Cryoglobulinaemia in rabbits hyperimmunized with a polyvalent pneumococcal vaccine.

During the anamnestic response, the sera of four rabbits immunized with a polyvalent pneumococcal vaccine contained large amounts of cryoglobulins belonging to the G and M immunoglobulin classes. These cryoglobulins appeared to possess antibody activity since they agglutinated a suspension of the pneumococci that was used for immunization.

Rapid molecular detection of macrolide resistance.

BACKGROUND: Emerging antimicrobial resistance is a significant threat to human health. However, methods for rapidly diagnosing antimicrobial resistance generally require multi-day culture-based assays. Macrolide efflux gene A, mef(A), provides resistance against erythromycin and azithromycin and is known to be laterally transferred among a wide range of bacterial species. METHODS: We use Recombinase Polymerase Assay (RPA) to detect the antimicrobial resistance gene mef(A) from raw lysates without nucleic acid purification. To validate these results we performed broth dilution assays to assess

Haemophilus influenzae, Pneumococci, group A streptococci and Staphylococcus aureus: sensitivity of outpatient strains to commonly prescribed antibiotics.

The degree of resistance of Haemophilus influenzae, pneumococci, group A streptococci and Staphylococcus aureus to phenoxymethylpenicillin, ampicillin, doxycycline, erythromycin and chloramphenicol was investigated with the MIC-method (plate-dilution technique). Each bacterial group consisted of 100 outpatient strains. Seven strains of H. influenzae were beta-lactamase-producing. Of the 100 haemophilus strains, 58% were sensitive and the rest intermediate sensitive to erythromycin. 99% were sensitive to doxycycline and all strains were sensitive to chloramphenicol. Essentially all pneumococcal

Filamentous capsulated streptococci from the human respiratory tract: chemical and immunochemical characterization of a glycoprotein capsular antigen of provisional binary capsular type 87.

A filamentous alpha-hemolytic streptococcus of provisional capsular type 87 isolated from the human respiratory tract has been shown to be binary capsulated. One of the capsular antigens appears to be a glycoprotein; the other appears to be a polysaccharide. Transformation reactions with deoxyribonucleic acid from streptococcus type 87 and a number of noncapsulated pneumococci yielded transformed pneumococci with either a glycoprotein capsule or a polysaccharide capsule, but not with both. Capsular precipitin (quellung) reactions were observed when streptococcus type 87 was treated with homolo

Real-world effectiveness of pneumococcal vaccination in older adults: Cohort study using the UK Clinical Practice Research Datalink.

BACKGROUND: The 23-valent pneumococcal polysaccharide vaccine (PPV23) is recommended for UK older adults, but how age moderates effectiveness is unclear. METHODS: Three annual cohorts of primary-care patients aged≥65y from the Clinical Practice Research Datalink selected from 2003-5 created a natural experiment (n = 324,804), reflecting the staged introduction of the vaccine. The outcome was symptoms consistent with community-acquired pneumococcal pneumonia (CAP) requiring antibiotics or hospitalisation. We used the prior event rate ratio (PERR) approach to address bias from unmeasured confou

NORMAL FLORA OF THE NOSE, THROAT, AND LOWER INTESTINE OF DOGS.

Clapper, W. E. (The Lovelace Foundation for Medical Education and Research, Albuquerque, N.M.) and G. H. Meade. Normal flora of the nose, throat, and lower intestine of dogs. J. Bacteriol. 85:643-648. 1963.-An attempt was made to isolate and identify the complete normal flora of the rectum, nose, and throat of beagles. For primary isolation, 12 different kinds of media were used. Incubation of blood agar plates and slants anaerobically, and of thioglycolate broth aerobically, allowed the growth of obligate anaerobes. From the rectal specimens, 20 species of bacteria and 10 species of fungi wer

Gram's stains of tracheal secretions predict neonatal bacteremia.

The presence of bacteria in tracheal secretions stained by the Gram method was evaluated as a method of predicting neonatal bacteremia. The presence of bacteria had a 74% sensitivity and a 47% predictive accuracy in identifying neonates with bacteremia before 12 hours of age. The specificity in predicting newborns without bacteremia was 98%. In the same neonates, an immature neutrophil-total neutrophil ratio of 0.2 or more had a 77% sensitivity in predicting neonates with bacteremia. Neonates with bacteria in their blood and tracheal aspirates, who died shortly after birth, had pneumonia on po

Epidemiology of childhood invasive pneumococcal disease in Australia: a prospective cohort study.

