NADPH oxidase 3 inhibition preserves hearing in mice after stereotactic radiosurgery.

Stereotactic radiosurgery (SRS) is a standard treatment for vestibular schwannoma but is associated with a substantial risk of progressive and irreversible hearing loss. NADPH oxidase 3 (NOX3), an enzyme predominantly expressed in the inner ear, has been identified as a key source of oxidative stress underlying cochlear injury. Using a mouse model of SRS-induced hearing loss, we found that genetic deletion of Nox3 preserved auditory function and protected cochlear structures, including hair cells, synaptic ribbons, and spiral ganglion neurons. Complementary experiments using local delivery of

GATA4-Driven Transcription of HtrA1 Promotes Cellular Senescence in Ménière's Disease and Age-Related Audio-Vestibular Dysfunction.

Ménière's disease (MD), a chronic inflammatory disorder with age-related increased incidence, exhibits poorly understood pathogenesis and limited therapeutic options. Here, we demonstrate that cellular senescence, marked by mitochondrial damage, reactive oxygen species accumulation, and senescence-associated secretory phenotype (SASP), is prevalent in the vestibular tissue of MD patients and an endolymphatic hydrops mouse model. The transcription factor GATA4 is upregulated in MD and mice, and its genetic deletion in hair cells alleviates LPS-induced audio-vestibular dysfunction and cellular s

Comorbid Auditory and Visual Dysfunction: From Pathogenic Genes to Gene Therapy.

Hearing and vision are the most important sensory functions. Genetic studies have revealed that specific genetic mutations can concurrently induce auditory and visual dysfunction. Comorbid auditory and visual impairment limits mutual sensory compensation, thereby severely delaying speech, cognitive, and intellectual development in affected pediatric patients and imposing a profound burden on their families. In this review, we summarize 23 genes currently recognized to be associated with both auditory and visual impairment and classify them according to their underlying pathogenic mechanisms. F

Compound Danshen Dripping Pills: A potential anti-oxidative drug for the treatment of noise-induced hearing loss in mice.

Acoustic damage is a common cause of hearing loss, and excessive oxidative stress has been identified as a key pathology in noise-induced hearing loss (NIHL). Despite its widespread occurrence, there are currently no effective therapeutic drugs available for the treatment of NIHL. Therefore, antioxidant therapy is widely used for the treatment of NIHL. Compound Danshen Dripping Pills (CDDP) is a traditional medicine with antioxidant, anti-inflammatory and microcirculatory improvement properties. However, it remains unknown whether CDDP has a hearing-protective function. In this study, we demon

Functional Correlation of Two Novel Nonsense POU4F3 Mutations Causing Late-Onset Progressive Nonsyndromic Hearing Loss in DFNA15 Families.

BACKGROUND: POU4F3 mutations cause DFNA15, an autosomal dominant nonsyndromic hearing loss. POU4F3 encodes a transcription factor crucial for inner ear hair cell development and maintenance. OBJECTIVE: To identify and functionally characterize novel POU4F3 mutations in two Chinese families with late-onset progressive hearing loss. METHODS: Massively parallel DNA sequencing (MPS) was performed on affected individuals from two unrelated Chinese families. Sanger sequencing validated mutations and confirmed co-segregation. Functional analyses included protein expression analysis by Western blots

Multifocal intravestibulocochlear and intracranial vestibular schwannomas: illustrative case.

BACKGROUND: Eighth cranial nerve schwannomas represent a rare pathological condition, and their sporadic unilateral multifocal presentation is even more uncommon. Here, the authors present a rare case of multifocal intravestibulocochlear and intracranial vestibular schwannomas (VSs) and review analogous cases in the existing literature. OBSERVATIONS: A 54-year-old female patient presented to the authors' clinic in March 2024 with complaints of left-sided hearing loss, tinnitus, and vertigo. Gadolinium-enhanced MRI revealed a Koos grade III VS localized in the cerebellopontine angle (CPA) with

Computer models and artificial intelligence increase the fidelity and efficiency of the in vitro models for hearing loss.

