Cryo-EM structure of human TMEM45A with a bound 2-Hydroxypalmitate.

The orphan transmembrane protein 45 A (TMEM45A) is highly expressed in keratinocytes as well as cancer cells under hypoxic conditions, where it contributes to metastasis and chemoresistance. The structure of human TMEM45A, determined with single-particle cryo-electron microscopy (cryo-EM) to 3.27 Å, reveals a homotetrameric assembly of seven-transmembrane-helix protomers. The first six transmembrane helices (TMHs) of each protomer assemble to form a central hydrophobic tunnel containing a 2-hydroxypalmitate.

Cryo-EM reveals the molecular basis oflaminin polymerization and LN-lamininopathies.

Laminin polymerization is the major step in basement membranes assembly. Its failures cause laminin N-terminal domain lamininopathies including Pierson syndrome. We have employed cryo-electron microscopy to determine a 3.7 Å structure of the trimeric laminin polymer node containing α1, β1 and γ1 subunits. The structure reveals the molecular basis of calcium-dependent formation of laminin lattice, and provides insights into polymerization defects manifesting in human disease.

Pterygodermatites (Mesopectines) niameyensis n. sp. (Nematoda: Rictulariidae), a parasite of Mastomys natalensis (Smith, 1834) (Rodentia: Muridae) from Niger.

Pterygodermatites (Mesopectines) niameyensis n. sp. is described from Mastomys natalensis in Niamey/Niger (West Africa). It differs from other species of same subgenus by the morphology of the head, which presents 4 simple cephalic papillae and nearly axial oral opening, a number of caudal papillae, precloacal cuticular formations, and the spicule length/body length ratio. Scanning electron microscopy shows the presence of 2 pairs of lateral sensory structures for male worms.

Extracellular vesicle-mediated transfer of membranous components from the highly malignant T24 urinary carcinoma cell line to the non-malignant RT4 urinary papilloma cell line.

This communication reports the first experimental evidence that in the bladder cancer model, membranous components labelled with the DiO dye and the cholera toxin subunit B can be transported from highly malignant (T24) to non-malignant (RT4) cells by extracellular vesicles. Taking into account the presence of stable membranous nanostructures found by scanning electron microscopy, we suggest a possible uptake mechanism in recipient cells through fusion with highly curved membranous regions.

Denuded congenital lesions: recessive dystrophic epidermolysis bullosa.

Recessive dystrophic epidermolysis bullosa (Hallopeau-Siemens type) (RDEB-HS) is a rare severe mechanobullous disorder resulting from a defect in collagen VII. Patients with RDEB-HS present with generalized blistering and denudation of the skin at birth and have mucosal involvement. The repeated blistering leads to scarring, which may be deforming and result in serious complications. Transmission electron microscopy is currently the gold standard for diagnosis of RDEB-HS.

Ultrastructural and electrophysiological correlation of the genotypes of NCL.

Several genetically different, but clinically similar childhood forms of neuronal ceroid lipofuscinosis are now recognized. Accurate diagnosis is important so that appropriate genetic advice can be given, molecular analysis can be undertaken, and prenatal testing can be considered. A combined clinical, electrophysiological, and histological (light and electron microscopy) approach offers the most reliable means of diagnosis in the majority of patients. Patients with an unusual presentation will also be identified.

The surgical pathologist's approach to fine needle aspiration.

The diagnostic process begins with triage of the FNAB, depending on the patient's problem. Portions of the specimen can then be set aside for appropriate immunocytochemistry, hormone receptors analysis, electron microscopy, flow cytometry, or molecular studies. Microscopic evaluation, as in surgical pathology, begins with scanning objective examination of tissue architecture, followed by study of cellular characteristics, and finally the nuclear features. Cytopathology and surgical pathology are no longer distinct entities.

Carcinoid tumor of the thymus: a unique neoplasm of the mediastinum.

In this report, we describe a patient with a thymic carcinoid and the evolution of his disease for the past 5 years. Due to the distinct nature of thymic carcinoids, their unique biologic and endocrine properties, and the variety of available treatment options, it is important to differentiate this entity from other mediastinal tumors. The use of immunohistochemical studies or electron microscopy by the pathologist permits more accurate differentiation and diagnosis.

Benign cutaneous Degos' disease.

We report the case of a 44-year-old woman with Degos' disease who also had a lupus anticoagulant. Electron microscopy of the mature lesions showed interwoven tubular structures within the endothelial cells, as have been observed in previous cases of Degos' disease. Four years after her first cutaneous lesions, there is no evidence of involvement of other organs. Aspirin (300 mg daily) has arrested the cutaneous disease.

Uncombable hair, retinal pigmentary dystrophy, dental anomalies, and brachydactyly: report of a new patient with additional findings.

A new ectodermal dysplasia syndrome was reported by Bork et al. in 1987 (Hautarzt 38:342-347). The syndrome consisted of hypotrichosis with the typical SEM (scanning electron microscopy) changes of uncombable hair, retinal pigmentary dystrophy, juvenile cataract, oligodontia, brachydactyly with brachymetacarpia; it was inherited as an autosomal dominant trait. We describe a sporadic case and add further clinical findings to expand the spectrum of this rare syndrome.

