Anti-auxin treatment and co-expression networks reveal auxin involvement in the transition to ripening in sweet cherry fruits.

The transition from development to ripening is governed by several hormones that interplay with abscisic acid (ABA) in non-climacterics, including auxin. In sweet cherry (Prunus avium), auxin has been shown to promote fruit coloration and ABA accumulation, in contrast to other non-climacteric models. Thus, this work aims to explore the role of auxin during the transition to ripening in sweet cherry by assessing the global gene expression response to an auxin homeostasis disturbance. For this, we used 5d, a fluorescently labeled compound with previously reported anti-auxin activity, which was a

Fungal endophytes relieve the growth-defence trade-off of their plant hosts through the production of bioactive alkaloids.

A central paradigm in plant biology is that there is a trade-off between growth and defence. We propose that Epichloë fungal endophytes relieve this trade-off in plants by the fungal-derived production of antiherbivore alkaloids. Lolium perenne plants without (nil) and with Epichloë LpTG-3 sp. strain AR37 able (wild type (wt), ΔidtA) and unable (ΔidtM) to produce bioactive indole diterpene alkaloids were subjected to an exogenous application of gibberellin (GA) hormone followed by a challenge with Rhopalosiphum padi aphids. The GA-derived plant growth promotion increased the susceptibility to

Molecular Basis of Phenotypic Plasticity: Understory-Induced DNA Methylation and Phytohormone Shifts Optimize Atractylodes lancea (Thunb.) DC Quality.

Atractylodes lancea (Thunb.) DC. (A. lancea) is a crop with medicinal and food properties. Currently, the majority of A. lancea available on the market is cultivated in extensive agricultural fields. This led to phenotypic variation in the plant and a reduction in the content of active ingredients. However, the cultivation of the understory can facilitate the phenotype of A. lancea. This, in turn, aligns with the characteristics of "excellent shape" and "high quality." Nonetheless, there is a paucity of research focusing on the phenotypic changes and molecular mechanisms underlying quality for

Nitric oxide interacts with brassinosteroid signalling to regulate plant growth and development in an organ-specific manner.

Nitric oxide (NO) is a plant gasotransmitter that regulates plant growth and development by interacting with different regulatory pathways. Among these, brassinosteroids (BRs) have become candidate phytohormones that interact antagonistically and closely with NO to regulate plant growth. Seedling growth in NO-deficient mutants is enhanced and hyper-responds to BR treatments, while NO overaccumulator mutants show some developmental defects. Interestingly, this hyper-response was also observed in mutants that had both constitutively activated BR signalling and reduced NO levels. BR signalling mu

Microplastics affect food intake and the expression of appetite regulators and nutrient transporters in pond loach (Misgurnus anguillicaudatus).

Microplastics (MPs) are emerging pollutants in freshwater ecosystems, posing risks to aquatic organisms. This study examines the effects of dietary MP exposure on food intake, appetite regulation, and nutrient transporter expression in pond loach (Misgurnus anguillicaudatus), a sediment-feeding facultative air breathing freshwater fish. Fish were exposed to small (250-300 μm), medium (425-500 μm), and large (710-850 μm) polyethylene microspheres for two weeks. Food intake was significantly reduced in fish fed small and medium MPs, but not large MPs, suggesting size-dependent ingestion effects.

Roles of auxin in the inhibition of shoot branching in 'Dugan' fir.

Shoot branching substantially impacts vegetative and reproductive growth as well as wood characteristics in perennial woody species by shaping the shoot system architecture. Although plant hormones have been shown to play a fundamental role in shoot branching in annual species, their corresponding actions in perennial woody plants are largely unknown, in part due to the lack of branching mutants. Here, we demonstrated the role of plant hormones in bud dormancy transition toward activation and outgrowth in woody plants by comparing the physiological and molecular changes in the apical shoot ste

CREBH Systemically Regulates Lipid Metabolism by Modulating and Integrating Cellular Functions.

Cyclic AMP-responsive element-binding protein H (CREBH, encoded by CREB3L3) is a membrane-bound transcriptional factor expressed in the liver and small intestine. The activity of CREBH is regulated not only at the transcriptional level but also at the posttranslational level. CREBH governs triglyceride metabolism in the liver by controlling gene expression, with effects including the oxidation of fatty acids, lipophagy, and the expression of apolipoproteins related to the lipoprotein lipase activation and suppression of lipogenesis. The activation and functions of CREBH are controlled in respo

Phytohormone-dependent plant defense signaling orchestrated by oral bacteria of the herbivore Spodoptera litura.

A vast array of herbivorous arthropods live with symbiotic microorganisms. However, little is known about the nature and functional mechanism of bacterial effects on plant defense responses towards herbivores. We explored the role of microbes present in extracts of oral secretion (OS) isolated from larvae of Spodoptera litura, a generalist herbivore, in phytohormone signaling-dependent defense responses in Arabidopsis thaliana (Arabidopsis). In response to mechanical damage (MD) with application of bacteria-free OS (OS- ) prepared by sterilization or filtration of OS, Arabidopsis leaves exhibi

Stories of Salicylic Acid: A Plant Defense Hormone.

Salicylic acid (SA) is a key plant hormone required for establishing resistance to many pathogens. SA biosynthesis involves two main metabolic pathways with multiple steps: the isochorismate and the phenylalanine ammonia-lyase pathways. Transcriptional regulations of SA biosynthesis are important for fine-tuning SA level in plants. We highlight here recent discoveries on SA biosynthesis and transcriptional regulations of SA biosynthesis. In addition, SA perception by NPR proteins is important to fulfil its function as a defense hormone. We highlight recent work to give a full picture of how NP

Expression of mosquito microRNA Aae-miR-2940-5p is downregulated in response to West Nile virus infection to restrict viral replication.