Hearing loss affects millions worldwide and is driven by diverse etiologies, including genetic mutations, ototoxic drug exposure, noise trauma, and aging. To investigate underlying mechanisms and test potential therapies, in vitro models, such as immortalized auditory hair cell lines, cochlear explants, and inner ear organoids, have become indispensable. However, these models face limitations in physiological relevance, scalability, and reproducibility. Recent advances in computer modeling, machine learning (ML), and deep learning (DL) offer powerful tools to enhance the accuracy, efficiency,

Therapeutic restoration of miR-96 prevents hearing loss in mice through modulation of noise-induced and genetic pathways.

Hearing loss often arises from impairments in multiple genes, complicating therapeutic development. MicroRNAs, as master regulators, offer promising targets for complex diseases. We explored miR-96's roles in hair cell (HC) function and noise-induced hearing loss (NIHL), finding that miR-96 -/-, not miR-96 +/- , mice exhibited progressive hearing loss due to gene regulatory network dysregulation from miR-96 loss in HCs not spiral ganglion neurons (SGNs). Viral-mediated delivery of miR-96 into the inner ear partially rescued hearing of miR-96 -/- mice. Tamoxifen-induced depletion of miR-96 in a

Elasticity and Thermal Stability are Key Determinants of Hearing Rescue by Mini-Protocadherin-15 Proteins.

Protocadherin-15 is a core protein component of inner-ear hair-cell tip links pulling on transduction channels essential for hearing and balance. Protocadherin-15 defects can result in non-syndromic deafness or Usher syndrome type 1F (USH1F) with hearing loss, balance deficits, and progressive blindness. Three rationally engineered shortened versions of protocadherin-15 (mini-PCDH15s) amenable for gene therapy have been used to rescue function in USH1F mouse models. Two can successfully or partially rescue hearing, while another one fails. Here we show that despite varying levels of hearing re

Deletion of the Ebf1, a mouse deafness gene, causes a dramatic increase in hair cells and support cells of the organ of Corti.

Following up on our previous observation that early B cell factor (EBF) sites are enriched in open chromatin of the developing sensory epithelium of the mouse cochlea, we investigated the effect of deletion of Ebf1 on inner ear development. We used a Cre driver to delete Ebf1 at the otocyst stage before development of the cochlea. We examined the cochlea at postnatal day (P) 1 and found that the sensory epithelium had doubled in size but the length of the cochlear duct was unaffected. We also found that deletion of Ebf1 led to ectopic sensory patches in the Kölliker's organ. Innervation of the

Hearing restoration by gene replacement therapy for a multisite-expressed gene in a mouse model of human DFNB111 deafness.

Gene therapy has made significant progress in the treatment of hereditary hearing loss. However, most research has focused on deafness-related genes that are primarily expressed in hair cells with less attention given to multisite-expressed deafness genes. MPZL2, the second leading cause of mild-to-moderate hereditary deafness, is widely expressed in different inner ear cells. We generated a mouse model with a deletion in the Mpzl2 gene, which displayed moderate and slowly progressive hearing loss, mimicking the phenotype of individuals with DFNB111. We developed a gene replacement therapy sys

Exploring the application of stem cell technology in treating sensorineural hearing loss.

Sensorineural deafness mainly occurs due to damage to hair cells, and advances in stem cell technology, especially the application of induced pluripotent stem cells (iPSCs) and adult stem cells, provides new possibilities for hair cell regeneration. This review describes the basic knowledge of stem cells and their important applications in regenerative medicine, as well as recent progress in stem cell research in the field of hair cell regeneration, especially the induced differentiation of hair-like cells. At the same time, we also point out the challenges facing current research, including d

SLC26A4-AP-2 mu2 interaction regulates SLC26A4 plasma membrane abundance in the endolymphatic sac.

