Respiratory and autonomic dysfunction in congenital central hypoventilation syndrome.

The developmental lineage of the PHOX2B-expressing neurons in the retrotrapezoid nucleus (RTN) has been extensively studied. These cells are thought to function as central respiratory chemoreceptors, i.e., the mechanism by which brain Pco2 regulates breathing. The molecular and cellular basis of central respiratory chemoreception is based on the detection of CO2 via intrinsic proton receptors (TASK-2, GPR4) as well as synaptic input from peripheral chemoreceptors and other brain regions. Murine models of congenital central hypoventilation syndrome designed with PHOX2B mutations have suggested

Cancer evolution: mathematical models and computational inference.

Cancer is a somatic evolutionary process characterized by the accumulation of mutations, which contribute to tumor growth, clinical progression, immune escape, and drug resistance development. Evolutionary theory can be used to analyze the dynamics of tumor cell populations and to make inference about the evolutionary history of a tumor from molecular data. We review recent approaches to modeling the evolution of cancer, including population dynamics models of tumor initiation and progression, phylogenetic methods to model the evolutionary relationship between tumor subclones, and probabilisti

CHZ868, a Type II JAK2 Inhibitor, Reverses Type I JAK Inhibitor Persistence and Demonstrates Efficacy in Myeloproliferative Neoplasms.

Although clinically tested JAK inhibitors reduce splenomegaly and systemic symptoms, molecular responses are not observed in most myeloproliferative neoplasm (MPN) patients. We previously demonstrated that MPN cells become persistent to type I JAK inhibitors that bind the active conformation of JAK2. We investigated whether CHZ868, a type II JAK inhibitor, would demonstrate activity in JAK inhibitor persistent cells, murine MPN models, and MPN patient samples. JAK2 and MPL mutant cell lines were sensitive to CHZ868, including type I JAK inhibitor persistent cells. CHZ868 showed significant act

The value of animal models to study immunopathology of primary human Sjögren's syndrome symptoms.

Sjögren's syndrome (SjS) is a complex chronic autoimmune disease of multifactorial etiology that results in eventual loss of secretory function in the exocrine glands. The challenges towards finding a therapeutic prevention or treatment for SjS are due primarily to a lack of understanding in the pathophysiological and clinical progression of the disease. In order to circumnavigate this problem, there is a need for appropriate animal models that resemble the major phenotypes of human SjS and deliver a clear underlying biological or molecular mechanism capable of defining various aspects for the

Genetically modified pigs to model human diseases.

Genetically modified mice are powerful tools to investigate the molecular basis of many human diseases. Mice are, however, of limited value for preclinical studies, because they differ significantly from humans in size, general physiology, anatomy and lifespan. Considerable efforts are, thus, being made to develop alternative animal models for a range of human diseases. These promise powerful new resources that will aid the development of new diagnostics, medicines and medical procedures. Here, we provide a comprehensive review of genetically modified porcine models described in the scientific

Alcoholic liver disease and exacerbation by malnutrition and infections: what animal models are currently available?

Alcoholic liver disease remains a frequent and serious problem for increasing numbers of patients. Research has expanded our molecular understanding of the cellular basis of disease progression; however, translation into therapy is still hampered by a lack of suitable animal models for alcoholic liver disease, as well as from consequences of related liver damage due to malnutrition, hepatitis C virus infection, or abuse of other substances. Many patients with liver disease do not simply consume too much alcohol; they also suffer from comorbidities such as obesity or viral hepatitis, and/or may

Understanding the mechanisms of human tubal ectopic pregnancies: new evidence from knockout mouse models.

Ectopic pregnancy, a worldwide health problem, is potentially life-threatening and occurs in approximately 1.5-2% of all pregnancies in the western world; however, the precise mechanisms underlying the initiation and development of tubal ectopic pregnancy are unknown. Tubal abnormalities and dysfunction, such as altered contractility or abnormal ciliary activity, have been speculated to lead to tubal ectopic pregnancy. To elucidate the cellular and molecular mechanisms of the tubal transport process, several knockout (KO) mouse models have been developed. This review summarizes what has been l

3,3'-Diindolylmethane ameliorates muscle atrophy by modulating mitochondrial function and calcium homeostasis.

BACKGROUND: Sarcopenia is a chronic degenerative condition characterized by the progressive loss of muscle mass and function. Current therapeutic strategies exhibit limited therapeutic efficacy and present substantial challenges in ensuring sustained patient compliance. Although mitochondrial dysfunction is a confirmed biomarker of skeletal muscle atrophy, the exact molecular mechanisms governing this process have yet to be fully elucidated. Notably, store-operated calcium entry (SOCE) not only serves as a pivotal mechanism in mitochondrial calcium signaling but has also been recently identifi

Developing models for the diagnosing of ulcerative colitis and prognosis of anti-TNF-α non-response based on neutrophil extracellular trap-associated genes.

BACKGROUND: Neutrophil extracellular traps (NET) play a pivotal role in the pathogenesis of ulcerative colitis (UC) and may contribute to the impaired response to anti-tumor necrosis factor alpha (TNF-α) therapies. However, the functional implications of NET-associated genes in UC remain poorly understood. This study aims to identify key NET-associated molecular signatures in UC, develop diagnostic models based on NET-related biomarkers, and construct predictive models for response to anti-TNF-α therapies (infliximab and golimumab). METHODS: NET-associated genes were obtained from the Kyoto E

How Does the Study MD of pH-Dependent Exposure of Nanoparticles Affect Cellular Uptake of Anticancer Drugs?

