therapies. Sex hormones are known to strongly influence cardiac remodeling through intracellular signaling pathways, yet their underlying mechanisms remain unclear. To address this need, we developed and validated a logic-based systems biology model of cardiomyocyte hypertrophy that, for the first time, incorporates the effects of both estradiol (E2) and testosterone (T) alongside well-established hypertrophic stimuli (Strain, angiotensin II (AngII), and endothelin-1 (ET-1)). We qualitatively validated the model to liter

CBCT-based online adaptive radiotherapy of the prostate bed: first clinical experience and comparison to nonadaptive conventional IGRT.

PURPOSE: Conventional image-guided radiotherapy (IGRT) of the prostate bed is challenged by the varying anatomy due to dynamic changes of surrounding organs such as the bladder and rectum. This leads to changed dose coverage of target and surrounding tissue. The novel online adaptive radiotherapy (oART) aims to improve target coverage as well as reduce dose exposure to surrounding healthy tissues by daily reoptimization of treatment plans. Here we set out to quantify the resulting changes of this adaptation for patients and treatment team. METHODS: A total of 198 fractions of radiotherapy of

Data-driven analyses of motor impairments in animal models of neurological disorders.

Behavior provides important insights into neuronal processes. For example, analysis of reaching movements can give a reliable indication of the degree of impairment in neurological disorders such as stroke, Parkinson disease, or Huntington disease. The analysis of such movement abnormalities is notoriously difficult and requires a trained evaluator. Here, we show that a deep neural network is able to score behavioral impairments with expert accuracy in rodent models of stroke. The same network was also trained to successfully score movements in a variety of other behavioral tasks. The neural n

Occipital Condyle Fracture Extending to the Inferior Part of the Clivus.

Although rarely reported in the literature, serious occipital and condylar fractures have been diagnosed more often with the widespread use of computed cranial tomography in traumas. In this paper, a 16-year-old female with a left occipital fracture extending from the left occipital condyle anterior of the hypoglossal canal to the inferior part of the clivus is presented. The fracture which had caused a neurological deficit was cured with conservative treatment. For delayed hypoglossal nerve paralysis due to swelling within the canal, methylprednisolone was started, and a complete cure was att

Evaluation of tilted cone-beam CT orbits in the development of a dedicated hybrid mammotomograph.

A compact dedicated 3D breast SPECT-CT (mammotomography) system is currently under development. In its initial prototype, the cone-beam CT sub-system is restricted to a fixed-tilt circular rotation around the patient's pendant breast. This study evaluated stationary-tilt angles for the CT sub-system that will enable maximal volumetric sampling and viewing of the breast and chest wall. Images of geometric/anthropomorphic phantoms were acquired using various fixed-tilt circular and 3D sinusoidal trajectories. The iteratively reconstructed images showed more distortion and attenuation coefficient