BACKGROUND: The widespread use of pneumococcal conjugate vaccines (PCV) has changed the epidemiology of invasive pneumococcal disease (IPD) in children globally. METHODS: Multicentre prospective audit of IPD episodes from five paediatric hospitals in Australia over 5.5 years between 2016 and June 2021. Children (<18 years) with Streptococcus pneumoniae isolated from a sterile site were included. RESULTS: There were 377 IPD episodes in 375 children: 338 (90%) had received ≥3 PCV doses; 42 (11%) had IPD risk factors. The most common presentations were complicated pneumonia (254, 67%), bacterae…

Investigating the Role of Invasive Streptococcus Constellatus Infection in Severe Systemic Disease Manifestations: A Case Report.

We hope to add to the literature evidence regarding the increasing morbidity associated with an invasive infection by a normal body commensal, Streptococcus constellatus (S. constellatus). An increasing amount of literature documentation of intra- and extracranial disease manifestations following a systemic infection by this micro-organism is noted. We describe the findings of severe, necrotizing right lung disease and possible septic brain emboli in a 54-year-old gentleman in whom microbiological investigations suggest Streptococcal pneumonia, S. constellatus, and mixed anaerobes as possible

Giant Purulent Pericarditis with Cardiac Tamponade Due to Streptococcus intermedius Rapidly Progressing to Constriction.

Purulent pericardial effusion, although rare, is a life-threatening condition usually produced by the extension of a nearby bacterial infection locus or by blood dissemination in the immune-suppressed subjects or in the course of cardiothoracic surgery. Because clinical features of purulent pericardial effusion are often nonspecific, it can cause delay in diagnosis. Therefore, a high index of suspicion is required for timely diagnosis and management. Herein, we describe a case of giant purulent pericardial effusion due to Streptococcus intermedius with the history of bronchiectasis and pneumon

Pyomyositis in south Indian children.

Thirty-two children with pyomyositis were studied. In 28 children, 23 boys and 5 girls, Staphylococcus aureus was the aetiological agent. The strains isolated were resistant to penicillin. The muscles of the thigh, back and upper arm were most frequently involved. Eight children had infections in other parts of the body, namely pneumonia, empyema, pericarditis, meningitis, osteomyelitis and arthritis. Two children died. At follow-up one child had chronic osteomyelitis. In four neonates, beta haemolytic streptococcus was the causative organism. All were septicaemic. One infant died.

Urinary pneumococcal serotype detection among children with and without community-acquired pneumonia.

BACKGROUND: Urinary antigen detection (UAD) assays can address diagnostic challenges with culture-based identification of S. pneumoniae. We aimed to evaluate the utility of Pfizer's UAD1 and UAD2 assays for pneumococcal serotype surveillance in children with community acquired pneumonia (CAP) or upper respiratory tract infections (URI). METHODS: From March 2021-December 2023, children 3 months to 5 years who presented to the Children's Hospital Colorado Emergency Department with respiratory symptoms were enrolled as CAP or URI; healthy children served as controls. Nasal swabs were tested for

A systematic algorithm using 16S ribosomal RNA for accurate diagnosis of pneumonia pathogens.

Over half of community-acquired pneumonia cases are caused by a few dozen bacterial species, and accurate identification of these pathogens is essential for effective treatment. In this study, we developed a reliable diagnostic method using 16S ribosomal RNA (16S rRNA) sequencing, considering intra-species variation, the need to differentiate Streptococcus pneumoniae from oral α-hemolytic streptococci, and applicability to the battlefield hypothesis, which helps distinguish true pathogens from commensal organisms that are not causative pathogens. We designed specific primers and a BLAST wrappe…

Community-acquired pneumonia caused by mixed aerobic bacteria.

Mixed bacterial pneumonia caused by organisms other than anaerobes has been infrequently reported. We describe six cases and review the literature. Two patients had co-infection with S pneumoniae and L pneumophila. Two were infected with S pneumoniae and K pneumoniae and the others simultaneously harbored M tuberculosis and N asteroides. The first two sets of patients had bacteria isolated from usually sterile sites (blood and lung), while the latter harbored repeatedly isolated organisms not usually felt to be part of the normal respiratory flora. Mixed infection may help explain the substant

Pneumococcal pneumonia is driven by increased bacterial turnover due to bacteriocin-mediated intra-strain competition.

Using chromosomal barcoding, we observed that >97% of the Streptococcus pneumoniae (Spn) population turns over in the lung within 2 days post-inoculation in a murine model. This marked collapse of diversity and bacterial turnover was associated with acute inflammation (severe pneumococcal pneumonia), high bacterial numbers in the lungs, bacteremia, and mortality. Intra-strain competition mediated by the blp locus, which expresses bacteriocins in a quorum-sensing-dependent manner, was required for each of these effects. Bacterial turnover from the activity of Blp-bacteriocins increased the rele…