The lack of knowledge about the uptake of NPs by biological cells poses a significant problem for drug delivery. For this reason, designing an appropriate model is the main challenge for modelers. To address this problem, molecular modeling studies that can describe the mechanism of cellular uptake of drug-loaded nanoparticles have been conducted in recent decades. In this context, we developed three different models for the amphipathic nature of drug-loaded nanoparticles (MTX-SS-γ-PGA), whose cellular uptake mechanism was predicted by molecular dynamics studies. Many factors affect nanopartic

Dissecting the impact of molecular T-cell HLA mismatches in kidney transplant failure: A retrospective cohort study.

INTRODUCTION: Kidney transplantation is the optimal treatment in end-stage kidney disease, but de-novo donor specific antibody development continues to negatively impact patients undergoing kidney transplantation. One of the recent advances in solid organ transplantation has been the definition of molecular mismatching between donors and recipients' Human Leukocyte Antigens (HLA). While not fully integrated in standard clinical care, cumulative molecular mismatch at the level of eplets (EMM) as well as the PIRCHE-II score have shown promise in predicting transplant outcomes. In this manuscript

Data-Driven Prediction of Ultraviolet-Visible Spectra From Molecular Structure Using Attention-Based Graph Neural Networks.

Predicting UV-vis spectra from molecular structure is an important but relatively underexplored task. In this study, we developed a deep learning approach based on graph neural networks (GNNs) to directly predict UV-vis spectra from SMILES representations. Several GNN architectures were evaluated, among which Attentive Fingerprint achieved the best performance with an R2 of 0.8630, mean absolute error of 0.0389, root mean squared error of 0.0740, cosine similarity of 0.9589, Pearson correlation coefficient of 0.9521, and distance metric of 1.5758 on the test set. Further analysis showed that t

Proteomic mapping of differentially vulnerable pre-synaptic populations identifies regulators of neuronal stability in vivo.

Synapses are an early pathological target in many neurodegenerative diseases ranging from well-known adult onset conditions such as Alzheimer and Parkinson disease to neurodegenerative conditions of childhood such as spinal muscular atrophy (SMA) and neuronal ceroid lipofuscinosis (NCLs). However, the reasons why synapses are particularly vulnerable to such a broad range of neurodegeneration inducing stimuli remains unknown. To identify molecular modulators of synaptic stability and degeneration, we have used the Cln3 -/- mouse model of a juvenile form of NCL. We profiled and compared the mole

Gabapentin Differentially Modulate c-Fos Expression in Hypothalamus and Spinal Trigeminal Nucleus in Surgical Molar Extraction.

The study on the efficacy of oral analgesics reported that no single class of drug is effective in post-surgical dental pain. Pain following removal of third molar is most commonly used and widely accepted acute pain model for assessing the analgesic effect of drugs in humans. Reports demonstrated that analgesic efficacy in the human dental model is highly predictive. The high incidence of false-negative findings in analgesic investigations hinders the process of molecular discovery. Molecular mechanism of post-surgical pain is not known. More importantly, the animal model for postoperative de

Spin-selectivity effect of G-quadruplex DNA molecules.

Chirality-induced spin selectivity has been attracting extensive interest in recent years and is demonstrated in a variety of chiral molecules, all of which arise from inherent molecular chirality. Here, we first propose a theoretical model to study the spin-dependent electron transport along guanine-quadruplex (G4) DNA molecules, connected to two nonmagnetic electrodes, by considering the molecule-electrode contact and weak spin-orbit coupling. Our results indicate that the G4-DNA molecular junctions exhibit pronounced spin-selectivity effect, and the asymmetric contact-induced external chira

Polycystic kidney disease: genes, proteins, animal models, disease mechanisms and therapeutic opportunities.

An increased understanding of the genetic, molecular and cellular mechanisms responsible for the development of polycystic kidney disease has laid out the foundation for the development of rational therapies. Many animal models where these therapies can be tested are currently available. This review summarizes the rationale for these treatments, the results of preclinical trials and the prospects for clinical trials, some already in early phases of implementation.

Preclinical and Virtual Models of Mucosal Melanoma: Bridging Translational Gaps in a Rare and Lethal Cancer.

Mucosal melanoma (MM) is a rare and lethal subtype of melanoma, disproportionately affecting Asian populations and exhibiting distinct clinicopathological and genetic features compared to cutaneous melanoma (CM). Often diagnosed at advanced stages, MM shows poor responses to conventional therapies, and no standardized treatment regimen currently exists. Progress in preclinical modeling, including cell lines, patient-derived xenografts (PDXs), organoids (PDOs), and comparative animal models-has provided valuable tools for studying MM pathogenesis and therapeutic resistance. Yet these models rem

Multi-target QSAR modelling for identification of novel inhibitors of class I HDACs.

In this study, the multi-target QSAR (mt-QSAR) models were constructed which can predict the inhibitory activity of compounds against various class I HDACs isoforms under different experimental conditions. Models based on mt-QSAR classification (a linear model and six non-linear models) were constructed using 1215 compounds obtained from the ChEMBL database by the Box - Jenkins moving average method using 13 deviation descriptors. The high predictive performance was found in the non-linear models based on Support Vector Classification, Random Forest and Gradient Boosting with accuracies exceed

Insights and challenges on animal models for progressive multiple sclerosis.

Progressive multiple sclerosis (PMS) remains a major clinical challenge due to its complex pathophysiology and limited therapeutic options. While disease-modifying therapies have significantly improved prognosis for relapsing forms of multiple sclerosis (RMS), effective treatments for the progressive, neurodegenerative phase are lacking. Animal models have been instrumental in elucidating the molecular mechanisms underlying multiple sclerosis (MS) pathology and have guided the drug development field for MS, yet their ability to faithfully replicate the development of PMS remains debated.
