Maladaptive self-focused attention and default mode network connectivity: a transdiagnostic investigation across social anxiety and body dysmorphic disorders.

Maladaptive self-focused attention (SFA) is a bias toward internal thoughts, feelings and physical states. Despite its role as a core maintaining factor of symptoms in cognitive theories of social anxiety and body dysmorphic disorders (BDDs), studies have not examined its neural basis. In this study, we hypothesized that maladaptive SFA would be associated with hyperconnectivity in the default mode network (DMN) in self-focused patients with these disorders. Thirty patients with primary social anxiety disorder or primary BDD and 28 healthy individuals were eligible and scanned. Eligibility was

Neuroimaging in anxiety disorders.

Neuroimaging studies using functional magnetic resonance imaging (fMRI), positron emission tomography (PET) and single-photon emission computed tomography (SPECT) to evaluate neurofunctional and neurochemical alterations related to the generation and control of affect in patients with anxiety disorders are reviewed. We performed a meta-analysis of symptom provocation studies, where neural activity was measured using fMRI, PET or SPECT to test the hypothesis that prefrontal regions modulate amygdala activity. Data revealed that reactivity in the amygdala was enhanced in patients with phobia as

Multifunctional Vertical Bulk-Heterojunction Transistors With Amorphous GaOx for Neuromorphic Perception-to-Decision.

Multifunctional neuromorphic hardware has emerged as a promising paradigm to transcend the physical separation of sensing, memory, and computing inherent in traditional von Neumann architectures. The central challenge lies in developing integrated devices with high functional density for in-situ fusion of sensing, memory, and computing. Here, we propose a multifunctional vertical bulk-heterojunction transistor (MVBT) based on amorphous GaOx dielectric layer, integrating four functions within a single device: deep-ultraviolet (DUV) photodetection, threshold neuronal firing, broadband optoelectr

Deep learning reveals endogenous sterols as allosteric modulators of the GPCR-Gα interface.

Endogenous intracellular allosteric modulators of GPCRs remain largely unexplored, with limited binding and phenotype data available. This gap arises from the lack of robust computational methods for unbiased cavity identification, cavity-specific ligand design, synthesis, and validation across GPCR topology. Here, we developed Gcoupler, an AI-driven generalized computational toolkit that leverages an integrative approach combining de novo ligand design, statistical methods, Graph Neural Networks, and bioactivity-based ligand prioritization for rationally predicting high-affinity ligands. Segmentation of suspicious densities in digital mammograms.

State-of-the-art algorithms for detection of masses in mammograms are very sensitive but they also detect many normal regions with slightly suspicious features. Based on segmentations of detected regions, shape and intensity features can be computed that discriminate between normal and abnormal regions. These features can be used to discard false positive detections and hence improve the specificity of the detection method. In this work two different methods to segment suspect regions were examined. A number of different implementations of a region growing method were compared to a discrete dy

Behavioral Outcomes and Neural Network Modeling of a Novel, Putative, Recategorization Sound Therapy.

The mechanisms underlying sound's effect on tinnitus perception are unclear. Tinnitus activity appears to conflict with perceptual expectations of "real" sound, resulting in it being a salient signal. Attention diverted towards tinnitus during the later stages of object processing potentially disrupts high-order auditory streaming, and its uncertain nature results in negative psychological responses. This study investigated the benefits and neurophysiological basis of passive perceptual training and informational counseling to recategorize phantom perception as a more real auditory object. Spe

Modeling the volume of tissue activated in deep brain stimulation and its clinical influence: a review.

Deep brain stimulation (DBS) is a neuromodulatory therapy that has been FDA approved for the treatment of various disorders, including but not limited to, movement disorders (e.g., Parkinson's disease and essential tremor), epilepsy, and obsessive-compulsive disorder. Computational methods for estimating the volume of tissue activated (VTA), coupled with brain imaging techniques, form the basis of models that are being generated from retrospective clinical studies for predicting DBS patient outcomes. For instance, VTA models are used to generate target-and network-based probabilistic stimulati

Auditory-limbic-cerebellum interactions and cognitive impairments in noise-induced hearing loss.

AIMS: This study aimed to explore the neural substrate of hearing loss-related central nervous system in rats and its correlation with cognition. METHODS: We identified the neural mechanism for these debilitating abnormalities by inducing a bilateral hearing loss animal model using intense broadband noise (122 dB of broadband noise for 2 h) and used the Morris water maze test to characterize the behavioral changes at 6 months post-noise exposure. Functional magnetic resonance imaging (fMRI) was conducted to clarify disrupted functional network using bilateral auditory cortex (ACx) as a seed.

Where did you "left" Piazza del Popolo? At your "right" temporo-parietal junction.