Decreased presence or activity of human SLC26A4 at the plasma membrane is a common cause of hearing loss. SLC26A4 (Pendrin) is necessary for normal reabsorption of endolymph, the fluid bathing the inner ear. We identified the μ2 subunit of adaptor protein 2 (AP-2) complex required for clathrin-mediated endocytosis as a protein-partner of SLC26A4 involved in regulating its plasma membrane abundance. We showed that, in the endolymphatic sac, where fluid reabsorption occurs, SLC26A4 is localized along the apical microvilli of mitochondria-rich cells, in contact with the endolymph, and associated

A Novel Mutation Located in the N-Terminal Domain of MYO15A Caused Sensorineural Hearing Loss.

BACKGROUND: MYO15A is one of the common genes of severe-to-profound sensorineural deafness. Mutations in this gene can cause both pre- and post-lingual hearing losses. In this study, a novel MYO15A variant (c.2482C>T) was identified to be associated with autosomal recessive non-syndromic hearing loss (ARNSHL) in a Chinese Uighur family. METHODS: To examine the effects of the MYO15A mutation on the morphology and function of the derived hair cell-like cells, two iPSCs were generated separately from the proband and a mutation-negative family member and those were then induced to hair cell-like

Lessons learned in delayed identification of a misplaced electrode array in the vestibule.

To report a case of cochlear implantation with a misplaced electrode array in the vestibule and the causes for the delay in identification. A 23-year-old male with left single-sided deafness underwent cochlear implantation. The intraoperative assessment did not reveal any major red flags of electrode array misplacement. He did not display any vestibular symptoms postoperatively but showed poor speech performance, even though the aided tone audiometry revealed good sound detection thresholds. High-resolution computed tomography (HRCT) showed that the entire perimodiolar electrode array was situ

Sublethal effects of acidified water on sensorimotor responses and the transcriptome of zebrafish embryos.

Acidification of freshwater due to human activities is a widespread environmental problem. Its effects on the sensorimotor responses of fish, particularly during embryonic stages, may affect population fitness. To address this, zebrafish embryos were exposed to water at pH 7, 5 and 4.5 (adjusted with HCl) for 120 h. Acidic water did not increase mortality or cause obvious morphological abnormalities but reduced the size of the inner ear organs (otic vesicle and otolith) and the eye lens. It also suppressed ion uptake (Na+, Ca2+, K+) and induced embryonic acidosis. Behavioral tests at 4 or 5 da

In Utero Gene Therapy and its Application in Genetic Hearing Loss.

For monogenic genetic diseases, in utero gene therapy (IUGT) shows the potential for early prevention against irreversible and lethal pathological changes. Moreover, animal models have also demonstrated the effectiveness of IUGT in the treatment of coagulation disorders, hemoglobinopathies, neurogenetic disorders, and metabolic and pulmonary diseases. For major alpha thalassemia and severe osteogenesis imperfecta, in utero stem cell transplantation has entered the phase I clinical trial stage. Within the realm of the inner ear, genetic hearing loss significantly hampers speech, cognitive, and

Histologic and ultrastructural study of intracranial Gaucheroma causing deafness in a patient with Gaucher disease type 3: Effects of substrate reduction therapy.

Hearing loss is frequently associated with Gaucher disease (GD). Gaucher cells are enlarged reticuloendothelial cells containing glucocerebroside in the lysosomes due to deficiency of the glucocerebrosidase. Gaucheromas consist of accumulated Gaucher cells. Gaucher cells accumulate in variable tissues including the liver, spleen, bone marrow, and the middle ear and the mastoid causing conductive hearing loss. Neurons and astrocytes in the central nervous system are affected in neuronopathic GD leading to sensorineural hearing loss. Gaucheromas can develop even in patients treated with enzyme r

Presynaptic Nrxn3 is essential for ribbon-synapse maturation in hair cells.

Hair cells of the inner ear and lateral-line system rely on specialized ribbon synapses to transmit sensory information to the central nervous system. The molecules required to assemble these synapses are not fully understood. We show that Nrxn3, a presynaptic adhesion molecule, is crucial for ribbon-synapse maturation in hair cells. In both mouse and zebrafish models, the loss of Nrxn3 results in significantly fewer intact ribbon synapses. We show in zebrafish that, initially, Nrxn3 loss does not alter pre- and postsynapse numbers but, later, synapses fail to pair, leading to postsynapse loss