Corneal argyrosis associated with silver soldering.

We report a patient who developed corneal argyrosis secondary to occupational silver soldering. Clinically, the cornea was notable for a green-brown discoloration localized to Descemet's membrane by slit-lamp biomicroscopy. Silver particles were identified within the anterior three eighths of Descemet's membrane by light and electron microscopy and energy-dispersive x-ray microanalysis. To our knowledge, the association between corneal argyrosis and silver soldering has not been previously reported.

[A malignant salivary lymphoepithelial lesion with a poorly differentiated epidermoid carcinoma. Apropos a case].

A case of malignant lymphoepithelial lesion of the submandibular gland is reported in a 71-years-old woman. The carcinoma showed a poorly differentiated pattern and its epidermoid nature was obvious after immunohistochemistry and electron microscopy. The epithelial nests were lying in an abundant lymphoid stroma consisting of small lymphocytes. A recurrence occurred ten months after surgical treatment. This observation is compared with those already related in the literature.

Muscle pathology correlates with permanent weakness in hypokalemic periodic paralysis: a case report.

We present a morphological follow-up in a case of familial hypokalemic periodic paralysis with progressive weakness. At age 12 years muscle biopsy revealed mild vacuolar changes. Seventeen years later, after the patient had developed permanent weakness, light and electron microscopy disclosed tubular aggregates in about 15% of the fibers and medium-grade myopathic alterations. We describe correlation of muscle pathology with permanent weakness in hypokalemic periodic paralysis.

[Primary carcinoma of the pituitary gland with Cushing's syndrome and metastases. Apropos of a case with review of the literature].

We report the case of a 25 year old man with pituitary carcinoma with Cushing's syndrome. Though the diagnosis was initially proposed, it was only confirmed by the appearance of meningeal and lymphatic metastases. These were secretory, as confirmed by immunocytochemistry and electron microscopy. These tumours are rare, whether they secrete or not, and can be confirmed only by the existence of metastases, most often in the brain or the liver.

Herpes-like viral dermatitis in a beluga whale (Delphinapterus leucas).

Approximately 3.5 mo following its capture, a beluga whale (Delphinapterus leucas) developed focal pale gray skin lesions. These lesions persisted for at least 8 mo. A biopsy from one of these sites revealed epithelial intranuclear inclusions. Herpes-like viral particles were seen by transmission electron microscopy. The eventual regression of skin lesions and lack of other clinical signs suggests the virus was only mildly pathogenic in this animal.

Unusual inclusions in mature polymorphonuclear neutrophils of cyclic neutropenia.

The bone marrow of an 11-year-old girl with cyclic neutropenia was examined by electron microscopy. In the nonneutropenic phase, mature polymorphonuclear neutrophils had numerous bundles of filamentous inclusions in both cytoplasms and nucleus. Primary granules were slightly increased in number. Similar filament-like inclusions also have been reported in some cases of hemopoietic dysplasia; however, our case showed no abnormality in chromosomes, no thrombocytopenia, and no leukemic change.

[The vernal plaque, a complication of so-called spring conjunctivitis].

The vernal ulcer is a peculiar corneal complication of vernal conjunctivitis. Clinically, the ulcer is oval, grey, and resistant to medical treatment. Histological study by light and electron microscopy reveal the lesion to be composed of fibrin deposits adherent to a well-preserved Bowman's membrane. These deposits impede lateral progression of the epithelium, thus, opposing spontaneous healing. Resolution of the ulcer can be obtained, however, by superficial keratectomy.

Recent advances in the morphology of myositis.

Myositis in man may be divided into infectious and non-infectious forms. The myopathologist more often deals with the latter forms which comprise dermatomyositis/polymyositis, inclusion body myositis, mixed connective tissue disease/collagenoses, and granulomatous myopathies. Modern morphological techniques as enzyme-histochemistry, electron microscopy, immunohistology, and morphometry are of different value in various forms of myositis, but are often indispensable techniques in up-to-date diagnostic work up of a myositis.

The pathogenesis of tendolipomatosis; an electron microscopical study.

The development of tendolipomatosis has been studied both by light and scanning electron microscopy in the tendons of healthy persons killed in accidents. Lipid cells are rarely seen in the intact tendon. In the early stages of tendolipomatosis lipid cells are found deep and singly but in the later phases lipocytes are seen to disrupt the continuity of collagen fibres and tendon bundles and so reduce the strength of the tendon.

Herpes simplex virus and congenital malformations.

We have reported a case of transplacentally transmitted herpes simplex virus (HSV) infection in association with both congenital malformations and other serious abnormalities, including facial abnormalities, microcephaly, cerebral atrophy, and microscopic cranial calcifications. Before death, the infant showed marked neurologic deficits, seizures, and respiratory distress. Serum IgM and complement fixing antibodies to HSV were elevated at birth. Light and electron microscopy and immunofluorescence studies confirmed the presence of the virus.