Humans have an advanced ability to recall places and scenes from memory, which they scan with their "mind's eye" in order to cope with daily demands such as navigating and learning new environments. In the striking neurological syndrome called representational or imaginal neglect, patients with right brain damage fail to report contralesional, left-sided elements of familiar places relative to their imaginary vantage point. Notwithstanding descriptions of single cases of representational neglect dissociated from perceptual or visual neglect, it is still unknown whether this disorder is due to

Independence and coherence in temporal sequence computation across the fronto-parietal network.

Time processing requires distributed and coordinated cortical dynamics, yet how multiple brain areas flexibly switch between coherent and independent temporal representations remains unclear. Using mesoscale two-photon calcium imaging, we simultaneously recorded neuronal populations in the secondary motor cortex and posterior parietal cortex of mice performing a novel alternating-interval timing task. Both areas encoded elapsed time through similar high-dimensional sequential activity, and decoding analyses revealed both coherent temporal errors shared across areas and independent errors confi

In Silico Drug Repositioning Identifies SYK Kinase Inhibitors as Potential Neuroprotective Agents for Ataxia-Telangiectasia.

Ataxia-Telangiectasia (AT) is a rare neurodegenerative disorder characterized by progressive neuronal loss and chronic neuroinflammation. Emerging evidence indicates that aberrant overexpression of the adaptor protein TYROBP promotes sustained recruitment and activation of spleen tyrosine kinase (SYK), contributing to pathogenic inflammatory signaling in AT. In this study, we applied a drug repositioning strategy to identify clinically approved compounds that inhibit SYK activity. An integrated in silico workflow was employed, combining convolutional neural network (CNN)-based molecular dockin

Neural computational modeling reveals a major role of corticospinal gating of central oscillations in the generation of essential tremor.

Essential tremor, also referred to as familial tremor, is an autosomal dominant genetic disease and the most common movement disorder. It typically involves a postural and motor tremor of the hands, head or other part of the body. Essential tremor is driven by a central oscillation signal in the brain. However, the corticospinal mechanisms involved in the generation of essential tremor are unclear. Therefore, in this study, we used a neural computational model that includes both monosynaptic and multisynaptic corticospinal pathways interacting with a propriospinal neuronal network. A virtual a

A three-terminal MoTe2 artificial synapse with electrical and optical dual-mode plasticity.

Artificial synaptic devices capable of integrating sensing, memory, and computing functions are highly desirable for next-generation neuromorphic systems. However, most reported artificial synapses rely on single-mode electrical or optical stimulation, limiting their ability to achieve multimodal perception and adaptive information processing. Here, we demonstrate a three-terminal artificial synaptic device based on MoTe2 flakes that enables both electrical and optical synaptic modulation within a unified device architecture. The device exhibits bidirectional electrical synaptic plasticity thr

Development of a multimodal fully automated ensemble model to predict EGFR mutation and efficacy of EGFR-TKI in non-small cell lung cancer.

BACKGROUND: Drugs for systemic therapy to advanced or recurrent non-small cell lung cancer (NSCLC) are determined by the type of driver oncogenes and programmed death-ligand 1 (PD-L1) immunostaining. In this study, we investigated the potential for artificial intelligence to assist in detecting epidermal growth factor receptor (EGFR) mutation, which is one of the most frequent and important driver oncogenes, and predicting the efficacy of EGFR-tyrosine kinase inhibitors (TKIs) in patients with NSCLC. METHODS: We built an ensemble model that combined machine learning models (logistic regressio

Biologically-constrained spiking neural network for neuromodulation in locomotor recovery after spinal cord injury.

Presynaptic inhibition after spinal cord injury (SCI) has been hypothesised to disproportionately affect flexion reflex loops in locomotor spinal circuitry. Reducing gamma-aminobutyric acid (GABA) inhibitory activity increases the excitation of flexion circuits, restoring muscle activation and stepping ability. Conversely, nociceptive sensitisation and muscular spasticity can emerge from insufficient GABAergic inhibition. To investigate the effects of neuromodulation and proprioceptive sensory afferents in the spinal cord, a biologically constrained spiking neural network (SNN) was developed.

Combining machine learning and iterative experiments to keep pace with emerging viral variants of concern.

Modeling and predicting viral mutations before they emerge plays a crucial role in pandemic preparedness, enabling the early identification of emerging variants of concern (VOCs) and guiding timely updates to vaccines, diagnostic tests, and therapeutic strategies. However, existing machine learning models and large-scale experiments lose their predictive power as viral variants evolve further from the original strains in sequence space. Here, we present a scalable framework that integrates random forest and neural network machine learning models with targeted high-throughput experimentation to