UNLABELLED: West Nile virus (WNV) is an enveloped virus with a single-stranded positive-sense RNA genome from the Flaviviridae family. WNV is spread by mosquitoes and able to infect humans, causing encephalitis and meningitis that can be fatal; it therefore presents a significant risk for human health. In insects, innate response to RNA virus infection mostly relies on RNA interference and JAK/SAT pathways; however, some evidence indicates that it can also involve microRNAs (miRNAs). miRNAs are small noncoding RNAs that regulate gene expression at posttranscriptional level and play an importan

The RBFOX1 regulatory gene network contributes to major depressive disorder, anxiety, irritability and neuroticism.

Major depressive disorder (MDD) is a complex psychiatric disorder frequently comorbid with anxiety disorders, suicide attempts, and personality traits such as irritability and neuroticism. RBFOX1, which encodes a splicing factor, is a known risk gene for MDD, and has also been associated with anxiety and neuroticism. However, the working mechanisms of how RBFOX1 influences these conditions are not yet understood. Here, we aim to shed light on the effects of RBFOX1 and its regulated genes on MDD, and the traits anxiety, irritability, neuroticism, as well as suicide attempt using genome-wide ass

Single-nucleus atlas of cell-type specific genetic regulation in the human brain.

Genetic risk variants for common diseases are predominantly located in non-coding regulatory regions and modulate gene expression. Although bulk tissue studies have elucidated shared mechanisms of regulatory and disease-associated genetics, the cellular specificity of these mechanisms remains largely unexplored. Here we present a comprehensive, single-nucleus multi-ancestry atlas of genetic regulation of gene expression in the human prefrontal cortex, comprising 5.6 million nuclei from 1,384 donors of diverse ancestries. Through multi-resolution analyses spanning eight major cell classes and 2

5'-flanking regions of camel milk genes are highly similar to homologue regions of other species and can be divided into two distinct groups.

The concentrations of individual casein and whey proteins in camel milk differ markedly to respective protein concentrations in bovine milk. The ratio of beta-casein to kappa-casein is considerably higher in camel milk. beta-Lactoglobulin is absent, but whey acidic protein and peptidoglycan recognition protein have been detected. Genomic sequences upstream to milk-protein genes, which are known to regulate the expression of milk proteins to a great extent, were determined for 10 camel milk-protein genes and compared to respective sequences in other mammals. Multiple sequence alignment showed c

Integrative Transcriptomic Profiling Identifies TNF and IL1B as Candidate Key Early-Response Genes in Macrophages Infected with Smooth Brucella Using a Comprehensive Bioinformatic Approach.

Smooth Brucella are the main pathogenic bacteria that threaten human health and food safety. The early stage of smooth Brucella and macrophage interaction is an important phase, and smooth Brucella species elicit a dramatic transcriptional response in infected macrophages. However, the key transcriptional events are still obscure. This study aimed to identify key candidate response pathways and genes in macrophages infected with smooth Brucella at the early interaction stage. Three gene expression profiles including GSE21117, GSE5202, and GSE8385 were retrieved from the NCBI GEO database, and

Hippocampal transcriptome profiling in a 22q11.2 deletion syndrome mouse model: comparison with human schizophrenia.

22q11.2 deletion syndrome (22q11.2DS) confers one of the highest genetic risks for schizophrenia, yet the molecular mechanisms remain incompletely understood. We performed comprehensive RNA sequencing of the dorsal hippocampus in Df1/+ mice, a 22q11.2DS model, integrating behavioral assessment and cross-species comparison with human schizophrenia postmortem data. Df1/+ mice exhibited selective contextual fear memory impairment without gross locomotor deficits. Transcriptomic analysis using integrated over-representation and gene set enrichment approaches revealed upregulation of synaptic signa

Identification of genes particularly sensitive to lipopolysaccharide (LPS) in human monocytes induced by wild-type versus LPS-deficient Neisseria meningitidis strains.

Lipopolysaccharide (LPS) in the outer membrane of Neisseria meningitidis plays a dominant role as an inflammation-inducing molecule in meningococcal disease. We have used microarray analysis to study the global gene expression after exposure of human monocytes for 3 h to wild-type N. meningitidis (10(6)), LPS-deficient N. meningitidis (10(6) and 10(8)), and purified N. meningitidis LPS (1 ng [33 endotoxin units]/ml) to identify LPS-inducible genes. Wild-type N. meningitidis (10(6)) induced 4,689 differentially expressed genes, compared with 72 differentially expressed genes induced by 10(6) LP

A computational strategy for finding novel targets and therapeutic compounds for opioid dependence.

Opioids are widely used for treating different types of pains, but overuse and abuse of prescription opioids have led to opioid epidemic in the United States. Besides analgesic effects, chronic use of opioid can also cause tolerance, dependence, and even addiction. Effective treatment of opioid addiction remains a big challenge today. Studies on addictive effects of opioids focus on striatum, a main component in the brain responsible for drug dependence and addiction. Some transcription regulators have been associated with opioid addiction, but relationship between analgesic effects of opioids

Expression and clinical value of key m6A RNA modification regulators in tuberculosis.

BACKGROUND: N6-methyladenosine (m6A), the most prevalent and reversible post-transcriptional RNA modification, is involved in the progression of various diseases. Nonetheless, the role of m6A modification in Tuberculosis (TB) pathogenesis remains unknown. Here, we investigated the general expression patterns and potential functions of m6A regulators in TB. METHODS: The differentially expressed m6A genes between the healthy and TB groups were evaluated using the public Gene Expression Omnibus (GEO) database, and quantitative real-time PCR (qRT-PCR) was used to test the expression of key m6A re
